Antimicrobial Activities of White, Red, and Extruded Ginsengs with Different Extraction Conditions

  • White, red, and extruded ginsengs were studied against 8 strains of food-borne pathogens and/or food spoilage microorganisms. The ginseng powders were extracted with different extractants and screened for antimicrobial activity using the disc diffusion and broth dilution techniques. The results showed that the yield of extraction was higher with increase of aqueous solution content and temperature. Preliminary screening revealed that the red ginseng extracts were most active, that has been found to be highly effective against all tested microbe except Listeria monocytogenes. Moreover, Bacillus subtilis has shown highly susceptible, which the diameters of inhibition zone values of 28 extracts were between 7 and 14 mm. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) recorded for the different crude ginseng extracts against microorganism using ranged from 6.25 to 100 mg/mL, indicated that the methanol extract of ginseng were more effective than ethanol and water extracts. The 60% methanol extract of red ginseng had the greatest effects against B. subtilis with MIC and MBC values at 6.25 mg/mL.

Effects of Special Mouth Care with an Aroma Solution on Oral Status and Oral Cavity Microorganism Growth in Elderly Stroke Patients (아로마 용액을 이용한 구강간호가 뇌졸중 노인의 구강상태와 구강 세균 집락에 미치는 효과)

  • Purpose: This study was conducted to examine the effect of oral care with an aroma solution on oral status and oral cavity microorganism growth in elderly patients with stroke. Methods: A non-equivalent control group, with a pretest-posttest design was used in this study. The participants were assigned to the experimental group (n=30) that received oral care with an aroma solution or the control group (n=31) that received 0.9% saline solution. To identify the effect of the experimental treatments, objective/subjective assessments of oral status and oral cavity microorganism growth were performed using the oral assessment guide, oral perception guide, and oral swab culture. Data were analyzed using Chi-square test, Fisher's exact test, and t-test with the SPSS version 21.0 program. Results: The objective oral status was significantly lower in the experimental group than in the control group (t= -3.64, p<.001). There was no significant difference between the subjective oral status of the experimental group and control groups (t= -1.24, p=.109). Oral microorganism growth was significantly lower in the experimental group than in the control group (t= -7.39, p<.001). Conclusion: These findings indicate that special mouth care using an aroma solution could be an effective oral health nursing intervention for elderly patients with stroke.

Cloning and Characterization of a Gene Encoding $\gamma-Butyrolactone$ Autoregulator Receptor from Saccharopolyspora erythraea

  • A gene encoding a $\gamma-butyrolactone$ autoregulator receptor was cloned from Saccharopolyspora erythraea, and the biochemical characteristics, including the autoregulator specificity, were determined with the purified recombinant protein. Using primers designed for the conserved amino acid sequence of Streptomyces $\gamma-butyrolactone$ autoregulator receptors, a 120 bp S. erythraea DNA fragment was obtained by PCR. Southern and colony hybridization with the 120 bp fragment as a probe allowed to select a genomic clone of S. erythraea, pESG, harboring a 3.2 kb SacI fragment. Nucleotide sequencing analysis revealed a 615 bp open reading frame (ORF), showing moderate homology (identity, $31-34\%$; similarity, $45-47\%$) with the $\gamma-butyrolactone$ autoregulator receptors from Streptomyces sp., and this ORF was named seaR (Saccharopolyspora erythraea autoregulator receptor). The seaR/pET-3d plasmid was constructed to overexpress the recombinant SeaR protein (rSeaR) in Escherichia coli, and the rSeaR protein was purified to homogeneity by DEAE-Sephacel column chromatography, followed by DEAE-ion-exchange HPLC. The molecular mass of the purified rSeaR protein was 52 kDa by HPLC gel-filtration chromatography and 27 kDa by SDS-polyacrylamide gel electrophoresis, indicating that the rSeaR protein is present as a dimer. A binding assay with tritium-labeled autoregulators revealed that rSeaR has clear binding activity with a VB-C-type autoregulator as the most effective ligand, demonstrating for the first time that the erythromycin producer S. erythraea possesses a gene for the $\gamma-butyrolactone$autoregulator receptor.

