건칠의 항주름 효과와 미백효과 연구 (The effect of wrinkle care and whitening by Rhus verniciflua Stokes)

  • Objectives : Recently, the demands for the effective and safe depigmentating and anti-aging agents of the skin have increased due to the medical, pharmaceutical and cosmetic reasons. The aim of this study is to search new materials from the traditional herbal medicines which inhibit the aging process of skin in vitro.Methods : Human dermis cell (HS68) was used to test the effect of Rhus verniciflua Stokes (RVS). 80% ethanol or water extracts were screened for their inhibitory activities against elastase. Elastase inhibition effect was tested by microplate reader instrument. And MMP-1 suppression effect of RVS was tested by western blot. These cells were investigated the viability by MTS assay. And also the inhibition effect of tyrosinase by RVS was tested.Results : RVS (final concentrstion 1 ㎎/㎖ appeared over 30% of inhibition of elastase activity. So we are investigated anti wrinkle effects of Rhus verniciflua Stokes look through MMP-1 inhibition activity, also Extracts of RVS showed higher anti-tyrosinase activity than arbutin as final concentration 1 mg/ml. These results suggest that herbal medicines could be strong potential sources of inhibition of anti-aging and whitening effects for the skin.Colclusions : RVS was the best suppressor candidate of elastase activity among other control oriental drugs. It was found that RVS did not have toxicity to cells. We found proper concentration of RVS to treat in HS68 culture and investigated not only wrinkle care effect but also whitening effect of RVS.

오미자 유산균 발효물의 주름개선 효과 (Anti-Wrinkle Effect of Schizandra chinensis Baillon Fermented with Lactobacillus plantarum)

  • 새로운 주름개선제 성분을 찾기 위해서 본 연구에서는 사람 피부 섬유아세포의 세포독성, 콜라겐 생합성, matrix metalloproteinase-I (MMP-1) 및 elastase 저해활성에 대한 Lactobacillus plantarum으로 발효된 오미자 발효액의 주름개선 효과를 평가하였다. 먼저, 오미자 추출물은 L. rhamnosus으로 $37^{\circ}C$에서 1일 동안 발효하였다. 발효물의 세포독성은 cytopathic effect reduction 방법에 의해 평가하였다. 콜라겐 생합성에 대한 발효물의 영향은 procollagen type-IC peptide EIA kit에 의해 평가하였으며, matrix metalloproteinase-I(MMP-1)에 대한 발효물의 영향은 Matrix Metalloproteinase-1 Biotrack activity Assay Kit에 의해 평가하였다. Elastase inhibition assay는 기질로써 N-Suc-$(Ala)_3$-nitroanilide을 사용하여 기질 반응에 의해 평가하였다. 결과로써 오미자 발효물은 사람 피부 섬유아 세포에 대해 $100{\mu}g/mL$의 농도에서 세포독성을 나타내지 않았다. 또한 오미자 발효물은 콜라겐 생합성을 촉진시켰으며, MMP-1의 저해 효과를 나타내었다(p < 0.05). Elastase inhibition assay에서 오미자 발효물의 $IC_{50}$$36.4{\mu}g/mL$이었다. 그러므로 본 연구에서 오미자 발효물은 주름개선 효과를 보유하고 있으며, 이것은 피부의 주름개선을 위해 사용가능하리라 사료된다.

곤달비의 주름개선효과 (Anti-wrinkle Effect of Ligularia stenocephala)

  • In the previous study, we reported the antioxidative activity and inhibition of melanogenesis of Ligularia stenocephala extracts. In this study, inhibitory effect on elastase and production type I procollagen were investigated. The compounds which was investigated were 4-hydroxyacetophenone, vanillin, chlorogenic acid, caffeic acid isolated from Ligularia stenocephala. They were slightly mild on elastase inhibition activity but 4-hydroxyacetophenone, vanillin exhibited good inhibition activity on collagen production. These results suggest that Ligularia stenocephala may have potential as an anti-aging ingredient in cosmetics.

