Cellulase and xylanase are main hydrolysis enzymes for the degradation of cellulosic and hemicellulosic biomass, respectively. In this study, our aim was to develop and test the efficacy of a rapid, high-throughput method to screen hydrolytic-enzyme-producing microbes. To accomplish this, we modified the 3,5-dinitrosalicylic acid (DNS) method for microwell plate-based screening. Targeted microbial samples were initially cultured on agar plates with both cellulose and xylan as substrates. Then, isolated colonies were subcultured in broth media containing yeast extract and either cellulose or xylan. The supernatants of the culture broth were tested with our modified DNS screening method in a 96-microwell plate, with a $200{\mu}l$ total reaction volume. In addition, the stability and reliability of glucose and xylose standards, which were used to determine the enzymatic activity, were studied at $100^{\circ}C$ for different time intervals in a dry oven. It was concluded that the minimum incubation time required for stable color development of the standard solution is 20 min. With this technique, we successfully screened 21 and 31 cellulase- and xylanase-producing strains, respectively, in a single experimental trial. Among the identified strains, 19 showed both cellulose and xylan hydrolyzing activities. These microbes can be applied to bioethanol production from cellulosic and hemicellulosic biomass.

Polylactic acid (PLA) has been highlighted as an alternative renewable polymer for the replacement of petroleum-based plastic materials, and is considered to be biodegradable. On the other hand, the biodegradation of PLA by terminal degraders, such as microorganisms, requires a lengthy period in the natural environment, and its mechanism is not completely understood. PLA biodegradation studies have been conducted using mainly undefined mixed cultures, but only a few bacterial strains have been isolated and examined. For further characterization of PLA biodegradation, in this study, the PLA-degrading bacteria from digester sludge were isolated and identified using a polymer film-based screening method. The enrichment of sludge on PLA granules was conducted with the serial transference of a subculture into fresh media for 40 days, and the attached biofilm was inoculated on a PLA film on an agar plate. 3D optical microscopy showed that the isolates physically degraded the PLA film due to bacterial degradation. 16S rRNA gene sequencing identified the microbial colonies to be Pseudomonas sp. MYK1 and Bacillus sp. MYK2. The two isolates exhibited significantly higher specific gas production rates from PLA biodegradation compared with that of the initial sludge inoculum.
The fungi associated with termites secrete enzymes such as laccase (multi-copper oxidase) that can degrade extracellular wood matrix. Laccase uses molecular oxygen as an electron acceptor to catalyze the degradation of organic compounds. Owing to its ability to transfer electrons from the cathodic electrode to molecular oxygen, laccase has the potential to be a biocatalyst on the surface of the cathodic electrode of a microbial fuel cell (MFC). In this study, a two-chamber MFC using the laccase-producing fungus Galactomyces reessii was investigated. The fungus cultured on coconut coir was placed in the cathode chamber, while an anaerobic microbial community was maintained in the anode chamber fed by industrial rubber wastewater and supplemented by sulfate and a pH buffer. The laccase-based biocathode MFC (lbMFC) produced the maximum open circuit voltage of 250 mV, output voltage of 145 mV (with a $1,000{\Omega}$ resistor), power density of $59mW/m^2$, and current density of $278mA/m^2$, and a 70% increase in half-cell potential. This study demonstrated the capability of laccase-producing yeast Galactomyces reessii as a biocatalyst on the cathode of the two-chamber lbMFC.
As the development of national lands increase, the necessity to develop less favorable terrain also increases. It also becomes more difficult and complicated to provide access that is both economical and safe from contributing to soil erosion on roadsides and to londscape damages. Because of the increased and justified emphasis upon environmental degradation, proper stabilization of the cut-and bank slopes of roadsides, plant sites and building sites in Korea requires careful planning as well as execution of the constructions. All fill slopes should be compacted to a degree consistent with design standards and material properties. Drainage facilities should be provided to prevent damaging concentrations of surface runoff and to avoid high pore pressures in cuts and fills. All surfaces of cut-fill slopes should be revegetated with suitable species as soon as possible after construction. To stabilize the cut-fill slopes of highway roadsides should be considerded the factor of stabilization as well as landscape conservation especially. Such basic influences as the effects of vegetations on water balance of rain fall, on the control of surface erosion and on the surface slope failures are briefly discussed in the report.

