  • A more rapid and efficient method to extract RNA from Ecklonia cava Kjellman (Laminariales, Phaeophyceae) was introduced in this study. Each step of the procedure was evaluated and the optimal concentration of each chemical in the lysis solution was determined. Tissue pulverization with PVPP and β-mercaptoethanol in the lysis solution were not essential for RNA extraction of this species. The highest yield and purity of E. cava RNA were obtained by the lysis solution containing 1% CTAB, 1 M NaCl, 0.7% PVP, 10mM EDTA and 100mM Tris-Cl (pH 9.0). Approximately 8μg of RNA was obtained from 200 mg of ground tissue. The ratios of the absorbance at 260 nm and 280 nm were from 1.6 to 1.8 and those of at 230 nm and 260 nm were from 1.8 to 2.0. The extracted RNAs obtained in this study turned out to have a sufficient quality for cDNA synthesis.

  • Leaf mesophyll protoplasts from five cultivars of tobacco (N. tabacum L.) were cultured. The protoplasts did not survive in culture medium containing Murashige and Skoog inorganic salts for over 6 days. NH4NO3 and FeSO4.Na2EDTA concentration of Murashige and Skoog medium were toxic in tobacco leaf mesophyll protoplast culture. Therefore we investigated optimum condition in Murashige and Skoog medium. High plating efficiency was obtained by reducing the concentrations of NH4NO3 and FeSO4.Na2EDTA to 1/3 and 1/10, respectively, on the supplemented with 5$\mu$M IAA, 0.5 $\mu$M 2.4-D 5 $\mu$M BAP. Plants were regenerated from protoplast-derived calluses.

  • Objectives: To determine the effect of root canal irrigants on the hydrophobicity and adherence of Staphylococcus epidermidis (S. epidermidis) to root canal dentin in vitro. Materials and Methods: Root dentin blocks (n = 60) were randomly divided into 4 groups based on the irrigation regimen: group 1, saline; group 2, 5.25% sodium hypochlorite (NaOCl); group 3, 5.25% NaOCl followed by 17% ethylenediaminetetraacetic acid (EDTA); group 4, same as group 3 followed by 2% chlorhexidine (CHX). The hydrophobicity of S. epidermidis to root dentin was calculated by cell surface hydrophobicity while the adherence was observed by fluorescence microscopy, and bacteria were quantified using ImageJ software (National Institutes of Health). Statistical analysis of the data was done using Kruskal-Wallis test and Mann-Whitney U test (p = 0.05). Results: The hydrophobicity and adherence of S. epidermidis to dentin were significantly increased after irrigating with group 3 (NaOCl-EDTA) (p < 0.05), whereas in group 4 (NaOCl-EDTA-CHX) both hydrophobicity and adherence were significantly reduced (p < 0.05). Conclusions: The adherence of S. epidermidis to dentin was influenced differently by root canal irrigants. Final irrigation with CHX reduces the bacterial adherence and may impact biofilm formation.

  • In order to choose a suitable soil extractant for the prediction model of heavy metal content in brown rice, four extractants-0.1 M HCl, 0.1 M $HNO_3$, 0.1 M $NH_4$-oxalate and 0.001 M 2Na-EDTA, were compared by analyzing 84 soil and 45 brown rice samples collected from paddy fields adj-acent to five old zinc-mining sites. Content of cadmium and zinc in brown rice had the highest correlation coefficient to 0.001 M 2Na-EDTA and 0.1 M HCl extractants, respectively. However, the lead content in brown rice was significantly correlated with only 0.1 M $NH_4$oxalate solution. For the simultaneous prediction of zinc, cadmium and lead in brown rice, 0.1 M $NH_4$-oxalate solution was the most effective. On the multiple analysis by using various chemical characteristics of soils, pH and calcium content of soils were effective variables for the estimation of cadmium content in brown rice, while CEC and magnesium content were more effective for the estimation of zinc content in brown rice. Furthermore, for the estimation of lead content in brown rice, factors such as pH, CEC, calcium, magnesium, potassium and organic matter content were important variables in the multiple regression equation.

