Infection Control of Computed Radiography Portable in Radiology (영상의학과 이동촬영장비의 감염 관리)

  • The purpose of this study is to prevent infection in the hospital by computed radiography portable and to provide basic data on infection-related education by investigating bacterial contamination level of computed radiography portable equipment using IP cassette. The results suggest that IP cassette No. 1 is infected with CNS and VRE, no. 2 with CNS, No. 3 with CNS and Pseudomonas aeruginosa, No. 4 with CNS, No. 5 with CNS and Bacillus sp., and No. 6 with enterococcus faecium. Enterococcus faecium and bacillus sp. were detected from the IP reader and Acinetobacter baumannii was detected on the mobile handle; Bacillus sp. on the control buttons, CNS and Bacillus sp. from the irradiation control handle, Acinetobacter baumannii on the x-ray generation switch, and CNS on the barcode scanner. In addition, Bacillus sp. Acinetobacter baumannii was found on the IP cassette mobile table and CNS and bacillus sp. were found on the lead apron. Acinetobacter baumannii and CNS were detected from the medical gloves worn by a radiological technologist during radiography. This suggests that IP cassette should be sterilized after use as it can hand over bacteria to IP reader and IP mobile table. Medical gloves that are in direct contact with patients should also be replaced after using them once and other supplies such as x-ray generation switch and lead apron should thoroughly be sterilized to prevent infection due to radiography as they are in a lot of contact with patients.

Antibiotic Susceptibility and Genetic Diversity of Enterococci Isolated from Clinical Specimens (임상검체에서 분리한 장구균의 항생제 감수성 및 유전적 다양성)

  • The purpose of this study was to investigate the distribution of Enterococci isolated from clinical specimens, and identify the aspect of antibiotic susceptibility and analyze the genetic difference by executing Rep-PCR over the strains resistant to aminoglycoside-typed antibiotics. From an assortment of the clinical specimens, 100 strains were isolated. The collection consisted of 49 strains of E. faecalis, 34 strains of E. faecium, 9 strains of E. avium, 4 strains of E. gallinarum, 3 strains of E. casseliflavus, and 1 strain of E. hirae. Ninety five were isolated from inpatients, and five strains were isolated from outpatient. Most of the E. faecalis and E. faecium were originated from urine, pus, and sputum. Most Enterococci showed 80% resistance to the cephalosprin-typed antibiotics. E. faecium showed the high resistance to all the antibiotic substances. One tenths of Enterococci showed the resistance to vancomycin. And also, most Enterococci showed the high resistance to amikacin and gentamicin as aminoglycoside-typed antibiotics. Genetic diversity of the resistant strains to aminoglycoside estimated using Rep-PCR was not significanty different.

Identification of Bacteriocin-producing Lactic Acid Bacteria from Kimchi and Partial Characterization of their Bacteriocin

  • Nineteen strains of bacteriocin-producing lactic acid bacteria were isolated from 432 Kimchi samples, and identified by the comprehensive biochemical and morphological tests verifying their cellular fatty acid composition. Using partially purified bacteriocins from these isolates, their inhibitory activities against other lactic acid bacteria and some pathogens, and sensitivity to enzyme and heat treatments were tested. The isolates were identified as Lactobacillus plantarum (2 strains), L curvatus (2 starins), L brevis (2 strains), Enterococcus faecium (6 strains), Leuconostoc mesenteroides subsp. mesenteroides (1 strain) and Lactobacillus sp. (6 strains). The bacteriocins produced by E. faecium strains provided the broadest spectrum of inhibition, affecting against other Gram-positive bacteria including lactic acid bacteria and health-threatening bacteria such as Clostridium perfringens and Listeria monocytogenes. The bacteriocins of Lactobacillus sp., L plantarum and L brevis strains were capable of inhibiting many strains of the lactic acid bacteria, whereas those of L curvatus and L mesenteroides subsp. mesenteroides strains were only inhibitory to a few strains. Generally, the inhibitory activities of both E. faecium and Lactobacillus sp. strains were greater than those of other producer strains. The bacteriocins from the isolates were sensitive to several proteolytic enzymes, and those of L curvatus and L mesenteroides subsp. mesenteroides were also sensitive to lipase and $\alpha$-amylase as well as to proteolytic enzymes. The bacteriocins from the strains of Lactobacillus sp. and a strain of L. brevis were resistant to autoclaving.

