Development and Application of Enzyme Immunoassay for Endosulfan Residue Analysis (Endosulfan 과 그 분해산물의 Enzyme Immunoassay에 의한 분석법의 개발과 응용)

  • An enzyme immunoassay(EIA) was developed for the analysis of insecticide endosulfan and its degradation products. The sensitivity and specificity of the antibody produced were examined. Optimal conditions in the ELISA system for residue analysis were also discussed. A mixed suspension of endosulfan-hemocyanin conjugate(ES-KLH) 1.1 mg/ml and Freund's adjuvant was injected subcutaneously to white rabbits and then collected antisera were tested for titers by indirect ELISA(1/24,000). Because of difficulties in the synthesis of endosulfan peroxidase conjugate, amine derivative of endosulfan-diol was synthesized and it showed 40% of conjugate yields(2mg/ml of conjugate). the highest sensitivity obtained enzyme-conjugate was a concentration of 200ng/ml. The detection limit of endosulfan in ELISA system was 5 ppb on the standard curve. In application of ELISA for residue analysis, the recoveries were really 100% both in the spiked soil and apple sample regardless of endosulfan concentration treated. On the other hand, chlorinated hydrocarbons of similar structure with endosulfan showed low cross-reactivity$(2.2%{\sim}29.2%).$

Biodegradation Pathways of Polychlorinated Biphenyls by Soil Fungus Aspergillus niger (Polychlorinated Biphenyl의 토양 미생물 Aspergillus niger에 의한 생분해 경로)

  • As of many organochlorine pesticides, polychlorinated biphenyls are ubiquitous organic contaminants, which can be found in the most environmental matrices. Their toxic effects include endocrinedisrupting activity. Most researches with these toxicants performed with mixtures of congeners, namely Aroclor and related study has been done in complex environmental matrix, rather than single biosystems or pure congeners. 5 congeners were synthesized and their fates in pure microbial culture (Aspergillus niger) were determined in this study. Among biphenyl and synthetic congeners, biphenyl, PCB-1 (2-chlorobiphenyl), and PCB-3 (4-chlorobiphenyl) were rapidly transformed to hydrophilic metabolites, followed by PCB-38 (3,4,5-trichlorobiphenyl), while the degradation of PCB-126 (3,3',4,4',5-pentachlorobiphenyl) was not observed. The amounts of transformation for biphenyl, PCB-1, PCB-3, and PCB-38 were 65, 38, 52, and 2% respectively. The major metabolites of the above congeners were identified as mono- and di-hydroxy biphenyls, which are known to give adverse endocrinological effects.

Effects of Plants, Rhizobacteria and Physicochemical Factors on the Phytoremediation of Contaminated Soil (오염 토양의 식물상 복원효율에 미치는 식물, 근권세균 및 물리.화학적 인자의 영향)

  • Phytoremediation is an economic and environmentally friendly technique to remediate contaminated-soil. In this study, the effects of plants, rhizobacteria and physicochemical factors on phytoremediation have been reviewed. For successful phytoremediation, the selection of plants is primarily important. To remediate soil contaminated with petroleum hydrocarbon, raygrass (Lolium multiflorum lam), white mustard, vetch (Vicia villosa), tall fescue (Festuca arundinacea), legumes, poplar, and Pine (Pinus densiflora) were mainly applied, and the removal efficiency of petroleum hydrocarbon were ranged 68 to 99%. Corn (Zea mays), raygrass (Lolium multiflorum lam), vetch (Vicia villosa), mustard, clover (Trifolium repens), and tall fescue (Festuca arundinacea) were used for the removal of polycyclic aromatic hydrocarbon, and their removal efficiencies were 50-98%. Rhizobacteria play significant roles for phytoremediation because they can directly participate in the degradation of contaminant as well as promoting plants growth. The following rhizobacteria were preferred for phytoremediation: Azospirillum lipoferum, Enterobactor cloacae, Azospirillum brasilense, Pseudomonas putida, Burkholderia xenovorans, Comamonas testosterone, Pseudomonas gladioli, Azotobacter chroococcum, Bacillus megaterium, and Bacillus subtilis. Pysicochemical factors such as pH, temperature, nutrient, electron acceptor, water content, organic content, type of contaminants are consequential limiting factors for phytoremediation.

Simple Material Budget Modeling for a River-Type Reservoir (하천형 저수지의 단순 물질수지 모델링)

  • Simple material budget models were developed to predict the dry season water quality for a river-type reservoir in Paldang, Republic of Korea. Of specific interest were the total phosphorus (TP), chlorophyll ${\alpha}$ (Chl. ${\alpha}$), 5-day biochemical oxygen demand (BOD), and chemical oxygen demand (COD). The models fit quite well with field data collected for 20 years and have enabled the identification of the origins of organic materials in the reservoir. The critical hydraulic load that determines the usability of phosphorus for algal production appeared to be about $1.5m\;d^{-1}$. When a hydraulic load was smaller than the critical value, the concentrations of $Chl.{\alpha}$, COD, and BOD in the reservoir water became sensitive to internal algal reactions such as growth, degradation, and settling. In spite of the recent intensive efforts for organic pollutant removal from major point sources by central and local governments, the water quality in the reservoir had not been improved. Instead, the concentration of COD increased. The model analysis indicated that this finding could be attributed to the continuing increase of the algal production in the reservoir and the allochthonous load from non-point sources. In particular, the concentrations of COD and BOD of algal origin during 2000~2007, each of which is comprised of approximately one half of the total, were approximately 2.5 times higher than those observed during 1988~1994 and approximately 1.3 times higher than those between 1995~1999. The results of this study suggested that it is necessary to reduce the algal bloom so as to improve the water quality of the reservoir.

