An analysis of the enteropathogenic bacteria isolation during the last five years (최근 5년간 분리된 enteropathogenic bacteria)

  • Bacteriologic diagnosis of enteric infection remains to be an important role of clinical laboratory because of the prevalence of the infection. Often the determination of etiologic agent and its susceptibility to antibiotics are of vital importance for a proper management of the infection. In our previous paper, an analysis of the isolation of enteric pathogens for the years 1969-73 was reported to clarify the status of those years. The present analysis was made based on the data obtained during the years 1974-78, to see if any change of the status was rendered. 1. During the 5-year period, from the cultures of 7,308 stool or rectal specimens 833 patients yielded enteric pathogens: 468 Shigella, 295 Salmonella, 30 Vibrio parahaemolyticus and 40 enteropathogenic Escherichia coli(EPEC). 2. Of the 295 Salmonella, 271 were S. typhi Isolation of 12 S.paratyphi-A, 1 Salmonella group B, 4 group C, 5 group D and 2 group E meant a definite increase of these sero-groups, S. typhi was most frequently isolated in August and in December, and from 30- to 39-year-old patients. 3. Of the 468 Shigella, 10 were subgroup A, 338 subgroup B, 3 subgroup C and 117 subgroup D. Most of the subgroup B belonged to type 1,2, or 3. The proportion of S. sonnei decreased from 31.3% in 1974 to 18.2% in 1978. In foreign patients, S. sonnei remained to be the frequntly isolated species. Shigella isolation was frequent in August and in 2- to 5-year-old patients. 4. V. parahaemolyticus was isolated from 30 and EPEC from 40 patients. 5. Ninty-nine per cent and 99.5% of the S. typhi isolates were susceptible to chloramphenicol and to ampicillin respectively. 92.8% of S sonnei were susceptible to ampicillin. S. flexneri type 2 was notable for their markedely decreased proportion being susceptible to ampicillin: 84.4% in 1974 and 25.6% in 1978.

Two Enteropathogenic Escherichia coli Strains Representing Novel Serotypes and Investigation of Their Roles in Adhesion

  • Enteropathogenic Escherichia coli (EPEC), which belongs to the attaching and effacing diarrheagenic E. coli strains, is a major causative agent of life-threatening diarrhea in infants in developing countries. Most EPEC isolates correspond to certain O serotypes; however, many strains are non-typeable. Two EPEC strains, EPEC001 and EPEC080, which could not be serotyped during routine detection, were isolated. In this study, we conducted an in-depth characterization of their putative O-antigen gene clusters (O-AGCs) and also performed constructed mutagenesis of the O-AGCs for functional analysis of O-antigen (OAg) synthesis. Sequence analysis revealed that the occurrence of O-AGCs in EPEC001 and E. coli O132 may be mediated by recombination between them, and EPEC080 and E. coli O2/O50 might acquire each O-AGC from uncommon ancestors. We also indicated that OAg-knockout bacteria were highly adhesive in vitro, except for the EPEC001 wzy derivative, whose adherent capability was less than that of its wild-type strain, providing direct evidence that OAg plays a key role in EPEC pathogenesis. Together, we identified two EPEC O serotypes in silico and experimentally, and we also studied the adherent capabilities of their OAgs, which highlighted the fundamental and pathogenic role of OAg in EPEC.

Isolation of New CHO Cell Mutants Defective in CMP-Sialic Acid Biosynthesis and Transport

  • Sialic acid is a sugar typically found at the N-glycan termini of glycoproteins in mammalian cells. Lec3 CHO cell mutants are deficient in epimerase activity, due to a defect in the gene that encodes a bifunctional UDP-GlcNAc 2-epimerase/ManNAc kinase (GNE). Sialic acid modification on the cell surface is partially affected in these cells. We have mutagenized Lec3 CHO cells and isolated six mutants (termed C2m) deficient in the cell surface expression of polysialic acid (PSA). Mutant C2m9 was partially defective in expression of cell-surface PSA and wheat germ agglutinin (WGA) binding, while in the other five mutants, both cell-surface PSA and WGA binding were undetectable. PSA expression was restored by complementation with the gene encoding the CMP-sialic acid transporter (CST), indicating that CST mutations were responsible for the phenotypes of the C2m cells. We characterized the CST mutations in these cells by Northern blotting and RT-PCR. C2m9 and C2m45 carried missense mutations resulting in glycine to glutamate substitutions at amino acids 217 (G217E) and 256 (G256E), respectively. C2m13, C2m39 and C2m31 had nonsense mutations that resulted in decreased CST mRNA stability, and C2m34 carried a putative splice site mutation. PSA and CD15s expression in CST-deficient Lec2 cells were partially rescued by G217E CST, but not by G256E CST, although both proteins were expressed at similar levels, and localized to the Golgi. These results indicate that the novel missense mutations isolated in this study affect CST activity.

