고당질 식이시 n6 와 n3 불포화 지방산이 쥐의 혈장지질 저하기전에 미치는 영향 (Plasma Lipid-Lowering Effect of n6 and n3 Polyunsaturated Fatty Acids in Rats Fed High Carbohydrate Diet)

  • To compare the hypolipidemic effects of n6 linoleic acid. n3 $\alpha$-linolenic acid and n3 eicosapentaenoic acid in rats fed high carbohydrate(70% Cal) diet. male Sprague Dawley rats were fed different experimental diets for 6 weeks. which were different only in fatty acid composition. The dietary fats were beer tallow(BT) as a source of saturated fatty acid (SFA), corn oil(CO) for n6 linoleic acid(LA), perilla oil(PO) for n3 $\alpha$-linolenic acid(LL) and fish oil(FO) for n3 eicosapentaenoic acid(EPA) and docosahexaenoic acid (DHA) Plasma total cholesterol(Chol) level was increased by n6 LA but decreased by n3 LL and n3 EPA and most effectively reduced by n3 EPA. HDL-Chol level was raised by n6 LA, but there was no significant change in HDL-Chol levels by n3 LL and was lowered by n3 EPA. Plasma TG level was reduced by n6 LA, but lipogenesis in liver was not affected by n6 LA. However, plasma TG level was lowered by n3 LL and EPA. Both lipogenic enzyme activity and liver TC level were also decreased by n3 PUFA. The relative proportions of TG in VLDL was significantly lowered by n3 EPA. but the proportions of Apo B in VLDL was not changed by n3 EPA. Overall. the hypolipidemic effect was in the order of EPA+ DHA(n3) >LL(n3) >LA(n6) and fish oil and perilla oil rich in n3 PUFA may have important nutritional applications in the prevention and treatment of hypertriglyceridemia.

Eicosapentaenoic and Docosahexaenoic Acids Reduce Arachidonic Acid Release by Rat Kidney Microsomes

  • The effects of eicosapentaenoic (EPA, 20:5n-3) and docosahexaenoic acids (DHA, 22:6n-3) on the phospholipase $A_2$ ($PLA_2$)-mediated release of arachidonic acid (AA, 20:4n-6) were studied in kidney microsomes from rats fed diets containing sunflower oil (SO) or fish oil (FO) concentrate for 11 months. The amounts of AA released by the endogenous $PLA_2$ enzyme were significantly lower by 38% in the FO, compared to the SO-fed rats (23.2 nmol versus 60.7 nmol AA released/mg protein/h in the FO- and SO-treated groups, respectively). The FO-derived microsomes released less linoleic acid (LA, 18:2n-6) and adrenic acid (22:4n-6), but larger amounts of the n-3 fatty acids, including EPA, DHA, docosapentaenoic acid (DPA, 22:5n-3), and 20:4n-3 than the SO-derived microsomes. A similar replacement of the AA and adrenic acid with the n-3 fatty acids including EPA and DHA was also observed in the microsomal phospholipid fraction from the FO-fed rats relative to the SO-treated group. The results suggest that the $PLA_2$-mediated release of AA is reduced and that of EPA is increased in compensation for AA decline in kidney microsomes from FO-fed rats (0.7 nmol EPA/mg protein/h versus 22.7 nmol EPA/mg protein/h for the SO and FO-treated groups). Replacement of the n-6 with n-3 fatty acids may explain the reduced synthesis of the AA-derived prostaglandins and the concomitant rise in the EPA-derived prostaglandins observed in kidneys of FO-treated rats.

식이지방 수준에 따라 n6 와 n3 계 불포화지방산이 혈장 지질수준에 미치는 영향에 관한 비교연구 (Differential Effect of n6 and n3 Polyunsaturated Fatty Acids on Plasma Lipids in Rats Fed Low and High Fat Diets)

