Purification and enzymatic properties of a peroxidase from leaves of Phytolacca dioica L. (Ombú tree)

  • A peroxidase (PD-cP; 0.47 mg/100 g leaves) was purified from autumn leaves of Phytolacca dioica L. and characterized. PD-cP was obtained by acid precipitation followed by gel-filtration and cation exchange chromatography. Amino acid composition and N-terminal sequence of PD-cP up to residue 15 were similar to that of Spinacia oleracea (N-terminal pairwise comparison showing four amino acid differences). PD-cP showed a molecular mass of approx. 36 kDa by SDS-PAGE, pH and temperature optima at 3.0 and $50.0^{\circ}C$, respectively and seasonal variation. The Michaelis-Menten constant ($K_M$) for $H_2O_2$ was 5.27 mM, and the velocity maximum ($V_{max}$) $1.31\;nmol\;min^{-1}$, while the enzyme turnover was $0.148\;s^{-1}$. Finally, the presence of $Ca^{2+}$ and $Mg^{2+}$ enhanced the PD-cP activity, with $Mg^{2+}$ 1.4-fold more effective than $Ca^{2+}$.

Cultural and Rainfall Factors Involved in Disease Development of Fusarium Wilt of Sweet Potato

  • Environmental factors such as soil moisture, land management, and weather conditions affecting Fusarium wilt of sweet potato were investigated in major sweet potato cultivation regions in Korea. Fusarium wilt occurred mainly in reclaimed terracing lands, which are flattened and located in hilly to mountainous areas at the base of the mountain, in early seasonal cultivation regions. Disease severity was lower in reclaimed fields with natural slope. The development of Fusarium wilt in the fields was highly correlated with precipitation during planting period (r=-0.96**). Fusarium wilt was more severe in fields with less than 20 cm of available soil depth than in fields with over 20 cm of available soil depth. Greenhouse studies were consistent with field studies that less soil moisture content caused severe Fusarium wilt of sweet potato. These results indicate that low rainfall and moisture of soil with low effective soil depth during planting period are important environmental factors influencing the development of Fusarium wilt.

  • I-III-VI2 chalcopyrite compounds, particularly copper, indium, gallium selenide(Cu(InxGa1-x)Se2, CIGS), are effective light-absorbing materials in thin-film solar application. They are direct band-gap semiconductors with correspondingly high optical absorption coefficients. Also they are stable under long-term excitation. CIS (CIGS) solar cell reached conversion efficiencies as high as 19.5%. Several methods to prepare CIS (CIGS) absorber films have been reported, such as co-evaporation, sputtering, selenization, and electrodeposition. Until now, co-evaporation is the most successful technique for the preparation of CIS (CIGS) in terms of solar efficiency, but it seems difficult to scale up. CIS solar cells have been hindered by high costs associated with a fabrication process. Therefore, inorganic colloidal ink suitable for a scalable coating process could be a key step in the development of low-cost solar cells. Here, we will present the preparation of CIS photo absorption layer by a solution process using novel metal precursors. Chalcopyrite copper indium diselenide (CuInSe2) nanocrystals ranging from 5 to 20nm in diameter were synthesized by arrested precipitation in solution. For the fabrication of CIS photo absorption layer, the CuInSe2 colloidal ink was prepared by dispersing in organic solvent and used to drop-casting on molybdenum substrate. We have characterized the nanoparticless and CIS layer by XRD, SEM, TEM, and ICP.

동화지구 절계 수문량 재추정 (Reestimation of Hydrologic Design Data in Donghwa Area)

  • The fundamental study of hydrologic redesign of Donghwa area located in a sccond tributary of Seomjin river was performed. The amounts of hydrologic design were estimated using the available cumulated hydrology data provided by Korea Agricultural and Rural Infrastructure Corporation (KARICO). The management status of The water resources in Donghwa area was also widely surveyed. The probability rainfalls, probable maximum precipitation (PMP) and probability floods were estimated and subsequently their changes analyzed. The amount of 200 year frequency rainfall with l day duration was 351.1 mm, 2.5 % increased from the original design value, and The PMP was 780.2 mm. The concentration time was reestimated as 2.5 hours from existing 2.4 hours. Soil Conservation Service(SCS) method was used to estimate effective rainfall- The runoff curve number was changed from 90 to 78, therefore the maximum potential retention was 71.6 mm, 154 % increased from the original value. The Hood estimates using SCS unit hydrograph showed 8 % increase from original value 623 $m^3$/s to 674 $m^3$/s and The probable maximum Hood was 1,637 $m^3$/s. Although the Row rate at the dam site was increased, the Hood risk at the downstream river was decreased by the Hood control of the Donghwa dam.

가스분무성형 Cu-5Ni-10Sn 합금의 미세조직 및 시효강화 (Microstructural Feature and Aging Characteristics of Spray-Formed Cu-5Ni-10Sn Alloy)

  • In this study, Cu-5Ni-10Sn(wt%) spinodal alloy was manufactured by gas atomization spray forming, and the microstructural features and mechanical properties of Cu-5Ni-10Sn alloy have been investigated during homogenization, cold working and age-hardening. The spray formed Cu-5Ni-10Sn alloy consisted of an equiaxed microstructure with a mixture of solid solution ${\alpha}$-(CuNiSn) grains and lamellar-structure grains. Homogenization at $800^{\circ}C$ and subsequent rapid quenching formed a uniform solid solution ${\alpha}$-(CuNiSn) phase. Direct aging at $350^{\circ}C$ from the homogenized Cu-5Ni-10Sn alloy promoted the precipitation of finely distributed ${\gamma}$' or ${\gamma}-(Cu,Ni)_3Sn$ phase throughout the matrix, resulting in a significant increase in microhardness and tensile strength. Cold working prior to aging was effective in strengthening Cu-5Ni-10Sn alloy, which gave rise to a maximum tensile strength of 1165 MPa. Subsequent aging treatment slightly reduced the tensile strength to 1000-1100 MPa due to annealing effects.

