This experiment was carried out to study the optimal chilling period for breaking the physiological dormancy of dehisced E. senticosus seeds and to investigate the critical seeding date in Kangwondo. Storage at $5^{\circ}C$ for 85 days was most effective in breaking dormancy of dehisced seeds, but didn't germinated synchronously. Only 28.1% of undehisced seeds germinated after 125 days of low temperature storage. For improving seedling survival rate in the field, 50% shading was more effective than 30% shading. Delayed seeding date resulted in decreased seedling fresh weight, dry weight, leaf area and root length, but seeding date has no significant affecte on stem length.

Antimutagenic effect of Eleutherococcus senticosus Maxim. on the mutagenicity induced by 2-AF and Trp-P-1 was studied using the Ames test with Salmonella typhimurium TA98 and TA100. In S. typhimurium TA98, the methanol extract $(500\;{\mu}g/plate)$ of root, stem, and leaf of E. senticosus showed inhibitory effects of 72.8, 70.0, and 78.7% on the mutagenicity induced by 2-AF, and 69.2, 64.9, and 59.4% by Trp-P-1, respectively, whereas none was observed in S. typhimurium TA100. These results suggest that the methanol extract of E. senticosus inhibits a frame shift mutation. And then the methanol extract further fractionated by chloroform, butanol, and water. The chloroform fractions of root, stem, and leaf showed strong antimutagenic effects induced by 2-AF and Trp-P-1 in S. typhimurium TA98, whereas none was observed in the butanol and aqueous fractions. The chloroform fractions of root, stem, and leaf showed antimutagenic effects of $13{\sim]92%$ in a dose-dependent manner.
This study was carried out to elucidate the influence of gibberellin soak and seed dressing with Toros wp. on the dehiscence of Eleutherococcus senticossus seeds. $GA_3$ treatment was effective on promoting after-ripening seed of Eleutherococcus senticossus whose concentration was higher until $500\;mg{\cdot}L^{-1}$, after-ripening period became shorter. But rotting percentage increased gradually up above $500\; mg{\cdot}L^{-1}\;GA_3$After all, $300\;mg{\cdot}L^{-1}\;GA_3$ was the most effective treatment for promoting after-ripening seed. Dressing treatment with Toros wp. on seeds reduced the dehiscent rate of E. senticosius. by suppressing activity of fungi living on the endocarp surface of seed.

Eleutherococcus senticosus (Rupr. et Maxim.) Harms is a tree species that has been traditionally used as a source for oriental folk medicine. It is known to contain both Eeleutheroside E and chlorogenic acid. These two compounds are believed to be effective elements in the tree. In the present study, we analyzed the levels of the two compounds from the plants collected from various locations in Korea to find out whether environment affected the quantity of both compounds in the tissue. When the plants growing in their natural habitats and those growing in plantations were compared, the level of the two compounds showed different pattern. In the case of chlorogenic acid in roots, the plants growing in plantations are found to contain higher levels than did those from natural habitats. On the other hand, higher levels of eleutheroside E were found in the plants from natural habitats. However, when the compounds in the stems were compared, the plants growing in the natural habitats contained higher level in both compounds than did those from plantations. Although the level of both compounds in the leaf tissue varied with the places from which they were originated, it appeared to be less than that found in roots or stems. There exists a tendency that the level of eleutheroside E showed a positive correlation with the diameter of both roots and stems of the plants.