Variations of Hydrogen Production in the Presence of Heavy Metals During Anaerobic Fermentation of Food Waste (음식물쓰레기의 혐기성 소화 시 중금속에 따른 수소생산량의 변화)

  • In this study, variations of hydrogen production were investigated with food waste fermentation in the presence of heavy metals. Hydrogen production was 79.48 mL/g COD with fermentation of food waste. In the presence of 1 mg/L of zinc, the hydrogen production was decreased about 60%. When the copper is present, the production of hydrogen is severely inhibited, while the coexistence of copper with zinc relaxes the inhibition of copper and restores hydrogen production. Butyric acid or acetic acid was observed as the main species during hydrogen production. Klebsiella sp., Clostridium sp., and Dysgonomonas sp. were mainly appeared in the samples not containing heavy metals. However, Enterococcus sp. extremely influenced the hydrogen production activities of samples containing zinc or copper.

동물의 장에서 분리한 Enterococcus sp.의 특성 및 분말화

  • In order to develop a lactic acid bacterial powder which can be used as a probiotic for human and animal, a lactic acid bacteria which has high resistance against low pH and ox-gall, and shows a good growth inhibition against E. coli, was isolated from an animal intestine and characterized. The isolated strain was identified as Enterococcus faecium. It had more than 90% of survival at low pH for 2 hours and almost 100% of survival in the presence of 0.3% ox-gall. When co-cultured with E. coli in MRS broth, all of the E. coli cells were killed within 24 hours. The final powdered product of the isolated strain was manufactured after a freeze drying process using an industrial media, and then checked its stability. Its storage stability was 80% for 11 months at 18$\circ$C.

Characterization of a Human Intestinal Bacterium, Enterococcus sp. GLY Capable of Converting Glycyrrhizin to Glycyrrhetic Acid (감초의 주성분인 glycyrrhizin을 대사하는 사람 장내세균의 특성 연구)

  • Glycyrrhizin은 감초의 주성분으로 많은 생리활성을 나타내는 성분이다. 감초를 복용하게 되면, 글리시리진은 장내에 서식하고 있는 장내세균에 의해 glycyrrhetic acid로 대사된다. 하지만, 지금까지 대사반응에 관여하고 있는 균주가 분리 후 자원화되지 않았다. 본 연구를 통해 사람의 대변으로부터 glycyrrhizin을 대사하는 균주로 분리된 strain GLY의 16S rRNA 유전자를 분석한 결과, Enterococcus faecium과 99%의 상동성을 보여, Enterococcus faecium GLY (Genebank No. MH048988)로 명명하였다. 대사활성의 특성을 검토하기 위하여 time course, 기질의 농도에 따른 대사활성의 변화, 근연 균주와 대사활성 비교를 실시하였다. Time course 실험에서 GLY균주가 증식함에 따라 기질인 glycyrrhizin은 줄어들고, 대사산물인 glycyrrhetic acid는 새로 생성되었다. 또한, 기질의 농도에 따른 대사활성의 차이 검토를 위해 여러 농도를 처리하여 배양하였을 때, 1.0mM을 처리한 샘플에서 최대값의 대사물 농도를 보였다. Enterococcus faecium와의 근연균주를 이용하여 glycyrrhizin 대사활성을 측정한 결과, strain GLY 균주가 가장 큰 대사능을 가지고 있었다. 본 연구는 앞으로 glycyrrhizin 생체이용 및 대사 연구를 위한 기초연구가 될 것이며, 장내세균에 의한 생약성분의 대사를 이해하는데 도움이 될 수 있다.

