  • 최근에 임목의 영양번식체계나 우량묘목을 양성하기 위하여 임목의 체세포배를 이용해서 "인공종자" 형태로 보관하거나 연속배양기를 사용하여 계속적으로 체세포배를 생산하는 방법은 매우 중요한 기법에 속한다. 이러한 체세포 배양기법은 임목의 생물공학 뿐만아니라 체세포배 발생학, 세포 및 분자생물학 연구에 있어서 중요한 역할을 한다. 현재 침엽수 체세포 배양에 대한 복잡한 기작이 많이 밝혀지고 있으나 이러한 기초적 지식이 현실적으로 잘 응용되지 못하는 점이 임목생물공학의 발전을 저해하는 요소로 지적되고 있다. 본 총설에서는 이러한 기초적 지식을 침엽수 체세포배 배양의 응용적 연구에 이용하는데 초점을 맞추었다.

야생식물 Sicyos angulatus L.로부터 캘러스 유도 및 체세포배 발생 (Callus Induction and Somatic Embryogenesis from Sicyos angulatus L.)

  • 박과식물과 우수한 접목친화성을 가진 것으로 알려진 야생식물인 Sicyos angulatus L.(일명 '안동오이')의 화기, 자엽 및 잎 조직을 배양하여 캘러스유도 및 체세포배의 획득 가능성을 밝히고 기내배양에 의한 묘의 대량증식에 필요한 기초자료를 얻고자 본 실험을 수행하였다. 자엽이나 화기 절편체로부터 캘러스유도를 위한 가장 적합한 식물생장조절제는 MS 배지에 각각 2,4-D 2.0㎎/L + BA 0.1 ㎎/L와 2,4-D 1.0㎎/L + BA 0.1 ㎎/L를 첨가한 것이었다. 잎 및 자엽 절편체로부터는 체세포배 발생이 되지 않았으나 화기 절편체에서는 체세포배 발생이 관찰되었다. 화기로부터 유래한 캘러스를 NAA 1.0 ㎎/L+ kinetin 1.0 ㎎/L가 첨가된 1/2 MS기본배지에 배양하면 가장 효율적으로 체세포배를 획득할 수 있었고, 탄소원으로는 sucrose 20 g/L가 가장 효율적이었다.

고추 소포자를 이용한 반수체 및 배가반수체 생산 (Production of Haploid and Doubled Haploid Plants from Isolated Microspore Culture of Hot Pepper (Capsicum annuum L.))

  • Haploid/double haploid plants developed from isolated microspores can significantly accelerate plant breeding. Haploid plants can naturally double their chromosomes to create a pure homozygous line of diploid plants. We present a method for producing embryos from isolated microspores of hot peppers (Capsicum annuumL.). We analyzed the polyploidization levels of the regenerated plants. The donor plants produced the optimal stage of microspores following short-term growth under low-intensity light, which resulted in high rates of embryogenesis and cotyledonary embryogenesis. To find an efficient culture method, liquid, doubled-layer, and 2-step cultures were tested. Liquid culture yielded the highest number of embryos, whereas the highest efficiency for cotyledonary embryogenesis was afforded by the doubled-layer culture. When normal cotyledonary embryos were transplanted onto a regeneration medium, they developed into complete plants. From these, 208 plants were tested via flow cytometric analysis, and 35.6% and 72.7% of the chromosomes from the Milyang-jare and LV2319 genotypes, respectively, were found to be spontaneous double haploids. These results are the same as those obtained on analyzing horticultural characteristics, including the size of leaves and the size and shape of fruits. The present study provides information on the practical application of isolated microspore culture of hot peppers, factors that affect embryogenesis, and methods for polyploidy testing.

Effect of carbon sources on somatic embryogenesis and cotyledon number variations in carrot (Daucus carota L.)

