Evaluation of Different Culture Conditions of Clostridium bifermentans DPH-1 for Cost Effective PCE Degradation

  • Clostridium bifermentans strain DPH-1 has already been found to dechlorinate perchloroethylene (PCE) to cis-dichloroethylene (cis-DCE) via trichloroethylene (TCE). In this study, our investigation on different culture conditions of this DPH-1 strain was extended to find a more efficient and cost effective growth medium composition for this DPH-1 strain in bioremediation practices. Temperature dependency of strain DPH-1 showed that the growth starting time and PCE degradation at $15^{\circ}C$ was very slow compared to that of $30^{\circ}C$, but complete PCE degradation occurred in both cases. For the proper utilization of strain DPH-1 in more cost effective bioremediation practices, a simpler composition of an effective media was studied. One component of the culture medium, yeast extract, had been substituted by molasses, which served as a good source of electron donor. The DPH-1 strain in the medium containing molasses, in the presence of $K_{2}HPO_4\;and\;KH_{2}PO_4$, showed identical bacterial multiplication (0.135 mg protein $mL^{-1}h^{-1}$) and PCE degradation rates ($0.38\;{\mu}M/h$) to those of the yeast extract containing medium.

장구균 검출 배지 개발 (Development of selective media for Enterococci)

  • 분변오염의 지표가 되는 장구균을 검출하기 위하여 장구균 선택 배지를 개발하였다. 기존에 알려진 여러 장구균 검출배지 중 총 9종류(Enterococci Confirmatory agar, Azide dextrose agar, Bromocresol-purple azide agar, Esculin bile agar, Citrate azide tween carbonate agar, KF Streptococcus agar, BROLACIN agar, Kanamycin esculin azide agar, Membrane filter Enterococcus selective agar)의 배지를 선별하였고, 배지 성분을 적절히 조합하여 새로운 배지 Enterococcus Mixed medium을 개발하였다. 배지의 효과를 검증하기 위하여 장구균 6종류(Enterococcus avium KACC 10788, Enterococcus faecium KACC 11954, Enterococcus saccharolyticus KACC 10783, Enterococcus durans KACC 10787, Enterococcus faecalis KACC 11304, Enterococcus hirae KACC 10779)와 비장구균 3종류(Escherichia coli KACC 10005, Staphylococcus aureus subsp. aureus KACC 10768, Bacillus subtilis KACC 10111)를 사용하였다. 그 결과 새롭게 개발된 장구균 검출배지에서 비장구균의 생장이 억제된 반면에 장구균 6종류는 모두 생장하는 것을 확인하였다. 또 환경시료에서 장구균을 분리할 수 있는지 확인하기 위해 하천과 바다에서 채취해온 시료를 이용하여 실험한 결과 장구균의 생장을 확인하였다. 따라서 이 배지를 이용하면 수질 분변오염의 지표가 되는 장구균을 효과적으로 검출할 수 있음을 확인하였다.

하수중의 Salmonella spp. 분리방법에 관한 비교연구 (A Study on the Salmonella spp. isolation method in the sewage specimens)

  • This study was carried out in order to compare the methods for Salmonella spp. isolation from sewage and to settle the most appropriate conditions for that isolation. The direct enrichment method was more effective than the pre- enrichment method for Salmonella spp. isolation. The rate of detection was much higher when the specimens were enriched at $41.5^{\circ}C$ than when at $35^{\circ}C$. Usage of XLBG agar medium showed better results for Salmonella isolation than that of SS medium. It can be suggested that the most effective combination for Salmonella spp. isolation was the direct centrifugation(3,000 rpm 100m1)- direct enrichment($41.5^{\circ}$C)-usage of XLBG medium.

