A Histological Study of Skin on Some Amphibia Inhabitated Chiri Mt. and Moodeung Mt. (지리산과 무등산에 서식하는 한국산 양서류의 피부에 관한 연구)

  • For the purpose of this study was accumulating histological data of skin some amphibia near Chiri Mt. and Moodeung Mt. Analyzed Anura and Caudata were Rana nigromaculata, Rana rugosa, Rana catesbeiana, Hynobius leechii. The histological prepared skin of frogs were compared, of which were selected from dorsal and belly. Excretory glands were identified granular glands, mucous glands, serous glands, vacuoles and excretory ducts in epidermal and dermal tissue. And developing excretory glands, well developed excretory glands and post developing excretory glands were identified also. These results were significantly as basal data on the comparative epidermal skin histology on some Korean Amphibia. Probably these glands of amphibian skin could be infered which were adaptable structure to ecological suffered condition. Following study of these results were more considerable data for comparative histology, comparative anatomy and comparative physiology and ecology of Amphibia.

  • Ultrastructure of the excretory duct of the small ampullate gland in the orb web spider, Nephila clavata L. Koch are studied with light and electron microscopes. The small ampullate glands, located near the midline portion of the abdominal cavity, are connected with the spigots(large spinning tubes) on the middle spinnerets and composed of three parts which are the excretory duct, the storage sac and the convoluted tail. The excretory duct of this gland is enclosed by a thin layer of the outer connective tissues. By the morphology of the apical cuticles and internal textures of the epithelial cells, the duct is subdivided into two regions which are proximal duct region near the sac and distal duct region near the spinnerets. At the distal region of the ducts, the subcuticle which had the function of water removal form the progenetive silk material is well developed, whereas at the proximal region this cuticle disappeared and instead of these, endocuticle is developed. Moreover the epithelium of the distal duct region is composed of columnar epithelial cells, but at the proximal region the epithelium is changed to squamous or cuboidal forms. In the cytoplasm of the epithelial cells, rough endoplasmic reticula, Golgi comlexes and large secretory vesicles related to the production of the cuticular materials are well developed. And between the adjacent epithelial cells, specialized septate junction and desmosomes are formed along the plasma membrane.

  • The ultrastructure of the sac and tail portion of the large ampullate gland and production of the silk materials in the orb web spider, Nephila clavata L. Koch, are studied with electron microscope. Large ampullate glands, the largest glands among the seven kinds of silk glands in this species, are composed of three parts which are the excretory duct, the storage sac and the convoluted tail. The wall of the sac is composed of a single layer of columnar epithelial cells. In the cytoplasm of these cells several kinds of the secretory granules, which are commonly associated with the rough endoplasmic reticula and had characteristic crystalloid, are seen. According to the morphology and internal textures of these granules, the sac portion is subdivided into proximal(connected with the convoluted tail) and distal(connected with excretory duct) portion. Between these two portions, the proximal portion is longer than the distal by four times. Silk materials, being synthesized to the shape of secretory granules, within the glandular epithelial cell of the tail portion are released to the inner cavity by the mechanism of the eccrine secretion. These secretory granules are originated from the rough endoplasmic reticula of the glandular epithelial cells, whereas no Golgi complexes has been found in any of the cells which have been examined.

  • The ultrastructure and silk formational process of the flagelliform glands, one of the capture thread producing organs in the orb web spider (Nephila clavata L. Koch), have been studied with electron microscope. A pair of flagelliform glands consist of three distinct regions: excretory duct, storage sac and convoluted tail. This gland is connected to the large spinning tube (spigot) of the posterior spinnerets with two pairs of aggregate glands, and showed a characteristic spigot structure called "triad". The excretory duct consists of three long segments surrounded by the same sheath of connective tissue. The distal duct near the spinning tube bears the electron lucent subcuticles which have the functions of water removal and orientation of the silk fiber, whereas the proximal duct near the sac contains the cuticule precursor transported through the intercellular space. Cytoplasms of the sac and tail regions have well developed rough endoplasmic reticulum and contain the round secretory granules commonly, but the electron densities and internal textures of the granules do not coincided. Adjacent epithelial cells are connected with the specialized septate junctions, and secretory granules accumulated at the apical pole are released to the lumen by the apocrine secretion.secretion.

