Study on the Chromosome Size, Number and Shape by the Centromeric Index, Arm Ratio and Relative Length in Single Comb White Leghorns (단관백색레그혼순계에 있어 중심입지수, 등완비 및 상대적길이에 의한 염색체의 형태적 특징과 수에 관한 연구)

  • Chromosome size, number and shape were studied by the centromeric index, the arm ratio and the relative length of chromosome. The chromosomes of 50 early chick embryos which were derived from a pure line of Single Comb White Leghorns were examined. Using a colchicine, hypotonic treatment, fixation and air-drying technique, the clear prometaphase figures were obtained from the whole embryo. The results of the present investigation of chromosome pairs were as follows, 1. Pair 1 and 2; metacentric and submetacentric chromosomes which could be clearly distinguished from each other by size. 2. Pair 3 and 4: acrocentric chromosomes of similar length but the 4th pair had a distinct short arm which was not present in the 3rd. 3, Pair 5; metacentric sex chromosomes, 2 chromosome had relative 5th length but the W chromosome had slightly shorter length than 7th pair of chromosomes. 4. Pair 6; acrocentric chromosomes similar in shape to pair 3 but of little more than half the size. 5, Pair 7 and 8; acrocentric chrocentric but the 7th pairs had a definite short arm. 6. Pair 9; similar length to the 7, 8 pairs but had a medially placed centromere. 7. microchromosomes of 30 pairs ; nearly all acrocentric chromosomes which appeared as paired dots. The total number of diploid was appeared to 72-78. But a number of observations presented the total diploid number in 78 (58%). The inconstancy in number observed in this study was presumably due to the minute size of the microchromosomes. Thus, the modal numbers for the diploid chromosome was at least 78.

Association of Endometrial Polyps with Membranous Adhesions in Uterine Cavity

  • The membranous adhesions could induce implantation failure despite transplantation of high quality of embryo. Clinically, of the patients who have membranous filmy adhesions, endometrial polyps have been found in not infrequently. Thus this study was tried to evaluate the features of endometrial polyps and the effect of endometrial polyps on formation and extents of membranous adhesions in uterine cavity of infertile patients under hysteroscopy. A retrospective study was conducted on 34 infertile patients who were diagnosed as endometrial polyps with membranous adhesions during hysteroscopy from July 2008 to July 2011. Number, size, location and morophologic type of endometrial polyps were investigated. If needed, methylene blue solution was instillated to endometrial cavity to identify membranous adhesions. Then, associations between membranous adhesions with features of endometrial polyps were evaluated. Mean size of endometrial polyp was $1.6{\pm}0.6$ cm, the bigger of endometrial polyps was, the larger of extents of membranous adhesions. (p<0.05). Endometrial polyps were locate evenly in endometrial cavity as follows: anterior uterine wall, 39.1%; posterior uterine wall, 34.8%; lateral uterine wall, 26.1%; upper: 29.4%, middle: 32.4%, lower segment, 35.3%. Mean number of endometrial polyps was $2.26{\pm}1.3$. The pedunculated type was 37.7% and sessile type was 32.4%. There was no statistically significant association of location, number and morphologic type of endometrial polyps with membranous adhesions. In conclusion, hysteroscopy before in vitro fertilization on infertile patients was worthy because of removing of endometrial polyps and membranous adhesions.

Oviduct-specific Glycoprotein 1 Locus is Associated with Litter Size and Weight of Ovaries in Pigs

  • Oviduct-specific glycoprotein 1 (OVGP1) is implicated in playing a role in fertilization and early embryo development. In this study, we have obtained the sequence of intron 9 of OVGP1 gene in swine. Comparative sequencing of Meishan (a native Chinese breed) and Large White pig breeds revealed an A/T substitution at position 943. A PCR-EcoRI-RFLP assay was developed to detect this mutation. Polymorphism analysis in Qingping animals showed that pigs with BB genotype had lower number of piglets born alive (NBA) in multiple parities than pigs with AA (p<0.05) and AB genotype (p<0.01). In Large $White{\times}Meishan$ ($LW{\times}M$) $F_2$ offspring, the weight of both ovaries (OW) of the BB genotype was significantly lighter than that of AB (p = 0.05) and AA (p<0.01) genotypes. Analysis of the data also revealed that the mutation locus affected these two traits mostly by additive effects. These studies indicated that the polymorphism was associated with NBA and OW in two distinct populations and further investigations in more purebreds or crossbreds are needed to confirm these results.

