Isolation of Enterobacter Cloacae Producing Phytase and Medium Optimization of Its Production (Phytase를 생산하는 Enterobacter cloacae의 분리 및 효소 생산의 배지 최적화)

  • Phytase (myo-inositol hexakisphosphate phosphohydrolase: EC 3.1.3.8) hydrolyzes phytic acid (myo-inositol hexakisphosphate) to myo-inositol and monophosphates. In order to obtain phytase producing bacteria, many samples were collected from various soils. Among thirty-five phytase-producing strains, YH100 showed the highest phytase activity. In order to identify the selected YHlOO strain, the morphological and physiological characteristics were examined according to the method of Bergey's manual by 168 rRNA sequence, cellular fatty acids profile, O+C contents and physiological test using API 20E kit. The strain YH100 identified to be a genus of Enterobacter cloacae and was named as Enterobacter cloacae YHlOO. Optimum medium for the phytase production by the Entemhacter c!o([we YHlOO was composed of 2.0%(w/v) glucose, 1.0%(w/v) peptone, 1.0%(w/v) beef extract, 0.1 %(w/v) KCI. and 0.1 %( w/v) sodium phytate.

Decolorizing Characteristics of Crystal Violet by Enterobacter cloace MG82. (Enterobacter cloacae MG82에 의한 Crystal Violet의 탈색특성)

  • Decolorizing characteristics of crystal violet by Enterobacter cloace MG82, which can decolorize rapidly triphenylmethane dyes, were investigated. The higher growth and decolorization activity was shown at big ratio of dissolved oxygen in the medium. The decolorization activity of crystal violet revealed highest at the middle of lag phase. As the concentration of crystal violet was higher, the growth of E. cloacae MG82 and decolorizing activity of crystal violet by this strain were worse. The maximum concentration of crystal violet at which E. cloacae MG82 be able to grow was 375 ${\mu}$M. E. cloacae MG82 was not able to use the crystal violet itself as a sole carbon source. So, it was shown that growth of E. cloacae MG82 and decolorization activity of crystal violet by this strain needed addition of another energy sources except this dye.

Growth Inhibition of Water Extract of Schizandra chinensis Bullion on the Bacteria (오미자 물추출물의 세균증식 억제효과)

  • This study was conducted to find material having antibacterial activity. The effects of preservatives, antibiotics and oriental medicines on growth of teated microorganisms were investigated. The growth of all tested bacteria was inhibited by water extract of Schizandra chinensis. Antibacterial activity on the concentration of Schizandra chinensis extract was tested. The growth of Escherichia coli W3110, Enterobacter colacae MG82 and Salmonella typhimurium was extraordinarily inhibited by more than 0.2% concentration of Schizandra chinensis extract. The specific growth rate of Escherichia coli W3110, Enterobacter cloacae ME82 and Salmonella typhimurium under control conditin had mean values of 0.514(hr ̄), 0.381(hr ̄) and 0.489(hr ̄), respectively. When 0.2% of Schizandra chinensis extract was added, specific growth rates of Escherichia coli W3110, Enterobacter colacae MG82 and Salmonella typhimurium wre decreased, compared to contorl, in 1.26, 2.23 and 1.50 fold, respectivley. Minimal inhibiotory concentration of Schizandra chinensis extract was 0.25% on the tested microorganisms The growth of Enterobacter cloacae MG82 was more inhibited by Schizandra chinensis extract than other tested microorganisms.

The Antibacterial Action of Chinese Cabbage Kimchi Juice on Staphylococcus aureus, Salmonella enteritidis, Vibrio parahaemolyticus and Enterobacter cloacae (배추김치즙의 Staphylococcus aureus, Salmonella enteritidis, Vibrio parahaemolyticus 및 Enterobacter cloacae에 대한 항균작용)