THE CHANGES OF SALIVARY GLAND AFTER THE LIGATION OF THE EXCRETORY DUCT IN SUBMANDIBULAR GLANDS (악하선 분비관의 결찰 후 타액선 조직의 변화)

  • Obstructive adenitis of the salivary gland following salivary stone or infectious disease of the gland and surrounding tissues is a common disease. It is often difficult to decide whether to perform total excision of the gland or to consider conservative treatment. The present study was designed to investigate histological, histochemical changes of submandibular gland after ligating the excretory duct for identifying the results of gland duct obstruction. A group of 40 rat of Sprague-Dawley weighing about $200{\sim}220gm$ were used in the present study. 30 rats had ligation of the main excretory ducts of submandibular glands just at the exit from the glands. For controls, 10 rats had a sham operation without duct ligation. They were inducted into euthanagia state by intracardial Ketamine injection in 1, 2, 3, 5, 7, 14, 21, 28, 35 and 42 days after the ligation. In each ligation period, 3 animals were used for ligation and one animal was for control. The submandibular glands were dissected out at sacrifice and stained with H&E, PAS, mucicarmine stain and histological examination were carried out under the light microscope. After examination and comparison of all specimens, the results were as follows: 1. In the features of H&E stain, acini disappeared by degrees after the ligation of the excretary duct and interstitial cells were displaced into fibrous connective tissue. Salivary gland had been atrophied with enlarging ducts and proliferating ductal cells. 2. Through total experimental period, a lot of vessels were observed and the atrophy of serous gland was severer than that of mucous gland. 3. The deep portion of submandibular glands showed severe degeneration rather than superficial portion of them after the ligation. 4. The changes which had enlarged ducts and proliferating ductal cells were observed in entire gland and more prominent in serous gland than mucous gland after the ligation. 5. Although PAS and mucicarmine reactions were decreased gradually after the ligation with the lapse of time, since 2 to 3 weeks they were strong positive reactions on entire gland, especially on duct-like structure. So, we can suggest that salivary gland will be atrophied but, survived acini will be redistributed around the ducts.

  • Among the four kinds of silk glands in the funnel-web spider Agelena limbata, the ampullate gland for dragline silk production is the most predominate one in both sexes, and is composed of three functional parts-excretory duct, storage ampulla and convoluted tail regions. Two pairs of major ampullate glands send secretory ductules to the anterior spinnerets, and another two pairs of minor ampullate glands supply the middle spinnerets. There are no apparent differences between the major and minor ampullate glands not only the external spigots but also their internal silk glands. However, the microstructure is very unique in this spider, because each gland has spherical shaped storage sac with twig-like branched tails. Nevertheless, the wall of the secretory region is similarly composed of a single layer of epithelial cells. The mature secretory silks in glandular epithelium are closely packed and accumulated as electron-opaque vesicles. Most of the secretory products which originated from the rough endoplasmic reticula(rER) are grown up by fusion with the surrounding small vesicles however, the Golgi complex does not seem to play an important role in this process of secretion.

  • The ultrastructure of the excretory duct of the large ampullate gland in the orb web spider, Nephila clavain L. Koch are studied with light and electron microscopes. The excretory ducts of the large ampullate glands connected with the large spinning tubes(spigots) on the anterior spinnerets are basically composed of three superposed types of the layers which are inner cuticles, monolayered epithelial cells and peripheral connective cells. According to the morphological characteristics of the cuticles and internal textures of the epithelial cells, the long excretory ducts are subdivided into three(distal, middle and proximal) portions. Especially, at the distal portion of the ducts near the spinning tubes, the electron lucent subcuticles which had the functions of water removal and orientation of silk fibers are well distributed, whereas at the middle and proximal portions these layers disappeared and instead of these, endocuticles are developed. The endocuticle contains two types of bands, which are electron dense and electron lucent. And along the length of the cuticular stem in the excretory duct, these two alternating bands are twisted spirally. In the cytoplasm of the columnar epithelial cells of the distal portion, rough endoplasmic reticula and Golgj complexes, related to the production of the cuticular materials are well developed. Between the adjacent epithelial cells, specialized septate junctions and desmosomes are formed along the plasma membranes. At the proximal portion of the duct, densely accumulated secretory materials appeared, and these are released to the inner canal by the apocrine secretion.

