Anxiolytic Effects of Woohwangcheongsimwon in Mice

  • Woohwangcheongsimwon (WHCSW) is a traditional oriental medicinal fomula which has been clinically used for treating strokes, palpitation, loss of consciousness and anxiety. The purpose of this study was to characterize the putative anxiolytic properties of WHCSW using an elevated plus-maze (EPM) and hole-board test. Control mice were orally treated with an equal volume of vehicle (10% Tween 80 solution), and positive control mice were treated with diazepam (1 mg/kg, i.p.). In the EPM test, WHCSW significantly increased the percentage of time-spent in the open arms (200 mg/kg, P < 0.05) and the percentage of open arm entries (200 and 400 mg/kg, P < 0.05). WHCSW also significantly increased the number of head-dips in the hole-board test (200 mg/kg, P < 0.05). In addition, the anxiolytic properties of WHCSW examined in the EPM test were inhibited by flumazenil (10 mg/kg, i.p.), a GABA$_A$ antagonist. However, no changes in spontaneous locomotor activity or myorelaxant effects were observed versus 10% Tween 80 controls. These results suggested that WHCSW is an effective anxiolytic agent, and that its anxiolytic effects are mediated via GABA$_A$ receptors.

Modeling the Fate of Priority Pharmaceuticals in Korea in a Conventional Sewage Treatment Plant

  • Understanding the environmental fate of human and animal pharmaceuticals and their risk assessment are of great importance due to their growing environmental concerns. Although there are many potential pathways for them to reach the environment, effluents from sewage treatment plants (STPs) are recognized as major point sources. In this study, the removal efficiencies of the 43 selected priority pharmaceuticals in a conventional STP were evaluated using two simple models: an equilibrium partitioning model (EPM) and STPWIN$^{TM}$ program developed by US EPA. It was expected that many pharmaceuticals are not likely to be removed by conventional activated sludge processes because of their relatively low sorption potential to suspended sludge and low biodegradability. Only a few pharmaceuticals were predicted to be easily removed by sorption or biodegradation, and hence a conventional STP may not protect the environment from the release of unwanted pharmaceuticals. However, the prediction made in this study strongly relies on sorption coefficient to suspended sludge and biodegradation half-lives, which may vary significantly depending on models. Removal efficiencies predicted using the EPM were typically higher than those predicted by STPWIN for many hydrophilic pharmaceuticals due to the difference in prediction method for sorption coefficients. Comparison with experimental organic carbon-water partition coefficients ($K_{ocs}) revealed that log KOW-based estimation used in STPWIN is likely to underestimate sorption coefficients, thus resulting low removal efficiency by sorption. Predicted values by the EPM were consistent with limited experimental data although this model does not include biodegradation processes, implying that this simple model can be very useful with reliable Koc values. Because there are not many experimental data available for priority pharmaceuticals to evaluate the model performance, it should be important to obtain reliable experimental data including sorption coefficients and biodegradation rate constants for the prediction of the fate of the selected pharmaceuticals.

Effects of Glycyrrhizae Radix on Repeated Restraint Stress-induced Neurochemical and Behavioral Responses

  • Glycyrrhizae radix (GR) is an herbal medicine that is commonly used in the East Asia for treating a variety of diseases, including stomach disorders. The objective of the present study was to examine the anti-stress effects of GR on repeated stress-induced alterations of anxiety, learning and memory in rats. Restraint stress was administered for 14 days (2 h/day) to the rats in the Control and GR groups (400 mg/kg/day, PO). Starting on the eighth day, the rats were tested for spatial memory on the Morris water maze test (MW) and for anxiety on the elevated plus maze (EPM). We studied the changes of the expressions of cholineacetyl transferase (ChAT) and tyrosine hydroxylase (TH) in the locus coerleus (LC) using immunohistochemistry. The results showed that the rats treated with GR had significantly reduced stress-induced deficits on their learning and memory on the spatial memory tasks. In addition, the ChAT immunoreactivities were increased. Gor the EPM, treatment with GR increased the time spent in the open arms (p<0.001) as compared to that of the control group. Moreover, GR treatment also normalized the increases of the TH expression in the LC (p<0.001). In conclusion, administration of GR improved spatial learning and memory and reduced stress-induced anxiety. Thus, the present results suggest that GR has the potential to attenuate the behavioral and neurochemical impairments caused by stress.