Purification Technology in Closed Water like a Reservoir and Pond using Oxygen Solubilized Device and Standardized Microorganism Culture System (산소용해수와 미생물제재를 이용한 호소 및 폐쇄수역의 정화기술)

  • The oxygen solubilized device(O.S.D) and standardized microorganism culture system is more efficient than physical and chemical purification techniques in closed water. This study was to determine how the O.S.D and standardized culture system is efficient in purification capacity in closed water based on the lab scale and pilot plant. In the batch test, inducing the quantitative results from pilot plant operation condition, removal efficiency of COD and TN were about 48.3% and 35% respectively, while SS and chlorophyll-a were 94.9% and 68.7%. The pilot plant results showed that suspended solid(SS) and chlorophyll-a removal efficiency were 60% and 59% respectively, due to coagulation characteristics by standardized culture. Total nitrogen(TN) and total phosphorus(TP)showed good effect for the purification of target pond water quality from field data. Additionally, released velocity was determined in control condition of $5.31mgPO{_4}^{3-}{\cdot}m^{-2}{\cdot}day^{-1}$ and $2486.8mgCOD{\cdot}m^{-2}{\cdot}day^{-1}$. Otherwise, phosphate and COD reflux in the aeration and microorganism condition was showed $-9.95mgPO{_4}^{3-}{\cdot}m^{-2}{\cdot}day^{-1}$ and $-397.88mgCOD{\cdot}m^{-2}{\cdot}day^{-1}$. This technology is the most effective not only removal of SS and chlorophyll-a but also control of phosphate and COD release which is very important phenomena in evaluating water quality in closed water like a reservoir and pond.

Growth Characteristics of Microorganism on Lapilli-Cement mixed Bioblocks (화산력-시멘트 혼합 바이오 블록의 미생물 생장 특성)

  • This research aims to utilize lapilli from the Mt. Baekdusan as environmently-friendly construction material. First of all, the neutralizing method for fabricating lapilli-cement-mixed bioblock was examined. And then, by use of the neuralized bioblock with microorganism for water purification, the growth inhibition effect against the pathogenic coliform bacillus was evaluated. The result regarding growth inhibition effect on pathogenic coliform bacillus indicates that the pretreatment condition, which is a concurrent procession with aqueous solution of 10% di-ammonium Phosphate after water curing, led to pH degraded below 10 which was the target value. Therefore it was concluded that the method was effective on bio-block neutralization. The microorganisms purifying water and di-ammonium phosphate were detected through the examination for microorganism existence on the bioblock, therefore it was concluded that the bioblock composed of lapilli and cement is able to be utilized in various structures as an environment friendly construction material.

Effect of Microbial Product on Microorganisms in Soil and the Growth of Leaf Lettuce (EM 활성액, 키토산 및 목초액 처리가 토양 미생물상의 변화 및 잎 상추의 생육에 미치는 영향)

  • This study was conducted to investigate the effect of microbial product on microorganisms in soil and the growth of leaf lettuce. The test material were treated with chitosan, wood vinegar and EM activity liquid, and treatment concentration was 50 times solution and 100 times solution level with foliar application. The results were summarized as follows : Among foliar application of microorganisms treatments diluted by chitosan 100 times solution level was effective considering growth of leaf lettuce as compared other dilutions and control plot. Change of microorganism number in the soil for cultivation of chinese cabbages and leaf lettuce was increased with microorganism treatment plot as compared with control plot. Specially chitosan 100 times solution level showes the most significant effect.

