Development of the Eggs and Pre-Leptocephalus Larvae by Natural Spawning of Artificially-Matured Japanese Eel, Anguilla japonica (인위적 성숙 유도된 뱀장어 Anguilla japonica의 자연산란에 의한 난발생과 Pre-leptocephalus 자어의 발달)

  • Embryonic and pre-leptocephalic larvae development of the eel, Anguilla japonica, are described following natural fertilization in the indoor tank of $23^{\circ}C$ water temperature. Following a routine hormone treatment technique for the brood stock, female eels were artificially matured by weekly intramuscular injections of salmon pituitary extracts (SPE) at a dosage of 20 mg/kg body weight (BW) for a total of 10-14 doses to induce ovarian maturation, while male eels received weekly intramuscular injections of human chorionic gonadotropin (HCG) at a dosage of 1 IU/g BW for a total of 6-10 doses to induce testicular maturation in a separate aquarium and induced natural spawning. Fertilized eggs of about 1.0 mm in diameter were pelagic and showed a typical discoidal cleavage. Hatching occurs 38 hrs after fertilization at a water temperature of $23^{\circ}C$. The newly hatched larvae measured about 3.0 mm in total length and the number of myomeres averages 42. Their mouths and anuses were opened at 4.5 days and the yolk sacs of the pre-leptocephalic larvae were almost absorbed at 6.5 days after hatching. Pre-leptocephalic larvae survive for 14.5 days. At this time they are $5.87{\pm}0.25mm$ in total length and have about 98 myomeres. However, morphological characterization of embryonic and pre-leptocephalic larvae were not different between natural fertilization and artificial fertilization by the dry method.

Residue level and pharmacokinetics of trichlorfon in the Japanese eel (Anguilla japonica) after bath treatment (Trichlorfon (TCF)의 약욕 투여에 따른 뱀장어 체내 약물 잔류량 및 약물동태학 연구)

  • This study performed a trichlorfon (TCF) residue and pharmacokinetic analysis with Japanese eels, Anguilla japonica, to obtain baseline data to establish the maximum residue level (MRL) of TCF in A. japonica. After dipping A. japonica in 30 ppm and 150 ppm of TCF at 28℃ and 18℃, drug residue in the body was analyzed with LC-MS/MS, and these results were further analyzed with the PK solver program to obtain the pharmacokinetic parameters of TCF in the serum, muscles, and liver. The maximum concentrations (Cmax) in the serum, muscles, and liver were 25.87-357.42, 129.91-1043.73, and 40.47-375.20, respectively, and the time to maximum concentration (Tmax) was 0.13-1.32h, 1.17-3.34h, and 0.14-5.40h, respectively. The terminal elimination half-life (T1/2) was 2.13-3.92h, 5.30-10.35h, and 0.65-13.81h, respectively. In the 30 mg/L concentration group, TCF was not detected in the serum of eels 96 hours after bathing, and was below the detection limit after 336 hours in muscle and liver. On the other hand, in the 150 mg/L concentration group, TCF was not detected in the serum of eels 336 hours after bathing, but was detected in muscle and liver at 336 hours. In conclusion, the results of this study would be useful in establishing the MRL of TCF in farmed A. japonica.

Effects of Water Temperature Changes on the Oxygen Consumption Rhythm in the Japanese eel, Anguilla japonica