  • We have launched an investigation for Embryonic Development, Larvae and Juvenile Morphology, of Buenos aires tetra in order to build basic data of Characidae and fish seeding production. We brought 50 couples of Characidae from Bizidduck aquarium in Yeosu-si, Jeollanamdo, from Korea on March of 2015. We put them in the tetragonal glass aquarium ($50{\times}50{\times}30cm$). Breeding water temperature was $27.5{\sim}28.5^{\circ}C$ (mean $28.0{\pm}0.05^{\circ}C$) and being maintained. The shape of fertilized egg was round shape, and it was adhesive demersal egg. The egg size was 0.63~0.91 mm (mean $0.74{\pm}0.07mm$, n=20). After getting fertilized egg, the developmental stage was gastrula stage, and embryo covered almost two-thirds of Yolk. Incubation was happened after 16 hours 13 minutes from gastrula stage, and the tail of juvenile came out first with tearing egg capsule. Immediately after the incubation, prelarvae had 3.78~3.88 mm length (mean $3.84{\pm}0.04mm$, n=5), and it had no mouth and anus yet. 34 days after hatching from the incubation, juvenile had 8.63~13.1 mm (mean $10.9{\pm}1.66mm$), and it had similar silver-colored body shape with its mother.

Reproductive Biology of Neocaridina denticulata denticulata and Latreutes planirostris (새뱅이와 넓적뿔꼬마새우의 생식생물학적 연구)

  • We investigated the fecundity, egg size, reproductive output, and breeding frequency and season of Neocaridina denticulata denticulata and Latreutes planirostris which inhabit extremely different habitats. The marine shrimp Latreutes planirostris produced more eggs had at a given carapace length than the freshwater shrimp N. d. denticulata. However, N. d. denticulata had a larger egg volume and greater reproductive output than L. planirostris. The monthly gonadosomatic index (GSI) of the freshwater shrimp began to increase in April and reached a maximum in May, suggesting a single breeding period. In contrast, the GSI of the marine shrimp exhibited two breeding peaks: May-June and September. In both shrimps, the regressions between carapace length and ovarian weight were significant at the non-eyed and eyed embryo stages. For both species, an analysis of covariance revealed significant difference between the two regressions in elevation, but not in slope. These results indicate the potential for multiple ovulations within the reproductive season.

Effects of Rearing Temperature on Larval Survival and Growth and on Reproductive Traits of Palaemon serrifer (Decapoda: Caridea: Palaemonidae)

  • Larvae of Palaemon serrifer were reared in the laboratory under three different temperature regimes ($15^{\circ}C$, $20^{\circ}C$, $25^{\circ}C$) to study the effects of rearing temperature on larval survival and growth, as well as other traits such as embryo volume, number of embryos (fecundity), incubation period, development. Mode and development period. Growth pattern was analyzed by measuring the molt increment and intermolt period. The intermolt period consistently increased with size and instar number and was shortest at $25^{\circ}C$. However, molt increments generally decreased with instar number. Number of embryos varied from 552 to 1355. The relationship between the number of embryos and carapace length was expressed by the equation (fecundity) y=2.7744x+0.208 ($R^2$=0.7961). Egg volume was a primary factor affecting other life-history traits. Egg volume was $0.078\;m^3$, which is relatively small thus embryos exhibited a relatively short incubation period and a comparatively short development period, and the nutritional mode was planktotrophic. Brood production was followed by a fast parturitional pattern. Most ovigerous females had mature ovaries when the parturial molt occurred soon after eclosion.

