As the first step to develop efficient means to control fungal spore bioaerosols, we designed, manufactured, and evaluated a fungal spore aerosol generator. We studied the physical and biological properties of the fungal spore bioaerosols on two common fungal species. The results demonstrated that the fungal spore bioaerosol generator effectively produces fungal spore bioaerosols.
Spore attachment of Ulva fasciata on agar and agaroses coating films showed various surfaces properties according to concentrations of agar or agaroses or via methanol. The highest number of spore attachment occurred in Sigma agarose coating film. Spore attachment on Bacto agar and other agaroses coating films showed the 12 ∼ 36 times less than Sigma agarose coated film. Comparison of spore attachment on 2.5% and 5% Seakem agarose and Bacto agar coating films differed in two concentrations while coating of 2.5% and 5% with NuSieve and Sigma agaroses did not differed. Spore attachment of coating made of 5% and 5% via 4% MeOH with Bacto agar and Nusieve agarose only differed. Overall, these results indicated agar and agarose coating films differed in spore attachment. Results of this work will be useful baseline for bioassay of antifouling activity of fouling organisms.
Arbuscular mycorrhizal fungi (AMF) spore propagation and long term maintenance is still a complicated technique for farmers. The use of AMF for their ability to promote plant growth and protect plants against pathogen attack and environmental stresses demands AMF propagation for large scale application. This study aimed to propagate AMF spores by trap culture technique and assess their ability to propagate with different host plants in a continuous plant cycle. Mycorrhizal inoculation by trap culture in maize resulted in longer shoots and roots than sudangrass plants. Increase in dry weight with higher percentage also was observed for maize plants. After first and second plant cycle, maize plants had the higher percentage of mycorrhizal response in terms of colonization and arbuscules than sudangrass. Maximum in spore count also achieved in the pots of maize plants. The results show that maize plant is more suitable host plant for AMF spore propagation and trap culture technique can be used effectively to maintain the AMF culture for long time.

Calcite forming bacteria (CFB) have been received increasing attention as a novel and environmental friendly strategy for the healing of concrete crack. Among the CFB, spore forming bacteria were proposed to overcome concrete condition (high pH, hydration heat, deicer). In this study, Lysinibaclillus sphaericus WJ-8 (WJ-8) isolated from concrete pavement was characterized. The WJ-8 was able to precipitate calcite at 10 mg/mL. When observed by scanning electron microscopy, WJ-8 showed spore formation and maximum spore yield was approximately 97.9%. Also response of spores against various environment stresses was examined. Approximately 83~97% of spores maintained their survivability at each three conditions ($60^{\circ}C$, 3 M NaCl and pH 12).

Salinity inhibits plant growth and restricts the efficiency of arbuscular mycorrhizal fungi. The selective uptake of nutrients from the soil and their effective transport to host roots make it essential for plant growth and development under salt stress. AMF spore associated bacteria shown to improve mycorrhizal efficiency under stress. Thus, this study aimed to understand the co-inoculation efficiency of AMF and SAB on maize growth and ion homeostasis under salt stress. Two AMF strains and one SAB were inoculated with maize either alone or in combination with one another. The results of our study showed that AMF and SAB co-inoculation significantly improved dry weight and nutrient uptake of maize under salt stress. Co-inoculation significantly reduced proline accumulation in shoots and Na+ accumulation in roots. Co-inoculation treatment also exhibited the high K+/Na+ ratios in roots at 25 mM NaCl. Mycorrhizal colonization showed positive influence for regulation of ZmAKT2, ZmSOS1 and ZmSKOR gene expressions, contributing to K+ and Na+ ion homeostasis. CLSM view showed that SAB were able move and localize into inter and intra cellular spaces of maize roots. In addition, CLSM view of AMF spores showed that gfp-tagged SAB also associated on the spore outer hyaline layer.