  • In order to investigate the effect of carbon sources on somatic embryogenesis and cotyledon number variations in carrot, embryogenic callus were cultured in the medium supplemented with various concentrations of sucroseor glucose, and with combination of 2% sucrose and various concentrations of mannitol or sorbitol. The maximum number of somatic embryos formed per flask (1,555.70) was obtained in the medium supplemented with 2% sucrose rather than glucose alone or a combination of mannitol or sorbitol and 2% sucrose, and the number of somatic embryos was decreased with the increasing of mannitol or sorbitol concentration. The frequencies of somatic embryos with two cotyledons were 35.14% for sucrose, 19.88% for glucose, 32.55% for mannitol + 2% sucrose, and 38.59% for sorbitol + 2% sucrose, respectively, and the frequencies of abnormal somatic embryos having 3 or more cotyledons were 64.86% for sucrose, 80.12% for glucose, 67.44% for mannitol + 2% sucrose, and 61.41% for sorbitol + 2% sucrose, respectively. Particularly, the frequency of somatic embryos with two cotyledons (59.16%) was the highest in the 2% sucrose medium compared to the frequency of abnormal somatic embryogenesis with three or more cotyledons, and the frequency gradually decreased with increasing concentration of glucose, mannitol or sorbitol. According to these results, it was found that the ratio of abnormal somatic embryo was higher than the normal somatic embryo in carrot, and was shown that somatic embryogenesis and the cotyledon number was affected by the concentrations of sucrose, glucose as carbon source, and mannitol and sorbitol as osmotic agents in culture medium.

Mind Bomb1 and DeltaD are Localized into Autophagosome after Endocytosis in Zebrafish during Neurogenesis

  • Endocytosis of the Notch ligand, DeltaD, by mind bomb1 is indispensable for activation of Notch in cell fate determination, proliferation, and differentiation during zebrafish neurogenesis. Loss of mind bomb1 activity as an E3 Ubiquitin ligase causes the accumulation of deltaD at the plasma membrane and results in the ectopic neurogenic phenotype by activation of Notch in early zebrafish embryogenesis. However, the regulatory mechanism of deltaD during neurogenesis is not identified yet. This study aims to analyze the pathway of mib1 and deltaD after endocytosis in vivo during zebrafish embryogenesis. Mind bomb1 and deltaD are co-localized into autophagosome and mutant form of mind bomb1 fails to cargo deltaD into autophagosomes. These findings suggest that mind bomb I mediates deltaD regulation by autophagy in an ubiquitin-dependent manner during zebrafish embryogenesis.

Effect of Plant Growth Regulators on Plant Regeneration Through Somatic Embryogenesis of Medicago sativa L.

  • An efficient plant regeneration system in alfalfa (Medicago sativa L.) through somatic embryogenesis was established. Embryogenic callus was obtained by culture of hypocotyl segments on MS medium with 0.02mg $L^{-1}$ IAA and 1.0mg $L^{-1}$ zeatin after 45 days of culture. Embryogenic calli were converted to the somatic embryos when transferred to either MS medium without plant growth regulators (PGRs) or MS medium containing various cytokinin (BA, kinetin and zeatin). Most of the somatic embryos were developed into plantlets on MS medium supplemented with 0.1 mg $L^{-1}$ kinetin. Also, secondary embryos appeared on the surface of primary embryo but they showed abnormal growth. Regenerated plantlets were transplanted to pots containing vermiculite and perlite for further analysis.

Plant Regeneration of Soybean Cultivars via Somatic Embryogenesis

  • Effective plant regeneration from immature cotyledons of soybean [Glycine max (L.) Merr.] cultivars was achieved via somatic embryogenesis. Somatic embryogenesis was performed with the cotyledons of immature embryos 14-20 d after flowering. Immature cotyledons of cv. Whangkeum were placed abaxial or adaxial side down on modified MS medium containing 20mg/L 2,4-D. The greatest number of somatic embryos, 1.2 per cotyledon, was produced from those of 4.0-4.9 mm in length which had been placed abaxial side down. Among cvs. Pecking, Whangkeum and Baekwoon, Pecking had the highest embryo induction efficiency with 4.3 somatic embryos per cotyledon in 20mg/L 2,4-D treatment and with 1.0 embryo per cotyledon in 8mg/L NAA treatment. Germinable globular somatic embryos were induced with the highest efficiency, 27.6%, in 20mg/L 2,4-D and were proliferated efficiently on liquid medium containing 10mg/L 2,4-D. The globular somatic embryos developed into germinable mature somatic embryos on medium containing 10 $\mu$M CoCl2, 9% sucrose, and 0.5% activated charcoal. These mature somatic embryos germinated on hormone-free mediu. After transfer to the soil, regenerated plants with seeds were obtained.

