With continuing demand for the development of new, effective and safe therapies, an investigation was carried out to test the efficacy of an antibacterial agent derived from marine edible seaweed. The methanolic extract of Ecklonia cava from marine edible seaweed evinced potential antibacterial activity against Enterococcus faecalis. Among five solvent-soluble fractions of E. cava methanolic extract, the ethyl acetate soluble extract (EtOAc) exhibited the strongest antibacterial activity, with a MIC value of $128{\mu}g/mL $ against E. faecalis strains. Furthermore, a synergistic antibacterial effect between an antibiotic and the EtOAc fraction was assessed using fractional inhibitory concentration (FIC) indices. A combination of ciprofloxacin and the EtOAc fraction resulted in a ${\sum}FIC_{min}$ range of 0.188 and ${\sum}FIC_{max}$ of 0.508 to 563, suggesting that the ciprofloxacin-EtOAc fraction of E. cava combination resulted in an antibacterial synergy effect against E. faecalis.
The mass cultivation of Ecklonia cava Kjellman was studied as a potential biomass source for the extract industry in Korea. Experiments were conducted to investigate the optimal conditions for artificial seed production and mass cultivation of this species. Maximum growth and young thalli development in the nursery culture area occurred at 2 m depth, whereas maximum growth of thalli in the main culture area occurred at 1 m depth. Production of E. cava was between 2.6 and 3.6 kg wet wt. $m^{-1}$ after depth control and removal of fouling organism, etc. The relationship between optimal water depth for culture and underwater irradiance during the E. cava cultivation was calculated as: y = -0.718x + 8.042 ($r^2$=0.976). The growth rates achieved in this trial indicate that E. cava cultures could produce and supply sufficient biomass.
Reactive oxygen species (ROS) play an important role in the pathogenesis of many human degenerative diseases such as cancer, aging, arteriosclerosis, and rheumatism. Much attention has been focused on the development of safe and effective antioxidants. To discover sources of antioxidative activity in marine algae, extracts from 17 kinds of seaweed were screened for their inhibitory effect on total ROS generation in kidney homogenate using 2',7'-dichlorofluorescein diacetate (DCFH-DA). ROS inhibition was seen in three species: UIva pertusa, Symphyocladia latiuscula, and Ecklonia stolonifera. At a final concentration of 25 $\mu\textrm{g}$/mL, U. pertusa inhibited 85.65$\pm$20.28% of total ROS generation, S. latiscula caused 50.63$\pm$0.09% inhibitory, and the Ecklonia species was 44.30$\pm$7.33% inhibition. E. stolonifera OKAMURA (Lam-inariaceae), which belongs to the brown algae, has been further investigated because it is commonly used as a foodstuff in Korea. Five compounds, phloroglucinol (1), eckstolonol (2), eckol (3), phlorofucofuroeckol A (4), and dieckol (5), isolated from the ethyl acetate soluble fraction of the methanolic extrclct of E. stolonifera inhibited total ROS generation.
The Induction of regeneration and maturation in the free-living gametophytes of Ecklonia stolonifera Okamura was studied at four temperatures (5, 10, 15, and $20^{\circ}C$), four levels of irradiance (5, 10, 20, and 40 ${\mu}mol\;m^{-2}s^{-1}$) and three photoperiods (14:10, 12:12, and 10:14 h L:D). Female gametophyte fragments were maintained in active regeneration without reaching sexual maturity under $5{\sim}10^{\circ}C$, 10 ${\mu}mol\;m^{-2}s^{-1}$, 10:14 h (L:D), whereas the conditions for male gametophytes were slightly different at $20^{\circ}C$, 40 ${\mu}mol\;m^{-2}s^{-1}$, 10:14 h (L:D). The sexual maturation of female and male gametophytes was facilitated at $15^{\circ}C$, 20 ${\mu}mol\;m^{-2}s^{-1}$, 14:10 h (L:D). These results provide basic information for controlling the regeneration and maturation of the free-living gametophytes for artificial seed production of E. stolonifera.

