Rotordynamic Design and Analysis of the Rotor-Bearing System of a 500Wh Flywheel Energy Storage Device (플라이휠 에너지 저장장치 회전체계의 동역학적 설계및 해석)

  • A 500Wh class high-speed Flywheel Energy Storage System (FESS) driven by a built-in BLDC motor/generator has been designed, which runs from 30000 to 60000rpm nominally. Due to the motor/generator inside, the flywheel rotor made of composites supported by PM/EM hybrid bearing system has a shape of bell or pendulum and thus requires accurate rotordynamic analysis and prediction of its dynamic behavior to secure the operating reliability. Rotordynamic analyses of the flywheel rotor-bearing system revealed that the bell shaped rotor has two conical rigid-body modes in the system operating range and the first conical mode, of which nodal point lies in the radial EM bearing position, can adversely affect the dynamic response of the rotor at the corresponding critical speed. To eliminate the possibility of wild behavior of the rotor, two guide bearings are adopted at the upper end of the rotor and motor/generator. It was also revealed that the EM bearing stiffness if 0.5~1.0E+6 N/m and damping of 2000 Ns/m are favirable for smooth operation of the system around the 2nd critical speed.

  • The equivalent circuits of Open End and Step discontinuity effects for Suspended Strip line are obtained. The circuit parameters for each discontinuity effect are represented by equations after optimization based on EM analysis results, and the coefficients of the equations are obtained by curve fitting for all impedance cases.

  • A Linear induction electromagnetic(EM) pump of liquid metal fast breeder reactor(LMFBR) is used for the purpose that the liquid metal of high temperature is transported by EM force. This paper evaluates magnetic flux density necessary for transporting liquid metal, using analytical model of the linear induction EM pump. Using the 2-D finite element method(2-D FEM), magnetic flux density is estimated in consideration of a geometric model, electric parameter, and velocity of liquid metal. From the viewpoint of hydrodynamics, the results can be used for flow analysis of the liquid metal.

  • Inhibition of the growth of human cancer cells by proteins secreted from 373-L1 cells was investigated in the present study. The growth of human cancer cells was inhibited by co-culture with 373-L1 cells under 10% FBS and DME, DME, GIT and serumless medium, respectively. The conditioned medium of cultured 373-L1 cells under serumless medium was concentrated 100-fold through an ultrafiltration cell with a 10,000 molecular weight cutoff at 4$^{\circ}C$ under positive pressure using nitrogen(373-L1 EM). 373-L1 EM inhibited the growth of HeLa, Hep G 2, KHOS-Np, A43l and MCF-7 cells. 3T3-L1 EM was purified with FPLC, DEAE-ion exchange chromatography and phenyl-sepharose chromatography. The major protein of 373-L1 EM has a molecular weight of 66,000-68,000 in SDS-PAGE analysis. The results suggest that the inhibitory activity of 373-L1 EM appears to be due to some protein(m.w.66,000-68,000) secreted by 373-L1 cells.

Influence of Growth Conditions on Plasmid DNA Production

  • The obtention of high yields of purified plasmid DNA is viewed as an essential issue to be considered towards efficient production of DNA vaccines and therapeutic plasmids. In this work, Escherichia coli $DH5\alpha$. bearing the pVAXI-LacZ plasmid was grown in a developed semi-defined medium at different temperatures and tryptone concentrations. Analysis of pDNA yields and E. coli morphology revealed that at higher temperatures (37 and $40^{\circ}C$), higher specific yields and E. coli filamentation were obtained. However, the best results were achieved when a lower tryptone concentration was used. This approach was shown to be a powerful tool to promote plasmid amplification, keeping the desirable plasmid structure, and favoring the attainment of quality. Our results suggest that by using tryptone alone as an amino acid source, pDNA amplification was improved and a specific yield of 20.43 mg pDNA/g dcw was achieved, proving that this strategy can improve pDNA yield even at a small scale.

Analysis of EM Penetration Problems in Complex Structures Using Finite-Difference Time-Domain Method (FDTD 방법을 이용한 복잡한 구조물에서의 전자파 침투 특성 해석)

  • In this paper, we analyzed the radiation patterns of a monopole antenna mounted on cylinder and EM penetration problems in the complex structures by using FDTD method associated with 3-D PML absorbing boundary condition. In order to validate the proposed FDTD code, the radiation patterns of monopole antenna mounted on cylinders were compared with the exact Carter's solutions. As a results, the predicted radiation pattern exhibited excellent agreement with exact solution. And the FDTD code is applied to analyze the EM penetration problems in complex structures, Blackhawk helicopter. As the plane wave is excited, a significant amount of energy penetrates the helicopter structure, and it is dependent on aperture/airframe interface.