Trichoderma koningii의 conidiospore로부터의 원형질체 분리에 관하여 (Isolation of protoplast from conidiospore of Trichoderma koningii)

  • Conditions for isolation of protoplasts from conidiospores of Trichoderma koningii ATCC 26113 were tested. Maximum production of conidial protoplasts was obtained by preincubation of conidiospores on liquid minimal medium for 8 1/2 hrs. and by reaction with cell wall lytic enzyme for 3 hrs. Among effective cell wall lytic enzymes (Driselase, p-Glucuronidase, Novozyme and Zymolyase), Driselase was the most effective one on the production of conidial protoplasts. The production of conidial protoplasts was also enhanced by addition of 2-Deoxy-D-Glucose $(25{\mu}g/ml)$ into liquid minimal medium. Over 70% of the initial swollen conidia, preincubated in liquid minimal medium supplemented with 2-Deoxy-D-Glucose $(25{\mu}g/ml)$, were converted to protoplasts by incubation with 2% (w/v) commercial lytic enzyme Driselase at $28^{\circ}C$ for 3 hrs. The reversion frequency of the conidial protoplasts was about 30 times (25-50%) higher than that of mycelial protoplasts (0.6-1.3%).

Synthesis of Bulk Medium with Negative Permeability Using Ring Resonators

  • This paper presents simple expressions for the effective permeability of bulk metamaterial consisting of ring resonators (RRs) or split ring resonators (SRRs) based on the convenient geometrical factors of the structure compared with wavelength. The resonant frequency dependence of the medium permeability, including loss effects, is analyzed in detail. Inverting the analysis equations, useful design (or synthesis) equations are derived for a systematic design process with some examples. This paper may particularly be useful for the design of a bulk metamaterial with a specific negative relative permeability at a desired frequency. The loss of metamaterials consisting of RRs (or SRRs) is also analyzed over a wide frequency band from 10 MHz to 10 THz.

  • The importance of reliability information management for an effective product quality management has been increased. Reliability management requires many different kinds of human and technical expertise and cost over the various discipline in the organization. However, it is difficult task to set up appropriate and effective life data analysis because of lacking engineers with reliability knowledge, statistical background and of funds in small and medium-sized enterprise. It is needed to develop a low cost implementation architecture of reliability information supporting system for small and medium-sized enterprise under web environment. In this paper, research efforts are presented to solve such problems of small and medium-sized enterprise under web environment.

Estimation of the effective dose of dental cone-beam computed tomography using personal computer-based Monte Carlo software

  • Purpose: To calculate the effective doses of cone-beam computed tomography (CBCT) using personal computer-based Monte Carlo (PCXMC) software (Radiation and Nuclear Safety Authority, Helsinki, Finland) and to compare the calculated effective doses with those measured using thermoluminescent dosimeters (TLDs) and an anthropomorphic phantom. Materials and Methods: An Alphard VEGA CBCT scanner (Asahi Roentgen Ind. Co., Kyoto, Japan) with multiple fields of view (FOVs) was used for this study. The effective doses of the scout and main projections of CBCT using 1 large and 2 medium FOVs with a height >10 cm were calculated using PCXMC and PCXMCRotation software and then were compared with the doses obtained using TLD-100 LiF and an anthropomorphic adult human male phantom. Furthermore, it was described how to determine the reference points on the Y- and Z-axes in PCXMC, the important dose-determining factors in this software. Results: The effective doses at CBCT for 1 large ($20.0cm{\times}17.9cm$) and 2 medium FOVs ($15.4cm{\times}15.4cm$ and $10.2cm{\times}10.2cm$) calculated by the PCXMC software were 181, 300, and $158{\mu}Sv$, respectively. These values were comparable (16%-18% smaller) to those obtained through TLD measurements in each mode. Conclusion: The use of PCXMC software could be an alternative to the TLD measurement method for effective dose estimation in CBCT with large and medium FOVs.

