Growth inhibition of hydrotrope-combined copper against Microcystis aeruginosa and evaluation of its toxicity (Microcystis aeruginosa에 대한 hydrotrope-combined copper의 생장억제 및 독성 평가)

  • Hydrotrope-combined copper (HCC) is a copper ($Cu^{2+}$)-based algicide, which is combined with a hydrotrope that keeps copper ion in solution to improve performance. This study assessed the growth inhibition effect of HCC against Microcystis aeruginosa which is one of the most common toxic cyanobacterium in eutrophic freshwater environment. Various HCC doses, ranging from 5.5 to $550{\mu}g/L$ as $Cu^{2+}$, were applied to either BG-11 or 1/4 diluted medium with low- or high-inoculum density of M. aeruginosa. Growth inhibition was monitored based on a decrease in chlorophyll-a content in culture medium during the incubation. Results showed that HCC significantly inhibited the growth of M. aeruginosa in a dose-dependent manner. In case of 1/4 diluted BG-11 medium, HCC dose as low as $5.5{\mu}g$ $Cu^{2+}/L$ completely inhibited the production of chlorophyll-a by M. aeruginosa. It was found that HCC did not induce any significant release of microcystin-LR from M. aeruginosa. Acute toxicity of HCC was tested using Daphnia magna, and the 24-h $EC_{50}$ value was 0.30 mg/L as $Cu^{2+}$ which was much higher than the actual inhibition dose. Ames test was performed using Salmonella enterica serovar Typhimurium TA100, and HCC showed no increase in the number of revertant colonies. The result suggested that HCC does not have any mutagenic potential in the aquatic environment. In addition, no genotoxic effect of HCC was also confirmed based on the SOS ChromoTest using Escherichia coli PQ37. Therefore, HCC could be used as a relatively safe and effective pre- and post-treatment agent to control hazardous algal blooming in aquatic environments.

  • Present contamination levels of soils along the major roadsides of Seoul, Korea were investigated, and base-line data were accumulated for future use. Topsoil $(1{\sim}5$ cm) and subsoil $(20{\sim}50$ cm) from five districts (Gangdong-, Gwangjin-, Nowon-, Seodaemun- and Seongdong-gu) were sampled. The collected samples were airdried, passed through 2-mm sieves, and analyzed to determine the physicochemical properties including pH, EC, CEC, exchangeable cations (Ca, Mg, K, and Na), and heavy metal contents (Cd, Cu, Pb, and Zn). Soil textures of topsoils and subsoils were mainly loamy sand and sandy loam, respectively. The range of pH was $4.5{\sim}10$.0 with an average of 7.5 for both topsoil and subsoil, which is much higher than that of the forest soils in Seoul. The ranges of 0.1 N HCI extractable Cd, Cu, Pb, and Zn contents for both topsoils and subsoils were $0.0l{\sim}l.19$, N.D. (not $detected){\sim}228$.99, $N.D.{\sim}352$.54, and $2.97{\sim}332$.96 mg $kg^{-1}$, respectively. Most of the average heavy metal contents were lower than the concern level of the Soil Environment Conservation Act of Korea, but were much higher than those of the , forest soils in Seoul. Some sites were higher in heavy metal contents than the concern levels; in particular, the average Cu content in Seongdong-gu was much higher than the concern level, 50 mg $kg^{-1}$. Careful management of the soil to prevent the aggravation of the present contamination level and the dissemination of contamination is highly recommended.

Geochemistry of Stream Water around the Abandoned Boeun Coal Mine, Hoenam Area (보은제일폐탄광 주변 하천수의 지구화학적 특징)

  • Stream water chemistry in the abandoned Boeun Jeil coal mine area was studied for a period of 3 months, including rainy and dry season. The stream waters were a nearly neutral and slightly alkali condition, and $Mg-SO_4$ type with Mg>Ca>Na>K and $SO_4>HCO_3>Cl>NO_3$. Chemical composition of the stream water was quite irregular during the experimental period. Concentrations of Na, K, $HCO_3$, U, Sr, and Cr decreased by $10{\sim}30%$ during rainy season, caused by dilution effects with rain. The concentration of Ca, Mg, $NO_3$, Cd, and Co increased during the rainy season, caused by more easily dissolved from bedrocks or mine drainage with slightly acidic condition than dry season. The stream water was enriched in Mg, Ca, $HCO_3$, $SO_4$, Al, Fe, Zn, Ni, Co, Cr, Cd, Sr and U. Concentrations of Na, Mg, Ca, $SO_4$, $HCO_3$, Fe, Zn, Ni, Sr, and U decreased linearly with distance from the mine adit. These elements were strongly controlled by dilution of unpolluted water influx and/or adsorption on the clay minerals and iron oxyhydroxide precipitates. This mine area exhibited two main weathering processes ; 1) oxidation with acidification derived from Fe sulphides, and 2) pH buffering due to Ca and Mg carbonate dissolution. This weathering processes were followed by adsorption of metals on iron oxyhydroxides and precipitation.