Nitrogen Removal in Column Wetlands Packed with Synthetic Fiber Treating Piggery Stormwater (축산단지 강우 유출수 처리를 위한 합성섬유충진 습지의 질소제거에 관한 연구)

  • A set of lab-scale polymer synthetic fiber packed column wetlands composing three columns (CW1, CW2 and CW3) with different hydraulic regimes, recirculation frequencies and pollutant loading rates, were operated in 2012. Synthetic fiber tested as an alternative wetland medium for soil mixture or gravel which has been widely used, has very high pore size and volume, so that clogging opportunity can be greatly avoided. The inflow to the wetland was artificial stormwater. All the wetlands achieved effective removal of TSS (94%~96%), TCOD (68%~73%), TN (35%~58%), TKN (62%~73%) and NH4-N (85%~ 99%). Particularly, it was observed that COD was released from the fiber during one distinct period in all wetlands. This was probably due to the degradation of polymer fiber, and the released organic matters were found to serve as carbon source for denitrification. In addition, with longer retention time and frequent recirculation, lower effluent concentration was observed. With higher pollutant loading rate, higher nitrification and denitrification rates were achieved. However, although organic matters were released from the fiber, the lack of carbon source was still the limiting factor for the system since the release persisted only for 40 days.

The Role of Heat Shock Protein 25 in Radiation Resistance

  • Overexpression of HSP25 delayed cell growth, increased the level of $p21^{waf}$, reduced the levels of cyclin D1, cylcin A and cdc2, and induced radioresistance in L929 cells. We demonstrated that extracellular regulated kinase (ERK) and MAP kinase/ERK kinase (MEK) expressions as well as their activation (phospho-forms) were inhibited by hsp25 overexpression. To confirm the relationship between ERK1/2 and hsp25-mediated radioresistance, ERK1 or ERK2 cDNA was transiently transfected into the hsp25 overexpressed cells and their radioresistance was examined. HSP25-mediated radioresistance was abolished by overexpression of ERK2, but not by overexpression of ERK1. Alteration of cell cycle distribution and cell cycle related protein expressions (cyclin D, cyclin A and cdc2) by hsp25 overexpression were also recovered by ERK2 cDNA transfection. Increase in Bc1-2 protein by hsp25 gene transfection was also reduced by subsequent ERK2 cDNA-transfection. In addition, HSP25 overexpression reduced reactive oxygen species (ROS) and increased expression of manganese superoxide dismutase (MnSOD) gene. Increased activation of NF-kB (IkB degradation) was also found in hsp25-overexpressed cells. Moreover, transfection of hsp25 antisense gene abrogated all the HSP25-mediated phenomena. To further elucidate the exact relationship between MnSOD induction and NF-kB activation, dominant negative $I-kB\alpha(I-kB\alpha-DN)$ construction was transfected to HSP25 overexpressed cells. $I-kB\alpha-DN$ inhibited HSP25 mediated MnSOD gene expression. In addition, HSP25 mediated radioresistance was blocked by $I-kB\alpha-DN$ transfection. Blockage of MnSOD with antisense oligonucleotides in HSP25 overexpressed cells, prevented apoptosis and returned the ERK1/2 activation to the control level. From the above results, we suggest for the first time that reduced oxidative damage by HSP25 was due to MnSOD-mediated down regulation of ERK1/2.

Thermal Decomposition of A New Insecticide KH-502 [O, O-Diethyl O-(1-phenyl -3-trifluoromethy-5-pyrazoyl) thiophosphoric acid ester] (신규(新規) 살충제(殺蟲劑)인 O, O, O-Diethyl-O-(1-phenyl-3-trifluoromethyl-5-pyrazoyl) thiophosphoric acid ester의 열(熱)에 의한 분해성(分解性))

