혼합세포를 이용한 Aerated Slurry Bioreactor에서의 in vitro Glutathione 생산 (In vitro Glutathione Production using Mixed Cells in an Aerated Slurry Bioreactor)

  • E. coli TGI/pDG7 $\alpha$ 와 빵효모의 혼합세포계에 의한 글루타치온 생산을 Aerated Slurry Bioreactor에서 수행하였다. 글루타치온 생산 효소는 34시간 동안 안정성을 보였으며, 글루타치온 수율은 4.6 mM을 유지함을 보였다. 고농도의 혼합세포를 이용한 Aerated Slurry Bioreactor에서 기질의 공급속도가 글루타치온 생산에 미치는 영향을 조사하였다. 세포가 회수되어 기질공급속도 5.2 mL/hr에서 글루타치온의 생산농도는 32시간에서 현탁반응보다 낮아짐을 보였고, 반면 세포가 회수되지 않은 기질공급속도 5.2 mL/hr에서는 22시간 동안 유지되었고 2.6 mL/hr에서는 42시간 동안 유지되었다. 글루타치온의 생산성은 기질공급속도가 10.4 mL/hr에서 25.7 mg/g wet $cell{\cdot}hr$이었으나, 2.6 mL/hr에서는 1.65 mg/g wet $cell{\cdot}hr$이었고 5.2mL/hr에서는 5.28 mg/g wet $cell{\cdot}hr$이었다. 계면활성제(Tween 80)을 사용한 경우, 글루타치온 생산시간을 25시간에서 45시간으로 연장되었다. 글루타치온 생산있어 산소가 세포계의 효소에 산소에 의해 영향을 미칠 것으로 판단되었고, 공기를 질소로 교체하여 글루타치온 생산시간을 40시간으로 연장시켰다.

Beneficial Effects of Dietary Antiacarcinogenic Conjugated Linoleic Acid(CLA) on the Performances of Laying Hens and Broilers

  • Effects of conjugated linoleic acid(CLA), known as an effective anticarcinogen in several aminal models, on the egg production and egg weight of laying hens, and the weight gains of broilers were investigated. CLA was synthesized from corn oil by the alkaline isomerization method and purified by the low-temperature precipitation method. Diets for laying hens and for broilers were synthesized to meet the specification of their NRC standard rationals. Two separated experiments(Experiment I and II) were conducted for laying hens. in experiment I, 45 hens(300 days of age) were divided into 15 hens per treatment group; each hen was housed in wired cage located in a temperature and humidity-controlled house and adopted to the control diet. One week later, each group was subjected to one the four treatment groups for 5 weeks : control, 1.0% CLA, 2.5% CLA and 5.0% CLA diets. Diet and water were ad libitum. The condition of experiment II was the same as that of experiment I except for the addition of 5% corn oil diet and the extension of feeding period to 7 weeks. Egg production, egg weight and feed intake were recorded every week. Forty-five broilers(10day of age) were adopted to the control diet for a week and then switched to the treatment diets for 5 weeks : control, 1.0% CLA, 2.5% CLA, 5% CLA and 5% corn oil, Body weight and feed intake of broilers were measured every week. Diets supplemented with various amounts of CLA enhanced the egg production and increased the egg weight regardless laying hen's age(150 days or 300 days) as compared to control diet. The most effective diet for the egg production and egg weight of young hens(150 days of age) was found to be 1.0% CLA diet, but relatively higher CLA diet(2.5% CLA) was required for old hens (300 day of age) to obtain similar results as seen in younger hens. All hens treated with CLA ate greater amount of feed than control hens. Broilers treated with various amount of CLA ate less feed as compared to control ones, but the body weight gain was greater than the control broilers. These results indicate that CLA enhanced the egg production and agg weight of laying hens, and increased the body weight gain of broilers with less diet consumption.

  • Most high efficiency silicon solar cells use a passivated selective emitter. It have been an important research subject for crystalline silicon solar cells for decades. It is being used in production for high efficiency solar cells. Most of the selective emitter process require expensive extra masking, etching steps, and a double diffusion process making selective emitters not cost effective. In this paper, we study method for single diffusion step selective emitter process as an alternative to not cost effective double diffusion process. Cost effective selective emitter that the efficiency should be increased significantly (mare than 0.2%) and that the process should simple, robust and cheap.

  • In the hot forging process, The forging defects that are caused by metal were strain, temperate, and inclusion. In this paper, the computer simulation analyzed the effective plastic strain and temperature behaviors. The quantitative analyses which proposed the effective mold design of S/CAM shaft was executed. The parameters of forging shape that affected on the optimize conditions that was calculated with simple equation were investigated. it is expected that the developed analysis model and design technique would greatly contribute to the drum brake optimal design considering temperature affected and material behaviors. This development could save more than 20% of production cost and reduced failure rate to more than 30%. By improving the life span of mold from 15,000 to 25,000, financial difficulty of company imposed on a mold manufacture could be overcome.