Osteoarthritis is thought to be induced by the ageing-related loss of homeostatic balance between degeneration and repair mechanism around cartilage tissue in which inflammatory mediators such as reactive oxygen species, cytokines and prostaglandins are prone to overproduction under undesirable physiological conditions. Phlorotannins are unique polyphenolic compounds bearing dibenzo-1,4-dioxin skeleton which are not found in terrestrial plants but found only in some brown algal species such as Ecklonia and Eisenia families. Phlorotanninrich extracts of Ecklonia cava including LAD103 showed significant antioxidant activities such as DPPH radical scavenging, ferric ion reduction, peroxynitrite scavenging, and inhibition of LDL oxidation, indicating their possible antioxidative interference both in onset and downstream consequences of osteoarthritis. LAD103 also showed significant down regulation of $PGE_2$ generation in LPS-treated RAW 246.7 cells, and significant inhibition of human recombinant interleukin-$1{\alpha}$-induced proteoglycan degradation, indicating its beneficial involvement in pathophysiological consequences of osteoarthritis, the mechanism of which needs further investigation. Since LAD103 showed strong therapeutic potentials in arthritic treatment through several in vitro experiments, it is highly encouraged to perform further mechanistic and efficacy studies.
Physicochemical properties and consumer perception of white pan bread as influenced by the addition of Ecklonia cava powder (ECP) were investigated. Freeze-dried Ecklonia cava were ground, sieved through a laboratory sieve and a fraction with particles less than $250{\mu}m$ was used. Amount of ECP added (0~3%) to the bread was found to affect the bread quality significantly (P<0.05). pH, bread height, and volume of the control was significantly higher than others (P<0.05) and decreased significantly (P<0.05) with the addition of ECP. Moisture content showed no significant differences (P>0.05). There were distinctive color changes with the addition of the powder: $L^*$- and $a^*$-values decreased but $b^*$-value increased significantly (P<0.05). The hardness of bread was found to increase but both cohesiveness and springiness showed a reverse trend with the addition of the powder. Consumer acceptance test indicated that ECP content 1% on wheat flour could be the recommended supplementation level for the consumers without sacrificing sensory quality.
Chromosomal aberration tests in vitro using Chinese hamster lung(CHL) cells were carried out to evaluate the possible role of the MeOH extract of Ecklonia stolonifera in modulating the chromosomal damage induced by Mitomycin C(MMC) and Benzo(a)pyrene(B(a)P), respectively. The MeOH extract of Ecklonia stolonifera(260$\mu\textrm{g}$/ml) reduced significantly the incidence of chromosomal aberration induced by treatment with B(a)P by 80%. The suppressive effect was much stronger than that of $\beta$-carotene, which is well known antimu-tagen. However, there was no marked decrease in the chromosomal aberration induced by MMC. In the tests involving chromosomal aberration induced by the treatment of the MeOH extract of Exklonia stlolnifera alone, there was no significant increase in comparison with the negative control. The results would seem to indicate that. at least under the conditions examined, the MeOH extract of Ecklonis stolonifera decreased the chromosomal aberrations induced by B(a)P in the CHL cells, but had little effect on the chromosomal aberration induced by MMC.

Ecklonia cava, an edible marine brown alga (Laminariaceae), is a rich source of bioactive compounds such as fucoidan and phlorotannins. Ecklonia cava extract (ECE) was prepared using 70% ethanol extraction and ECE contained 67% and 10.6% of total phlorotannins and dieckol, respectively. ECE treatment significantly inhibited receptor activator of nuclear $factor-{\kappa}B$ ligand (RANKL)-induced osteoclast differentiation of RAW 264.7 cells and pit formation in bone resorption assay (p <0.05). Moreover, it suppressed RANKL-induced $NF-{\kappa}B$ and mitogen-activated protein kinase signaling in a dose dependent manner. Downregulated osteoclast-specific gene (tartrate-resistant acid phosphatase, cathepsin K, and matrix metalloproteinase-9) expression and osteoclast proliferative transcriptional factors (nuclear factor of activated T cells-1 and c-fos) confirmed ECE-mediated suppression of osteoclastogenesis. ECE treatment ($100{\mu}g/ml$) increased heme oxygenase-1 expression by 2.5-fold and decreased intercellular reactive oxygen species production during osteoclastogenesis. The effective inhibition of RANKL-stimulated osteoclast differentiation and oxidative stress by ECE suggest that ECE has therapeutic potential in alleviating osteoclast-associated disorders.
This study was performed to determine the optimal ratio for preparing an extract comprising the Ecklonia cava and Hizikia fusiformis complex as a therapeutic material for alleviating inflammatory respiratory diseases. First, to examine the optimal ratio for preparing the complex (SD-EH), Ecklonia cava and Hizikia fusiformis extracts were used; four extracts with different mixing ratios were prepared. The effects of the SD-EH extract on MUC5AC mRNA expression in PMA-treated NCI-H292 cells were analyzed; it was confirmed that the MUC5AC expression was significantly reduced after treatment with the SD-EHA-001 (E(100) : H(0)), SD-EHB-001 (E(90) : H(10)), SD-EHC-001 (E(80) : H(20)), and SD-EHD-001 (E(70) : H(30)) extracts. Western blotting was used to determine whether the SD-EH extract affects the expression levels of COX-2 and MMP-9 in PMA-treated A549 cells. The protein expression levels of COX-2 and MMP-9 were significantly lower (p < 0.001) in the cells treated with the SD-EHC-001 (E(80):H(20)), SD-EHD-001 (E(70) : H(30)), and SD-EHE-001 (E(60) : H(40) extracts than in the cells treated with PMA alone. The SD-EHC-001 (E(80) : H(20)) extract markedly downregulated the expression levels of MUC5AC, COX-2, and MMP-9. Therefore, the SE-EH extract may serve as a potential therapeutic agent for treating inflammatory respiratory diseases.

The biomass of the brown seaweed, Ecklonia, was used to remove Cr(VI) from wastewater. Previously, Cr(VI) was removed through its reduction to Cr(III) when brought into contact with the biomass. In this study, the effects of ionic strength, background electrolytes, and Cr(III), Ni(II), Zn(II), and Fe(III) on the Cr(VI) reduction were examined. An increased ionic strength inhibited the Cr(VI) reduction. The presence of other heavy metals, such as Cr(III), Ni(II), or Zn(II), only slightly affected the Cr(VI) reduction, while Fe(III) enhanced the reduction. Although the above various parameters could affect the reduction rate of Cr(VI) by Ecklonia biomass, these effects were relatively smaller than those of pH and temperature. In addition, the previously derived rate equation was found to be applicable over a range of ionic strengths and with different background electrolytes. In conclusion, Ecklonia, bioniass may be a good candidate as a biosorbent for the removal of Cr(VI) from wastewaters containing various other impurities, and scale-up to a practical process may be accomplished using the previously derived rate equation.