An Easy, Rapid, and Cost-Effective Method for DNA Extraction from Various Lichen Taxa and Specimens Suitable for Analysis of Fungal and Algal Strains

  • Lichen studies, including biodiversity, phylogenetic relationships, and conservation concerns require definitive species identification, however many lichens can be challenging to identify at the species level. Molecular techniques have shown efficacy in discriminating among lichen taxa, however, obtaining genomic DNA from herbarium and fresh lichen thalli by conventional methods has been difficult, because lichens contain high proteins, polysaccharides, and other complex compounds in their cell walls. Here we report a rapid, easy, and inexpensive protocol for extracting PCR-quality DNA from various lichen species. This method involves the following two steps: first, cell breakage using a beadbeater; and second, extraction, isolation, and precipitation of genomic DNA. The procedure requires approximately 10 mg of lichen thalli and can be completed within 20 min. The obtained DNAs were of sufficient quality and quantity to amplify the internal transcribed spacer region from the fungal and algal lichen components, as well as to sequence the amplified products. In addition, 26 different lichen taxa were tested, resulting in successful PCR products. The results of this study validated the experimental protocols, and clearly demonstrated the efficacy and value of our KCl extraction method applied in the fungal and algal samples.

Fabrication of 50 to 1000 nm Monodisperse ZnS Colloids

  • Monodisperse ZnS colloidal particles with precisely specified diameters over a broad size range were synthesized by controlled aggregation. Sub-10nm ZnS seed crystals were first nucleated at ambient temperature and then grown at an elevated temperature, which produced large polydisperse colloidal particles. Subsequent rapid thermal quenching and heating processes induced a number of secondary nucleations in addition to growing the large polydisperse microparticles which were finally removed by centrifugation and discarded at the completion of the reaction. The secondary nuclei were then aggregated further at elevated temperatures, resulting in colloidal particles which exhibited a nearly monodisperse size distribution. Particle diameters were controlled over a wide size range from 50 nm to 1 μm. Mie simulations of the experiment extinction spectra determined that the volume fraction of the ZnS is 0.66 in an aggregated colloidal particle and the colloidal particle effective refractive index is approximately 2.0 at 590 nm in water. The surface of the colloidal particles was subsequently coated with silica to produce ZnS@silica core-shell particles.

Improvement of wear resistance of Zircaloy-4 by nitrogen implantation

  • Nitrogen implantation process has been applied for improvement of wear resistance of Z Zircaloy-4 fuel cladding materials. Nitrogen was implanted at 120 ke V to a total do range of 1xHP ions/cm2 to 8xlO17 ions/cm2 at various temperatures of 298"C to 676"C. The m microstructure changes by nitrogen implantation were analyzed by using TEM, XRD 뻐d A AES, cmd then wear behavior was evaluated by ball-on-disc wear testings at various loads a and sliding velocity under unlubricated condition. Nitrogen implantation produced ZrNx nitride above 4.37x1017 ions!cm2 as well as heavy d dislocations, which enhanced microhardness of the implanted surface of up to 900 Hk from 2 200 Hk of unimplanted substrate. Hardness was also found to be increased with increasing i implantation temperature and enhanced up to OOOHk at 620 "C. the wear resistance was g greatly improved with increasing total ion do않 as well as implantation temperature. The effective enhancement of wear resistance at high dose and tem야ratures is believed d due to significant hardening associated with high degree of precipitation of Zr nitrides and g generation of prismatic dislocation I$\infty$ps.infty$ps.

Process Development for the Enzymatic Hydrolysis of Food Protein: Effects of Pre-treatment and Post-treatments on Degree of Hydrolysis and Other Product Characteristics

  • An enzymatic process was developed to produce protein hydrolysater form defatted soya protein. Various unit operations were tried, and the effects of pre- and post-treatments on the product characteristics such as degree of hydroylsis (DH), free amino acid content (%FAA) and average molecular weight (MW) were investigated. The use of acid washes showed no difference in %DH. Increasing pH during pre-cooking gave lower %DH. Alkaline cooking made too much insoluble protein, thus the protein yield was too small. A better hydrolysis with more acceptable taste was obtained when the combination of Neutrase/Alcalase/Flavourzyme was used in place of Alcalase/Flavourzyme combination; Untoasted defatted soya was more effective on the proteolysis than toasted one. The MW of the evaporated and spray dried product was higher than that of undried product, due to precipitation of low-solubility components. When ultrafiltration and the product concentration carried out the product separation by reverse osmosis, the solubility and the taste of the product were improved. The difference between enzyme hydrolysate and acid hydrolysate was significant in free amino acid composition, especially in tyrosine, phenylalanine, glutamine and asparagine.

Formation of Magnetic Graphene Nanosheets for Rapid Enrichment and Separation of Methyl Orange from Water

  • Magnetic-graphene nanosheets have been synthesized via a simple effective chemical precipitation method followed by heat treatment. The composite nanosheets are super paramagnetic at room temperature and can be separated by an external magnetic field. The prepared magnetic-graphene nanosheets were characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM), Raman spectroscopy, and BET surface area analysis. The results demonstrated the successful attachment of iron oxide nanoparticles to graphene nanosheets. It was found that the attached nanoparticles were mainly $Fe_3O_4$. The magnetic-graphene nanosheets showed near complete methyl orange removal within 10 mintues and would be practically usable for methyl orange separation from water.