  • 식이지방의 수준에 따라 n6 와 n3 불포화지방산이 혈장 지질조성에 미치는 영향과 또 그 기전이 다른지를 연구하고자 Sprague Dawley 종 수컷쥐에게 저지방(LF, 10% Cal)식이와 고지방식이(HF, 40% Cal)를 각각 6주동안 투여하였으며, 사용된 기름은 포화지방산 급원으로는 쇠기름, n6 linoleic acid(LA) 급원으로 corn oil, n3 ${\alpha}-linolenic$ acid(LL) 급원으로 perilla oil, n3 eicosapentaenoic acid(EPA)와 docosahexaenoic acid(DHA) 급원으로 fish oil이었다. Ultracentrifugation 방법으로 VLDL fraction을 분리하여 thin layer chromatography에 의해서 화학적조성을 구하였다. Plasma cholestere 수준은 LF 와 HF 모두 n6 LA에 의해서는 오히려 증가되었고 n3 LL과 n3 EPA에 의해서는 감소되었으며, HF 에서 n3 EPA가 가장 cholesterol 저하효과가 있었다. HDL-Chol 농도는 n6 LA에 의해서 증가되었으나, HF 경우 n3 EPA에 의해서 유의성있게 감소되었다. Plasma TG 농도는 n3 EPA에 의해서 가장 감소되었고 간의 lipogenic enzyme 활성을 억제하였으며 VLDL fraction의 TG상대적양(%)이 유의성 있게 낮아졌다. 이때 LF 군에서는VLDL fraction의 apo-B의 상대적양(%)이 감소되지 않았으나 HF군에서 n3 EPA에 의해서 유의성 있게 낮았다. 그러므로 n3 EPA의 hvpotriglyceridemic effect는 간에서 lipogenesis를 억제하여 plasma VLDL로 TG 분비가 억제되었을 것이며, HF일 경우는 간에서 apo-B 생성도 억제되었다. 관상동맥성심장질환의 예방적 차원에서 평상시에도 쇠기름과 corn oil 보다는 n3 PUFA가 풍부한 들기름이나 생선을 더욱 이용하는 것이 바람직하다.

Dietary Reference Intake of n-3 polyunsaturated fatty acids for Koreans

  • This paper examines the process and evidence used to create the Dietary Reference Intake (DRI) of alpha-linolenic acid (ALA) and eicosapentaenoic acid (EPA) + docosahexaenoic acid (DHA) for Koreans. ALA (18:3n3) is an essential fatty acid, and EPA and DHA are known to have beneficial effects on cardiovascular disease risk and reduction of triglyceride levels. Various international organizations have suggested dietary recommendations for n-3 polyunsaturated fatty acids (PUFAs), including ALA, EPA, and DHA. A DRI for Koreans was established for the first time in 2020, specifically for the adequate intake (AI) of ALA and EPA + DHA. This recommendation was based on the average intake of ALA and EPA + DHA from the Korea National Health and Nutrition Examination Survey 2013-2017. For Korean infants, the AI of ALA and DHA was based on the fatty acid composition of maternal milk. Estimated average requirement and a tolerable upper intake level have not been set for n-3 PUFA due to insufficient evidence. In addition, the intake level of n-3 PUFA for prevention of chronic disease has also not been determined. Future studies and randomized controlled trials are required to establish the UL and to define the level for disease prevention.

Eicosapentaenoic Acid, Docosahexaenoic Acid 농축어유와 들깨유가 저지방 식이를 섭취한 흰쥐의 지질대사에 미치는 영향 (The Effect of Dietary Concentrated Oils of Eicosapentaenoic Acid, Docosahexaenoic Acid and Perilla Oil on lipid Metabolism in Rata Fed Low Fat Diet)

  • To compare the effect of three kinds of n-3 fatty acids-eicosapentaenoic acid(EPA), docosahexaenoic acid(DHA) and perilla oil (PO)-on serum and liver lipid levels and fatty acid composition of liver phospholipid(PL) at low fat level(5%, w/w), 4-weeks old Sprague-Dawley rats were fed with one of five different oil diets for 4 weeks. Beef tallow(BT) and corn oil(CO) was used as control for sturated or n-6 fatty acid respectively. Se겨m concentrations of cholesterol(TC) and phospholipid(PL) were lower in PO DHA and EPA groups than in BT and CO groups. HDL-cholesterol levels were higher in CO and PO groups than in EPA, DHA and BT groups. Liver PL concentrations were higher in DHA and EPA groups than in CO, PO and BT groups, but liver TC and heal PL and TC concentrations did not show any significant difference among groups. Hepatic fatty acid composition of phosphatidylcholine (PC) and phosphatidylethanolamine (PE), two major phospholipids in liver, reflected their dietary fatty acid composition. In PC and PE, total percentage own-6 series was higher in CO group than in any other groups, and that own-3 series was higher in DHA and PO groups than in EPA, CO and BT groups. Moreover, the ratio of 20 : 4/18 : 2 was lower in PO and DHA groups than in EPA, CO and BT groups. On the contrary, the percentage of C22 : 6 was lower in EPA, CO and BT groups than in PO and DHA groups. These results revealed that n-3 series(EPA, DHA and PO) were more effective in lowering um lipids than n-6 fatty acids or saturated fatty acid. Based on the results of fatty acid composition of hepatic phospholipid, we suggest that the dietary effect of PO and DHA on antiatherogenic characteristics seems to be similar extent. In addition, the effect of EPA might not be significantly different from that of BT or CO in the view of eicosanoids production from the precursor fatty acid. These difference of hepatic fatty acid composition might come from other characteristics of dietary oil as well as the type of unsaturation, not from the carbon chain length or the degree of unsaturation of n-3 fatty acid.