  • We investigated the effects of temperature changes on the oxygen consumption rhythm in Japanese eels, Anguilla japonica, using an automatic intermittent flow respirometer (AIFR). The endogenous rhythm of the oxygen consumption rate (OCR) in the eels (n = 18; 44-74 cm, 145-690 g), freshly collected by bag net from estuaries, was nearly synchronous with the tidal pattern of the estuarine collection site. The magnitude of mean OCR (mOCR) of eels showed variable range of 82.2 - 116.5 ml $O_2\;kg^{-1}ww \;h^{-1}$ under constant conditions. In case of increasing temperature from 25 to $38^{\circ}C$, the OCR of eels exhibited a gradually increasing trend with a rhythmic pattern until $36^{\circ}C$. Above $36^{\circ}C$ the rhythms of the OCR dampened and the OCR decreased rapidly at around $36-37^{\circ}C$. The OCR of the eels exhibited the maximum value at $38^{\circ}C$, and then it sharply decreased. The results suggested that the critical thermal maximum (CTM) regarding the endogenous rhythms of the eels was at around $36-37^{\circ}C$ when water temperature increased at $0.5^{\circ}C$/14 h following the acclimation at $25^{\circ}C$. In case of decreasing temperature ($0.5^{\circ}C$/14 h) from 25 to $0^{\circ}C$, the OCR of the eels displayed a abrupt decrease up to $23^{\circ}C$, and between at 23 and $20^{\circ}C$, there was an agitation which showed a slight increase in the OCR with a duration of 1-2 days. Below $9^{\circ}C$, the OCR rhythm of the eels showed a constant state regardless of temperature decreasing. These results suggest that the Japanese eel has an upper incipient lethal temperature at $36^{\circ}C$, with a lower thermal limit at $9^{\circ}C$. The biochemical aspects of the eels influenced by water temperature need to be further studied.

  • This study was carried out to examine the acute toxic effects of ammonia to the eel, Anguilla japonica in high temperature and pH levels by histopathological observations. The eels of 40 g average body weight were exposed to 4 different concentrations of total ammonia (0, 10, 20, 30 mg/$\ell$) for 24~120 hours. Each concentration was treated under 4 different levels of pH (7.5, 8.0, 8.5, 9.0) and each of these treatments was tested at 2 different temperatures ($27^{\circ}C$, $32^{\circ}C$). Histopathological changes in gill tissues were observed by hematoxylin-eosin stain. As increasing of pH (from 7.5 to 9.0), water temperature (from $27^{\circ}C$ to $32^{\circ}C$), total ammonia concentration (from 0 mg/$\ell$ to 30 mg/$\ell$) and exposure time (from 24 hours to 120 hours), gill discolorated to dark brown with the naked eye and gill tissues showed hypertrophy of gill lamellae, winding of the secondary gill lamellae, epitherial separation and necrosis histopathologically. When gill lamellae epithelium was separated from the blood vessels, gill discolorated to dark brown with the naked eye.

Upstream Behavior of Glass Eels (Anguilla japonica) in an Experimental Eel-ladder (실험 어도에서 실뱀장어의 소상 행동)

  • Upstream behavior of glass eels was examined in an experimental eel-ladder at a laboratory of the National Fisheries Research and Development Institute from March to May in 2008. The study was made under various environmental factors and conditions affecting the upstream migration of glass eels in order to design a functional ladder that would allow the passage of glass eels. The experimental eel-ladder consisted of an upper freshwater chamber and a lower sea water chamber; glass eels in sea water can move up to the upper freshwater chamber through the slope (eel-ladder) between them. The optimal condition of the eel-ladder was estimated by comparing the number of glass eels that moved upstream depending on various conditions. Since the glass eels actively moved up the slope to river water rather than to reservoir water or tap water, the experiment was realized using river water. A significantly higher number of glass eels moved up during the spring tide than during the neap tide, and during night than during the day. Upstream movement was significantly higher during high tide than during low tide. Glass eels effectively moved up through a slope of less than $30^{\circ}$ and water-flow velocity lower than 0.4 m/sec. The fish preferred a coarser ladder bed covered with small gravels, brush or carpet.

