Antitumor Activity of $\alpha$-Lactalbumin on the Tumor Cells ($\alpha$-Lactalbumin의 암세포 증식 저해효과에 관한 연구)

  • Bovine serum albumin(BSA), $\alpha$-lactalbumin($\alpha$-LA), $\beta$-lactoglobulin($\beta$-LG) and bovine immunoglobulin G (IgG) were investigated the cytotoxicity on tumor cell lines. $\alpha$-LA was showned a tendency of dose dependaent on cytotoxicity using WiDr. The growth of WiDr was inhibited 82% by 1mg/ml of $\alpha$-LA. However, IgG, BSA and $\beta$-LG were not shown the cytotoxicity on WiDr. When $\alpha$-LA was purified by using high pressure liquid chromatography(HPLC), the main component($\alpha$-LA) was eluted at 33.057 min and extremely small quantities eluted at 32.310 min. The cytotoxicity of main component (eluted at 33.057 min peak) was lower than commercial $\alpha$-LA. And the cytotoxic activity of hydrolyzed $\alpha$-LA and $\alpha$-LA treated with EDTA were lower than commercial $\alpha$-LA on tumor cells.

Development of Fusant Degrading Aniline and 4-chlorobiphenyl by Spheroplast Fusion between Pseudomonas sp. and Flavimonas oryzihabitans (Flavimonas oryzihabitans와 Pseudomonas sp.간 원형질체 융합에 의한 Aniline과 4-chlorobiphenyl 분해균주 개발)

  • Spheroplast cell fusions were performed with Flavimonas oryzihabitans degrading aniline and Pseudomonas sp. degrading 4-chlorobiphenyl to develope the new fusant degrading aniline and 4-CBP and its characters were investigated. F. oryzihabitans was induced to antibiotic marker ($Cm^r$ by NTG treatment for the fusants selection. The results of spheroplast formation and regeneration frequencies of the strains treated with lysozyme-EDTA were 99% and 5.0~6.6%, respectively. Fusion products were treated with 40% (v/v) PEG 6000 and fusion frequency was $3.16{\times}10^{-4} $. The DNA content of fusant, F22 was approximately 2-fold compared with parents. The fusant was stable, and showed the mixed biochemical characteristics of the parent strains. F22 was similar to parent for cell growth pattern and degrading capacity on 5 mM aniline but cell growth rate of F22 was 1.5-fold higher than that of the parent on 10mM aniline. However 4-CBP degrading ability of F22 was slightly lower than that of parental strain.

Characterization of the Outer Membrane-Associated 2-Furaldehyde Dehydrogenase from Klebsiella pneumoniae (Klebsiella pneumoniae 균주의 세포외막에서 분리한 2-furaldehyde dehydrogenase의 특성에 관한 연구)

  • An outer membrane-associated 2-furaldehyde dehydrogenase, catalyzing the oxidation of 2-furaldehyde to 2-furoic acid from Klebsiella pneumoniae was purified to homogeneity and characterized. The enzyme showed its highly specific dependency on $\beta$-$NAD^{+}$. Enzyme activity was monitored during purification by using substrate 2-furaldehyde and coenzyme $\beta$-$NAD^{+}$ by means of high performance liquid chromatography. The outer membrane was successfully collected by the methods of Percoll density gradient ultracentrifugation and ultracentrifugation after preferential solubilization of the membrane with $Mg^{2+}$ and Triton X-100. The enzyme was purified by the series of procedures including extraction of outer membrane protein with EDTA and lysozume, and fractionation by column chromatography on QAE-Sephades Q-50, and subsequently Sephadex G-100. The enzume showed its optimal activity at $85^{\circ}C$, pH 9.5, and in the presence of 1.5% (vol/vol) Triton X-100. The enzyme exhibited a native molecular size of 88,000 by nondenaturing polyacrylamide gel electrophoresis and had an apparent Km of 4.72mM for 2-furaldehyde.

