Microtubules and microfilaments are major cytoskeletal components in mammalian ova that provide the framework for chromosomal movement and cellular division. Extensive changes of cytoskeletal organization occur during maturation and fertilization. The changes in cytoskeletons are essential for the normal meiotic maturation and for the formation of the biparental diploid genome of the embryo, and thus are repeated at each cell cycle during embryonic development. Disturbance of the cytoskeletal organization could result in abnormal gamete development and early embryonic death.

Tasmania with its clean air, isolated from mainland Australia, has been producing high-quality agricultural products and has been continually developing organic farming since 1946 when the Living Soil Association of Tasmania(LSAT) was established. The organic farming movement in Tasmania has been actively advocated through three steps: the philosophical embryonic period, the movement diffusion period and the industrialised development period. The campaigns for informing about the connection between healthy soil and life unfolded during the embryonic period. This was followed by the birth of publicity of organic farming and the certification system through the dissemination of organic farming techniques and various events related to agriculture in the diffusion period when the Organic Gardening and Farming Society(OGFS) was established in 1972. In the industrialised development period, The Organic Coalition of Tasmania (OCT) which is representative of Tasmania was organised in 2000 and has been leading the organic farming industry. The organic farming movement in Tasmania not only limits the use of artificial agricultural chemical but pursues the quality of food, environment, the health of life including all animals and plants, the issue of development in rural society, social justice, and equity in understanding. It is far more holistic in its philosophy. The output of organic food accounts for 1 % of the total amount of agricultural production and 150 certified organic farms have managed with 5,000ha of land in 2010. The supply channels for organic foods vary from farmer's market, specialty stores, supermarket chains, local store to the cooperative community. Also the consumers' behaviour for organic foods has been establishing as an alternative life style. The education of the value and role of organic farming on the environment should be enlarged for the consumption of the organic food. In addition, organising for small farmers who act individually and the link with differentiated local food have still remained issues.

In eukaryotes, chromosomes undergo a series of complex and coordinated movements during cell division. The kinesin motor proteins, such as the chicken Chromokinesin, are known to bind DNA and transport chromosomes on spindle microtubles. We previously cloned a family of retrograde C-terminus kinesins in Caenorhabditis elegans that mediate chromosomal movement during embryonic development. Here we report the cloning of a C. elegans klp-12 cDNA, encoding an ortholog of chicken Chromokinesin and mouse KIF4. The KLP-12 protein contains 1609 amino acid and harbors two leucine zipper motifs. The insitu RNA hybridization in embryonic stages shows that the klp-12 gene is expressed during the entire embryonic development. The RNA interference assay reveals that, similar to the role of Chromokinesin, klp-12 functions in chromosome segregation. These results support the notion that during mitosis both types, the anterograde N-terminus kinesins such as KLP-12 and the retrograde C-terminus kinesins, such as KLP-3, KLP-15, KLP-16, and KLP-17, may coordinate chromosome assembly at the metaphase plate and chromosomal segregation towards the spindle poles in C. elegans.
Angiogenesis is regulated by a large number of molecules and complex signaling mechanisms. The G protein $G{\alpha}_{13}$ is a part of this signaling mechanism as an endothelial cell movement regulator. Gene expression analysis of $G{\alpha}_{13}$ knockout mouse embryos was carried out to identify the role of $G{\alpha}_{13}$ in angiogenesis signaling during embryonic development. Hypoxia-inducible response factors including those acting as regulators of angiogenesis were over expressed, while genes related to the cell cycle, DNA replication, protein modification and cell-cell dissociation were under expressed. Functional annotation and network analysis indicate that $G{\alpha}_{13}{^{-/-}}$ embryonic mice were exposed to hypoxic conditions. The present analysis of the time course highlighted the significantly high levels of disorder in the development of the cardiovascular system. The data suggested that hypoxia-inducible factors including those associated with angiogenesis and abnormalities related to endothelial cell division contributed to the developmental failure of $G{\alpha}_{13}$ knockout mouse embryos.
Could STS throw another-colored light on the Hwang's Affair, the scientific fraud committed by Hwang Woo-Suk and his research team in Korea? And could analytic tools of STS unfold another meanings which have been overlooked in most of the traditionally social-sciences-oriented analyses? In this essay, I try to answer these questions by analyzing the Hwang's Affair in the view of STS, especially by using some concepts of actor-network theory(ANT): movement, translation and displacement. I want to say that the Hwang's Affair seems to be a part of normal scientific activity, not an abnormal phenomenon, and as an evidence, focus on the similarities of their life styles between "pure/real scientist" Louis Pasteur and "impure/political scientist" Hwang Woo-Suk. I try to mobilize some concepts of ANT, especially movement, and find out why scientists came to move toward the opposed direction on the pure/real-impure/political line. I suggest that there exists "laboratory politics" as the key factor in this bifurcation. My tentative conclusion is that Pasteur can take a position to make his great world, so-called the Pasteurian world, owing to the success of "double movement" in which he treated his laboratory as a fulcrum to lift up the world, but Hwang degrades himself to "ugly scientific politician" due to the loss of the momentum of his movement; Hwang treated his laboratory only as the symbolic resources and in turn failed to solidify material entities, his real political resources, even though he knew the importance of laboratory.
Real time B-mode ultrasound was used to detect the early conceptus in 187 Korean native cattles between days 10 and 60 after last insemination. The ultrasound diagnostic findings were systemically confirmed by palpation per rectum after the 60th day of last insemination. The embryonic vesicle and the embryo proper within the veside were first visible on mean day fl and 23, respectively. The heartbeat of the embryo proper could be detected on day 26, and the limb buds, placentomes, amnion, fetal movement, umbilical cord, optic area and split hooves were first visible on day 33, 34, 34, 44.5, 45, 32 and 48, respectively. The mean length of embryo proper was 3.8mm on day 23 which later increased to 56. 6rnrn on day 60. When ultrasound was used to detect the conceptus between days 20 and 30 after insemination and palpation per rectum after the 60th day of insemination, the accuracy rates of pregnancy detection by ultrasound scanning at days 20, 22, 24, 26, 28, 30 were 44.4, 69.2, 78.6, 87.5, 90.0, 93.3%. In summary, the early pregnancy diagnosis of Korean native cattle with ultrasound appears high accuracy rates. It is considered that ultrasound can be used in veterinary practice well.
Plasma progesterone (P$_4$) concentrations were measured for confirming the estrus observation and for the early pregnancy diagnosis in 130 cows of small farmers. Ultrasonographic examinations were performed from day 30 after artificial insemination to establish the characteristic ultrasonographic appearances of gestational structures in each pregnant stages. Of the 130 cows inseminated, 111 cows (85.4%) were an ovulatory estrus, 12 cows (9.2%) were an unovulatory estrus, and 7 cows (5.4%) were the error of estrus detection, respectively. The accuracy for early pregnancy diagnosis in 111 ovulatory estrus cows achieved when the discriminatory concentration at day 21 after artificial insemination was placed at 3.0 ng-/ml in plasma, was 86.7 % for positive diagnosis and 100% for negative diagnosis, respectively. Pregnancy diagnosis by ultrasonography were performed to evaluate gestational structures from day 30 after artificial insemination in 83 cows. Pregnant cows were 72 of 83 cows. The characteristic ultrasonography of gestational structures in each gestational stages was as follows. The embryo proper was observed within anechoic fetal fluid between 28 and 40 days after insemination, and amnion and embryonic heartbeat was also detected in this period. Between days 41 and 50, embryo proper was detected as an discriminated from head and body, and forelimb buds and hindlimb buds were also observed in this period. Between days 51 and 60, an embryo proper was clearly discriminated from head and body, and fetal movement, forelimb buds and hindlimb buds were observed in this period. Between days 61 and 70, fetus was completely developed, and fetal skeleton, organs and cotyledon were observed. After day 71, each organs of fetus were rapidly developed and a fetus was partially observed in screen because fetus was too big and larger, These results indicate that plasma P$_4$ determination at days 0,6 and 21 after artificial insemination can be utilized for confirming the estrus observation and for early pregnancy diagnosis. Also, ultrasonography was reliable method for early pregnancy diagnosis at day 30 after artificial insemination.