Prediction of nonresponsiveness to mediumdose intravenous immunoglobulin (1 g/kg) treatment: an effective and safe schedule of acute treatment for Kawasaki disease

  • Purpose: Medium-dose (1 g/kg) intravenous immunoglobulin (IVIG) is effective in the majority of patients with Kawasaki disease (KD) but some patients who do not respond to medium-dose IVIG are at high risk for the development of coronary artery lesions (CALs). The purpose of this study was to identify the clinical predictors associated with unresponsiveness to medium-dose IVIG and the development of CALs. Methods: A retrospective study was performed in 91 children with KD who were treated with mediumdose IVIG at our institution from January 2004 to December 2013. We classified the patients into responders (group 1; n=68) and nonresponders (group 2; n=23). We compared demographic, laboratory, and echocardiographic data between the 2 groups. Results: Multivariate logistic regression analysis identified 6 variables as predictors for resistance to medium-dose IVIG. We generated a predictive scoring system assigning 1 point each for percentage of neutrophils ${\geq}65%$, C-reactive protein ${\geq}100mg/L$, aspartate aminotransferase ${\geq}100IU/L$, and alanine aminotransferase ${\geq}100IU/L$, as well as 2 points for less than 5 days of illness, and serum sodium level ${\leq}136mmol/L$. Using a cutoff point of ${\geq}4$ with this scoring system, we could predict nonresponsiveness to medium-dose IVIG with 74% sensitivity and 71% specificity. Conclusion: If a patient has a low-risk score in this system, medium-dose IVIG can be recommended as the initial treatment. Through this process, we can minimize the adverse effects of high-dose IVIG and incidence of CALs.

Development of a Selective Medium for Surveillance of Fusarium Head Blight Disease

  • Fusarium head blight (FHB), predominantly caused by Fusarium graminearum and F. asiaticum, is a significant fungal disease impacting small-grain cereals. The absence of highly resistant cultivars underscores the need for vigilant FHB surveillance to mitigate its detrimental effects. In 2023, a notable FHB outbreak occurred in the southern region of Korea. We assessed FHB disease severity by quantifying infected spikelets and grains. Isolating fungal pathogens from infected samples often encounters interference from various microorganisms. We developed a cost-effective, selective medium, named BGT (Burkholderia glumae Toxoflavin) medium, utilizing B. glumae, which is primarily known for causing bacterial panicle blight in rice. This medium exhibited selective growth properties, predominantly supporting Fusarium spp., while substantially inhibiting the growth of other fungi. Using the BGT medium, we isolated F. graminearum and F. asiaticum from infected wheat and barley samples across Korea. To further streamline the process, we used a direct PCR approach to amplify the translation elongation factor 1-α (TEF-1α) region without a separate genomic DNA extraction step. Phylogenetic analysis of the TEF-1α region revealed that the majority of the isolates were identified as F. asiaticum. Our results demonstrate that BGT medium is an effective tool for FHB diagnosis and Fusarium strain isolation.

Hohenbuehelia petaloides의 배양학적 특성에 관한 연구 (Studies on the Cultural Characteristics of Hohenbuehelia petaloides)

  • This study was carried out to obtain the basic data on artificial culture of Hohenbuehelia petaloides. The optimum medium are glucose peptone medium (GP), Hennerberg medium, Phellinus igniarius medium (PIM), Lentinus edodes medium (LEM), Czapek dox medium. The optimum condition for the mycelial growth was $30^{\circ}C$ and pH 6.0. The carbon sources such as dextrine, fructose and lactose were favorable to mycelial growth. The optimal concentrations of carbon sources are 10% dextrin and fructose. As nitrogen sources, tryptone, casamino acid and histidine appeared to be favorable. The optimal concentrations of nitrogen sources are 1% soy tone and 0.3% ammonium nitrate. The optimal concentration of yeast extract is 0.4%. The mineral nutrients of $KH_2PO_4$, $K_2HPO_4\;and\;MgSO_4{\cdot}7H_2O$ were effective and the optimal concentrations were 0.046, 0.1 and 0.05%, respectively.