Bacillus subtilis spores can be used for protein display to engineer protein properties. This method overcomes viability and protein-folding concerns associated with traditional protein display methods. Spores remain viable under extreme conditions and the genotype/phenotype connection remains intact. In addition, the natural sporulation process eliminates protein-folding concerns that are coupled to the target protein traveling through cell membranes. Furthermore, ATP-dependent chaperones are present to assist in protein folding. CotA was optimized as a whole-cell biocatalyst immobilized in an inert matrix of the spore. In general, proteins that are immobilized have advantages in biocatalysis. For example, the protein can be easily removed from the reaction and it is more stable. The aim is to improve the pH stability using spore display. The maximum activity of CotA is between pH 4 and 5 for the substrate ABTS (ABTS = diammonium 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonate). However, the activity dramatically decreases at pH 4. The activity is not significantly altered at pH 5. A library of approximately 3,000 clones was screened. A E498G variant was identified to have a half-life of inactivation ($t_{1/2}$) at pH 4 that was 24.8 times greater compared with wt-CotA. In a previous investigation, a CotA library was screened for organic solvent resistance and a T480A mutant was found. Consequently, T480A/E498G-CotA was constructed and the $t_{1/2}$ was 62.1 times greater than wt-CotA. Finally, E498G-CotA and T480A/E498G-CotA yielded 3.7- and 5.3-fold more product than did wt-CotA after recycling the biocatalyst seven times over 42 h.

The control of pathogenic microorganisms is important issue in human environments, especially in surface area. However, surface disinfection has not been fully researched. In this study, the surface disinfection under UV irradiation was performed to investigate the quantitative kinetics for Bacillus subtilis spore inactivation in several experimental conditions, such as light intensity, temperature and surface roughness. This study reports that UV light would apparently inactivates the microorganisms and the required IT value for 2 log (99%) Bacillus subtilis spore inactivation was found to be 14.5 $mJ/cm^2$ in plain surface, as predicted by the Delayed Chick-Watson model. When UV was irradiated, there were no significant temperature effects. However, the experimental result shows that the more increased IT values are required at larger surface roughness.
BACKGROUND: In order to select a fungicide that can effectively control anthracnose disease in Japanese plum fruit, mycelial growth inhibition effect and spore germination inhibition effect of six fungicides were tested in vitro against six isolates of Colletotrichum acutatum and five isolates of C. gloeosporioides that were isolated from diseased Japanese plum fruit. METHODS AND RESULTS: Inhibitory effects of fungicides on mycelial growth were investigated after inoculating each isolate on potato dextrose agar amended with four discriminatory concentrations of each fungicide for 7 days at $25^{\circ}C$. For spore germination inhibitory effect, each isolate of the Colletotrichum spp. was cultured in potato dextrose agar for 7-14 days at $25^{\circ}C$. After adjusting the concentration of spores of each isolate to $1{\times}10^6mL^{-1}$ by diluting with 0.025% PDB, the spore suspension was mixed with each fungicide (1:4, v/v), and $60{\mu}L$ aliquots were dispensed to sterile hole slide glass. Hole slide glasses were placed in a humidified box and incubated for 15 hours at $25^{\circ}C$. Then, spore germination was observed under an optical microscope. At recommended concentration of fungicide prochloraz manganese showed the highest mycelial growth inhibitory effect and dithianon showed the lowest mycelial growth inhibition. The $EC_{50}$ values for the inhibition of spore germination by dithianon and pyraclostrobin were $0.069-0.126{\mu}g/mL$ and $0.37-1.59{\mu}g/mL$, respectively. Although benomyl, prochloraz manganese, azoxystrobin, and tebuconazole did not inhibit the spore germination, they appeared to restrain mycelial growth by abnormal growth of germ tube and mycelium after germination. CONCLUSION: Dithianon seemed to have preventive effect. Prochloraz manganese, azoxystrobin, and tebuconazole were likely to have control effect. Pyraclostrobin is considered to have both preventive and control effect against anthracnose disease of Japanese plum fruit.
A lichen-forming fungus was successfully isolated by discharged spore method from Korean lichen (Hetero-dermia sp.) and cultivated in pure culture. The isolate JR0012 inhibited mycelial growth of several plant-pathogenic fungi. Mycelial growth of the four Pythium spp. tested was completely inhibited. Potato dextrose broth was found to be the medium favorable for large-scale production of antibiotics from the isolate. Anti-fungal substances produced in axenic culture were partially purified. This is the first report in Korea of lichen-forming fungus successfully isolated and which exhibited strong antifungal activity against plant-pathogenic fungi, especially the four Pythium spp..

In order to optimally induce sporulation and toxin formation in Bacillus thuringiensis, exhaustion of specific nutrients as well as resuspension experiments were tried.
