Structural changes of the endosperm of Panax ginseng C.A. Meyer from fertilization to germination were investigated by light microscope. The endosperm of the ginseng seed is cellular type. Since endosperm cells adjacent embryo continuously breakdown and disappear with the elongation of embryo, the real of endosperm is gradually decreased. As the anatropous ovules of immature seed with green seed coat developes more and more, ovary cells adjacent ovary cavity become abundant by the periclinal division, their size is decreased, hypotrophy of cell wall discern, and they are gradually differentiated in seed coat. Though embryo responds strongly to basic dye at the stage of completion of endosperm formation, tissue of endosperm responds to acidic dye positively Cell wall of embryo and endosperm are composed of primary cell wall not lignified. Endosperm cells adjacent embryo begin to breakdown in the endosperm tissue of indehiscent seed before the beginning of the after-ripening. Dehiscent seed of which seed coat is opened through after-ripening represent the form as a seedling in the result of embryo developments with the formation of organs; radicle, cotyledon, plumule. Umbilifom layer represents strong positive response to the toluidine blue and the basic function. Umbiliform layer that endosperm cells breakdown and disappear is observed clearly at the periphery of the embryo cotylemon, while slightly at the periphery of the radicle.
This study was undertaken to measure the concentrations of estradiol-$17{\beta}$, progesterone and testosterone, and to study their relationship with each other and with follicular size in individual buffalo ovarian follicles categorized as small (4 to 5 mm diameter), medium (6 to 9 mm diameter) and large (${\geq}10mm$ diameter). Steroid hormone concentrations varied markedly within follicles of each size category. Estradiol-$17{\beta}$ concentrations (pmol/ml) were positively related to follicular diameter (R = 0.34, n = 308, p < 0.001) and were significantly higher (p < 0.001) in large (1$118.46{\pm}30.25$), compared to those in medium follicles ($50.32{\pm}8.29$) which, in turn were significantly higher (p < 0.001) than those in small follicles ($19.70{\pm}$5.57). Progesterone and testosterone concentrations (pmol/ml) were not related to follicular diameter and were not different among small ($330.99{\pm}27.32$ and $17.68{\pm}2.44$ respectively), medium ($384.84{\pm}26.20$ and $36.47{\pm}4.55$, respectively) and large follicles ($253.25{\pm}32.23$ and $22.57{\pm}4.48$, respectively). Estradiol-$17{\beta}$ and progesterone concentrations were positively related (R = 0.39, n = 47, p < 0.01) in small, unrelated in medium and negatively related in large follicles (R = -0.59, n = 23, p < 0.01). There was no relationship between estradiol-$17{\beta}$ and testosterone concentrations in follicles of all the three size categories. Progesterone and testosterone concentrations were positively related in large follicles (R = 0.57, n = 18, p < 0.02). There was no relationship between the two hormones in small and medium sized follicles. When the follicles with estradiol-$17{\beta}$/progesterone molar ratios of > 1.00 were considered non-atretic, and the rest at different stages of atresia, 197/208(95%) follicles were found to be atretic.
배아란 동물이나 식물과 같은 다세포 생물의 초기 단계를 의미한다. 배아의 단계에서 다세포 생물의 기초적인 체제가 결정되기 때문에 배아는 개체발생의 기구를 연구하는 중요한 연구대상이 된다. 생물학자들은 배아 연구를 위해 대용량의 배아 이미지 데이터를 소유하고 있으며, 이러한 대용량 데이터 중 원하는 이미지를 효율적으로 검색하기 위해서는 데이터 구조화가 필요하다. 데이터베이스 구조화를 위해 주로 사용되는 방법으로 계층적 클러스터링이 있다. 그러나 기존의 계층적 클러스터링 방법은 데이터베이스를 트리 형태로 구조화 하는 과정에서 클러스터의 크기와 클러스터 내의 객체 수를 동시에 고려하지 못하기 때문에 결과 클러스터링 트리가 경사 트리일 가능성이 매우 높다. 경사 트리인 경우 사용자가 원하는 이미지를 검색하기 위해 트리를 순회할 때 많은 시간이 걸린다. 따라서 본 논문에서는 대용량의 배아 이미지 데이터를 경사 되지 않으며 균형 상태에 가까운 트리 형태로 구조화하기 위한 방안을 제시한다. 제안하는 방안은 데이터베이스 내에 저장된 배아 이미지를 그래프로 변환하고 반복적으로 그래프 분할 알고리즘을 적용하여 클러스터를 생성한다. 이 때 클러스터의 크기와 클러스터 내의 객체 수를 동시에 고려하여 특정 클러스터의 크기가 지나치게 커지거나 객체 수가 많아지는 것을 방지한다. 실험을 통해서 제안하는 방안의 우수성을 규명하고 시각화 툴을 제공하여 사용자가 원하는 배아 이미지를 쉽게 찾을 수 있도록 돕는다.
