Precursor Feeding Effects of Alkaloid Production in Suspension Cultures of Eschscholtzia californica (캘리포니아 양귀비(Eschscholtzia californica) 현탁세포배양에서 전구체가 알칼로이드 생성에 미치는 영향)

  • The accumulation of benzophenanthridine alkaloids, sanguinarine, chelerythrine, chelirubine and macarpine occurred in suspension cultures of Eschscholtzia californica. To increase alkaloid production, feeding experiments with the biosynthetic precursors, tyrosine, tyramine, L-dopa, dopamine with and without elicitation were studied. In feeding experiments with various precursors, the total alkaloid production was slightly increased. The precursor feeding with elicitation, however, increased total alkaloid production several times.

Elicitation and In Situ Recovery of Alkaloids in Suspension Cultures of California Poppy

  • Large and rapid increases in benzophenanthridine alkaloid production occured in suspension cultures of Eschscholtzia californica cells treated with elicitors. Response to different biotic elicitors showed that elicitors prepared from yeast extract, Collectotrichum lindemuthianum and Verticillium dahliae induced alkaloid formation. Highest alkaloid accumulation was obtained with $60\;\mu\textrm{g}$ of yeast extract elicitor per gram of fresh cell weight. In time course performance after elicitor addition, more than 40 hours were required to obtain saturated alkaloid accumulation. Compounded silicone fluid, an ideal accumulation phase for two-phase culture of E. californica, accumulated a large amount of alkaloids produced in a specific manner. Elicitation in two-phase culture clearly increased net alkaloid production as well as their concentrations in the accumulation phase.

Baicalin Production in Transformed Hairy Root Clones of Scutellaria baicalensis

  • A transformed hairy root clone of Scutellaria baicalensis was established following infection with Agrobacterium rhizogenes ATCC15834. Three root clones of S baicalensis were selected by growth habit and baicalin content. The most active strain-the SR-03 clone-was examined for its growth and baicalin content under various culture conditions. The root growth and baicalin content were maximized in a Schenk and Hildebrandt medium supplemented with 4 and 6% sucrose, respectively. The accumulation of baicalin in transformed hairy roots was enhanced through exposure to various elicitors. Elicitation was attained by the addition of methyl jasmonate, salicylic acid, and various concentrations of fungal cell wall elicitors to the medium. The accumulation of baicalin in the elicited cultures ranged from 10.5 to 18.3 mg/g dry weight of the roots, which was 1.5- to 3-fold the amount attained in controls.

  • Social Network Services (SNSs) have grown to be new and promising tools of marketing. By referring to researches done on e-mail viral marketing, this paper operationizes SNS viral marketing effectiveness to accurately reflect marketing success in SNS environment, and tries to identify its affecting factors. As potential affecting factors, fan size, advertisement type, existence of engagement elicitation and incentive are identified. By sampling real advertisement postings on Facebook, we showed that fan size, advertisement type, and engagement elicitation are factors affecting SNS viral marketing success. This research expanded the conventional model of viral marketing into SNS settings to improve understanding on SNS viral marketing. Motivation is discussed as an important factor, and this research showed that viral campaign can be more successful when it triggers internal motivation to engage, but not the external motivation. This research could also be a guide for practitioners on how to post a successful advertisement in SNSs.

  • In the early stages of a software project, it is critical to understand the needs of the customers and elicit their customer requirements. Various requirements elicitation methods have been proposed. However, existing methods still have the limitations such as a limited number of target customers, limited expression of customers' opinions, and difficulty in collecting the customers' opinions continuously. A novel method for eliciting customer requirements is proposed by utilizing a social network service (SNS), which is a shared source of raw information of the customers' needs and opinions. The proposed method is validated to show its effectiveness in overcoming the limitations of existing methods.

  • This study presents a new approach for expert opinion elicitation process to assess an uncertainty inherent in accident management. The need to work with rare event and limited data in accident management leads analysis to use expert opinions extensively. Unlike the conventional approach using point-valued probabilities, the study proposes the concept of fuzzy probability to represent expert opinion. The use of fuzzy probability has an advantage over the conventional approach when an expert's judgment is used under limited dat3 and imprecise knowledge. The study demonstrates a method of combining and propagating fuzzy probabilities. finally, the proposed methodology is applied to the evaluation of the probability of a bottom head failure for the flooded case in the Peach Bottom BWR nuclear power plant.

  • Objective : This study was performed to investigate the effect of Astragali Radix on leukocytopenia anaphysis with carcinostatic substance elicitation. Methods : We observe if there were differences in a general tendency by EAR (AR extracts) capacity in the Astragali Radix treated group and the cyclophosphamide treated group, a general tendency of recovery from leukocytopenia over lime and checked for liver and kidney toxicity. Results : In cyclophosphamide treated with EAR 1000mg/kg group, the recovery rate of leukocytopenia increased and there were no differences in liver and kidney functions. Conclusions : Astragali Radix was effective and safe in increasing rate of leukocytopenia anaphysis with carcinostatic substance elicitation and indicated possibility for clinically adaptation.