Anxiolytic-like Effects of Sanjoin-Tang Extracts and its Ingredients in the Elevated Plus-Maze in Mice

  • This study was carried out to evaluate the putative anxiolytic-1ike effects of the aqueous extracts of Sanjoin-tang (SJIT) and its ingredients using the elevated plus maze (EPM) test in mice. SJIT consists of five herbs, namely, Zizyphi Spinosi Semen (roasted), Glycyrrhizae Radix, Cnidii Rhizoma, Anemarrhenae Rhizoma, and Hoelen. The aqueous extracts of SJIT and each herbal drug were orally administered to ICR mice, 1 hr before evaluating behavioral activity in the EPM test, respectively. Repeated treatments (for 3 days) of the aqueous extract of SJIT (400 mg/kg) significantly increased time-spend in the open arms and arms entries into the open arms in the EPM test. Zizyphi Spinosi Semen (400 mg/kg), an ingredient of SJIT, significantly increased timespent in the open arms and arm entries into the open arms (P < 0.05). However, the other ingredient of SJIT did not show any anxiolytic-like behaviors. In addition, the anxiolytic-like effects of Zizyphi Spinosi Semen were blocked by pindolol (lO mg/kg), a $5-HT_{1A}$ receptor antagonist. These results suggest that Zizyphi Spinosi Semen (roasted) as an ingredient of SJIT plays a crucial anxiolytic role, and it acts via the serotonergic nervous system.

  • This paper presents a study on the accuracy position Controller design for the Composite Smoke Bomb Rotational driving system using a BLDC servo motor. Function of Smoke Bomb is blind in the enermy's sight so that need to high response. The BLDC servo motor controller was designed with DSP(TMS320VC33), IGBT(Insulated Gate Bipolar. Transistor), IGBT gate driver and CPLD(EPM7128). This paper implements those control with vector control and MIN-MAX PWM. Vector control requires information about rotor positions, a resolver should be used to achieve that. The main controller is implemented with a TMS320VC33 high performance floating-point DSP(Digital Signal Process) and PWM Generator is embodied using EPM7128.

Liriopis Tuber improves stress-induced memorial impairments in rats (구속 스트레스로 유발된 기억 손상에 대한 맥문동(麥門冬)의 신경보호 효능)

  • Objectives : We investigated the effects of Liriopis Tuber on the nervous protection. Methods : We measured the reaction behavior with EPM(Elevated Plus Maxe) and MWM(Morris Water Maze) after applying restraint stress to rats. Also, the degrees of AchE generation were measured with immunohistochemical method in the regions of hippocampus, and the degree of TH generation in the regions of VTA and LC, respectively. Results : 1. As the results of measuring how long EPM which reflects anxiety reaction stayed in the open arm, there was the trend which can suppress anxiety reaction in the MMD+stress group(Liriopis Tuber+stress) but no statistical significance. The counting results how many EPM passed between opened and closed arm showed suppression trend against a physical activity in the saline+stress group but there was no statistical significance. 2. According to the result of MWM, the saline+stress group showed the learning retardation which means increased time arriving at goal compared to ti1e normal group at the second and third day of measurement. On the contrary, a learning retardation was significantly decreased in the MMD+stress group. Among the probe trial test a memory loss occurred in the saline+stress group, meanwhile memory ability was notably increased in the saline-stress group. 3. The degree of TH generation was investigated at the VTA and LC respectively after test animals treated with drug. In the saline+stress group, TH-immunoreactive cells were significantly increased, and these stress-induced TH increases were suppressed in the saline+stress group at the VTA region. However, the saline+stress group did not show any significant difference. 4. the degree of AchE generation was investigated at the CA3 of hippocampus. The saline+stress group showed that AchE-immunoreactive cells were significantly decreased. Those stress-induced reductions of AchE cells were meaningfully recovered in the saline+stress group. Although the cells showed recovery trend in the region of CA1 of hippocampus, statistical significance was not observed. Conclusion : The results of our study indicate that Liriopis Tuber can improve spatial memory ability of rats applied a restraint stress. This improvement of the spatial memory ability is considered to have an efficacy of nervous protection that Liriopis Tuber suppresses the generation of AchE in the hippocampus region and enhances the generation of TH in the VTA.