Variation of cyanobacteria occurrence pattern and environmental factors in Lake Juam (주암호 유해남조류 출현양상과 환경요인 변화)

  • The study analyzed the relationship between harmful cyanobacteria and physicochemical factors in Lake Juam from 2005 to 2018. The research locations were designated St. 1 (Juam-Dam) and St. 2 (Sinpyong). Harmful cyanobacteria was found in four genera (Microcystis sp., Anabaena sp., Aphanizomenon sp., Oscillatoria sp.). The average standing crops of harmful cyanobacteria in both locations were 2,575 cells mL-1 and 2,557 cells mL-1 from 2005 to 2011. Since 2012, there has been a significant decrease that the measurements were 42 cells mL-1 and 82 cells mL-1 from 2012 to 2018. To analyze the reason for the decrease in harmful cyanobacteria, Pearson's correlation and t-tests were performed on data collected during the summer period (June-September). Pearson's correlation showed a significantly positive correlation with total nitrogen(TN), outflow, and storage and a negative correlation with electrical conductivity. T-tests were conducted in two different periods and showed decreases in total nitrogen, electrical conductivity, and residence time. The average rainfall was decreased from 263.3 mm (2005-2011) to 219.9 mm (2012-2018) and total nitrogen was decreased from 0.912 mg L-1 (2005-2011) to 0.811 mg L-1 (2012-2018) and the same variability was seen in TP (total phosphorus). Therefore, it seems that the low-rainfall decreased the nutrients (TN) and variability in the TP, resulting in a decrease in harmful cyanobacteria in Lake Juam.

Hydrochemical Characteristics of Natural Mineral Water in the Daebo and Bulguksa Granites (대보화강암과 불국사화강암지역 먹는샘물의 수리화학적 특성)

  • Groundwater quality of the natural mineral water was investigated in hydrochemical aspects in order to ensure that mineral water meets stringent health standards. There exist 20 mineral water plants in the Daebo granite and 4 mineral water plants in the Bulguksa granite, respectively. Both granite areas show some differences in water chemistry. The pH, EC, hardness, total ionic contents in groundwater of the Daebo granite area are higher relative to those of the Bulguksa granite area. The content of major cations is in the order of Ca>Na>Mg>K, while that of major anions shows the order of $HCO_3>SO_4$>Cl>F. The fact that the $Ca-Na-HCO_3$ type is most predominant among water types may reflect that the dissolution of plagioclase that is most abundant in granitic rocks plays a most important role in groundwater chemistry. Representative correlation coefficients between chemical species are variable depending on geology. In the Daebo granite area, $Ca-HCO_3(0.84),{\;}Mg-HCO_3(0.81),{\;}SiO_2-Cl(0.74),{\;}Na-HCO_3(0.70)$ show relatively good correlationships. In the Bulguksa granite area, fairly good correlationships are found among some components such as K-Mg(0.93), $K-HCO_3(0.92)$, Mg-Cl(0.92), $Cl-HCO_3(0.91)$, and K-F(0.90). According to saturation index, most chemical species are undersaturated with respect to major minerals, except for some silica phases. Groundwater is slightly undersaturated with respect to calcite, whereas it is still greatly undersaturated with respect to dolomite, gypsum and fluorite. Based on the phase equilibrium it is clear that groundwater is mostly in equilibrium with kaolinite and becomes undersaturated with respect to feldspars, evolved from the stability area of gibbsite during water-rock interaction. While the activity of silica increases, there is no remarkable increase in the acivities of alkali ions and pH, which indicates that some amounts of silicic acid dissolved from silica phases as well as feldspars were provided to groundwater. It is concluded that chemical evolution of groundwater in granite aquifers may continue to proceed with increasing pH.