Isolation and Characterization of a Butyric Acid Bacterium from Infant Feces (장내 항세균성 낙산균의 분리 및 특성)

  • To find bacteria which can inhibit growth of enteropathogenic Clostridium perfringens ATCC 13124, spore forming butyric acid bacteria were isolated from 26 fecal samples of infants. Fourteen strains were found to be antagonistic to the enteropathogen and five of them produced butyric acid. A strain which produced the highest butyric acid was selected and identified as Clostridium butyricum. This organism sporulated in SM medium in 36 hours with optimum rates at 37$^{\circ}C$ and at pH 5.5. The spores tolerated well at high heat and acidity, and possible application of Clostridium butyricum as intestinal controller was discussed.

Epidemiological Survey on Piglet diarrhea in eastern Chonnam province (전남동부지방의 설사자돈에 대한 역학조사)

  • This study was perfomed to examine the distribution of causative agent of piglets diarrhea in eastern Chonnam province from February 1994 to March 1995. The causative agents of diarrhea were examined by bacterial culture test, parasitological test and serological test against PED, TGE and Rota. The 35 isolated E. coli were tested for antibiotic sensitivity. The results obtained were as follows. 1. The incidence of 81 piglets with diarrhea was most prevalent as 39.5% in the age of 2 to 4 weeks, compare to 34.6% in the age of 5 to 8 weeks and 16.0% under 1 weeks and 9.9 % in the age of 9 to 11 weeks after birth. 2. The incidence of 81 piglets diarrhea showed bacterial diarrhea(75.3%), viral diarrhea (35.8%) and parasitological diarrhea(18.5%). When compared the isolation frequency of each etiological agent, enteropathogenic E. coli was most prevalent as 55.5% in bacterial diarrhea, rotavlrus enteritis as 18.5% in viral diarrhea and trichuriasis as 13.6% in parasitological diarrhea. 3. The complicated infection of piglets was most prevalent as 41.7% in rotavirus enteritis with enteropathogenic E coli in 24 complicated piglets diarrhea. 4. In antibiotic sensitivity test, isolates showed moderatly resistance to Tobramycin, Amikacin, Imipenem, Trimethoprim/Sulfamethoxazole, Gentamicin, Ampicillin but sensitivty to Ticarcillin/K. The 30 E. coli isolate showed multiple drug resistances in 3 different antibiotics.

The effects of Torilis fructus extracts against enteropathogenic Escherichia coli in Piglets (사상자 추출물의 자돈 병원성 대장균 억제 효과)

  • The antibiotic chemotherapy produces sometimes side effects and fails to eliminate bacterial infection. The occurrence of strains resistant to antibiotics would be expected to increase, and it is nowadays important to search for non-antibiotic substances. We are aimed to evaluate the effects of Torilis fructus extracts against enteropathogenic Escherichia coli (E. coli) in Piglets. The piglets were divided with three groups; Negative control group, E. coli-infected positive control group, and the Torilis fructus extracts treated group with E. coli infection. During the study period, we compared clinical signs, weight increase rate, fecal scores, gross findings between the treated group and non treated group. After necropsy, necropsy findings and histopathological findings were conducted with the comparison between the groups. As the results of this study, the Torilis fructus extracts additive showed the effects on the suppression of E. coli-induced lesions. On the basis of this study results, our data suggest that the Torilis fructus extracts additive have the antimicrobial effects. The Torilis fructus extracts additive could be used as the alternative material for antimicrobial feed additives.

