Plant Regeneration from Immature Embryo and Bulb Scale Tissue of Hippeastrum hybridum (아마릴리스의 미숙배와 인편조직으로부터 식물체 재분화)

  • Immature hybrid embryos of H. hybridum, 'Picottee', 'White Christmas', 'Eldorado', 'Origin', 'Red Lion', 'elstar', 'Crypsy' were cultured on the MS medium supplemented with various concentrations of 2,4-D, NAA, BA and TDZ. Among the treatments, NAA were more effective for the shoot regeneration and bulblet formation than other treatment. Addition of 0.5 ㎎/L NAA was effective for bulblet induction from explant Shoot regeneration was most effective on the medium with 1.0㎎/L NAA and 2.0 ㎎/L TDZ. The addition of 1.0-2.0㎎/L TDZ induced numerous shoots per explant but strongly inhibited root development when compared to 1.0-2.0㎎/L BA. When bulb scale segments of 'Star Van Holland' was incubated, bulblet formation was the most effective on MS medium with 0.5㎎/L NAA.

Effect of Polyamines, Salt Strength, Sucrose, and Gelling Agents on plant Regeneration from Meristem Culture of Aloe spp. (알로에 생장점 배양시 식물체 재분화에 미치는 Polyamine, 염류농도, 당 및 Gelling Agent의 효과)

  • This study was carried out to investigate the effect of polyamines, salt strength. sucrose and gelling agents on the regeneration of plantlets by meristem culture of Aloe arborescens Mill. and Aloe vera L.. Shoot multiplication was more effective when 10mg/ l spermine in Aloe arborescens and 1mg/ l spermidine in Aloe vera added into MS medium than when other polyamines were treated into media. A quarter strength of MS medium was effective for rooting of shoots regenerated. Higher concentration of sucrose (45g/ l) was more effective for shoot regeneration. Addition of 4g/ l gelrite into the medium was effective for induction of multiple shoots from Aloe than that of agar or other concentrations of gelrite. When plantlets regenerated from meristem culture were transferred to pot. survival rate of plantlets was 80% on perlite and was 95% on vermiculite. respectively.

Effects of Cold Pretreatment and Medium Composition on Anther Culture Initiation in Strawberry

  • Callus culture initiation of strawberry (Fragaria${\times}$ananassa Duch.) was investigated at different Murashige and Skoog (MS) medium strengths, types and concentrations of plant growth regulators, and incorporating a cold pretreatment period to determine the optimal nutritional and environmental conditions. No high quality callus was induced on MS media without auxin regardless of medium strength. When 6-benzylaminopurine (BA) was combined with indole acetic acid (IAA), naphthalene acetic acid (NAA), and 2,4-dichlorophenoxyacetic acid (2,4-D), high quality callus were highly induced compared to medium supplemented with auxin alone. When $0.5mg{\cdot}L^{-1}$ BA was combined with IAA, NAA, and 2,4-D, high quality callus induction was more effective than the medium supplemented with the other BA concentrations. The best combination of auxin and cytokinin for high quality callus induction was $1.0mg{\cdot}L^{-1}$ NAA and $0.5mg{\cdot}L^{-1}$ BA. Although the differences in callus induction were not significant, high quality callus induction at half strength MS medium was more effective than at full strength medium. When $30g{\cdot}L^{-1}$ sucrose was added to the half strength MS medium, the rate of high quality callus induction increased. The optimum cold pretreatment temperature and period for high quality callus induction were $4^{\circ}C$ and 72 h, respectively. Regeneration rate of high quality callus increased in MS medium supplemented with thidiazuron.

  • Multiple shoots obtained in MS medium suppler with 5.0 mg/L BA though shoot-tip culture. The frequency of vitrified shoot was lower on Bacto-agar medium than on Gelrite as gelling agent. Addition of activated charcoal at concentrations of 0.1~0.3% reduced vitrification and markedly increased shoot growth, and formation and growth of roots, but significantly reduced the number of shoots formed. The ratio of fresh weight to dry weight was decreased by increasing light intensity and agar concentration. Eight-tenths times of macroelement of MS medium was observed to be effective for shoot formation. Addition of IAA effectively promoted shoot formation in both shoot tip and node-bud explants. Supplement of 5.0 mg/L BA, 0.3 mg/L IAA to MS medium was most effective in shoot proliferation on shoot tip and node-bud explants.Multiple shoots obtained in MS medium suppler with 5.0 mg/L BA though shoot-tip culture. Effective Medium Theory for Particulate Composite Materials Carrying Elastic Waves

  • In this paper, two major theories of effective medium for composite materials are studied, which provide the average dynamic mechanical properties of particulate composites carrying elastic waves on the basis of the concept of self-consistent scattering of waves. These two theories are also compared in detail with each other to delineate the difference in the underlying ideas and their limitation of applicability. Furthermore, prospective directions for potential improvement of the theories are found. Numerical results for three particulate composites are given and discussed.