Effect of Deoxygenizer on the Suppression of Lipid Deterioration of Boiled and Dried-Anchovy Engraulis japonica

  • The effects of deoxygenizer (Deoxy) and sodium-erythorbate (Na-ery) on the changes in fatty acid compositions were investigated to prevent the loss of n-3 polyunsaturated fatty acids in lipid of bolied and dried-anchovy during storage. After storage for 5 months, docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) of total lipid (TL) in Deoxy group decreased only $3.0\%\;and\;0.5\%$, respectively, compared to those of before storage. However, those in Control group decreased 9.5\%\;and\;2.3\%,$ respectively. In the case of Na­ery group, the percentages of DHA and EPA decreased were lower than those of Control. Most of DHA and EPA in n was lost in the early stage of storage. Total DHA remained in phospholipid (PL), triglyceride (TG), and free fatty acid (FFA) fractions after storage for 5 months were $98\%,\;66\%\;and\;62\%$ in Deoxy, Na-ery and Control group, respectively, and total EPA remained was slightly high level compared with those of DHA. The loss of DHA was more in PL than in TG. Consequently, deoxygenizer was recognized as a good material to prevent the loss of DHA and EPA of the boiled and dried-anchovy during storage.

황소개구리에서 추출한 지방의 지방산 조성 (Fatty Acid Compositions of Lipids Extracted from Bullfrogs)

  • 본 연구에서는 황소개구리(Rana catesbeiana Shaw)가 불포화지방산의 주요 공급원이 될 수 있다는 점에 착안하여, 국내에서 서식하고 있는 황소개구리에서 지방을 추출하여 지방산 조성을 분석하였다. 황소개구리의 다리와 몸통에는 1%(w/w)이하의 지방이 함유되어 있었으며, 계절에 따른 변화를 관찰할 수 없었다. 황소개구리 다리에서 분리한 지방에는 26~31%(w/w) 포화지방산, 16~24% 모노엔산, 30~40% 고도불포화지방산이 함유되어 있었으며, 몸통에서 분리한 지방에는 23~28% 포화지방산, 29~44% 모노엔산, 16~30% 고도불포화지방산이 함유되어 있었다. 황소개구리에서 추출한 지방의 주요 지방산은 palmitic acid, oleic acid, linoleic acid이었다. 다리에서 분리한 지방에는 3~8% eicosapentaenoic acid(EPA)와 6~10% docosahexaenoic acid(DHA)가 함유되어 있었으며, 몸통에서 분리한 지방에는 1~3% EPA 와 DHA가 각각 함유되어 있었다.

Growth and Fatty Acid Composition of Juvenile Olive Flounder Paralichthys olivaceus Fed Diets Containing Different Levels and Ratios of Eicosapentaenoic Acid and Docosahexaenoic Acid

  • This study was carried out to investigate the influences of dietary levels, ratios and sources of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) on the growth and fatty acid compositions of juvenile olive flounder Paralichthys olivaceus. Sixteen diets containing five levels of EPA (0.5%, 1.0%, 1.5%, 2.0%, and 4.0%), five levels of DHA (0.5%, 1.0%, 1.5%, 2.0% and 4.0%), three ratios of EPA/DHA (75/25, 50/50 and 25/75), two levels of squid liver oil (5% and 10%) and a control diet containing 5% soybean oil were hand-fed to triplicate groups of fish (average weight, $9.7{\pm}0.3g$) for 8 weeks. Survival, specific growth rate, feed efficiency and protein efficiency ratio of fish were not affected by dietary EPA and DHA levels or ratios. Also, the dietary treatment had no significant effect on the lipid and protein contents of muscle and whole body of fish. A corresponding increase in the EPA and DHA contents of fish occurred with increasing EPA and DHA levels in their diets. Our results suggest that juvenile olive flounder require a dietary EPA level of approximately 0.32% in the presence of 0.74% DHA for suitable survival and growth, and that EPA and DHA levels in fish muscle can increase to as much as 32% and 53%, respectively, of the total fatty acid content.