A STUDY ON MORPHOLOGIC CHANGES OF DENTINAL SURFACES BY DENTIN CONDITIONING AGENTS (상아질표면처리제에 의한 상아질표면의 형태변화에 관한 연구)

  • The purpose of this study is to observe the morphological changes of prepared dentin surfaces by 5 dentin conditioning agents. Freshly extracted 48 healthy human molars were used in this study. The teeth were stored at $4^{\circ}C$ physiologic saline solution befor experiment. The teeth were cross-sectioned to expose dentin below 3.0mm at the cusp tip and above 2.0mm at the cemento-enamel junction with Crystal Cutter (MC411 D, Maruto Co., Japan). The specimens were then divided into 12 groups. The sectioned dentin surfaces in group 1, 3, 5, 7, 9, and 11 were prepared with No. 301 diamond point under air-water spray and those in group 2, 4, 6, 8, 10, and 12 were prepared with No. 700 carbide bur. The prepared dentin surfaces were conditioned with Nitric acid, Citric acid, Poly acrylic acid, EDTA, and Phosphoric acid. All the specimens were gold-coated with Eiko ion coater (Eiko-engineering Co.) and observed in Hitachi S-2300 Scanning electron microscope at 20 KV. The following results from this study were obtained; 1. The dentinal smear layers prepared with diamond point were compacted than those prepared with the carbide bur. 2. The dentinal smear layers prepared with diamond point or carbide bur were thick but after treatment of dentin conditioning agents smear layers were removed almost. 3. Irrespective of the uses of the diamond point or the carbide bur the morphological changes of dentin surfaces treated with the same conditioning agents were similar. 4. Treatment of nitric acid and EDTA was a little effect in removing dentinal smear layer. 5. Treatment of citric acid and phosphoric acid removed the smear layer very effectively and showed dissolution of peritubular dentin and opening of dentinal tubules.

  • It was investigated to develop the technology for simultaneous removal of $SO_2/NO$ in flue gas using liquid homogeneous catalyst. Test was carried out using a bench scale and a pilot scale experiment. The investigation led to the following results: 1) Removal efficiency of $SO_2$ gas showed good results regardless of operating condition. Removal efficiency of NO gas, however, proportionally increased with higher packing height, lower concentration and larger injection rate of catalyst 2) The optimum design parameters for simultaneous removal of $SO_2/NO$ gas using Fe(II)-EDTA catalyst were as follow: HTU(height of transfer unit) = 0.5 m, liquid gas ratio = 20 $L/m^3$, NTU (number of transfer unit) = 3 stages, cross dimension of scrubber=0.025 $m^2$ 3) The removal efficiencies of $SO_2$ and NO were 95% and 81%, repletely. 4) The high HTU is advantageous on removal of the NO, but the excessive HTU diminishes operating efficiency. Consequently, it is important to decide the HTU of optimum.

Studies on Secretion of Catecholamines Evoked by Panaxadiol in the Isolated Rabbit Adrenal Gland (파낙스디올의 가토적출부신의 카테콜아민분비 작용에 관한 연구)

  • The effect of panaxadiol (PD). an active component of Korean ginseng saponins on the secretion of catecholamines (CA) from the rabbit adrenal gland and its mode of action were investigated. PD ($400\;{\mu}g$) increased significantly the secretion of CA from the isolated perfused rabbit adrenal gland. PD-induced secretion of CA was reduced markedly by treatment with atropine. CA secretion induced by Ach or PD was potentiated by physostigmine treatment. Chlorisondamine inhibited CA secretion of PD or Ach. Perfusion of PD ($400\;{\mu}g$) for 30 minutes enhanced the secretory activity of CA by Ach. Ouabain weakened the secretory rsponse caused by PD but enhanced the response by Ach. Adenosine treatment resulted in marked increase in CA secretion by PD or Ach. Perfusion with calcium free Krebs solution containing 5 mM EDTA for 30 minutes completely blocked the secretory effect induced by Ach and also weakened that evoked by PD. It is suggested that PD causes the secretion of CA from the rabbit adrenal gland by a calcium dependent exocytotic process. The secretory effect of PD is due to the stimulation of cholinergic muscarinic and nicotinic-receptors present in the adrenal gland partly by direct action on the chromaffin cell.