Effect of a Freshwater Bivalve (Unio douglasiae) and a Submerged Plant (Potamogeton crispus) on the Growth Inhibition of a Cyanobacterium Oscillatoria sp. (담수패류(Unio douglasiae)와 침수식물(Potamogeton crispus)의 유해 남조 Oscillatoria sp. 성장억제 효과)

  • This study examined the inhibition effects of a freshwater bivalve (Unio douglasiae) and a submerged plant (Potamogeton crispus) on the cyanobacterial bloom (Oscillatoria sp.). The experiment were conducted in aquarium $(50cm{\times}65cm{\times}120cm)$ with lake sediments in the bottom of the aquarium in 10 cm thick. Before the experiments, artificial cyanobacterial bloom was induced with the addition of lake sediment and CB medium. Total 12 transparent acrylic cylinders (${\Phi}19cm$, height 40 cm) were placed in the aquarium, and within which bivalves and plants were placed in various conditions such as the control (C), plant addition (P:5 stems), mussel addition (U:2 individuals), and both mussel and plant addition (PU: the same quantity as used in each treatment). The experiment was conducted in triplicate during 7 days. pH, dissolved oxygen (DO), electric conductivity (EC), salinity, cyanobacterial cell density, chlorophyll-${\alpha}$ concentration, and mussel filtering rate were monitored daily. At the end of the experiment, total phosphorus (TP), total nitrogen (TN), and plant height and weight were measured. Overall, a large degree of cyanobacterial growth inhibition appeared in both P and U treatments, and the effect was highest in the U treatment, followed by P and PU. The combined treatment of both U and P did not show any synergic effects compared to the effect in separated treatment. In all enclosures of the treatments chlorophyll-${alpha}$ (Chl-${alpha}$) concentration decreased until 36 hours after the additions of the plants and mussels. In contrast, Chl-${alpha}$ concentrations increased in PU enclosures after 36 hours. The same trend was shown in the cell density of Oscillatoria. pH and DO gradually decreased until 120 and 144 hours, respectively, in the P and PU enclosures. TP concentration increased in the mussel enclosures (U and PU), while TN concentration largely decreased in the plant enclosures (P and PU). Our results suggest that applied bivalve (Unio) and submerged plant (Potamogeton) seemed to have a potential effect on the growth inhibition of cyanobacteria, but their combined application may have an antagonistic effect to diminish the degree of the inhibition.

A Analysis of Trophic Structure in Lake Namyang Using the Ecopath Modelling (Ecopath 모델을 이용한 남양호의 영양구조 분석)

  • The purpose of this study was to describe quantitatively trophic structures and to analyze energy flows in the Lake Namyang using the Ecopath with ecosim (Walter et al., 1997). The sampling and analyses were carried out at 6 sampling sites of the Lake Namyang during May and November in 2007. A total of 10 groups were considered in this study (detritus, macrophytes, phytoplankton, zooplankton, zoobenthos, Cyprinus carpio, Carassius cuvieri, Carassius auratus, Pseudobagrus fulvidraco and other fishes) to assess the trophic relationship, energy flows and interactions between them. As a result, it was concluded that Lake Namyang was consisted of primary producers (Detritus, Macrophytes, Phytoplankton), primary consumers (Zooplankton, Zoobenthos, Cyprinus carpio, Carassius cuvieri, Carassius auratus, Other fishes) and secondary consumer (Pseudobagrus fulvidraco). The total system throughput was estimated at $14.1\;kg\;m^{-1}\;year^{-1}$ including a consumption of 39%, exports of 21%, respiratory flows of 12% and flows into detritus of 28%. MTI analyses indicate that Pseudobagrus fulvidraco have positive impact on Cyprinus carpio, Carassius cuvieri and Carassius auratus. On the other hand, other fishes have negative impact on Cyprinus carpio, Carassius cuvieri and Carassius auratus. All the functional groups except detritus had a negative impact on themselves and this may show within-group competition for the same resources.

Effects of Compost Leachate on Growth and Yield of Leaf Lettuce in Hydroponic Culture (퇴비단 여과액비가 수경재배에서 상추의 생육과 수량에 미치는 영향)