  • Antibacterial activities of Chinese cabbage kimchi juice were determined against Staphylococcus aureus ATCC 6538p, Salmonella enteritidis ATCC 13076, Vibrio parahaemolyticus ATCC 17802 and Enterobater cloacae ATCC 13047. It was found out that 2% content of Chinese cabbage kimchi juice reduced the colony numbers of Salmonella enteritidis ATCC 13076, Enterobater cloacae ATCC 13047, Staphylococcus aureus ATCC 6538p and Vibrio parahemolyticus ATCC 17802 by 63%, 72%, 76% and 90%, respectively. 6% content of Chinese cabbage kimchi Juice completly inactivated Vibrio parahaemolyticus. But at the same content (6%) of Chinese cabbage kimchi juice, colony number of Salmonella enteritidis ATCC 13076 Enterobater cloacae ATCC 13047 and Staphylococcus aureus ATCC 6538p were reduced to 11%, 10% & 9%, respectively.10% Chinese cabbage kimchi juice had 100% inhibitory effect on all test bacteria. Therefore, Chinese cabbage kimchi has effective antibacterial activities against Staphylococcus aureus ATCC 6538p, Salmonella enteritidis ATCC 13076 Vibrio parahaemolyticus ATCC 17802 and Enterobacter cloacae ATCC 13047.

  • We investigated the prevalence of extended spectrum ${\beta}$-lactamase (ESBL) genes and 16S rRNA methyltransferase genes to study antimicrobial resistance mechanisms of Enterobacter cloacae strains isolated from a university hospital in the Chungcheong province of Korea. Eight of the bacteria strains involved in this study contained CTX-M-15 type ESBL. Among 8 strains harboring the ESBL gene, 3 strains also harbored armA gene. The three isolates showed resistance to antimicrobial agents belonged to third cephalosporin, aminoglycoside, and fluoroquinolones. Furthermore, interspecies plasmid transfer of the antimicrobial resistant genes may induced horizontal spreading of the genes and emergence of multidrug resistant bacteria. Therefore, surveillance for existence of antimicrobial resistance determinants is important to prevent distribution of antimicrobial resistant strains.

Production of Biogenic Amines by Microflora Inoculated in Meats

  • The effects of microorganisms inoculated in beef, pork and chicken on the production of various biogenic amines (BA) were examined. Acinetobacter haemolyticus, Aeromonas hydrophila subsp. hydrophila, Alcaligenes faecalis subsp. faecalis, Bacillus cereus, Bacillus subtilis, Enterobacter aerogenes, Enterobacter cloacae, Escherichia coli, Lactobacillus alimentarius, Lactobacillus curvatus, Leuconostoc mesenteroides subsp. Mesenteroides, Proteus mirabilis, Proteus vulgaris, Pseudomonas aerugina, Salmonella enteritidis and Salmonella typhimurium were inoculated into beef, pork and chicken and incubated for 24 h at optimum temperatures of each bacterium. In ground beef, total amount of amines (TAA) produced was highest in the sample inoculated with Bacillus cereus, followed by Enterobacter cloacae. In ground pork, TAA was highest in the sample inoculated with Alcaligenes faecalis, followed by Enterobacter cloacae, Proteus vulgaris and Bacillus cereus. TAA of chicken breast was highest in the sample inoculated with Alcaligenes faecalis, followed by Bacillus cereus and Lactobacillus alimentarius while in chicken leg was the sample inoculated with Proteus vulgaris, followed by Enterobacter aerogenes, Enterobacter cloacae and Alcaligenes faecalis. Among biogenic amines produced, cadaverine (CAD) was detected at the highest level, followed by putrescine (PUT) and tyramine (TYM), their order being reversed by the kind of microorganism in beef and pork. In chicken breast and leg, CAD level was still the highest but PUT, TYM or PHM was the second highest, depending upon the kind of microorganism inoculated. In total, Alcaligenes faecalis, Enterobacter cloacae and Bacillus cereus were ones that produced a larger amount of BAs regardless of meat sources from different species.

