Lipid Specificity for Membrane Oxidation Catalyzed by Cytochrome c : An EPR Study

  • Decay of the spin label attached to cytochrome c or to stearic acid has been measured by electron paramagnetic resonance (EPR) spectroscopy to monitor membrane oxidation induced by cytochrome c-membrane interaction. Binding of cytochrome c sequestered the acidic phospholipids and membrane oxidation was efficient in the order linoleic oleic>stearic acid for a fatty acid chain in the acidic phospholipids. The spin label on cyt c was destroyed at pH 7 whereas that on stearic acid embedded in the membrane was destroyed at pH 4, presumably due to different modes of cyt c-membrane interaction depending on pH. Interestingly, cyt c also interacts with phosphatidylethanolamine, an electrically neutral phospholipid, to cause rapid membrane oxidation. Both EPR and fluorescence measurements indicated that electrostatic interaction is at least partially responsible for the process.

  • Alnine pellets were installed in a nuclear power plant for one or two operation cycles and measured by electron spin resonance (ESR) spectrometers for dosimetry. Dose and "x/y ratio", i.e., satellite peak over main center peak ratio, were measured for the returned alanine dosimeters from the nuclear power plant and compared to the values of reference alanine dosimeters exposed only to gamma rays. The variation of the x/y ratio change depended on the population of radicals from each radiation component with different LET. The gamma dose in a mixed radiation field was estimated by an additive gamma ray irradiation experiment and the measured dose rate at specified locations in the containment building.

Analytical Characteristics of Electron Spin Resonance for Identifying Irradiated Ramen Soup with Radiation Sources (방사선 조사된 라면수프의 조사선원에 따른 전자스핀공명 분석특성)

  • The identification characteristics of irradiated commercial Ramen soup were investigated depending on radiation sources and doses by electron spin resonance (ESR) spectroscopy. Two commercial powder soups (RS-1, RS-2) were irradiated at 0 to 20 kGy under ambient conditions by both a Co-60 gamma irradiator and an electron beam (EB) accelerator, respectively. Crystalline sugar-induced multi-component signals with g-values of 2.010/2.011, 2.006, 2.002 and 1.999 were detected in the irradiated Ramen soup (RS-1, RS-2), whereas $Mn^{2+}$ signals were observed in non-irradiated samples, thereby distinguishing each other. Under the same analytical conditions, the intensity of ESR signals was higher in EB-irradiated samples than the gamma-irradiated ones. Determination coefficients ($R^2$) between irradiation doses and corresponding ESR responses were above 0.9665 in all the samples, and the magnetic field of specified g-value remained constant. The predominant ESR signals of $g_2$ (2.010-2.011) and $g_3$ (2.002) increased with corresponding doses of irradiation ($R^2$= 0.9750-0.9981).

  • Electron Spin Resonance (ESR) spectroscopy has been used to detect the presence of radiation-induced free radicals in biological samples since the mid 1950s and to irradiate foods containing cellulose, crystalline sugar, and bone. Therefore, we analyzed the ESR spectrum of irradiated infant formula and its ingredients in this study. Samples were irradiated with 2 different radiation sources of $^{60}Co$ gamma rays and electron beams (EBs), and the absorbed doses were 0, 1, 3, 5, and 7 kGy. ESR measurements were performed under normal atmospheric conditions using a JEOL JES-FA100 spectrometer equipped with an X-band bridge. Irradiated infant formula showed anunsymmetrical spectrum ($g_1$=2.0050, $g_2$=2.0006); in contrast, non-irradiated samples showed asymmetrical spectrum. The ingredients of irradiated samples showed a multi-component ESR signal in glucose and lactose and a singlet-type spectrum in milk powder (g=2.0050). $R^2$ of the dose-response curve showed a fine linearity of over 0.95 across the entire sample. We also compared the spectra of identical samples irradiated with $^{60}Co$ gamma rays and EBs, because EBs can be used for food irradiation in foreign countries, although this is not permitted in Korea. However, we could not find any significant differences according to the types of radiation source. Thus, ESR spectroscopy can be used to detect irradiated infant formula and several types of primary ingredients in this formula.

  • Substituted anthrasemiquinone radical anions, which were formed by laser flash photoreduction of the substituted anthraquinone (1,4-DHAQ, 1,5-DHAQ, 1,8-DHAQ, 1,8-DCAQ), were studied by the electron spin resonance spectroscopy in the mixed solvents of the 2-propanol and the triethylamine. Hyperfine splitting constants (hfsc) were determined by simulating the ESR spectra. Extended Huckel Molecular Orbital (EHMO) calculations were performed to obtain $\pi$ spin densities for these quinones. The hyperfine splitting constants could be assigned to specific protons on the basis of $\pi$ spin densities.