Histopathological study of chronic nitrite toxicity on the japanese eel, Anguilla japonica (아질산의 만성중독증에 의한 참장어(뱀장어)의 병리조직학적 연구)

  • The research was carried out to examine the chronic toxic effects of nitrite on the Japanese eel, Anguilla japonica by neans of histological observations. Young eel, 10.8g mean body weight. were exposed to 6 different concentrations of nitrite(1, 5, 10, 20, 30 and 40ppm) for 10 weeks. Each concentration was treated under 5 different levels of pH(5.5, 6.0, 6.5, 7.0 and 7.5) and each of these treatment was tested at 2 different temperature regimes($25^{\circ}C$ and $30^{\circ}C$). Proper concentration of nitrite was made by $NaNO_2$ and proper pH levels were made by the combinations of 0.1M $KH_2PO_4$ and 0.1M $NaHCO_3$. Histopathological test of gill tissues were made along with the test of the formation of thrombocystes and chloride cells on the gill filaments. At the lower pH levels, mucus secretion from the gill was incrased as the nitrite concentration increased. As the level of nitrite increased the number of chloride cells on the gill filament were decreased. Most of the remained chloride cells were observed only at the terminar part of the gill filament at 40ppm of nitrite. Degeneration of gill tissues were observed when nitrite levels were over 10ppm along with detachement and sweption of the epithelial cells of the gill lamellae. Shrunken gill lamellae and formation of thrombosis in the capillaries of gill lamellae were also observed. When temperature goes higher at the higher level of nitrite, necroses in the gill lamellae was increased. At the lower than 10ppm of nitrite, degeneration of gill lamellae was occured at the beginning of the test period but regenerated later. Negative effects of nitrite on the growth of young eel was started between 5~10ppm at the pH level of 7.0 and 7.5. Thrombosis formation were also started at this level. The safety concentration of nitrite at the pH levels of 7.0 and 7.5 on the small eel seems to be 1ppm. Thrombosis and gill lamella detachment and necrosis in the gill capillaries were not observed at this level. Chloride cells were appeared the whole part of the gill filament.

Effect of TBT and PAHs on CYP1A, AhR and Vitellogenin Gene Expression in the Japanese Eel, Anguilla japonica

  • Gene expressions of cytochrome P4501A (CYP1A), aryl hydrocarbon receptor (AhR) and vitellogenin (Vg) by endocrine disruptors, benzo[${\alpha}$]pyrene (B[a]P) and tributyltin (TBT) were examined in cultured eel hepatocytes which were isolated from eels treated previously with B[a]P (10 mg/kg) or estradiol-$17{\beta}$ (20 mg/kg) in vivo, and the relationship between CYP1A, AhR and Vg genes were studied. When the cultured eel hepatocytes were treated with B[a]P ($10^{-6}-10^{-5}M$) the gene expressions of CYP1A and AhR were enhanced in a concentration-dependent manner. However, when treated with TBT ($10^{-9}-10^{-5}M$) the gene expressions of CYP1A and AhR were suppressed at high concentrations ($10^{-6}-10^{-5}M$), while having no effects at low concentrations ($10^{-9}-10^{-7}M$). Gene expression of Vg was also suppressed by TBT in a concentration-dependent manner in cultured eel hepatocytes which was previously treated in vivo with estradiol-$17{\beta}$.

Development of the Slurry Type Diet for the Growing Leptocephalus, Eel Larvae (Anguilla japonica) (뱀장어 자어용 액상사료 개발)

  • This feeding experiment was conducted to investigate the effects of slurry type diet on growth performance and survival rate of growing leptocephalus, eel larva. We need to find new materials of diets for rearing eel larvae. Test diets were formulated with the eggs of the shark, fish soluble concentrate, soybean peptide and fish protein hydrolysate. Fish (average length 6 mm) were fed 3 slurry type diet(A, B and C) based on shark egg for 5 times per day. During feeding experiment, survival rates were significantly different among 3 slurry type diets. Total protein, lipid, moisture, ash and free amino acids contents were analyzed for slurry type diets. Leptocephalus fed the C slurry type diet was grown up to $38.0{\pm}9mm$ at 150 days. But all leptocephalus fed B slurry type diet were died at 100 days, reaching $16.4{\pm}8mm$. This results suggest that basic information for diet development of eel leptocephalus.