  • The compost leachate was dark-colored solution that leaches out of the bottom of the compost pile. The compost leachate was rich in nutrients and can potentially used in plant culture. In the organic production, commercial liquid fertilizer was used to insure the availability of nutrients during the formation of the yield. The cost of supplemental liquid fertilizer could be reduced by developing a fertilizer based on animal fertilizer. This experiment was conducted to investigate the effect of different combination of compost leachate and conventional inorganic solution in hydroponic culture for lettuce growth. Six different treatments were applied. The compost leachate(CL) and nutrient solution(NS) were mixed by six different mixing ratios of 0:100, 20:80, 40:60, 60:40%, 80:20 and 100%:0% based on nitrogen content. The chemical nutrient solution was the solution of National Horticulture Research Station for the growth of leaf lettuce. The concentration of nutrient solution was adjusted 1.5 mS/cm in EC. The compost leachate was low in phosphorus(P), calcium(Ca), magnesium(Mg), but rich in potassium(K). The plant height of lettuce treated with CL 20+NS 80% was similar with 100% NS of control plot. Plant height was highest in the plot of CL 20+NS 80%. The treatment of 100% compost leachate was lowest in the growth characteristics of leaf lettuce. Number of leaf was very low in 100% compost leachate compared with plot of chemical nutrient solution. In the beginning of growth stage, SPAD value was reduced in plot treated with CL100%, but CL20+NS80% plot was higher compared to 100% compost leachate. SPAD value of leaf lettuce leaves was decreased as the amount of CL was increased. The dry weight of lettuce were 107.4, 104.2g in plot of NS 100% and CL 20%+NS80%, respectively. The leaf number and plant weight were decreased at high application plots of compost leachate. The leaf lettuce showed lowest in the plot treated with 100% compost leachate, and the growth of lettuce severely decreased after application of 100% CL treatment. The results showed that compost leachate can be use as liquid fertilizer for the organic hydroponic production. The mixture solution of 20% of compost leachate and 80% of nutrient solution could be used as a nutrition solution in hydroponic culture of leaf lettuce. Our studies have shown that is possible to produce using compost leachate, although growth is slower than when using a conventional inorganic hydroponic solution.

Composting Methods for Pig Sludge and the Stabilized Investigation of Crop Cultivation (돈분의 자원화 퇴비 제조 방법 및 작물 재배 안전성 검정)

  • This study was carried out development a new composting system to lower copper and zinc concentration in plg sludge compost and conduct an inquiry into the possibility of crop cultivation. The concentrations of nitrogen, phosphorus and pH averaged 4.4%, 6.3% and 7.57, respectively, which were higher concentrations than in commercial organic fertilizers, and the concentrations of copper and zinc averaged 805 and 1,704 mg/kg, respectively, which were beyond the heavy metal concentration limit in byproduct compost. Hydrated citric acid I lowered the concentrations of copper and zinc by 58% and 97%, respectively and hydrated oxalic acid II lowered the concentrations of copper and zinc by 48% and 56%, respectively in pig sludge compost. Lower concentrations of copper and zinc in pig sludge resulted from the enhanced hydrated-citric acid concentration in organic acid solution mixed with distilled water. The concentrations of copper and zinc were 330, and 41 mg/kg in the pig sludge treated with 100% hydrated citric acid. Agitation composting system stabilized the compost earlier than the stationary composting system, in which the stabilization condition was confirmed by higher temperature by $4^{\circ}C$ at highest temperature and 7 days earlier cooling down after highest temperature. The levels of germination index (G.I) 80 were obtained 15 and 20 days after composting in agitation and stationary composting system, respectively. The concentrations of copper and zinc were 2.4 and 4.26 mg/kg respectively in soils amended with pig sludge compost after removing process of heavy metals by citric acid, but 8.0 and 22.37 mg/kg, respectively in soils amended with Pig Sludge. The concentrations of heavy metals was highest in com cultivated in soils amended with pig sludge. The copper and zinc concentrations In corn leaves were 75.2 and 50.56 mg/kg respectively, which were 4 and 2 fold higher than the com cultivated in soils amended with pig sludge compost after heavy metal removing process by hydrated citric acid.

Environmental Evaluation of the Productivity and Functional Fructo-oligosaccharides of Yacon Cultivated in Various Regions of Korea (우리나라 재배지역별 야콘 생산성 및 기능성 프락토올리고당 평가)

  • The purpose of this study was to investigate the effects of envrionmental factors on the ecological responses, yield, and quality properties and physicochemical characteristics of yacon cultivated in various regions of Korea. Experiments were carried out in eight regions from 2010 to 2013. The temperature range in Jinbu, Bonghwa, Cheolwon, and Gangneung during the growth period of yacon cultivation was $17.5-24.6^{\circ}C$. The total yield and marketable yield of tuberous root in Jinbu were 4,065 and 3,196 kg/10a, respectively. The sugar content of yacon tuberous roots comprised 0.11-0.20% fructose, 0.11-0.37% glucose, 0.39-0.68% sucrose, 0.07-0.37% reducing sugar, and 7.03-9.62% fructo-oligosaccharides. The content of fructo-oligosaccharides, which is a functional substance, was the highest in yacon cultivated in Jinbu. Based on the productivity and functional fructo-oligosaccharides, the optimum areas to cultivate yacon are Jinbu and Bonghwa, which are located in the middle-highland zone (500-560 m) and have a suitable growth temperature of $18-25^{\circ}C$. The results of the present study indicate that temperature could be the factor with the greatest influence on the root growth of yacon.
