Injectable Gel Type Formulation of Hydrated Egg Phosphatidylcholine and Hyaluronate for Local Drug Delivery

  • Injectable gel composed of egg phosphatidylcholine (egg PC), hyaluronate (HA) and water was formulated for local drug delivery. The lamellar liquid crystalline structure of the egg PC/water system did not change by adding HA in the formulation. However, egg PC/HA/water gel was more resistant to erosion than the egg PC/water gel. The egg PC/HA/water and egg PC/water gels containing model drugs, tetracycline and sudan IV were prepared to perform in vitro and in vivo drug release experiments. In vitro release of tetracycline was sustained in the gel type formulations. The release rate of hydrophobic sudan IV was extremely slow. More than 99% of sudan IV remained inside the gel after 5 days. In vivo release of drugs from the air pouch model in Balb/c mice shows that lipophilic sudan IV remained for more than 10 days whereas tetracycline remained for 1 day in the pouch. The compatibility of the gels was also examined by histopathology. The gels did not cause any adverse inflammatory effect in the air pouch.

Complexation of Amphotericin B With Egg Phosphatidylcholine Liposomes

  • The complexation and physical characteristics of egg phosphatidylcholine (PC) liposome containing amphotericin B(AmB) were investigated through circular dichrosim(CD) spectra, the size distribution, the turbidity change, and the calcein release. CD spectra of AmB-containing egg PC mxture exhibited a positive peak around 330 nm indicative of complexation of AmB and four negative peaks. The positive peak increased up to $2.2{\;}millidegree/{\mu}g$ AmB as AmB contents increased up to 12% (w/w), suggesting that AmB-phospholipid complexation was promoted by the antibiotics. The effective diameter of liposomesa by dynamic light scattering decreased from 450 nm to 220 nm as the amount of AmB in liposomes increased from o to 30% (w/w). The complexation may be responsible for the reduction in size. On the other hand, at around 1 mN deoxycholate (DOC), the reltive turbidities of 5 and 10% (w/w) AmB-containing liposome suspension were less than 1 probably due to the soblubilization of the complex, while those of pure PC liposome suspension were larger than 1 at the same concentration. Deoxycholate-induced release of liposomes, indicating the intercalation of the drug into the bilayers. Therefore, it is concluded that in AmB/eggPC/water system, AmB-phospholipid complexcoexists with AmB-containing liposomes.

Interaction of Human Globin with Hemin-Containing Liposome

  • The globin in cytosol receives the heme in mitochondria and folds to the hemoglobin within erythrocyte. Two mechanisms have been proposed that the heme was transfered post-translationally or cotranslationally to the globin. In this research, how the globin in cytosol receives post-translationally the heme from membranes was studied according to pH and phospholipid composition. Globins dissolved in various pH buffer solutions $(PH\;3\~7)$ were rapidly added into the bulk of egg phosphatidylcholine $100\%\;or\;60\%$ liposomes containing hemin in pH 7 buffer solution. Hemin was very highly transferred to globin at pH 4 and 6. Also, hemin was more efficiently transfered to globin in egg phosphatidylcholine $100\%$ than in $60\%$ liposomes.

Effects of Phosphatidylcholine and Phosphatidylethanolamine from Egg Yolk on Thermal Oxidation of Canola Oil (달걀 노른자에서 분리한 포스파티딜콜린과 포스파티딜에탄올아민이 카놀라유의 가열산화에 미치는 영향)

  • The principal objective of this study was to assess the effects of phosphatidylcholine (PC) and phosphatidylethanolamine (PE) extracted from egg yolk on the oxidation of tocopherol-stripped canola oil and its browning, as well as their content changes during 12 hr of heating at $180^{\circ}C$. PC and/ or PE contents in the oil were measured at 200, 500, 1,000, or 2,000 ppm. PL contents in the oil and oil browning were determined by high performance liquid chromatography (HPLC) and spectrophotometry, respectively. The oil oxidation was evaluated by the combination of fatty acid composition, conjugated dienoic acid content, and p-anisidine value. PC was degraded at a slower rate than PE during heating and the co-presence of PE reduced its rate of degradation. PE increased oil browning more profoundly than PC did. PC significantly reduced oil oxidation during heating; however, we noted a possible antagonism between PE and PC in reducing the oil oxidation. Egg yolk PC was a better antioxidant in oil oxidation during heating.