  • This experiment was conducted to determine factors affecting callus formation of some varieties of red clover. Growth regulators. basal medium, medium pH, agar concentration, and plant age for explant were investigated. The result obtained were as follows. As auxin source, 0.1 mg/ l picloram was found to be effective on callus formation. Callus formation was the highest on PC medium among several growth media. The callus produced on PC medium was more friable than those produced on the other media. The medium pH 5.8 gave the best response for callus formation. The effective seedling age for callus formatiom was around 5-7 days old. In agar concentration, 0.5-0.8%(WN) was suitable for callus formation.

Effects of Composition and Concentration of Media on the Acclimation of the Shoots of Scrophularia buergeriana Miquel Produced in Bioreactor (생물반응기 생산 현삼 신초의 순화에 미치는 배지의 영향)

  • This study was carried out to investigate the optimum composition and concentration of medium for the acclimation of the shoots derived from bioreactor culture of Scrophularia buergeriana. Rooting and growth of the shoots cultured in bioreactor for 4 weeks were better on liquid full strength MS medium while one-tenth of MS medium was good for rooting and growth of the shoots cultured in bioreactor for 6 weeks. Leaf expansion was more effective in the shoots cultured for 6 weeks on solid MS medium than in those cultured for 4 weeks. No leaf expanded in liquid medium. Vitrification rate was higher in full strength solid MS medium than in half strength MS medium. It is concluded that acclimation of the shoots is more effective on the half strength solid MS medium containing 1.2% agar.

  • Six Sigma Management has been introduced as a management strategy by leading companies and some small and medium-sized companies in Korea. But it is not easy for small and medium-sized companies to implement this new management system because of their business environment such as poor manpower or lack of time, etc. This paper proposes an effective implementation model of the Six Sigma Management for small and medium-sized companies using the management by process. First, We review the concept of Six Sigma Management and consider the problems of introducing the system for small and medium-sized companies. And then, Six Sigma Management model for small and medium-sized companies named SIX SIGMA-s is presented. Finally, we present a case study of applying SIX SIGMA-s for a medium- sized company.

Effects of Medium Components and Growth Regulators on Callus Development and Shoot Regeneration from Shoot Explants of Black Locust (Robinia pseudoacacia)

  • Effects of growth regulators and medium components were tested for shoot multiplication and callus growth from shoot explants of black locust. During shoot multiplication, callus growth at the cut end of shoot explants proceeded shoot development. The basal callus growth seemed to be a function of both mineral components and cytokinin supplemented in the medium. Maximum callus growth was induced by 0.5${\mu}$M BAP and the callus growth decreased as the level of BAP increased. Positive correlations were found between basal callus growth, and shoot multiplication and growth. Shoot multiplication was greatest on BSM medium (black locust shoot culture medium) supplemented with 1 $\mu$M BAP. With medium containing high nitrogen content, both shoot multiplication and growth were significantly enhanced. A new BRM medium was the most effective for rooting of black locust among three rooting media tested.

Effect of Germanium Treatment in Culture Medium on Germanium Absorption by Callus Induced from Brown Rice (배지내 게르마늄 처리가 현미 유도 캘러스의 게르마늄 흡수에 미치는 영향)

  • This study was carried out to investigate the effect of Germaniwn(Ge) treatment in the culture media on the Ge absorption by the callus induced from brown rice cv. Dongjinbyeo. MS medium was more effective on the growth ratio of callus and the content of Ge and some inorganic elements except K in callus than N$_{6}$ medium. The more Ge treatment in the N$_{6}$ or MS medium, the more Ge absorption by the callus, but the growth ratio of callus and the content of Ca, Mg, and K in callus were decreased. The content of Fe, Mn, Zn, and Cu was increased under treatment up to 100~200mg /$\ell$ Ge, but tended to be decreased under treatment more than that of Ge concentration. Under treatment less than 150mg /$\ell$ Ge, GeO$_2$(inorganic Ge) was more effective on the Ge absorption by callus than Ge-132(organic Ge), but Ge-132 was more effective on the Ge ab-sorption by callus and the activity of callus in case of treatment more than 150mg /$\ell$ Ge. The lower pH of culture medium, the higher Ge content in the callus. When callus was cultured on medium supplemented with Ge and 0.1~1.0mM of citric acid or myo-inositol, content of Ge and some inorganic elements in callus, as well as growth and dry weight of callus, were tend to increase in comparison to control, but myo-inositol was more effective on them than citric acid.cid.