Program Development to Evaluate Permeability Tensor of Fractured Media Using Borehole Televiewer and BIPS Images and an Assessment of Feasibility of the Program on Field Sites (시추공 텔리뷰어 및 BIPS의 영상자료 해석을 통한 파쇄매질의 투수율텐서 계산 프로그램 개발 및 현장 적용성 평가)

  • A computer program to numerically predict the permeability tensor of fractured rocks is developed using information on discontinuities which Borehole Televiewer and Borehole Image Processing System (BIPS) provide. It uses orientation and thickness of a large number of discontinuities as input data, and calculates relative values of the 9 elements consisting of the permeability tensor by the formulation based on the EPM model, which regards a fractured rock as a homogeneous, anisotropic porous medium. In order to assess feasibility of the program on field sites, the numerically calculated tensor was obtained using BIPS logs and compared to the results of pumping test conducted in the boreholes of the study area. The degree of horizontal anisotropy and the direction of maximum horizontal permeability are 2.8 and $N77^{\circ}CE$, respectively, determined from the pumping test data, while 3.0 and $N63^{\circ}CE$ from the numerical analysis by the developed program. Disagreement between two analyses, especially for the principal direction of anisotropy, seems to be caused by problems in analyzing the pumping test data, in applicability of the EPM model and the cubic law, and in simplified relationship between the crack size and aperture. Aside from these problems, consideration of hydraulic parameters characterizing roughness of cracks and infilling materials seems to be required to improve feasibility of the proposed program. Three-dimensional assessment of its feasibility on field sites can be accomplished by conducting a series of cross-hole packer tests consisting of an injecting well and a monitoring well at close distance.

Anxiolytic activity of standardized extract of Korean ginseng - a study on exploratory behavior

  • The roots of the plant Korean ginseng have been extensively used in the traditional Chinese herbal medicine. We investigated the standardized extract of Korean ginseng on animal models of anxiety based on exploratory behavior. Korean ginseng extract (KGE) (3, 10 and 30 mg/kg) was administered intra-peritoneally. The anxiolytic activity was studied using elevated plus maze (EPM) paradigm, light/dark apparatus (LDA), open field apparatus (OFA) and the hole board apparatus (HBA). Diazepam (1mg/kg) was used as a standard anxiolytic drug. In EPM, KGE (10 mg/kg) significantly (P < 0.05) increased the time spent in open arms and the number of entries in open arms. In LDA, KGE (10 mg/kg) increased the number of transitions. In OFA, KGE (3 and 10 mg/kg) significantly increased (P < 0.05) the number of squares traversed. In HBA the number of head pokes were significantly increased with KGE (3 and 10 mg/kg). KGE at all selected doses did not affect the motor coordination. Thus, the study suggests that saponin containing standardized Korean ginseng extract possess anxiolytic activity.

Antidepressant Effects of Gammakdaejo-Tang on Repeated Immobilization Stress in the Ovariectomized Female Rats

  • Gammakdaejo-Tang (GMT) is a traditional oriental medicinal formula, a mixture of 3 crude drugs, and it has been clinically used for treating mild depressive disorders. The purpose of the study was to examine the effect of Gammakdaejo-Tang (GMT) on repeated stress-induced alterations of learning and memory on a passive avoidance test (PAT) test and also the anxiety-related behavior on the elevated pulse maze (EPM) in ovariectomized female rats. We assessed the changes in the reactivity of the cholinergic system by measuring the immunoreactive neurons of choline acetyltransferase (ChAT) in the hippocampus after behavioral testing. The rats were exposed to the immobilization (IMO) stress for 14 days (2hours/day), and Gammakdaejo-Tang (400 mg/kg, p.o.) was administered 30 min before IMO stress. Treatments with GMT caused significant reversals of the stress-induced deficits in learning and memory on a working memory test, and it also produced an anxiolytic-like effect on the EPM, and increased the ChAT reactivities (p<0.001, respectively). These results suggest that Gammakdaejo-Tang might prove to be an effective antidepressant agent.

Analysis and design of a FSK Demodulator with Digital Phase Locked Loop (디지털 위상고정루프를 이용한 ESK복조기의 설계 및 성능 분석)

  • In this paper, FSK(Frequency Shift Keying) demodulator which is widely used for FH-SS system is designed and the experimental results are analyzed. The performance of the ADPLL(All-digital Phase-Locked-Loop), which is the main part of the demodulator circuit, is analyzed by the computer program. Using Maxplus-II tool provided by altera. co., ltd, each part of the ADPLL is designed and all of them is integrated into EPM7064SLC44-10 chip. And the simulation results are compared with the characteristics of the implemented circuits for analysis. There is about 2${\mu}\textrm{s}$ difference in time constant of the PLL. This difference is not critical in the demodulator. And the experimental results show that the transmitted data is well demodulated when the phase difference between the FSK modulated signal and the reference signal is about 180 degree.