Background : The purpose of this study was to investigate the anti-inflammatory properties of stems and leaves of Eleutherococcus senticosus (Rupr. & Maxim.) Maxim. (ES) from Gangwon-do. Methods and Results : Stems and leaves of ES were collected from two areas in Gangwon-do: Cheorwon-gun and Samcheok-si. Samples were extracted with water by using the pressurized liquid extraction method and with 70% prethanol A by using the heat reflux extraction method. The anti-inflammatory effects of ES were evaluated through the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazoliumbromide(MTT), lactate dehydrogenase(LDH) assay, nitric oxide(NO) assay, enzyme-linked immunosorbent assay(ELISA), and Western blot analysis in RAW 264.7 macrophages stimulated with lipopolysaccharide (LPS). 1) Results showed that ES leaf extractions were not cytotoxic at a concentration of up to 30 ㎍/㎖. The leaves of 70% prethanol A extractions of ES(30 ㎍/㎖) inhibited NO, interleukin-6(IL-6), and tumor necrosis factor-α(TNF-α) production and decreased the protein level of cyclooxygenase 2(COX-2). There was no significant change in the protein level of inducible nitric oxide synthase(iNOS). The stem extractions of ES did not exhibit anti-inflammatory effects. Conclusions : In this study, the leaves of 70% prethanol A extractions of ES demonstrated anti-inflammatory effect on RAW 264.7 macrophages. The 70% prethanol A extractions have a relatively higher anti-inflammatory effect on RAW 264.7 macrophages than water extractions.
A method for long-term conservation of somatic embryos of Eleutherococcus senticosos was described. Suspension cultured globular somatic embryos were successfully conservated for 36 months at $4^{\circ}C$. The embryos resumed growth within two weeks when returned to MS liquid medium containing $0.2\;mg/{\ell}$. 2,4-dichlorophenoxy acetic acid. The optimal condition for cell proliferation was achieved when somatic embryos cultured at $32^{\circ}C$ in 1/3 MS liquid medium, and about 1.2 g of embryogenic cell was induced from 150 globular embryos after 6 weeks of suspension culture. The embryogenic cells produced from these somatic embryos exhibited normal plant regeneration on auxin-free medium.
The aim of this study was to described the genetic structure of Eleutherococcus senticosus in Korea. We investigated 10 patches, which are eight Korean patches and two foreign patches come from Russia and China growing at Korean habitat, using ISSR(inter-simple sequence repeats) markers. In ISSR PCR, the overall percentage of polymorphic ISSR amplicons was 76% and the mean number of amplicons per ISSR primer was 11.5, which were higher than the RAPD results for the some cultivars collected in Korea(Kim et al., 1998) ; 57% and 5.7, respectively. So ISSR markers provide more powerful tool than RAPD markers for the investigation of genetic variation in E. senticosus. There are relatively high genetic variation among patches as 62.8%, but low variation within eight Korean patches. Such pattern of genetic variation, which is not ordinary in other tree species, may be result from the narrow and limited habitats and the asexual reproduction of this species at the natural stands in Korea. Although the small sample size in this study seemed to be resulted in the high genetic variation among patches, the overall genetic interpretation of this study might not be much affected on the basis of the characteristics of the distribution and the reproduction system of E. senticosus. Analysis of genetic distance between all pairs of the patches did not reveal any trends with regard to geographic distance, which was confirmed by the results obtained from AMOVA(analysis of molecular variance) and PCA(principal component analysis). These results suggest that, in addition to the preservation of the natural stands, the conservation of larger number of patches with small number of individuals per patch is more effective for the ex situ conservation and for maintaining the genetic diversity of E. senticosus in Korea than smaller patches with large number of individuals.
Experiments were conducted to find the optimal acclimatization conditions for Eleutherococcus senticosus plantlets regenerated from bioreactor cultured somatic embryos, various acclimatizing conditions were compared regarding both survival rate and growth of the plantlets. Among the various temperature and artificial soil tested, the highest survival rate (88%) was observed when plantlets were acclimatized in Klasmann bed soil at $10^{\circ}C$ When in vitro plantlets directly transplanted to field environment, shading treatment was necessary and 50% shading was more effective than 30% shading. Transplanting season were also important for successful acclimatization of in vitro cultured plantlets, transplanted on March 20 with 50% shading exhibited the best survival rate and further growth.
Purpose: This study was performed to investigate the effects of immune cell activation and the antitumor effect for the combination of treatment with X-irradiation and E/eutherococcus senticosus Maxim Root (ESMR) on mouse tumor cells. Materials and Methods: ESMR (250g) was extracted with 80% methanol, concentrated under decompression and lyophilized. To determine whether ESMR is able to activate the immune cells or not, the proliferation of splenocytes in vitro and the number of B cells and T cells in splenic lymphocytes in ESMR-pretreated mice were evaluated. X-irradiation was given to the mouse fibrosarcoma tumor cells (FSa II) by 250 kv X-irradiation machine. The cytotoxicity of ESMR was evaluated from its ability to reduce the clonogenecity of FSa II cells. In X-irradiation alone group, each 2, 4, 6 and 8 Gy was given to FSa II cells. In X-irradiation with ESMR group, 0.2 mg/ml of ESMR was exposed to FSa II cells for 1 hour before X-irradiation. Results: The proliferation of cultured mouse splenocytes and thymocytes were enhanced by the addition of ESMR in vitro. The number of B cells and T cells in mouse splenic lymphocytes was significantly increased in ESMR pretreated mice in vivo. In FSa II cells that received a combination of 0.2 mg/ml of ESMR with X-irradiation exposure, the survival fraction with a dose of 2, 4 and 6 Gy was $0.39{\pm}0.005$, $0.22{\pm}0.005$ and $0.06{\pm}0.007$, respectively. For FSa II cells treated with X-irradiation alone, the survival fraction with a dose of 2, 4 and 6 Gy was $0.76{\pm}0.02$, $0.47{\pm}0.008$ and $0.37{\pm}0.01$. The difference in the survival fraction of the mouse FSa II cells treated with and without ESMR was statistically significant (p<0.05). Conclusion: Treatment with ESMR increased cell viability of mouse splenocytes in vitro and especially the subpopulation of B cells and T cells in splenocytes in ESMR-pretreated mice. However, treatment with ESMR did not increase the level of Th and Tc subpopulations in the thymocytes. Treatment with the combination of ESMR and X-irradiation was more cytotoxic to mouse tumor cells than treatment with X-irradiation alone; this finding was statistically significant.