Genetic Approach to Identify Critical Factors for Mouse Early Embryogenesis

  • Development of the mammalian pre-implantation embryos has unique features, such as a slow and unsynchronized cell division, compaction, and eventual formation of blastocysts with inner cell mass and trophectoderm. In order to have a clue on molecular mechanisms that reside in mouse early development, we suppressed expression of early embryo-specific genes with RNAi and observed their development in vitro. We observed developmental defects in embryos microinjected with dsRNAs for Oct4 or Nanog among the tested genes. Careful examinations revealed that development of the most of the Oct4- or Nanog-suppressed embryos were arrested at the morula stage. These results suggest that the Oct4 and Nanog activities are also required for embryogenesis earlier than the blastocyst stage.

Somatic embryogenesis and plant regeneration in zygotic embryo explant cultures of rugosa rose

  • Rugosa rose (Rosa rugosa) is cultivated as a garden flower and an important genetic resource for the breeding of roses (R. hybrida). This study describes culture conditions for high frequency plant regeneration from zygotic embryo explants via somatic embryogenesis in rugosa rose. Mature zygotic embryo, cotyledon, and radicle explants formed embryogenic calluses at frequencies of 38, 6.7, and 8.8% when cultured on half-strength Murashige and Skoog medium (${\frac{1}{2}}MS$) supplemented with 2.26, 9.05, and $9.05{\mu}M$ 2,4-dichlorophenoxyacetic acid, respectively. Embryogenic calluses produced numerous somatic embryos, which then developed into plantlets on ${\frac{1}{2}}MS$ without growth regulators. Regenerated plantlets were grown to whole plants in a growth chamber.

납과 아연이 말똥성게(Hemicentrotus pulcherrimus)의 수정 및 배아 발생에 미치는 영향 (Effects of Lead and Zinc on the Fertilization and Embryo Development of the Sea Urchin (Hemicentrotus pulcherrimus))

  • 본 연구는 말똥성게(Hemicentrotus pulcherrimus)의 생식세포 및 pluteus 유생을 이용하여 중금속인 납(lead, Pb)과 아연(zinc, Zn)의 독성을 조사였다. H. pulcherrimus 배우자 및 배아에 미치는 Pb과 Zn의 독성은 각각 31, 63, 125, 250, 500 ppb 및 16, 31, 63, 125, 250 ppb의 농도에서 조사하였다. 0.5 M KCl 용액을 이용하여 방란 및 방정을 유도하였고, 수정률 및 정상 배아발생률의 조사는 수정 후 각각 10분 및 64시간째 관찰하여 시행하였다. Pb 노출 시 수정률은 대조군과 비교하여 유의적인 변화가 없었다. 그러나 정상 배아발생률은 농도가 높을수록 농도의존적으로 유의적인 감소를 보였다. Zn을 노출시켰을 경우 수정률과 정상 배아발생률은 농도가 높을수록 농도의존적인 유의적 감소를 나타냈다. H. pulcherrimus의 정상 배아 발생에 대한 독성치는 각각 Pb (반수 영향농도 ($EC_{50}$) 45.13 ppb, 95% Cl 40.12~50.05 ppb), Zn (반수영향농도 ($EC_{50}$) 19.82 ppb, 95% Cl 18.26~21.31 ppb)로 나타났다. 또한 Pb과 Zn의 무영향농도(NOEC)는 각각 <31.25 ppb 및 <15.63 ppb로 나타났고, 최소영향농도(LOEC)는 31.25 및 15.63 ppb로 나타났다. 본 연구 결과, H. pulcherrimus의 초기 배아발생 과정은 Pb과 Zn 등의 중금속에 높은 민감성을 보인다. 따라서 H. pulcherrimus는 해양생태계 위해 평가를 위한 시험생물로서 사용이 가능하다고 사료된다.