  • Serial ultrasonographic examinations were daily performed on 12 Shih-tzu bitches from 15 days after ovulation until parturition to determine the time of first detection and ultrasonographic appearance of the fetal and extra-fetal structures and to determine the size of gestational structures. Gestational age was timed from the day of ovulation (day 0), which was estimated to occur when plasma progesterone concentration was first increased above 4.0 ng/ml. The gestational length in 12 Shih-tzu bitches was 62.1 (range:60-64) days. The initial detection of the fetal and extra-fetal structures were: gestational sac at days 19.2 (16-21); zonary placenta in the uterine wall at days 25.9 (24-28); yolk sac membrane at days 24.1 (22-26); amniotic membrane at days 26.6 (24-28); embryo initial detection at days 22.3 (21-24); heartbeat at days 23.8 (23-25); limb buds at days 29.3 (27-31); fetal movement at days 32.3 (31-34); stomach at days 32.6 (29-34); urinary bladder at days 33.1 (29-35); skeleton at days 35.9 (34-38) and kidney at days 45.8 (45-48).

  • Using morphometric technique, the present study was made of the kidney of rabbit embryo in the 13 days of the gestation. Their section areas of the renal tubules and the glomerular cells were compared with the those of adult rabbit, 1. In the developing renal tubule, the tubular section areas were larger than that of the adult, but the nuclear section areas of tubular calls were smaller than the adult. The microvilli of their cell surface were poorly developed, and their cytoplasm contained well developed mitochondria with euchromatic nucleus. 2. In the developing glomeruli. the nuclear section area of the cells of the parietal layer, the podocytes, and the endothelial cells were similar size, but their nuclei were larger than those of the adult. The cytopalsm of the podocytes contained well-developed rough endoplasmic reticulum, Golgi apparatus, and round mitochondria.

  • Ultraviolet irradiation of the vegetal hemisphere of the fertilized frog egg prior to first cleavage resulted in alterations in neural morphogenesis. The sensitivity to UV irradiation dropped drastically at the point of 2/3 time lapse between fertilization and the appearance of the first cleavage furrow. Scannining electron micrographs revealed a decrease in size of the cells on the surface of the irradiated embryos. However, ectoderm exchange frafts indicated that the ectoderm of the irradiated embryo retains its capaciiy to form a completely normal neural morphology and epidermal surface. These facts were interpreted in terms of a decrease in the capacity for invagination during gastrulation, and subsequent primary embryonic induction.

Determination of the Synthetic Time and the Transport Pattern of Vicilin and Legumin in Ginseng Endosperm Cell Using Double Immunogold Labeling (이중 면역금입자 표지법을 이용한 인삼 배유세포내 Vicilin과 Legumin의 합성시기 및 수송방식)

  • Vicilin and legumin, the storage Proteins of seed, were Purified from ginseng (Panax ginseng C.A. Meyer) endosperm cells. They were immunized in rabbits, and antibodies were raised respectively. Using these two antibodies, double immunogold labeling of vicilin and legumin was carried out to determine the gap of synthetic time and the transport pattern of vicilin and legumin in the ginseng endosperm cells. Vicilin and legumin were synthesized at the same time at early embryo developmental stage. They were secreted from the Golgi bodies and accumulated into the small vacuoles. As the endosperm cells developed, vicilin and legumin localized in the small vacuoles were gradually transported toward the large central vacuole where they were stored. Protein bodies were derived from the vacuoles filled with proteins and distributed in the endosperm cells of mature red seed. Protein bodies were various in size from 1 to 8 ${\mu}{\textrm}{m}$ in which vicilin and legumin were mixed each other. The number of small particles labeled on the vicilin was greater than that of large particles labeled on the legumin in the protein bodies indicating that the amount of vicilin is higher than that of legumin in the protein bodies.