The study on the causal agent of Streptococcicosis (Lactococcus garvieae), isolated from cultured marine fishes (해산 양식어류로부터 분리된 연쇄구균증의 원인균, Lactococcus garvieae에 대한 연구)

  • The purpose of this study was to investigate the microbiological characteristics and the distributions of the bacteria causing streptococcicosis occurred in marine fish farm, Korea. Many kinds of cultures fishes suffered from the disease accompanied with typical symptoms, including darkening of the skin, exophthalmia, petechiae inside of the opercula and distended abdomen. The isolates from the diseased fishes were compared with Lactococcus garvieae by biochmical, biophysical and serological methods and polymerase chain reaction(PCR) assay. We isolated 35 strains of the geuns Streptococcus from the diseased olive flounder, Paralichthys olivaceus, yellow tail, Seriola quinqueradiata and Korean rockfish, Sebastes schlegeli. 15 strains out of the isolates were identified to L. garvieae and the others were not because of their different biochemical and biophysical charateristics. Seven strains of the isolates were agglutinated by rabbit serum raised against L. garvieae $KG^+$ phenotypic cells(ATCC49156)as a reference strain. Twenty-one strains of the isolates identified to L. garvieae since they were formed the expected band through performing PCR assay using specific primers, pLG-1(5'-CATAACAATGAGATCGC-3') and pLG-2(5'-GCACCCCGCGGTTG-3'). In the present study, it showed that L. garvieae was a dominant strain causing streptococcicosis in the tested area due to occurrence of 21 strains as L. garvieae out of all the isolates, 9 atrains as Streptococcus sp. and 5 strains as Enterococcus sp.

The Bacterial Contamination in Glasses for Vision Correction (시력 교정용 안경의 세균 오염)

  • Purpose: Recently, bacterial contamination of equipment and accessories required for vision correction has become a main causal factor in ophthalmic diseases. Thus, We investigated on both the actual condition of bacterial contamination from glasses of vision correction. Methods: Investigation of microorganisms was carried out with a group of 145 glasses wearers, composed of 36 elementary school students, 37 middle school students, 38 high school students, 10 college students, and 32 aged men. Results: Seventeen species of bacteria are detected from glasses of vision correction: B. cereus, B. licheniformis, Bacillus sp., CNS, Enterococcus sp., Escherichia coli, Proteus sp., Pseudomonas sp., Serretia sp., Streptococcus sp., Staphylococcus epidermidis, Staphylococcus aureus, Streptococcus hemolyticus,, Acinetobacter sp., Enterobacter cloacae, GNR, and Pseudomonas aeruginosa. Among 17 species of bacteria, there are some potential causative agents for keratitis, corneal ulcer, Acute dacryocystitis, Orbital cellulitis, Periphlebitis retinae, Marginal blepharitis, and Acute conjunctivitis. Enterobacter cloacae, Pseudomonas aeruginosa and Staphylococcus epidermidis cause keratitis. Pseudomonas sp., and Staphylococcus aureus cause corneal ulcer. Staphylococcus aureus causes acute dacryocystitis, orbital cellulitis, periphlebitis retinae, marginal belpharitis. Streptococcus hemolyticus causes acute conjunctivitis. Conclusions: In summation, it is verified that hazardous, opportunistic and infectious microorganisms exist in glasses for vision correction. Ophthalmic diseases are predicted. Therefore, supplementary research on the development of a cleaning solution to cleanse the infection and of an effective method to remove microorganisms is required.

Clostridium difficile-associated Intestinal Disease and Probiotics

  • Probiotics are traditionally defined as viable microorganisms that have a beneficial effect in the prevention and treatment of pathologic conditions when they are ingested. Although there is a relatively large volume of literature that supports the use of probiotics to prevent or treat intestinal disorders, the scientific basis behind probiotic use has only recently been established, and clinical studies on this topic are just beginning to get published. Currently, the best studied probiotics are lactic acid bacteria, particularly Lactobacillus and Bifidobacterium species. Other organisms used as probiotics in humans include Escherichia coli, Streptococcus sp., Enterococcus sp., Bacteroides sp., Bacillus sp., Propionibacterium sp., and various fungi, and some probiotic preparations contain more than one bacterial strain. Probiotic use for the prevention and treatment of antibiotic-associated diarrhea caused by Clostridium difficile induced intestinal disease as well as for other gastrointestinal disorders has been discussed in this review.