Evaluation of Antioxidantive and Antimicrobial Activities of Garlic Stem and Red Cabbage, and Their Application to Pork Patties during Refrigerated Storage (붉은 양배추와 마늘종 추출물의 항산화 및 항균활성 평가와 냉장저장 중 돈육패티에 이용)

  • This study was carried out to investigate antioxidative and antimicrobial activities of ethanol extracted garlic stem and red cabbage, and their application to pork patties during refrigerated storage. Total phenolic compounds of ethanol extracted garlic stem and red cabbage were 3.13 g/100 g and 3.99 g/100 g, respectively. 1,1'-diphenyl-2-picryhdrazyl (DPPH) radical scavenging activity of garlic stem extracts increased up to 0.05%. Iron chelating activities of both garlic stem and red cabbage increased up to 0.5%, and red cabbage up to the concentration of 1% had similar iron chelating activity to those of EDTA. Pork patties containing garlic stem had higher Hunter L and b values, (p<0.05), whereas those with red cabbage had highest Hunter a values, resulting in similar value to those of control (p>0.05). Thiobarbituric acid reactive substances (TBARS) of pork patties with garlic stem were lower than those of the control (p<0.05). In addition, antioxidantive activities of pork patties with red cabbage were higher than those with garlic stem. No differences in microbial contents were observed between the control and treatments. In conclusion, garlic stem and red cabbage extracts might be used as natural antioxidants to be used in the manufacture of functional meat products.

  • 접근성이란 특정 목적을 가진 통행자가 한 지점에서 다른 지점으로 얼마나 쉽게 갈 수 있는가 를 나타내는 척도를 말한다. 따라서 접근성은 주로 공간적인 거리나 시간이 가장 중요한 요인으로 작용하며 최근에는 공간적 거리보다는 접근시간이 더 중요한 접근성 척도로 사용되고 있다. 본 연구는 GIS공간분석기법과 접근성이론을 접목하여 도시시설의 적정후보지 평가방법을 정립한 후 현재 검토되고 있는 수도권 도심공항터미널 입지 후보지 중 어떤 후보지가 이용들의 총통행시간을 최소화하는 후보 지인가에 대해 평가해보았다. 평가 결과 다음과 같은 사실을 밝힐 수 있었다. 첫째, 수도권 전체로 볼 때 평균접근시간이 가장 양호한 후보지는 쌍문터미널(현 간이 도심공항터미널)이고, 2위는 현 삼성동 도심공항터미널과 강남고속터미널, 3위는 동서울터미널과 남서울터미널 순으로 나타났다. 둘째, 수도권전체 이용자들의 총통행시간을 최소로 하는 도심공항터미널 후보지는 접근성이 가장 양호한 쌍문동터미널이 아니라 현 서울역이 1위이고, 2위는 강남고속 터미널, 3위는 용산 역으로 나타났다. 셋째, 터미널 후보지간의 가중평균접근 시간은 도심인 서울시내에 입지한 후보지와 수도권 외곽에 입지 한 후보지간에 큰 차이가 없으나 총 이용자 접근시간은 서울시에 입지한 후보지보다 외곽의 후보지가 훨씬 높게 나타나 뚜렷한 차별성을 보이고 있다. 넷째, 최적후보지 1,2,3순위 모두 서울도심과 강남도심에 입지한 지역들로 나타나 교통의 접근성 보다는 아직 인구밀집도가 주요 도시시설의 입지결정에 주요한 결정요인 되는 것으로 나타났다.해석 시스템을 구축할 예정이다. 추후에는 하수도 관망해석 컴포넌트와 하수도 업무 컴포넌트와의 통합부분에 대한 연구가 진행되어야 할 것이다.7.0로 하고 표준(標準) EDTA 용액(溶液)을 소량(少量)넣고 8N-KOH로 pH $12{\sim}13$으로 하고 N-N 희석분말(稀釋粉末)을 지시약(指示藥) 으로써 표준(標準) EDTA 용액(溶液)으로 적정(滴定)하여 Ca 치(値)를 얻었다. Ca와 Mg의 합계결정치(合計決定値)와 Ca 적정치(滴定値) 차(差)로 Mg 치(値)를 얻었다. 음(陰) ion 구분(區分)으로부터 상법(常法)에 의하여 $MgNH_4PO_4$의 침전(沈澱)을 만들어서 HCl에 녹키고 일정량(一定量)의 표준(標準) EDTA 용액(溶液)을 넣어 pH 7.0로 한다음 완충액(緩衝液)으로 pH 10으로 하고 BT 지시약(指示藥)을 써서 표준(標準) Mg $SO_4$용액(溶液)으로 적정(滴定)하여 P 치(値)를 얻었다. 본법(本法)으로 Na-phytate를 분석(分析)한 결과(結果) Na-phytate의 분자식(分子式)을 $C_6H_6O_{24}P_6Mg_4CaNa_2{\cdot}5H_2O$라고 하였을 때의 이론치(理論値)에 비(比)하여 P가 98.9% Cark 97.1%, Mg가 99.1%이고 통계처리(統計處理)한 결과분석치(結果分析値)와 이론치(理論値)는 잘 일치(一致)된다. 그러나 종래법(從來法)에 의(依)한 분석치(分析値)는 이론치(理論値)에 비(比)하여 P가 92.40%, Cark 86.80