Swarming Differentiation of Vibrio vulnificus Downregulates the Expression of the vvhBA Hemolysin Gene via the LuxS Quorum-Sensing System

  • Swarming has proven to be a good in vitro model for bacterial surface adherence and colonization, and the swarming differentiation of a bacterium has been shown to be coupled with changes in the expression of virulence factors associated with its invasiveness, particularly in the early stages of infection. In this study, we attempted to determine whether the expression of vvhA, which encodes for hemolysin/cytolysin (VvhA), is either upregulated or downregulated during the swarming differentiation of V. vulnificus. The insertional inactivation of vvhA itself exerted no detectable effect on the expression of V. vulnificus swarming motility. However, in our lacZ-fused vvhA transcriptional reporter assay, vvhA expression decreased in swarming V. vulnificus as compared to non-swarming or planktonic V. vulnificus. The reduced expression of vvhA in swarming V. vulnificus increased as a result of the deletional inactivation of luxS, a gene associated with quorum sensing. These results show that vvhA expression in swarming V. vulnificus is downregulated via the activity of the LuxS quorum-sensing system, suggesting that VvhA performs no essential role in the invasiveness of V. vulnificus via the adherence to and colonization on the body surfaces required in the early stages of the infection. However, VvhA may playa significant role in the pathophysiological deterioration occurring after swarming V. vulnificus is differentiated into planktonic V. vulnificus.

In Vitro Susceptibility of Diarrhea-Causing Escherichia coli to 9 Antibacterial Agents in Clinical Use (최근 분리된 장내 병원성 대장균의 항균제 감수성)

  • To determine the prevalence of antibiotic resistance in fecal E. coli and to investigate possible associations between antibiotic resistance and other plasmid-mediated virulence properties, antibiotic disk susceptibility tests for nine antibiotics were done on 141 strains of E. coli isolated from diarrheal children and well controls. Eighty two percent of the test strains were resistant to one or more antibiotics. Antibiotics to which the test strains were most resistant in descending order were ampicillin (85%), trimethoprim/sulfamethoxazol (60%), and cephalothin (55%). Seventy nine percent of these resistant strains were resistant to two or more antibiotics. All 141 test strains were sorted into enterotoxigenic E. coli (ETEC), enteropathogenic E. coli (EPEC), enteroadherent E. coli (EAEC) and non-pathogenic E. coli and the percentages of strains resistant to multiple antibiotics were compared. Among ETEC regardless of its source, multiple drug resistance was more frequent in strains producing heatstable enterotoxin (ST) only than in strains producing only heat-labile enterotoxin (LT) or both. In EAEC, multiple resistance was more frequently associated with strains isolated from diarrheal patients than with those from well controls. The major antibiotic resistance patterns possessed by multiple resistant enteropathogenic strains were $SXT^R$ $AM^R$, $CR^R$, and $SXT^R$ $AM^R$ $CR^R$. Of 28 ST- producing $SXT^R$ ETEC, 26(96%) were also resistant to ampicillin and 17 (61%) were resistant to cephalothin. The similar pattern was observed in EAEC and EPEC as well. This study has important implications for the treatment of E. coli diarrhea with antibiotics because it is possible that dissemination of virulence could occur under the force of selective antibiotic pressure. In addition, this study suggests that the in vivo efficacy of SXT in treating diarrheal illness be reevaluated.

Comparison of Biochemical Identification to Detect Pathogenic Escherichia coli in Fresh Vegetables (신선편이 엽채류의 병원성 E. coli 검출을 위한 생화학적동정법 비교 분석)

  • The objective of this study was to isolate pathogenic Escherichia coli from fresh vegetables with selective media and Petrifilm, and identify a suspicious colony using biochemical identification. Twenty gram of lettuce, twenty gram of cabbage and ten gram of sprout were prepared, and a 5-strain mixture of pathogenic E. coli (Enterohemorrhagic E. coli NCCP11142, Enterotoxigenic E. coli NCCP14037, Enteropathogenic E. coli NCCP14038, Enteroaggregative E. coli NCCP14039, Enteropathogenic E. coli NCCP15661) was inoculated to obtain 1, 2 and 3 log CFU/g. Eighty to ninety milliliter of buffered peptone water (BPW) was placed and pummeled for 60 s. As a results, the Petrifilm method was all positive, but enrichment method of qualitative analysis was negative except for 3-log CFU/g inoculated lettuce. Regarding biochemical identification of pathogenic E. coli, the identification rates were dependent on type of methods and vegetables; lettuce: API 20E 100% (44/44), Microgen GNA 100% (44/44) and Food System 66.7% (10/15), cabbage: API 20E 64.7% (22/34), Microgen GNA 50% (16/32) and Food System 60% (9/15), sprout: API 20E 65.1% (28/43), Microgen GNA 62.3% (27/43) and Food System 53.3% (8/15). These results could be useful in determining an appropriate method to detect pathogenic E. coli in fresh vegetables.