These experiments were carried out to determine the effect of pregnancy in bisected embryo. The embryos of ICR mouse were microsurgically bisected at morula and blastocyst stage using microsurgical blade attached a micromanipulator. These bisected embryos without zona pellucida were cultured up to blastocyst stage and cell count and diameter of stained blastomere, and transferred pseudopregnant mice. And the development of these bisected embryos was compared with the results of production of young of the corresponding intact embryos or cell stage. When the bisected mouse embryos were cultured in vitro for 20 to 24 hours in morula stage(77.2%) or 3 to 6 hours in blastocyst stage(84.1%), them were developed to the expanded blastocyst stage. There were no significant(P<0.05) differences in the development rate of bisected embryos between in morula and blastocyst stages. The embryo size of blastocyst developed in vitro from bisected embryo was small(P<0.05)than intact embryo. However, the number of blastomeres with bisected embryo (24.7+1.3and 21.5+1.2 respectively) were significantly(P<0.05) reduced, compared with that of intacted embryos(36.3+1.1 and 41.4+1.2 respectively). When compared with the result of pregnancy rate(63.6%) after surgical transfer of bisected morulae, a similar result(65.4%) was obtained with bisected blastocyst stage(P< 0.05). However, production of youngs (38.8%) after transfer of bisected morula, a similar result (38.1%) was obtained with bisected blastocyst stage (P<0.05).
The objective of this work was to determine the effect of corpus luteum (CL) grade on pregnancy rate after embryo transfer in Korean cattle and we found that CL development was linked to pregnancy rate. The in vivo derived blastocyst-stage embryos were transferred to 15 recipients synchronized in the estrus cycles. Based on size and palpable characteristics, CLs were categorized into three grade. The grade three CL is not to be identified by rectal palpation. The pregnancy rates tended to increase with the increase in CL size of recipients. In grade one, two, and three, the pregnancy rates were 62.5%, 50.0%, and 0%, respectively. This result suggests that pregnancy rates after embryo transfer might be affected by the CL status of recipients.
Hordeum bulbosum을 이용한 보리 반수체육종법의 실용화를 체계화하기 위해 일련의 실험을 실시하였으며, 유배의 적취시기, 유배의 크기에 따른 배배양 반응, 배지 및 환경조건 등을 조사하였다. 1. 유배의 적취시기는 배발육과 식물체 발생률을 고려할 때 수분 후 17일이 적기였다. 2. 식물체 발생에 유리한 유배의 크기는 0.9mm내외였다. 3. 식물체 발생에 효과적인 배지 및 배양조건은 MS배지의 암상태에서 발생시킨 후 명상태로 하는 것이 정상 식물체 발생률(3%)이 높았으나, 작업상의 편리성을 고려한다면 B5배지에서 24시간의 일장처리조건이 좋았다. 4. 반수체는 정상 2배체에 비하여 배 및 식물체의 발생이 현저히 부진하였다.
This study was carried out to compare the actual size(length and height) of ovaries, follicles and corpora lutea of Korean native cow with those on sonograms. We used 3 different probes(3.5 MHz abdominal probe, 6.5 MHz transvaginal probe and 5.0 MHz transrectal probe) and a calipher for measurements of ovaries, follicles and corpora lutea on sonograms and actual size. Under water immersion, 157 ovaries were scanned with 3 probes and measured in actual size and compared each other. The average height and width of ovaries of Korean native cows were 17.40$\pm$3.99 and 34.23$\pm$6.02mm, respectively. In comparison of height, length of ovaries and preovulation follicles, we found that image with a transvaginal probe was nearly the same as the actual size(p<0.01), but with an abdominal probe the image was appeared larger than the actual size. In measurement(diameter) of preovulation follicles the transvaginal probe was proven to be more accurate to the actual size than other probes and in corpus luteum measurement all probes were accurate. In the comparison of number of follicles by different size ranges, there was no statistical difference in the count of follicles over 10 mm in diameter between the transvaginal probe and naked eyes.

Somatic embryos were induced from immature cotyledons and cultured on a MS medium containing 40mg/L 2,4-D. The maximum induction of embryos was obtained from immature cotyledons in a size of 3-4mm, and the highest frequency was obtained in the induction medium at pH 7.0. For embryo development, embryogenic tissues were transferred to a MSM6AC and MSM6 media. Developing embryos were placed at 27$^{\circ}C$with dim light (20$\mu$$molm^{-2}$$s^{-1}$) provided by cool fluorescent tubes (3-D wavelength light is better than standard light). Somatic embryos were clearly developed from globular stage to cotyledonary stages. The color of embryo may be a useful parameter for estimation of embryo quality. When the embryo becomes mature, embryo will be ready for desiccation in order to induce roots and shoots of embryos.
Four pluriparous Korean black goat does were superovulated with FSH and mated with fertile bucks. Anesthetized animals were placed in lateral recumbency, then size 8 Foley catheter was inserted into the uterus through the cervix under the vaginal speculum and the balloon was inflated to fix the catheter in the uterine body. The opposite end of the catheter was connected to a 3-way and a flushing medium was infused into the uterus. Modified Dubecco's PBS with 1% FBS was used as the flushing medium. Four goats were allocated in two groups depending on the type of medium infusion into uterus. Injection group; the flushing medium was injected into uterus and the infused medium was collected by to-and-fro method using a syringe. Gravity-flow group; the flushing medium was allowed to enter the uterus by gravity flow by lifting the medium bottle and drained out of the uterus into a collecting tube. All four goats had catheter inserted through the cervix and uteri flushed successfully. The volume (recovery rate) of recovered medium varied considerably from 87 ml/200 ml (43.5%) to 148 ml/160 ml (92.5%). Nine embryos/ova in total were recovered from Gravity-flow group goats. Although the embryo recovery rate was low, the possibility of a transcervical embryo recovery in Korean black goat had been proven in this preliminary experiment.