  • The fine structure of the aggregate glands-one of the capture thread producing organs-in the orb web spider, Nephila clavata L.Koch, is studied with light and electron microscopes. Gluey capture threads or sticky spirals of the orb web are originated from the silks of two flagelliform glands and four aggregate glands which are connected to the posterior spinnerets, and the arrangement fo their spigots(large spinning tubes) shows a charaterstic form called "triad". The aggregate galnd is composed of large and multilobed secretory portion and thick excretory duct surrounded by large irregular nodules. The excretory duct of the aggregate galnds basically consists of three superposed types of cells which are inner columnar epithelium, nodule-forming cells and outer connestives. The cuticles of the proximal duct near the secretory portion are composed of endocuticle and exocuticle, whereas ghe distal duct near the spinning tubes has a electron lucent subcuticle which had the functions of water removal and orientation of silk fibers. In the cytoplasmic process of the large and irregular nodule-forming cells surrounded by invaginations of the plasma membranes, numerous mitochondria and glycogen particles are contained. The maturational level of the nodule cells is perceived from the appearence of these cell inclusions. The secretory portion of the glands which porduce the secretory silk material shows two layers of the cells which are simple cuboidal epithelium and several connective layers. In the cytoplasm of the glandular epithelial cell, rough endoplasmic reticulums are well developed, and two types of secretory granules are observed. ULTRASTRUCTURAL STUDY ON THE EFFECTS OF RETROGRADE INFUSION OF WATER-SOLUBLE CONTRAST MEDIA ON THE RABBIT SUBMANDIBULAR GLAND (수용성조영제의 주입이 가토악하선조직에 미치는 영향에 관한 전자현미경적 연구)

  • The author observed the effects of retrograde infusion of water soluble contrast media (Tele- brix 30) on the rabbit submandibular glands and compared the effects of different degrees of filling. 26 rabbits were divided into 2 groups of 12 each as experimentals and I group of 2 as normal controls. One experimental group was filled with 0.2㎖ and the other with 0.4㎖. Right submandibular gland of each rabbit was infused with contrast media and left one with physiologic saline as a experimental control, at a constant rate of 0.12㎖/min. using an infusion pump via the main excretory duct. Immediately after the infusion of contrast media, oblique lateral radiographs of the glands were made with occlusal film in order to confirm the glandular filling. The rabbits were sacrificed after varying periods (1, 8, 24 hours and 3, 6, 10 days) and the tissues were prepared for light and electron microscopic examination. The results were as follows: 1. In glands filled with 0.2㎖ contrast media, the initial changes were a few vacuole formation in the acini and slight dilation of the intralobular duct. The moderately severe changes such as vacuole formation in the acini, the abnormal substructure within the secretory granule, dilation of acinar and intercalated duct lumen, scalloping of striated duct lumen and inflammatory cell infiltrate were observed at 3 days. The general appearance was successively recovered, so the tissue had a normal appearance at 10 days. 2. In glands filled with 0.4㎖ contrast media, the most prominent alterations such as severe acinar atrophy, decreased number of secretory granules, proliferation of connective tissue stroma and pronounced inflammatory cell infiltrates appeared at 6 days. Although the general appearance returned to be almost normal at 10 days, acinar cells showed some atrophy and decreased secretory granules. 3. In glands subjected to 0.4㎖ infusion, the alterations were more severe and the recovery was slower than those seen in the glands to 0.2㎖ infusion.

  • Ultrastructure of the poison secreting organ in the spiders, Agelena limbata Thorell and Nephila clavata L. Koch were studied using scanning and transmission electron microscopes. The venom glands located its secretory sac portion in cephalothorax and excretory duct in the fang of chelicera are one pair of simple alveolar glands composed of three kinds of basic tissues-outer spiral musculature, middle myoepithelium and inner glandular epithelium. The muscle cells of the venom gland junctioned with the motor nerve endings at neuromuscular contact area are composed of smooth muscle fibers, whereas the myoepithelial cells between the musculature and inner glandular epithelium have compact collagenous fibers within the cytoplasm. The glandular epithelial cells which arranged along the concentrical location are subdivided into basal light cells and apical dark cells according to electron densities of their cytoplasms.