  • With treatment of Kitosan, Wood vinegear and EM(effective microoganism) which farmers call it as substance in fertilizing, conditioning and disease control substances, authors in vestigated on microorganisms change in soil and ealy growth characteristics of tomato. The results were summarized as follows: Among foliar application of kitosan, wood vinegear and EM(effective microoganism) treatments diluted by chitosan 500 times solution level was effective considering growth of tomato as compared other dilutions and control plot. Change of microorganism number in the soil for cultivation of tomato was increased with microorganism treatment plot as compared with control plot. Specially chitosan 500 times solution level showes the most significant effect.

Field Applicability Evaluation Using Effective Microorganism Brewing Cycle for Contaminated Soil in Water Retention Basin (복합발효미생물을 이용한 하천유수지 오염토의 현장적용성 평가)

  • In this study, by using a Effective Microorganisms Brewing Cycle, it confirmed the purification effect of pollutants that are adsorbed on the basins stench removal and retarding soil. On the basis of on-site application test, a soil decontamination system will be suggested. Using a Effective Microorganisms Brewing Cycle, the odor concentration is reduced 2.5 times than that of natural purification treatment method. It was measured and found that the quality of the pore water discharged from the soil is improved. In addition, it was found that a composite of copper and lead with the fermentation microorganisms adsorbed on soil particles from the surface of the stirred experiments lagoon mixed soil is reduced to 65% and 66%, respectively, The TPH organic component was confirmed that the reduction effect of 85%. Restoration of reservoir contaminated soils using the effective microorganism brewing cycle needs to be more developed and implemented as a long-term purification system. This study may be a good reference of developing more complete microorganism brewing system which will efficiently reduce the odor and soil contamination based on optimal stirring and mixing ratio of the compound solutions and contaminated soils in reservoir.

Anti-inflammatory Effects of Effective Microorganism Fermentation Substance on Atopic Dermatitis-like NC/Nga Mouse Model (아토피 피부염 유사 NC/Nga 마우스 모델에서 유용 미생물 발효물질의 항염증효과)

  • Atopic dermatitis (AD) is a chronically relapsing pruritic inflammatory skin disease. To find new anti-inflammatory products for skin inflammatory disease such as AD and contact dermatitis, we produced the effective microorganism fermentation substance (EM-S) by fermentation of medicinal plants with effective microorganisms including photosynthetic bacteria, lactic acid bacteria and yeast, screened the effects of EM-S on NC/Nga model mice. Murine AD-like skin lesions were made by painting Dermatophagoides farinae (Df) extract. Topically applied EM-S significantly reduced clinical severity score, ear thickness and histological grade in AD-like NC/Nga mouse model by Df antigen sensitization. In addition, the serum IgE and Th2 chemokine levels (TARC/CCL17, MDC/CCL22 and CTACK/CCL27) were significantly reduced by EM-S. Futhermore, skin tissue expressions of Th2 chemokines were significantly reduced by EM-S. These results demonstrate that topical application of EM-S may be improve the AD-like skin lesion by suppressing IgE and Th2 chemokines.

Role of oyster mushroom as an alternative fermentation source in the growth of white-spotted flower chafer(Protaetia brevitarsis seulensis) (사료 발효원으로 느타리버섯 종균이 흰점박이꽃무지 생육에 미치는 영향)

  • Effective microorganisms (EM), which are sources of fermentation byproducts in herbal medicine, were compared with oyster mushrooms(OM) to identify alternative larval feeds for white-spotted flower chafer (Protaetia brevitarsis seulensis). The nutritional composition of each fermented feed was analyzed. The content of crude protein, crude fat, and fiber was higher in the OM fermented feed, except for crude ash. No difference in nutritional content of larvae based on fermented feed was observed. A comparative weekly analysis of the live weights of larvae was based on the fermented feed used. The average weight was significantly higher in the feeds using EM and OM from third week of observation. The larval survival rate in the presence of fermented feed was 96.7% compared with 9.8% with non-fermented feed. The results suggest that fermented food source is essential for the growth of white-spotted flower chafer, and OM was a stable alternative to EM as a fermentation source for the survival of white-spotted flower chafer.