Although Pacific mackerel (Scomber japonicus) is an important commercial species in Korea, its recruitment mechanism remains largely unknown. Diel vertical positioning of larvae in the water column, which is affected by their specific gravity and the surrounding water density, may help to provide an understanding on recruitment success through predator avoidance and prey availability. The specific gravity measurement on Pacific mackerel eggs and larvae would seem to be essential information necessary to learn about the transport process from spawning to nursery grounds, and consequently the recruitment success. Eggs were artificially fertilized, and larvae were fed with rotifer when their mouths opened 3-4 days after hatching. We conducted the experiment using a density gradient water column to measure the ontogenetic changes in specific gravity from fertilization to 10 days after hatching. Egg specific gravity was stable during most of the embryonic period, but a sudden increase to $1.0249g\;cm^{-3}$ happened just before hatching. However, the specific gravity of newly hatched larvae was much lighter ($1.0195g\;cm^{-3}$), and specific gravity tended to increase continuously after hatching. Comparison of specific gravity with seawater density reveals that eggs and newly hatched larvae can float in the surface layer of the ocean. For the later period of the experiment, the specific gravity showed a cyclic diel pattern: the highest in the evening while the lowest at dawn. The fullness of larval stomach may be responsible for the observed differences in specific gravity, because stomach fullness was lower (40-60%) at midnight, and higher (80-85%) in evening. The diel pattern of specific gravity might provide clues regarding how larvae match the diel vertical migration of prey organisms.
Life can be kept in suspended animation either before fertilization at oocyte stage or after fertilization at different stages of embryonic development for a variety of reasons. It not only has potential applications in fertility preservation and management in human but also has important roles in the preservation and management of animal genetic resources, low-cost international movement of selected genetics, and rapid dissemination of germplasm through assisted reproductive technologies (ART) and genetic engineering. Currently, slow-freezing and vitrification are the two approaches by which oocytes and embryos can be cryopreserved for long-term storage. Both of these methods have their own advantages and disadvantages but allow the cryopreservation of oocytes and embryos with comparable efficiency. Vitrification of oocyte and embryos, although proven successful just 13 years after slow-freezing, is generally considered an emerging technology and appears to slow gain acceptance in both animal and human ART despite having controversial storage and contamination issues. In this manuscript, we discuss the basic techniques of oocyt/embryo cryopreservation and review the current status and recent developments in vitrification.