  • In ethanol fermentations with Saccharomyces sake, phosphatidylcholine-egg albumin as a supplement in fermentation media was much more effective in enhancing ethanol production than linoleic acid-ergosterol. It came from the differences in alcohol-tolerance between egg albumin and ergosterol. The egg albumin was supposed to function as a nutrient rather than to form protective layers around the cells against ethanol.

  • Lecithin is a naturally occurring group of phospholipids found in nearly every living cell and has been widely used as the ingredient of functional foods. Lecithin has high content of phosphatidylcholine(PC), pharmaceutical material which promotes metabolism through the cell membrane. This study was carried out to improve the present inconvenient analytical method of PC in law for health & functional foods. The commodities used in this experiment, were two kinds of egg yolk and eight kinds of soybean lecithin functional foods. PC was separated with isocratic elution with hexane : isopropanol : D.W (30:60:8) through silica column (2.1$\times$150 mm) by HPLC with Evaporative Light-Scattering Detector (ELSD). The flow rate of the eluent was 0.5 ml/mim and infect volume was 10ul. The neubilizer temperature of detector was $60^{\circ}C$, drift tube temperature of that was $75^{\circ}C$ and gas flow was 30 psi. Quantification was carried out by external standardization. Limit of quantification was 0.15ppm. Lecithin contents of egg yolk and soybean Products were > $66\%$ and > $81\%$), respectively. Phosphatidylcholine contents of egg yolk and soybean products were > $74\%$ and > $18\%$, respectively.

  • To increase skin permeability of LMWH (Low Molecular Weight Heparin), ultradeformable liposomes were developed. Ultradeformable liposomes were developed by Egg phosphatidylcholine (Egg-PC) and edge activator. Entrapment efficiency, vesicle size and zeta potential of vesicles were determined and characterized for deformability and stability. Transepidermal permeation of LMWH was compared to saturated aqueous control in vitro. The steady-state flux and its maximum time were calculated from the flux curves. (omitted)

  • For continuous ethanol production In an immobilized cell reactor consisting of Saccharomyces sake, feedings of one tenth to three tenths times diluted fermentation media were effective for maintaining the high ethanol productivity and physical stability of immobilized beads. In case two tenths times dilltued one of the fermentation medium supplemented with egg albumin hydrolysate(0.5%) and phosphatidylcholine(0.5%) was fed, a maximum ethanol productivity of $69 g/\ell$-hr was attained at a dilution rate of$1.1lhr^{-1}$, and it was 50% higher than that of the two tenths times diluted one of the fermentation medium without any supplement, $46 g/\ell$-hr.

Reduction of Cell Membrane Toxicity of Amphotericin B Using Liposome System (리포좀계를 이용한 Amphotericin B의 세포막 독성 저하)

  • Lipsome system composed of egg phosphatidylcholine was employed to reduce the membrane toxicity of Amphotericin B(Amp. B). Liposomal Amp. B, which showed a free drug equivalent antibiotic effect on fungi, displayed a remarkable reduction of toxicity of the drug against the membrane of red blood cell than that of fungizone which has been used in clinical treatment, and it shows conspicuously lowered toxicity on red blood cells. However liposomal Amp. B which contains cholesterol as a component of liposome lowered the antibiotic effect and toxicity than that phosphatidylcholine liposome. This due to the affinity between Amp. B and cholesterol. In addition to this, ${\beta}$-glucuronidase from snail juice crude enzyme reveals synergistic effect on liposomal Amp. B and free Amp. B. We also obtained positively raised antibiotic effect, when enzyme which is coupled with palmitic acid using NHSP inserted into liposome bilayer From these results, we suppose that the use of liposomal system in the case of Amp. B shows increasing antibiotic effect and dramatically lowered toxicity, thus, we think that we can solve the problem of Amp. B toxicity, which cause hesitate of clinical use.