하수도 업무지원을 위한 하수도 관망해석 컴포넌트 설계에 관한 연구

  • 국가지리정보체계사업을 시작으로 여러 응용분야에서 지리정보시스템을 구축하고 있으며, 특히 지자체에서는 도시정보화 차원에서 UIS(Urban Information System;도시정보시스템)를 구축하고 있다. 지자체의 업무들은 지자체의 상황에 따라 조금씩 차이를 보이지만 업무 단위로 따져 볼 때 공통적으로 수행되는 업무들이 많으며, 지리정보 관련 업무 또한 예외가 아니다. 지금까지 각 지자체의 도시정보시스템은 공통업무에 대하여 중복개발이 불가피 하였으며, 재사용성을 기대하기 어려웠다. 이런 비효율을 극복하는 방안으로 컴포넌트 기술이 대두되었으며, 컴포넌트 기술을 이용하여 업무변화에 유연하고 재활용을 극대화할 수 있는 업무 컴포넌트 개발이 필요한 시점이다. 본 논문은 지자체의 지리정보 관련 업무의 하나인 하수도 업무 중 하수배출용량 산정을 위한 하수도 관망해석 컴포넌트 설계를 제시하였다. 하수도 관망해석 컴포넌트는 하수배출용량을 산정하고, 하수 배출용량을 토대로 통수능이 부족한 하수관거를 추출하며, 통수능 부족 관거에 대하여 적정한 교체 관경 결정을 지원할 수 있도록 설계하였다 컴포넌트 설계과정은 표준업무를 지원할 수 있도록 업무분석을 수행하였으며, 이의 결과물로 하수도 관망해석 알고리즘을 도출하였으며, 알고리즘을 기반으로 하수도 관망해석 업무를 수행하는 하수도 관망해석 컴포넌트를 설계하고 설계 내용을 UML(Unified Modeling Language)로써 명세화 하였다. 현재 설계에 따라 하수도 관망해석 컴포넌트가 개발되고 있으며, 개발된 컴포넌트를 이용한 하수도 관망해석 시스템을 구축할 예정이다. 추후에는 하수도 관망해석 컴포넌트와 하수도 업무 컴포넌트와의 통합부분에 대한 연구가 진행되어야 할 것이다.7.0로 하고 표준(標準) EDTA 용액(溶液)을 소량(少量)넣고 8N-KOH로 pH $12{\sim}13$으로 하고 N-N 희석분말(稀釋粉末)을 지시약(指示藥) 으로써 표준(標準) EDTA 용액(溶液)으로 적정(滴定)하여 Ca 치(値)를 얻었다. Effect of microwave heating and salts addition on pH and acidity of Kakdugi during fermentation (깍뚜기 발효중 순간 가열과 염첨가가 pH변화에 미치는 영향)

  • Addition of salts or their mixtures and microwave heating were studies for their effects on Kakdugi fermentation. The Kakdugi, a Korean fermented Chinese radish, were prepared by salting in 15% NaCl solution and fermented at $25^{\circ}C$. From the results, a first order reaction relationship was found between salt permeated into the radish and time during brining in $5{\sim}10%$ NaCl solution. Addition of 0.05M KCl into 15% NaCl brining solution or microwave heating of salted radish for 3 minutes showed a little decreasing effect on Kakdugi fermentation rate while beating for 1 or 2 minuted resulted a rather increase. When three different salt mixtures in the concentration range of $0.001{\sim}0.01\;M$ were added into half fermented Kakdugi(appr. pH 4.4), the fermentation was greatly controlled based on pH change. Among the salt mixtures, mixture III$(Na_2HPO_4,\;Na_2PO_4,\;NaNO_2,\;Ca\;EDTA,\;Sod.\;citrate)$ showed a most significant effect where the time required to reach pH 4.0 after addition was extended by more than 6 folds when it was compared to the control method.