Identification Characteristics of Irradiated Dried Red Pepper during Storage by the Analysis of Electron Spin Resonance and Hydrocarbons (전자스핀공명 및 Hydrocarbon 분석에 의한 방사선처리 건고추의 저장 중 검지특성)

  • Electron spin resonance (ESR) and hydrocarbon characteristics were analyzed to establish identification conditions for irradiated dried red pepper. The ESR spectroscopy for 4 different parts (powder, pericarp, seed, stem) of the samples showed that irradiated samples signaled (g=2.024, 2.006, 1.987) a pair of peaks from a cellulose radical at intervals of 6 mT, which were not found on the non-irradiated samples. The ESR signals increased in directly proportion to the irradiation doses, which were still detectable after 12 weeks of storage at room temperature. The GC-MS analysis of hydrocarbons after fat extraction and separation by florisil column chromatography revealed that hydrocarbons, such as 1-tetradecene (14:1), 1,7,10-hexadecatriene (16:3), 1,7-hexadecadiene (16:2), 1-hexadecene (16:1), 6,9-heptadecadiene (17:2), and 8-heptadecene (17:1), were detected only from the irradiated samples immediately after irradiation and 8 months of storage. They linearly increased with the dose of irradiation, suggesting them as radiation-induced markers for irradiated dried red pepper.

  • The identification characteristics of inadiated dried onion and garlic at 1, 4 and 7 kGy were investigated by analyzing their photostimulated luminescence (PSL), thermoluminescence (TL) and electron spin resonance (ESR). The PSL results showed that the photon counts of non-irradiated dried onion were less than 700 (negative) and those of irradiated samples at 1kGy or more were over 5000 (positive), making it possible to discriminate non-irradiated onions from irradiated ones. However, the intermediate counts (700-5000) were observed in irradiated garlic at 1 and 4 kGy, even though positive counts were measured in 7 kGy group. Radiation-induced TL glow curves $(TL_1)$ were measured at $150^{\circ}C$ range in both irradiated onion and garlic samples at 1 kGy or more, which could identify irradiated groups. Furthermore, determination of TL ratios $(TL_1/TL_2)$ for both samples could verify the above results. It was found, however, that ESR spectroscopy was not suitable for the detection of irradiated dried onion and garlic.

FTIR characterization and antioxidant activity of water soluble crude polysaccharides of Sri Lankan marine algae

  • Polysaccharides of marine algae exhibit different structural characteristics and interesting biological functions. In this study, crude polysaccharides (CP) of eleven Sri Lankan marine algae obtained through hot water extraction and ethanol precipitation were investigated for DPPH, alkyl, and hydroxyl radical scavenging activities using electron spin resonance spectrometry and for intracellular reactive oxygen species scavenging activity in the Chang liver cell line. Characterization of CPs was done by Fourier transform infrared (FTIR) spectroscopy and by analysis of the monosaccharide composition. Time-dependent density functional theory quantum-chemical calculations at the RB3LYP/6-31G(d,p) level for constructed dimeric units of the corresponding polysaccharides were used to resolve the FTIR spectra. CPs from Chnoospora minima showed the highest DPPH and alkyl radical scavenging activities and higher intracellular reactive oxygen species scavenging effects for both AAPH and $H_2O_2$ induced ROS production in "Chang" cells. The major polysaccharide constituent in C. minima CP was identified as fucoidan and it displayed a higher sulfate content. The degree of sulfation of these polysaccharides suggests a positive correlation with the observed antioxidant properties.

In vivo ESR measurement of free radical reaction in living mice

  • Recently, free radicals such as active oxygen species, nitric oxide, etc are believed to be one of the key substances in physiological and pathological, toxicological phenomena, and oxidative damages, and all organism have defencing system against such as free radicals. Formation and extinction of free radicals may be regulated through bio-redox system, in which various enzymes and compounds should be involved in very complicated manner. Thus, direct and non-invasive measurement of in vivo free radical reactions with living animals must be essential to understand the role of free radicals in pathophysiological phenomena. Electron spin resonance spectroscopy (ESR) is very selective and sensitive technique to detect free radicals, but a conventional ESR spectrometer has large detect in application to living animals, since high frequent microwave is absorbed with water, resulting in generation of high fever in living body. In order to estimate in vivo free radical reactions in living whole animals, we develop in vivo ESR-CT technique using nitroxide radicals as spin probes. Nitroxide radicals and their reduced forms, hydroxylamines, are known to interact with various redox systems. We found that! ! the signal decay due to reduction of nitroxyl radicals is influenced by aging, inspired oxygen concentration, ischemia-referfusion injury, radiation, etc. In the present paper, I will introduce in vivo ESR technique and my laboratory recent results concerning non-invasive evaluation of free radical reactions in living mice.

A Study on Generative Characteristics of Radicals in Aqueous Solutions of Humic Acids using Electron Spin Resonance (전자기공명기법을 이용한 Humic Acid 용액에서의 라디칼 발생 특성에 관한 연구)

  • We studied to determine quantitatively the radical species generated from humic acid (HA) solutions by irradiation(>400 mn). The formation of radical species from HA solutions was investigated with ESR spectroscopy. We gave ESR spectrum with g-value 2.0048 and line width 0.559mT, coincided with those of the semiquinone radical. 0.1 mg/L HA solution generated the radicals of $1.2{\times}10^{-6}M$, increased with increasing HA concentration. Also, pH and ionic strength effect of the amount of the semiquinone radical generate from HA solution. In this study, we have found that the singlet oxygen affects the semiquinone radicals generation in HA solution.