왕겨초액의 대량생산과 항노화 및 미백 효과 (Mass Production of Chaff-vinegar and Its Effect of Anti-Aging and Whitening)

  • 본 연구는 왕겨를 이용한 항산화, 항노화에 효과적인 초액을 대량생산하여 기능성을 검증하고 왕겨초액 내의 기능성물질을 분석하여 사업화의 가능성을 타진하고자 하였다. 정제된 왕겨초액을 분획하여 기능성이 확인된 부분을 HPLC 기기를 이용하여 확인한 결과 caffeic acid 외에 7종의 polyphenol 유래물질에서 기능성을 확인하였다. 기능성 화장품소재 시험으로는 세포 생존율 측정(MTT assay), 주름개선 (collagenase inhibition assay, elastase inhibition assay), 미백(tyrosinase inhibition assay), 항산화 시험(DPPH free radical scavenging assay, SOD-like activity, xanthine oxidase inhibition assay)이 실시하였다. 피부주름형성은 콜라겐의 합성과 분해가 중요한 원인으로 작용한다고 알려져 있지만, 최근 많은 연구에서 엘라스틴의 재형성과 분해 또한 주름형성 기전에서 중요한 작용을 하는 것으로 보고되고 있다. 왕겨초액은 $100{\mu}L/mL$의 농도에서 collagenase 저해효과가 100%, elastase 저해효과가 55% 이상으로 주름개선 효과가 우수하였다. 왕겨초액은 $100{\mu}L/mL$의 농도에서 tyrosine에 대한 tyrosinase 저해효과가 80% 이상으로 미백효과가 우수하게 나타났다. DPPH radical 소거율, xanthine oxidase 저해와 SOD-like 활성을 이용하여 항산화 효과를 측정하였다. 왕겨초액 $100{\mu}L/mL$의 농도에서 DPPH radical 소거율이 80%, xanthine oxidase 저해효과가 80%, 그리고 SOD-like 활성이 100%를 나타내었다. 따라서 본 연구를 통하여 정제 왕겨초액을 이용한 산업화를 위한 가능성을 타진하여 효과가 좋음을 확인할 수 있었다. 이와 같은 결과로 왕겨초액을 이용한 기능성 화장품의 소재로 뿐만 아니라, 식품 및 의약외품의 품목허가를 통하여 고부가가치의 상품화소재로의 가능성이 있다고 사료됩니다.

사람의 백혈구 내에 있는 Elestase: 순수부리, 금속이온의 화학량, 그리고 Chelating 효과에 의한 활성도 조절 (Human Neutrophil Elastase: Rapid Purification, Metal binding Stoichiometry and Modulation of the Activity by Chelating Agents)

  • 사람의 혈액으로부터 Elastase를 분리하는 새로운 방법을 개발하여 순도 높은 효소를 얻고 이 효소에 의하여 발생되어지는 질병의 예방과 치료에 응용하기 위하여 이 효소의 근본적인 성질규명을 시도하였다. 정제 방법으로는 Sephodex G-75를 이용한 1회의 액체 크로마토그라피와 HPLC을 이용한 2회의 코마토그라피를 거치는 2단계 방법을 사용하였다. 이때 얻어진 효소는 분자량이 $26,000{\sim}29,700$ 사이에 있는 3개의 다른 분자량을 가진 물질로 확인되었으며, 항체 반응에서 사람의 결구내 Elastase의 특성을 나타내었다. 함유하고 있는 미량의 금속이온을 분석한 결과 정제하는 단계에 따라 Zn 이온의 효소분자에 대한비는 증가하였으며 가장 순수한 분자내의 효소분자와 Zn 이온과의 비는 1 : 2였다. 이 효소는 chelating agent에 의하여 활성도를 잃게 되었으며, 반대로 chelating agents에 의하여 활성도를 잃고 있는 반응 medium에 그 chelaing agents의 농도를 초과하는 2가 이온 즉 Zn 이온, Ca 이온, 그리고 Mn 이온을 넣으면 그 활성도는 원상복귀되나 Mg이온에 의하여는 그 활성도가 원상회복 되지 않았다. 이 모든 성질을 종합하면 사람의 Neutrophil granule에 있는 Elastase는 지금까지 알려진 바와 같이 Serine pretense임과 동시에 metalloenzyme으로 제고되어야 한다고 제안한다. 나아가서chelating agents에 의하여 이 효소의 활성도를 조정할 수 있다는 것은 이 효소에 의하여 일어나는 질병의 치료에 응용할 수 있는 가능성도 보여준다.

