  • Metabolomics is the study of changes in the metabolic status of an organism as a consequence of drug treatment, environmental influences, nutrition, lifestyle, genetic variations, toxic exposure, disease, stress, etc, through global or comprehensive identification and quantification of every single metabolite in a biological system. Since most chronic diseases have been demonstrated to be linked to nutrition, nutritional metabolomics has great potential for improving our understanding of the relationship between disease and nutritional status, nutrient, or diet intake by exploring the metabolic effects of a specific food challenge in a more global manner, and improving individual health. In particular, metabolite profiling of biofluids, such as blood, urine, or feces, together with multivariate statistical analysis provides an effective strategy for monitoring human metabolic responses to dietary interventions and lifestyle habits. Therefore, studies of nutritional metabolomics have recently been performed to investigate nutrition-related metabolic pathways and biomarkers, along with their interactions with several diseases, based on animal-, individual-, and population-based criteria with the goal of achieving personalized health care in the future. This article introduces analytical technologies and their application to determination of nutritional phenotypes and nutrition-related diseases in nutritional metabolomics.

Water quality management by bio-purification of bivalve, Mytilus galloprovincialis, in Masan Bay (이매패의 생물정화 기작을 이용한 마산만의 수질개선방안)

  • Masan Bay is a representative semi-closed bay acted as a sedimentation reservoir with a slow current velocity and a poor water circulation in Korea. The pollutants from terrestrial sources into the Masan Bay have apparently environmental pollution problems, such as eutrophication, red tied, and hypoxia. In this study, An ecological modeling work was performed to estimate the material circulation including the growth of bivalve in ecosystem. Furthermore, the effect of water purification was calculated by filter feeding bivalve to particulate organic matter just like COD and phytoplankton. And Water quality management strategy by bio-purification of bivalve is derived through selection of location, quantification of bivalve aquaculture farm. The results showed that the optimum location for bivalve farming is where phytoplankton accumulation by physical processes is maximized and the optimum density and area of bivalve are 35 individuals $m^{-3}$ and ca. 500 hectare, respectively. When assuming conditions for the optimum growth of bivalve, COD could decrease by up to 18% even without other reduction of pollution loads.

  • The purpose of this study is to clarify the challenges and suggest regulatory improvement strategy by investigating the Japanese building maintenance system which had been conducted for decades. The efficiency and problems of the current building maintenance system of Korea are analyzed by stake holders and purpose of management. The results show that several strategies are important to improve the quality of building maintenance. They include check list focusing on the purpose of maintenance, efficient and fair criteria, rational checking period considering owner's budget, cooperation with professionals in facilities, and promotion of professional companies participation. The results imply that the challenges should be solved systematically rather than individually.

  • 오염토양을 관리하는데 있어 현재 가장 중요하게 인식되는 부분은 오염물질이 정화되었다는 판정을 '어떤 기준으로 내리는 것이 합리적인가?'하는 것이다. 오염된 토양을 '어느 정도의 수준으로 복구를 하여야 하는가?'에 대한 의문이 학계와 산업계를 중심으로 꾸준히 제기되어 왔으며 미국의 경우, 환경청(Environmental Protection Agency) 등을 중심으로 그러한 물음에 대한 해답을 찾기 위한 본격적인 연구 및 조사활동이 활발히 진행되고 있다. 지금까지는 오염물질의 토양잔류농도와 획일화된 규제기준을 비교하여 오염여부를 판단한 후 복구여부 및 수준이 결정되어 왔는데 이러한 방법에 문제점이 있으며 현실적이지 못하다는 과학적 증거들이 발견, 축적되면서 이에 대한 대안이 모색되기 시작하였고 그 중의 하나가 '위해성에 근거한 복원전략(risk-based remediation strategy, RBRS)' 이라고 할 수 있다. RBRS는 오염토양의 정화기준을 결정하는데 있어 기존의 화학적 방법(즉, 토양잔류농도)뿐만 아니라 생물학적, 독성학적 방법도 같이 사용함으로써 오염물질의 실질적인 이용성에 따른 생독성을 평가하는 방법이다. RBRS의 가장 큰 장점은 오염물질의 종류뿐만 아니라 오염 지역의 특수성(예: 토지이용용도, 토양 및 지하수 특성, 수용체 집단의 특성 등)을 고려하여 오염물질의 위해성(risk) 여부를 판단하며 그 결과에 의거하여 오염지역의 정화 실시 여부 및 수준을 결정한다는 것이다. 따라서 RBRS에 의한 정화 및 복구사업은 과학적, 합리적, 경제적인 바탕 위에서 시행된다고 볼 수 있다. 이러한 장점들을 이유로 우리나라에서도 향후 오염지역의 정화에 RBRS와 같은 개념이 이용될 것으로 예상되며 학계를 중심으로 이에 대한 인식과 이해의 폭을 넓혀 나가야 할 것이다.