Optimization of Cu, Hg and Cd removal by Enterobacter cloacae by ferric ammonium citrate precipitation

  • Iron precipitating organisms play a significant role in the formation of ferric hydroxide precipitate, which acts as strong adsorbent for toxic metal. In this respect four different iron precipitating cultures were isolated from Hutti gold mine surface winze water sample on citrate agar medium. The best isolate was screened out for metal removal study on the basis of fast visual iron precipitation. The selected isolate was identified as Enterobacter sp. based on routine biochemical tests and Biolog GN microplate results and as Enterobacter cloacae subsp. dissolvens by 16S rRNA gene sequence analysis (GenBank accession number EU429448). Influence of medium composition, medium initial pH, the influence of inoculum size, effect of various media and ferric ammonium citrate concentration were studied on metal removal in shake flask experiments. Under the optimized conditions studied, E. cloacae showed $94{\pm}2$, $95{\pm}2$ and $70{\pm}2%$ of cadmium, copper and mercury removal from a simulated waste in shake flask studies. In lab scale column reactor more than 85% of copper and mercury removal was achieved.

Medium Composition of Enterobacter cloacae YJ-1 for Maximizing Hydrogen Production (수소생산 증진을 위한 Enterobacter cloacae YJ-1의 배지조성)

  • In order to maximize hydrogen production by Enterobacter cloacae YJ-1, anaerobic hydrogen producing bacteria, the medium composition was optimized. Glucose was better than other carbon sources in hydrogen production and its production was 975.4 mL/L at $2\%$ (w/v) for 48 h. Organic nitrogen sources were more effective than inorganic nitrogen sources and also yeast extract among organic nitrogens was the most effective in hydrogen production. Among metal ions, $Na_2MoO_4$ was most effective, and its production was 1753.3mL/L at $0.04\%$ (w/v). Addition of amino acid was very effective with compare to another components of medium, and cystein was most effective among them. Under the optimum medium obtained in batch culture, semi-batch culture in order to produce continuous hydrogen was run. The highest hydrogen production was earned at $3\%$(w/v) of glucose and the amount was 2215.4 mL/L.

Microbes causing abnormal Takju fermentation in traditional wheat-based Nuruk: A case study (밀누룩으로 제조한 전통탁주의 이상발효 원인 규명: 사례분석)

  • In this article, we report a case study on the cause of abnormal fermentation of Takju based on a microbial community analysis of wheat-based Nuruk and Takju using next-generation sequencing. The microbial community showed notable changes during alcoholic fermentation. Members of the genera Aspergillus and Staphylococcus were dominant in Nuruk, whereas Saccharomyces, Pediococcus, Bacillus, Lactobacillus, and Enterobacter were predominant during Takju fermentation. In particular, members of the Enterobacter and Bacillus genera that were present were identified as the opportunistic pathogens B. wiedmannii and E. cloacae. The relative abundance of two species increased in Mitsul-3, wherein the concentration of organic acid rapidly decreased, and in Deotsul-1, in which raw materials were added. Furthermore, the relative abundance of lactic acid bacteria (LAB) in both the Nuruk and Takju was very low. Therefore, this result showed that the abnormal proliferation of E. cloacae and B. wiedmannii in Takju was due to the absence of LAB.

The Production of Folic Acid by Microorganisms Isolated from Fermenting Corn Meal (옥수수 가루 발효 과정에서 분리한 미생물에 의한 Folic Acid의 생산)

  • Twenty-five out of 35 strains isolated from fermented corn meal produced folic acid. Bacillus licheniformis strain 6 and Enterobacter cloacae strain 18 produced the largest quantities of 1830$\pm$271 ng and 1350$\pm$161 ng per 100$m\ell$ of the assay broth, respectively. B. licheniformis produced maximum yields when initial pH values were 6,7, or 8 and were incubated at 35$^{\circ}C$ for 5 days. The initial pH (range 4-8) had no effect on folic acid production by E. cloacae; 55 $^{\circ}C$ for 5 days was optimal for this bacterium. Added carbohydrates had no effect on the production of total folic acid in the bacterial cells in pure or mixed cultures. However, in their growth media, carbohydrates enhanced the production of free and total folic acid by E. cloacae and in the mixed cultures. Added carbohydrates had no significant (P < 0.05) effect on the production of free and total folic acid by B. licheniformis.