Inhibitory effects of medicinal plants on elastase activity and biological activities in the active Plant extracts

  • Inhibitory effects of 200 medicinal plants on elastase activity were screened. Among the 200 plants, six plant extracts exhibited more than 65% of inhibition on elastase activity by their total methanol extracts at 1, 000$\mu\textrm{g}$/$m\ell$ as a final concentration. For six active plants including Areca catechu, Cinnamonum cassia, Myristica frograns, Curcumma longa, Alponia katsumadai, and Dryopteris crassirrhizoma, the inhibitory effects of their comsituents were examined on the activity of human leukocyte elastase, hyaluronidase and lipid peroxidation. In lipid peroxidation assay using TBA method, three of the 6 plants including Curcumma longa, Areca Catechu and Alponia katsumadai exhibited more than 70% of inhibition at the concentration of 1, 000$\mu\textrm{g}$/$m\ell$, but only one plant, Areca Ctechu showed high inhibitory effect on hyaluronidase activity. The results suggest that medicinal plants showing several biological activities may be potent inhibitors of anti-aging process on skin and that might be useful for application in cosmetics.

Purification and Characterization of Serine Protease Inhibitors from Dolichos lablab Seeds; Prevention Effects on Pseudomonal Elastase-Induced Septic Hypotension

  • Three kinds of serine protease inhibitors, members of the Bowman-Birk trypsin inhibitor, were purified from Dolichos lablab seeds and named Dolichos protease inhibitor 1, 2 and 3 (DI-1, DI-2 and DI-3), respectively. Each inhibitor showed a single band with gel mobility at around 15.9, 12.1 and 14.6 kDa on 20% SDS-PAGE under reducing conditions. To characterize inhibitory specificity, the inhibition constant (Ki) for these inhibitors was measured against several known serine proteases. All three Dolichos protease inhibitors (DI-1, DI-2 and DI-3) inhibited the activity of trypsin and plasmin, but had no effect on thrombin and kallikrein (either for human plasma kallikrein or for porcine pancreas kallikrein). DI-1 inhibited chymotrypsin most effectively (Ki = $3.6{\times}10^{-9}\;M$), while DI-2 displayed inhibitory activity for porcine pancreatic elastase (Ki = $6.2{\times}10^{-8}\;M$). Pre-treatment of the 33 mg/kg of DI-mixture (active fractions from $C_{18}$ open column chromatography that included DI-1, DI-2 and DI-3) inhibited the induction of pseudomonal elastase-induced septic hypotension and prevented an increase in bradykinin generation in pseudomonal elastase-treated guinea pig plasma. Also, the increase of kallikrein activity, by injection of pseudomonal elastase, was inhibited by the pretreatment of the DI-mixture in a guinea pig. Since the DI-mixture had no inhibitory effect on kallikrein activity when Z-Phe-Arg-MCA was used as a substrate in vitro, its inhibitory activity in the pseudomonal elastase-induced septic hypotension model might not be due to a direct inhibition of plasma kallikrein in the activation cascade of the Hageman factor and prekallikrein system. These results suggest that the Dolichos DI-mixture might be used as an inhibitor in pathogenic bacterial protease-induced septic shock.

한약재 복합 추출물이 항산화 및 항노화에 미치는 영향 (Anti-oxidation and Anti-aging Effect of Mixed Extract from Korean Medicinal Herbs)