  • Thermal decomposition was conducted to investigate the influence of the various factors on stability of a new insecticide, [O, O-Diethyl O-(1-phenyl-3-trifluoromethyl-5-pyrazoyl) thiophosphoric acid ester : KH-502], in view of those informations applicable for industrial exploitation. In the thermal decomposition experiment, KH-502 was, after mixing with Fe, Cu and adjustment of moisture and pH conditions, subjected to three temperatures, 25, 50, and $100^{\circ}C$. Results for stability, and degradation pattern of KH-502 from the above experiment can be summarized as follows: 1. Main products of the thermal decomposition when this was conducted in the closed system were identified as following five compounds:O, O, O-Triethylthiophosphoric acid(TEPA), 1-Phenyl-3-trifluoromethyl-5-ethoxypyrazole(PTMEP), 1-Phenyl-2-ethyl-3-trifluoromethyl-5-hydroxypyrazole(PETMHP), O, O-Diethyl O-(1-phenyl-3-trifluoromethyl-5-pyrazoyl)phosphoric acid ester(KH-502 oxo form), O, S-Diethyl O-(1-phenyl-3-trifluoromethyl-5-pyrazoyl)phospho rothiolate(S-ethyl KH-502). However, compounds such as oxo form and S-ethyl KH-502 were not identified when the thermal decomposition was proceeded in the open system. 2. KH-502 was stable at 25 and 50$^{\circ}C$, but it was decomposed at 100$^{\circ}C$ following the first-order kinetics at the early stages of decomposition. 3. Rate constants for the thermal decomposition of KH-502 at 100$^{\circ}C$ were in the orders of Cu powder addition 0.344>Cu plate addition 0.21>moisture addition 0.05>closed system=open system=iron addition=pH 5.5 adjustment 0.04>pH 8.5 adjustment 0.027 day$^{-1}$, representing KH-502 was decomposed fast at Cu powder treatment and slow at pH 8.5 adjustment. 4. Half-life for the thermal decomposition of KH-502 at 100$^{\circ}C$ was in the orders of Cu powder addition 2.02

Chemical Water Quality and Fish Community Characteristics in the Mid- to Downstream Reach of Geum River (금강 중.하류역의 이.화학적 수질특성 및 어류 군집특성)

  • The objectives of the study were to analyze chemical water quality and fish community structures in the downstreams of Geum River during 2004~2005. Water quality parameters showed that ionic dilution was evident during July~August due to a rainfall and river runoff by Asian monsoon. Mean concentrations of BOD showed a seasonal variation and were greater in the downstream than in the midstream. The total number of fish species observed was 34 (11 families) and 30.3% of them was Korean endemic species. The dominant species with > 20% of the total were Zacco platypus (25.9%) and Microphysogobio jeoni (21.5%) in the river. Exotic species observed was only one species, Carassius cuvieri with relative abundance of 1.8%. According to the structure analysis of fish community, species richness index was highest (3.714) in S6 and lowest in S4 (1.961), while species diversity index was highest in S2 (1.01) and lowest in S5 (0.507). Tolerant species dominated the fish community (49%), and the sensitive species were rare (4.4%), indicating a biological degradation of ecosystem. In the mean time, omnivore species was composed of 49% in the fish community and insectivore species was nearly same with the proportion of the omnivores.

Evaluation on Weathering Characterization on Rock Types Using Artificial Weathering Test (인공풍화시험을 이용한 암종별 풍화특성 평가)

  • For exposed slopes, the weathering degree over time has a major effect on the engineering properties of rocks and the slope stability. Rocks are gradually changed by weathering into soil over time, and the resulting physical, chemical and mechanical changes of rocks affect the engineering stability of the slope. However, there are not many ways to objectively evaluate the weathering degree of a slope. In this study, therefore, to investigate the weathering characteristics of rocks, granite, gneiss and shale distributed in the Chungbuk region were sampled by weathering stage and changes in their component minerals and tissues were investigated. Furthermore, artificial weathering was induced using the freezing and thawing test and quantitatively investigated through porosity and absorption rate. In addition, the changes of microcracks due to artificial weathering were evaluated through box fractal dimension ($D_B$). Through mineralogical study the phase change of constituting minerals, the growth of secondary minerals, the development of micro-cracks and the fabric changes due to weathering were observed. The mineralogical, chemical and engineering evaluations of the weathering degree through the experimental results in this study are expected to be useful for analyzing the weathering characteristics and causes by rock type and for proposing a methodology to evaluate the degradation of physical properties comparatively and quantitatively.

Political Ecology and Bioregionalism: New Directions for Geography and Resource-Use Management (정치생태학과 생물지역주의 - 지리학과 자원이용관리를 위한 새로운 방향 -)

  • This paper provides an overview of political ecology, a body of theory that focuses on the links between political and economic inequality on the one hand, and environmental degradation on the other. Adopting a tripartite classification scheme that identifies three political ecology traditions -'classical', 'democratic' and 'poststructuralist'- the discussion shows the need for a move within the poststructuralist tradition away from a narrow and quasi-idealistic focus on discourse to a more robust philosophical engagement with ontological and epistemological issues grounded in Gilles Deleuze's development of Nietzschean materialism. From there. the author draws on numerous examples from Canada, and surveys the available literature on 'bioregionalism', a relatively new intellectual tradition evolved from the North American environmental social movements of the 1970s and 1980s. The so-called 'bioregional approach' stresses that administrative units need to reflect (rather than transect) eco-geographical and cultural features. Bioregionalism is described and assessed as a potential pragmatic research framework for geographers and other planners wishing to respond proactively to the call for a revamped, poststructuralist political ecology. The paper concludes that a bioregional approach to political ecology avoids the weaknesses identified by certain critics, provides scope for consideration of fundamental philosophical ideas, and as such, represents a practical development of a poststructuralist political ecology.