n-3계 지방산 투여가 성장기 흰쥐의 간장 및 혈청 Triacylglycerol 농도에 미치는 영향 (Effects of ${\alpha}-linolenic$, eicosapentaenoic and docosahexaenoic acids administration on lowering of triacylglycerol level in the hepatic and serum of rats)

  • 식이성지방 n-6/n-3비(1 : 2) 및 포화지방산:단순불포화지방산:다가불포화지방산비(1:1:1)를 일정하게 조정한 조건하에서 n-3계 지방산, ${\alpha}-LA(C_{18:3}),\;EPA(C_{20:5}),\;DHA(C_{22:6})$를 함유시킨 식이를 조제하여 성장기 SD-계 흰쥐 수컷에 2주간 급여하여 간장, 혈청의 Triacylglycerol(TG)농도 및 간장 TG 합성에 관여하는 효소 활성에 대하여 검토하였다. 본 실험에 사용한 EPA 및 DHA 는 순도 98%로서 고도로 정제한 ethyl ester 형태로 식이에 첨가하였다. 그 결과, ${\alpha}-LA$군에 비하여 EPA군 및 DHA군에서 간장 및 혈청 TG 농도는 유의적으로 감소하였으나, 총콜레스테롤 농도는 유의차가 인정되지 않았다. 간장의 인지질 농도는 DHA군에서만 유의적으로 증가하였다. 간장 막 결합성 phosphatidate phosphohydrolas(PAP)성은 간장TG 농도와 같은 경향으로서 EPA와 DHA군에서 유의하게 감소하였으며 Diacylglycerol acyltransferase(DGAT) 활성은 각 군간에는 변화가 없었다. 따라서, 간장TG 농도와 간장 막결합성 PAP활성과의 사이에 높은 상관관계(r=0.84)를 나타내어 간장TG 합성억제가 인정되었다. 지방산 합성계 효소의 활성도는 EPA및 DHA군에서 감소하였다. 이상의 결과로부터 n-3계 지방산, EPA 및 DHA에 의한 간장 및 혈청 TG 농도의 감소는 막 결합형 PAE 활성과 저해에 의한 간장TG 합성 및 지방산 합성 저해가 관여하는 것이 시사되었다.

에이코사펜타엔산(EPA) 항산화활성 연구 (Eicosapentaenoic Acid (EPA) Antioxidant Activity Study)

  • 본 연구에서는 천연 생리활성물질인 에코사펜타엔산을 다량 함유한 흑해삼의 항산화 효과를 조사하여 기능성 화장품 원료로서의 우수성을 제시하였다. 세포내 산화질소(NO) 생성 억제 효과에 대해 해삼 추출물의 활성산소 제거능을 평가하였다. 해삼의 생리활성물질은 활성산소를 제거하는 항산화 효과가 있으며, 평가 결과 우수한 NO 생성이 억제됨을 확인하였다. 연구에 따르면 생리활성 물질은 낮은 농도에서도 높은 항산화 효능을 보여 항산화 효과, 면역 및 염증 반응 조절에 관여하는 것으로 나타났습니다. 해삼의 항산화 제거능은 해삼 추출물의 EPA(eicosapent aenoic acid)를 함유하는 페놀 성분의 항산화 효능에 크게 기여하는 것으로 생각된다. 해삼의 항산화 성분이 작용한다는 보고와 유사한 것으로 밝혀졌다. 또한 해삼 추출물에 다량 함유된 에이코사펜타엔산(EPA) 성분이 염증 활성 억제 효과에 관여하는 것으로 입증돼 상처 치유에도 탁월하다. 본 연구의 결과, 해삼 추출물에 함유된 페놀 성분의 효과가 항산화 활성과 밀접한 인과관계가 있음을 확인하였다.