For the base station or repeater which is in operation as part of a mobile communication system, any kind of degradation in antenna performance such as lowered gam or increased VSWR which maybe due to aging of antenna, connector corrosion or other environmental factors, is difficult to detect and react to immediately. In this paper, a VSWR measurement device which is capable of detecting the VSWR of a PCS forward path ($1,840\~1,870$MHz) antenna in operation in real time without any interruption in service is developed. Such regular measurement will allow the prediction of any serious failures and insure reliable operation of the antenna. All components of the antenna VSWR measurement device was researched so that the whole system could be produced and shown to be accurate in measuring the YSWR of an actual RF repeater in operation, proving it's effectiveness.
The decayed wood by Fomes pini (Thore) Lloyd required a smaller H factor than the sound wood for pulping to permanganate number 20. The H factors for the decayed wood pulping by the kraft and soda processes were reduced by 15% and 17%, respectively, in the presence of 1% anthraquinone. The wood components degraded by fungi are normally more readily solubilized in alkali than the corresponding components in sound wood. The nonphenolic ${\beta}$-O-4 type lignin model compound, veratrylglycerol-${\beta}$-guaiacyl ether(I), and phenolic model compound, syringylglycerol-${\beta}$-syringyl ether(III), were degraded by the white-rot fungi to yield ${\alpha}$-guaiacoxy-${\beta}$-hydroxypropioveratrone(II) from the former and ${\alpha}$-syringyloxy-${\beta}$-hydroxypropiosyringone(IV) from the latter. Structures of the degradation products indicated that C ${\alpha}$-oxidation could occur with white-rot fungi. It has been shown that the alkaline cleavage of ${\beta}$-aryl ether bonds in the lignin units is accelerated by the presence of ${\alpha}$-carbonyl groups.

Rainfall is one of the most active forces that cause soil erosion. The action of rain on the soil exerts an erosive power caused by the impact of the drops, which fall with variable speed and kinetic energy, depending directly on the diameter of the drop. The objective of this study is to determine algorithms capable of estimating rainfall erosivity for the region of Maceió-AL. For this purpose, erosion rains were collected between 2003 and 2006 using a RD-69 disdrometer, which continuously and automatically measures rainfall distribution in a range of 1 min. The determination of algorithms in the form of power equation to estimate was adjusted with one and two independent variables (amount of rainfall, duration and maximum intensity).

As the genetically modified microorganisms (GMMs) and their recombinant plasmid DNAs could be released into natural environments, their stabilities and impacts to indigenous microorganisls have become very importhant research subjects concerning with environmental and ecological aspects. In this study, the genetically modified E. coli CU103 and its recombinant pCU103 plasmid DNA, in which pcbCD genes involving in degradation of biphenyl and 4-chlorobiphenyl were cloned, were studied for their survival and stability in several different waters established under laboratory conditions. E. coli CU103 and its host E. coli XL1-Blue survived longer in sterile distilled water (SDW) and filtered autoclaved river water (FAW) than in filtered river water (FW). A lot of extracellular DNAs were released from E. coli CU103 by lytic action of phages in FW and the released DNAs were degraded by DNase dissolved in the water. Such effects of the factors in FW on stability of the recombinant pCU103 plasmid were also observed in the results of gel electrophoresis, quantitative analysis with bisbenzimide, and transformation assay. Therefore, the recombinant plasmids of pCU103 were found to be readily liberated from the genetically modified E. coli CU103 into waters by normal metabolic processes and lysis of cells. And the plasmid DNAs were quite stable in waters, but their stabilities could be affected by physicoKDICical and biological factors in non-sterile natural waters.

A cross linked polyethylene (XLPE) material has been widely used to develop a distribution power cable due to its excellent electrical characteristics and mechanical strength. However, several problems such as environmental disruption, electrical aging, thermosetting property, and impurities which cause degradation also arise. Therefore, a novel dielectric material should be developed to substitute for the XLPE. Several kinds of polyolefin materials to substitute for the conventional dielectric material, XLPE are developed and A cylindrical rod to cylindrical rod electrode system made with stainless steel is used to perform the experiments according to the ASTM D-149 protocol under an AC and Impulse input voltage condition. The experimental results are calculated by the Weibull distribution method and analyzed by an Finite Element Method(FEM). Finally, the dielectric characteristics of the conventional XLPE and novel polyolefin are experimented compared with each other in this study.

Traffic volume increases according to highway expansion and industrial development which causes repetitive defect and durability degradation on pavement. The research of quality assurance system used abroad has introduced Korea. Korea Expressway Corporation (KEC) has developed a Quality Performance Index (QPI) that quantitatively assesses the level of quality of the final product, and practical applications. Assessment factor on concrete pavement consisted of pavement thickness, compressive strength, IRI and spacing factor. Assessment factor on concrete pavement is determined by empirical evaluation factor from abroad. In this study, analysis of evaluation factors of concrete pavement by using pavement life prediction simulation and measured data were evaluated with consideration of feasibility of the assessment factor. Pavement life, performance and durability are affected by pavement thickness, compressive strength, IRI and spacing factor in assessment factor on concrete pavement, QPI.