SoEM: a novel PCR-free biodiversity assessment method based on small-organelles enriched metagenomics

  • DNA metabarcoding is currently used for large-scale taxonomic identification to understand the community composition in various marine ecosystems. However, before being widely used in this emerging field, this experimental and analytic approach still has several technical challenges to overcome, such as polymerase chain reaction (PCR) bias, and lack of well-established metabarcoding markers, a task which is difficult but not impossible to achieve. In this study, we present an adapted PCR-free small-organelles enriched metagenomics (SoEM) method for marine biodiversity assessment. To avoid PCR bias and random artefacts, we extracted target DNA sequences without PCR amplification from marine environmental samples enriched with small organelles including mitochondria and plastids because their genome sequences provide a valuable source of molecular markers for phylogenetic analysis. To experimentally enrich small organelles, we performed subcellular fractionation using modified differential centrifugation for marine environmental DNA samples. To validate our SoEM method, two marine environmental samples from the coastal waters were tested the taxonomic capturing capacity against that of traditional DNA metabarcoding method. Results showed that, regardless of taxonomic levels, at least 3-fold greater numbers of taxa were identified in our SoEM method, compared to those identified by the conventional multi-locus DNA metabarcoding method. The SoEM method is thus effective and accurate for identifying taxonomic diversity and presents a useful alternative approach for evaluating biodiversity in the marine environment.

Production of 4-Ethyl Malate through Position-Specific Hydrolysis of Photobacterium lipolyticum M37 Lipase

  • Microbial lipases are used widely in the synthesis of various compounds due to their substrate specificity and position specificity. 4-Ethyl malate (4-EM) made from diethyl malate (DEM) is an important starting material used to make argon fluoride (ArF) photoresist. We tested several microbial lipases and found that Photobacterium lipolyticum M37 lipase position-specifically hydrolyzed DEM to produce 4-EM. We purified the reaction product through silica gel chromatography and confirmed that it was 4-EM through nuclear magnetic resonance analysis. To mass-produce 4-EM, DEM hydrolysis reaction was performed using an enzyme reactor system that could automatically control the temperature and pH. Effects of temperature and pH on the reaction process were investigated. As a result, 50℃ and pH 4.0 were confirmed as optimal reaction conditions, meaning that M37 was specifically an acid lipase. When the substrate concentration was increased to 6% corresponding to 0.32 M, the reaction yield reached almost 100%. When the substrate concentration was further increased to 12%, the reaction yield was 81%. This enzyme reactor system and position-specific M37 lipase can be used to mass-produce 4-EM, which is required to synthesize ArF photoresist.

Effects of Aloe-emodin and Emodin on Proliferation of the MKN45 Human Gastric Cancer Cell Line

  • Aloe-emodin (1, 8-dihydroxy-3-hydroxyl-methylanthraquinone; AE) and emodin (1,3,8-trihydroxy-6-methylanthraquinone; EM) are anthraquinone derivatives that have been detected in some medical plants and share similar anthraquinone structures. AE and EM have been shown to exhibit anticancer activities in various cancer cell lines; however, the inhibitory effects of these derivatives on the growth of cancer cells were previously reported to be different. Gastric cancer is the second most common cause of cancer cell death worldwide. In the present study, we examined the inhibitory effects of 0.05 mM AE and 0.05 mM EM on the proliferation of the MKN45 human gastric cancer cell line. The proliferation of MKN45 cells was significantly inhibited in AE- and EM-treated groups 24 h and 48 h after treatment. Furthermore, the inhibitory effects of EM were stronger than those of AE. The cell cycle of MKN45 cells were arrested in G0/G1 phase or G0/G1 and G2/M phases by AE and EM, respectively. However, an analysis of intracellular polyamine levels and DNA fragmentation revealed that the mechanisms underlying cell death following cell arrest induced by AE and EM differed.

Electromagnetic Survey in Korea (한국의 전자탐사 현황)

  • Electromagnetic(EM) survey has been in use for over a half century as a standard routine for, mineral exploration in many parts of the world. But EM survey work and serious research effort were initiated in Korea only as late as in early 1980s, largely inspired by four pioneers who did their graduate studies in the U.S.A. in 1970s. Nevertheless domestic achievements in the field of EM survey are remarkable in the last two decades: the field operations and related interpretational skills appear to have reached a global standard, even compared with the most advanced in other countries, virtually in a whole spectrum of the method which includes magneto-tellurics(MT), Controlled Source Audio-frequency Magneto-tellurics(CSAMT), geomagnetic sounding, small loop survey systems, Very Low Frequency(VLF), Ground Penetrating Radar(GPR), time domain surveys, and noise analysis. Besides mineral exploration, EM survey has been applied in Korea to hydrogeology, geotechnical engineering, non-destructive investigation of structures, unexplored ordnance(UXO) investigation, environmental monitoring, and archaeological investigation as well. Now that original contributions of several Korean geophysicists are found even in new frontiers such as high-frequency EM survey, investigation in time-domain EM field for buried metal objects and structures, and also modem data inversion scheme, it is duly hoped that they make some technical breakthrough to unravel still entangled knots of EM survey method in a forseeable future.