  • From environmental point of view, the foreign exchange crisis has motivated the Government to make the positive promoting policy for holding the foreign currency, leading to a series of small companies' bankruptcy due to foreign distribution companies' advance into our country, creating a new consuming culture. Though inaugurations have been vividly in progress in the aftermath of recession, reduced staffs and arranged layoff, their way gives priority to the reduced frame of the existing method rather tham the development or improvement of a new distribution. It is difficult to attain the sales goal unless a marketing analysis is not properly made, due to store managers' lack in expertism of management. In view of culture, the change of retail stores is imperative at the point that the type of consumers' purchase is rapidly changing and a more positive business system is needed. preventing an opportunistic loss of management through the analysis of outcome such as consumer management, sales management and account management by using computers. In view of design. the display in sale is to interpret products more charmingly, and should make interpretation accurately by selecting an important theme. For this, taking the store for valuables for instance. the progress on the effective foundation and store dinic business by presenting the design blue print can be made, and the strategy coping with the foreign distribution market's rush into Korea can be established. through the advanced store management.

Ozone Simulations over the Seoul Metropolitan Area for a 2007 June Episode, Part V: Application of CMAQ-HDDM to Predict Ozone Response to Emission Change (2007년 6월 수도권 오존모사 V - 배출량 변화에 따른 오존농도 예측 시 민감도기법 적용)

  • In this paper, we use the HDDM (High-order Decoupled Direct Method)-driven ozone sensitivity to predict change in ozone concentrations in response to domain-wide $NO_x$(Oxides of Nitrogen) and VOC (Volatile Organic Compound) emission controls over the Seoul Metropolitan Area during June 11~19, 2007. In order to validate the applicability of HDDM to $NO_x$ and VOC control scenarios, the HDDM results are compared to Brute Force Method (BFM). For VOC controls, NME (Normalized Mean Error) between BFM and HDDM remains less than 2% until the domain-wide VOC emissions are reduced by 80%. The NME for a 40% reduction in the domain-wide $NO_x$ emissions is less than 5% but increases abruptly after further reductions in the $NO_x$ emissions (i.e., 80% reduction). The results indicates that it may be inaccurate to use ozone sensitivity coefficients estimated at a given base emission condition in predicting ozone after $NO_x$ reductions larger than ~50% of the domain total in the SMA. Therefore, HDDM application on piecewise emissions is desirable to predict ozone response to emission controls with accuracy (i.e., truck emissions rather than the domain total). For computational efficiency, HDDM shows approximately 30% faster than the BFM sensitivity approach.

Educating restaurant owners and cooks to lower their own sodium intake is a potential strategy for reducing the sodium contents of restaurant foods: a small-scale pilot study in South Korea

  • BACKGROUND/OBJECTIVES: This study was conducted to evaluate the feasibility of a sodium reduction program at local restaurants through nutrition education and examination of the health of restaurant owners and cooks.SUBJECTS/METHODS: The study was a single-arm pilot intervention using a pre-post design in one business district with densely populated restaurants in Seoul, South Korea. The intervention focused on improving nutrition behaviors and psychosocial factors through education, health examination, and counseling of restaurant personnel. Forty-eight restaurant owners and cooks completed the baseline survey and participated in the intervention. Forty participants completed the post-intervention survey. RESULTS: The overweight and obesity prevalences were 25.6% and 39.5%, respectively, and 74.4% of participants had elevated blood pressure. After health examination, counseling, and nutrition education, several nutrition behaviors related to sodium intake showed improvement. In addition, those who consumed less salt in their baseline diet (measured with urine dipsticks) were more likely to agree that providing healthy foods to their customers is necessary. This study demonstrated the potential to reduce the sodium contents of restaurant foods by improving restaurant owners' and cooks' psychological factors and their own health behaviors. CONCLUSIONS: This small pilot study demonstrated that working with restaurant owners and cooks to improve their own health and sodium intake may have an effect on participation in restaurant-based sodium reduction initiatives. Future intervention studies with a larger sample size and comparison group can focus on improving the health and perceptions of restaurant personnel in order to increase the feasibility and efficacy of restaurant-based sodium reduction programs and policies.