  • Objectives : The present study was designed to investigate effects of mixed extract from korean medicinal herbs (MIX) on oxidation/reduction reaction-related and aging-related enzyme in vitro. Methods : We performed MTT assay, collagenase inhibition assay, elastase inhibition assay, tyrosinase inhibition assay, DPPH free radical scavenging assay, SOD-like activity and xanthine oxidase inhibition assay. Results : Recently, many studies have reported that elastin is also involved in inhibiting or repairing wrinkle formation, although collagen is a major factor in the skin wrinkle formation. The MIX showed 97% inhibition of collagenase activity, and 64% inhibition of elastase activity at 1 mg/ml concentration of MIX, next only to positive control, which indicate good efficacy for anti-wrinkle ingredient. Also it's treatment showed 34% inhibition of tyrosinase activity, to relate whitening effect, at the same dose of MIX. Antioxidant activities were determined by DPPH radical scavenging, xanthine oxidase (XO) inhibiting activity and SOD-like activity. Also these scavenging, XO-inhibiting and SOD-like activities were measured in 91%, 80%, and 63% inhibition, respectively, at a treated dose of 1 mg/ml, compare to control. Conclusions : These results suggest that possibility of mixed korean medicinal herbs as a functional ingredient for anti-wrinkle and whitening, anti-oxidation and anti-aging cosmetic formula.

하엽(荷葉) 추출물이 항산화 효능 및 피부노화에 미치는 영향 (Antioxidant and Anti-wrinkling Effects of Extracts from Nelumbo nucifera leaves)

  • Objective : The purpose of this study was to investigate anti-aging and antioxidant effects of extracts of Nelumbo nucifera leaves (NN-L) using ethanol on skin .Methods : Each part of leaves(NN-L), flowers(NN-F) and stem(NN-S) was extracted with 70% ethanol. We performed radical scavenging assay(DPPH, ABTS+, Superoxide anion radical), elastase inhibition assay, collagenase inhibition assay. NN-L extracts were tested for cell viability(MTT assay), MMP-1 inhibition and MMP-1 protein expression on CCD-986sk cells (human fibroblast line).Results : Recently, many studies have reported that elastin is also involved in inhibiting or repairing wrinkle formation, although collagen is a major factor in the skin wrinkle formation. We measured its free radical scavenging activity, elastase inhibitory activity and expression of MMP-1 (matrix metalloprotease-1) in human fibroblast cells. Among the parts of Nelumbo nucifera, NN-L showed the highest antioxidant activities and in radical scavenging. DPPH, ABTS+ and Superoxide anion radical scavenging activity of NN-L at concentration of 1,000 μg/mL were 91.43%, 99.31% and 73.7% respectively. In vitro elastase and collagenase inhibition effects of NN-L at concentration of 1,000 μg/mL was 42.8% and 55.3% respectively. The ethanol extract of NN-L showed cell viability of 95.4% in 50 μg/mL concentration. In addition, The results from Western blot assay showed that NN-L decreased the expression of MMP-1 protein in a dose-dependent manner (by up to 35.0% at 50 μM).Conclusion : The findings suggest that the NN-L great potential as a cosmeceutical ingredient with antioxidant and anti-wrinkling effects.

Inhibition Effect Against Elastase, Collagenase, Hyaluronidase and Anti-oxidant Activity of Thinning Green Ball Apple

  • The thinning Green ball apple was extracted using water and ethanol and a phenolic concentration of thinning Green ball apple was $50-200{\mu}g/mL$. The water and ethanol extracts of thinning Green ball apple showed 94.69% and 92.24% 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity and 100.30% and 99.16% 2,2'-Azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity at phenolic concentration of $200{\mu}g/mL$, respectively. The water and ethanol extracts of thinning Green ball apple showed antioxidant protection factor of 1.76 antioxidant protection factor and 1.76 antioxidant protection factor, respectively. The water and ethanol extracts showed 101.46% and 99.64% anti-oxidative effect on thiobarbituric acid reactive substances at phenolic concentration of $200{\mu}g/mL$. Hence, the water and ethanol extracts of thinning Green ball apple can be considered a potential anti-oxidant. The water and ethanol extracts showed 33.28% and 32.14% hyaluronidase inhibition, respectively, at phenolic concentration of $150{\mu}g/mL$. The water and ethanol extracts showed 47.33% and 40.92% elastase inhibition and 46.19% and 65.58% collagenase inhibition at phenolic concentration of $200{\mu}g/mL$, respectively. About these experiments, thinning Green ball apple was found to exhibit anti-oxidation activity as well as hyaluronidase, elastase and collagenase inhibitory activities. Therefore, thinning Green ball apple can be considered a potential source for functional food.