  • Alteration of physical and chemical characteristics and the effect of removal of mineral substances in waterlogged archaeological woods by different cleaning processes were examined using oak wood(Quercus spp.) that was excavated from wetland near Gwangju, Korea. Cleaning methods employed in the present work were (1) tools, (2) deaeration, (3) EDTA and (4) ultrasonic cleaning, which are being currently applied in the field of preservation treatment. Cleaning process were performed independently or continuously. Composition of mineral substances in the waterlogged archaeological wood was almost same as the that of soil in which waterlogged archaeological woods were buried. In case of independent cleaning, tools cleaning efficiently removed the mineral substances on surface. Surface color become brighter after cleaning with EDTA. In contrast, deaeration and ultrasonic cleaning did not show any significant removal of mineral substances. In continuous cleaning process, tool cleaning as the first step treatment showed the same effect as shown in independent cleaning. Although deaeration as the second step cleaning did not remove the mineral substances, it could be assumed to contribute the infiltration of dimensional agents by homogenization of wood. EDTA treatment (the third step cleaning) removed the iron(Fe) and increased the whiteness of wood color. The ultrasonic treatment (the fourth step cleaning) removed the sodium(Na) remained after EDTA treatment and the fine mineral substances.

  • In this study, 1,2-hexanediol, ethylhexylglycerin, and glyceryl caprylate known as skin conditioning agents with antimicrobial activity in cosmetics were investigated for their antimicrobial activity against gram-positive and gram-negative bacteria when combining them with various ingredients in cosmetic formulations. Seven kinds of substances expected to improve the antibacterial activity in formulations were selected to blend with these three compounds.; 1,3-propanediol, 1,3-butanediol, glycerin, dipropylene glycol, niacinamide, EDTA-2Na, and ethanol. The checkerboard assay was conducted to confirm the antibacterial synergy of these seven substances and the three skin conditioning agents. Consequently, ethanol and EDTA-2Na were detected as the significant materials with synergistic effect as well as ethanol showed antibacterial synergy with all the three compounds. When seven of selected materials were mixed with glyceryl caprylate, there were synergistic or additive activity against S. aureus, gram-positive bacteria and it was suggested that the combination with 1,2-hexanediol and glyceryl caprylate was useful to control both gram-positive and gram-negative bacteria when there were four kinds of polyols. This study is meaningful in that it confirmed the antibacterial synergy of the skin conditioning agents with antimicrobial activity and the main raw materials frequently used in cosmetics, thereby contributing to prediction of the antibacterial activity of the skin conditioning agents in cosmetic formulations.

  • The $Ca^{++}$ and $Mg^{++}$ released during Ca, Mg-Na exchange on Kampo 78 montmorillonite which was treated with various concentrations of NaCl solution, were measured with EDTA titration metbod in the leaching solutions. Lanthanide montmorillonite was prepared with various neutral lanthanide ions from sodium montmorillonite in which the exchangeable ions are displaced from the exchanger, such as the displacement of $Na^+$ by $Ln^{3+}$ ions, Cation exchange capacity (CEC) is determined on remaining lanthanides in the leaching solutions with E. D. T. A titration method. As a results of this study, there were no difference of C. E. C in series of lanthanide contraction, but C. E. C depends on charge density of montmorillonite. When we conformed the structure of Ln-montmorillonite by X-ray diffraction. It was found that there was much difference of pattern between Na-montmorillonite and Ln-montmorillonite.

  • In nuclear power plants and other nuclear facilities the removal of cobalt from radioactive liquid waste is needed to reduce the radioactivity concentration in effluents. In liquid wastes containing strong organic complexing agents such as EDTA cobalt removal can be problematic due to the high stability of the Co-EDTA complex. In this study, the removal of cobalt from NaNO3 solutions using antimony oxide (Sb2O3) synthesized from potassium hexahydroxoantimonate was investigated in the absence and presence of EDTA. The uptake studies on the ion exchange material were conducted both in the dark (absence of UV-light) and under UV-C irradiation. Ca2+ or Ni2+ were included in the experiments as competing cations to test the selectivity of the ion exchanger. Results show that UV-C irradiation noticeably enhances the cobalt sorption efficiency on the antimony oxide. It was shown that nickel decreased the sorption of cobalt to a higher extent than calcium. Finally, the sorption data collected for Co2+ on antimony oxide was modeled using six different isotherm models. The Sips model was found to be the most suitable model to describe the sorption process. The Dubinin-Radushkevich model was further used to calculate the adsorption energy, which was found to be 6.2 kJ mol-1.