Heavy Metals Contents and Chemical Characteristics in Compost from Animal Manures (가축분 퇴비의 중금속 함량 및 화학적 형태별 특성)

  • Objectives: This study was conducted to evaluate the total heavy metals contents and chemical forms in the animal manure compost. Materials and methods: A total of 109 compost samples were collected throughout the country and classified into three groups in accordance with the raw materials; pig manure, poultry manure and mixed(pig+poultry+cattle) manure. The compost samples were analyzed for total metal content and sequential chemical extraction to estimate the quantities of metals. Results: Concentrations of Zn and Cu in several compost samples were higher than the maximum acceptable limits by the Korea Compost Quality Standards. Concentrations of Zn, Cu, and Cd in compost samples were 257~5,102, 68~1,243, and 0.02~2.54 mg/kg respectively, while Cr, Ni, As, and Pb were < 20 mg/kg. The concentrations of heavy metals in pig manure compost were higher than those of both the poultry and the mixed manure compost. The predominant forms for extracted metals were Cr, Ni, Zn, As, and Pb, residual; Cu, organic; and Cd, carbonate. Conclusions: Results indicate that the Zn and Cu contents in compost were higher than other heavy metals and the heavy metal contents were greater in pig manure compost followed by mixed and poultry manure compost. To prevent the accumulation of heavy metals in soil where animal manure compost is applied, strategy for reducing heavy metal concentrations in animal manure and compost must be considered.

Nrf2 Expression and Apoptosis in Quercetin-treated Malignant Mesothelioma Cells

  • NF-E2-related factor 2 (Nrf2), a basic leucine zipper transcription factor, has recently received a great deal of attention as an important molecule that enhances antioxidative defenses and induces resistance to chemotherapy or radiotherapy. In this study, we investigated the apoptosis-inducing and Nrf2- upregulating effects of quercetin on malignant mesothelioma (MM) MSTO-211H and H2452 cells. Quercetin treatment inhibited cell growth and led to upregulation of Nrf2 at both the mRNA and protein levels without altering the ubiquitination and extending the half-life of the Nrf2 protein. Following treatment with quercetin, analyses of the nuclear level of Nrf2, Nrf2 antioxidant response element-binding assay, Nrf2 promoter-luc assay, and RT-PCR toward the Nrf2-regulated gene, heme oxygenase-1, demonstrated that the induced Nrf2 is transcriptionally active. Knockdown of Nrf2 expression with siRNA enhanced cytotoxicity due to the induction of apoptosis, as evidenced by an increase in the level of proapoptotic Bax, a decrease in the level of antiapoptotic Bcl-2 with enhanced cleavage of caspase-3 and PARP proteins, the appearance of a sub-$G_0/G_1$ peak in the flow cytometric assay, and increased percentage of apoptotic propensities in the annexin V binding assay. Effective reversal of apoptosis was observed following pretreatment with the pan-caspase inhibitor Z-VAD. Moreover, Nrf2 knockdown exhibited increased sensitivity to the anticancer drug, cisplatin, presumably by potentiating the oxidative stress induced by cisplatin. Collectively, our data demonstrate the importance of Nrf2 in cytoprotection, survival, and drug resistance with implications for the potential significance of targeting Nrf2 as a promising strategy for overcoming resistance to chemotherapeutics in MM.

Artificial induction and isolation of cadmium-tolerant soil bacteria

  • Environmental pollution caused by various heavy metals is a serious global problem. To solve this problem, microbial bioremediation of contaminated metals has developed rapidly as an effective strategy when physical and chemical techniques are not suitable. In this study, cadmium (Cd)-tolerant soil bacteria were isolated via artificial induction in laboratory conditions instead of screening bacteria naturally adapted to metal-contaminated soils. Wild-type (WT) bacteria grown in uncontaminated soils were artificially and sequentially adapted to gradually increasing Cd concentrations of up to 15 mM. The resultant cells, named Soil-CdR15, survived at a Cd concentration of 10 mM, whereas WT cells failed to survive with 4 mM Cd on solid media for 2 d. In liquid media containing Cd, the SoilCdR15 cells grew with 15 mM Cd for 7 d, whereas the WT cells could not grow with 5 mM Cd. Both Soil-CdR15 and WT cells removed approximately 35% of Cd at the same capacity from liquid media containing either 0.5 or 1.0 mM Cd over 2 d. In addition to Cd, the Soil-CdR15 cells showed increased resistance to nickel, zinc, and arsenic compared to WT cells. The Soil-CdR cells were identified as Burkholderia sp. by partial sequencing of 16S rRNA. The data presented in this study demonstrate that isolation of heavy metal-tolerant microorganisms via artificial induction in laboratory conditions is possible and may be useful for the application of the microorganisms for the bioremediation of heavy metals.