저비용 응집제를 이용한 미세조류 응집 효율 비교 (A Comparative Study on Microalgae Recovery Rates in Response to Different Low Cost Bio-flocculant Applications)

  • In this study, low cost bio-flocculants, chitosan, cationic starch and Mg-sericite, were used as a flocculant to harvest freshwater microalgae, Chlorella vulgaris. Chitosan, cationic starch and Mg-sericite separated successfully >98% of C. vulgaris at following optimal parameters: 90 mg/L chitosan at pH 6-7, 70 mg/L cationic starch at pH 9-10 and 50 mg/L Mg-sericite at pH 4-5. A relatively high correlation coefficient (R2) of 0.9993 for chitosan, 0.9971 for catonic starch and 0.9924 for Mg-sericite was obtained. The investigated flocculants amount increased linearly with increasing the microalgae amount. The biopolymer, Mg-sericite, was more effective than that of other investigated flocculants. These results indicated that a bio-flocculants, chitosan, cationic starch and Mg-sericite, could prove to be an effective flocculant for economical production of microalgae biomass. In addition, Mg-sericite was more effective comparing to the other investigated flocculants.

  • Studies have shown that the future of the wine market rests on the effective and efficient changes in technology to the supply chain used by most of the major global players. In today's wine industry, companies are faced with the ever-shifting demand for their products, strict regulation and increasing price competition. Even at that, mature companies in the wine industry are succeeding by scaling up production, streamlining their supply chains, expanding into new geographic areas, implementing more efficient processes, cleverly marketing products, and focusing on ever closer relationships with suppliers, partners and customers. However, this paper looks at supply chain challenges in the wine industry from a global perspective presented in the inbound, manufacturing and outbound processes as well as offer effective solutions in order for companies to gain a competitive advantage and succeed on a global level.

Extracellular Proteinase를 생산하는 효모의 분리동정과 효소의 생산

  • A yeast strain TH65 producing a high level of proteinase under alkaline condition was isolated, and identified as Yarrowia lipolytica by morphological, physiological, and biochemical characteristics. In proteinase productivity, glycerol and glucose among tested carbon sources were very effective, and optimum concentration of glucose was 0.5%. Skim milk was found to be most effective nitrogen source in productivity, and its optimum concentration was 0.6%. But, cysteine, cystine and tryptophane decreased the proteinase productivity. Yeast extract was relatively effective at the range of 0.1-0.5%. The yeast showed maximum production of proteinase at 18$\circ$C, pH 9-11, and cultivation time of 36 hours.

  • In this study, the characteristics of single-component and other sealant were analyzed to provide repair methods and effective utilization of sealant due to crack generation. Humidity studies have shown that acrylic silicone is effective in many areas of fluidity and temperature change, and that polysulfide-silicon is most effective in resistance to linear expansion coefficient and external pressure. Therefore, the difference between drying shrinkage and thermal expansion coefficient between various building materials and sealant, as well as the use of sealant, is analyzed, and construction methods derived accordingly, and prevention of defects are considered to be part of the construction project.

Editorial : COVID-19 infection and ginseng: Predictive influenza virus strains and non-predictive COVID-19 vaccine strains

  • Vaccines help protect people from infections. However, Coronavirus 2019 (COVID-19) vaccinees often still become infected with COVID-19 variants (breakthrough infections) and may go on to suffer from long COVID symptoms due to short-lasting immunity and less-effective protection provided by available vaccines. Moreover, the current COVID-19 vaccines do not prevent viral transmission and ward off only about 15% of breakthrough infections. To prepare more effective vaccines, it is essential to predict the viral strains that will be circulating based on available epidemiological data. The World Health Organization recommends in advance which influenza strains are expected to be prevalent during influenza season to guide the production of influenza vaccines by pharmaceutical companies. However, future emerging COVID-19 strain(s) have not been possible to predict since no sound epidemiological information has been established. Thus, for more effective protection, immune stimulators alone or in combination with vaccines would be preferable to protect people from COVID-19 infection. One of those remedies would be ginseng, which has been used for potentiating immunity in the past.

The Effects of Xylose Treatment on Rumen Degradability and Nutrient Digestibility of Soybean and Cottonseed Meals

  • Two trials were conducted to evaluate the effect of xylose treatment on rumen degradability characteristics of DM, OM and CP and in vivo digestibility of DM, OM, CP and crude fiber (CF) of soybean meal (SBM) and cottonseed meal (CSM). In Trial 1, three ruminally cannulated Merino rams were used. Xylose treatments at both levels, 0.5 and 1%, decreased effective degradability of DM, OM and CP of SBM, whereas 0.5 and 1% xylose treatment of CSM did not show any effect on effective degradability of DM, OM and CP. By contrast, maximum potential degradabilities of DM, OM and CP of CSM seemed to be increased by 1% xylose treatment. It was concluded that xylose treatment was effective in protecting SBM proteins from degradation in the rumen, but the same treatment was not so effective for CSM protein. In trial 2, three Merino rams were used. With treatments, DM, OM, CP and CF digestibilities of SBM and CSM were not changed. Crude fiber digestibility was numerically increased by the treatments of 0.5 and 1% xylose of both SBM and CSM compared to untreated SBM and CSM but differences were not significant. In conclusion SBM proteins can be effectively protected from degradation in the rumen by xylose treatment, without negatively affecting in vivo digestibility of protein, whereas xylose treatment appeared to be less effective on protecting of CSM proteins.
