Autophagy Is a Potential Target for Enhancing the Anti-Angiogenic Effect of Mebendazole in Endothelial Cells

  • Mebendazole (MBZ), a microtubule depolymerizing drug commonly used for the treatment of helminthic infections, has recently been noted as a repositioning candidate for angiogenesis inhibition and cancer therapy. However, the definite anti-angiogenic mechanism of MBZ remains unclear. In this study, we explored the inhibitory mechanism of MBZ in endothelial cells (ECs) and developed a novel strategy to improve its anti-angiogenic therapy. Treatment of ECs with MBZ led to inhibition of EC proliferation in a dose-dependent manner in several culture conditions in the presence of vascular endothelial growth factor (VEGF), basic fibroblast growth factor (bFGF) or FBS, without selectivity of growth factors, although MBZ is known to inhibit VEGF receptor 2 kinase. Furthermore, MBZ inhibited EC migration and tube formation induced by either VEGF or bFGF. However, unexpectedly, treatment of MBZ did not affect FAK and ERK1/2 phosphorylation induced by these factors. Treatment with MBZ induced shrinking of ECs and caused G2-M arrest and apoptosis with an increased Sub-G1 fraction. In addition, increased levels of nuclear fragmentation, p53 expression, and active form of caspase 3 were observed. The marked induction of autophagy by MBZ was also noted. Interestingly, inhibition of autophagy through knocking down of Beclin1 or ATG5/7, or treatment with autophagy inhibitors such as 3-methyladenine and chloroquine resulted in marked enhancement of anti-proliferative and pro-apoptotic effects of MBZ in ECs. Consequently, we suggest that MBZ induces autophagy in ECs and that protective autophagy can be a novel target for enhancing the anti-angiogenic efficacy of MBZ in cancer treatment.

Expression of HYOU1 via Reciprocal Crosstalk between NSCLC Cells and HUVECs Control Cancer Progression and Chemoresistance in Tumor Spheroids

  • Among all cancer types, lung cancer ranks highest worldwide in terms of both incidence and mortality. The crosstalk between lung cancer cells and their tumor microenvironment (TME) has begun to emerge as the "Achilles heel" of the disease and thus constitutes an attractive target for anticancer therapy. We previously revealed that crosstalk between lung cancer cells and endothelial cells (ECs) induces chemoresistance in multicellular tumor spheroids (MCTSs). In this study, we demonstrated that factors secreted in response to crosstalk between ECs and lung cancer cells play pivotal roles in the development of chemoresistance in lung cancer spheroids. We subsequently determined that the expression of hypoxia up-regulated protein 1 (HYOU1) in lung cancer spheroids was increased by factors secreted in response to crosstalk between ECs and lung cancer cells. Direct interaction between lung cancer cells and ECs also caused an elevation in the expression of HYOU1 in MCTSs. Inhibition of HYOU1 expression not only suppressed stemness and malignancy, but also facilitated apoptosis and chemosensitivity in lung cancer MCTSs. Inhibition of HYOU1 expression also significantly increased the expression of interferon signaling components in lung cancer cells. Moreover, the activation of the PI3K/AKT/mTOR pathway was involved in the HYOU1-induced aggression of lung cancer cells. Taken together, our results identify HYOU1, which is induced in response to crosstalk between ECs and lung cancer cells within the TME, as a potential therapeutic target for combating the aggressive behavior of cancer cells.

Molybdenum-Based Electrocatalysts for Direct Alcohol Fuel Cells: A Critical Review

  • Direct alcohol fuel cells (DAFCs) have gained much attention as promising energy conversion devices due to their ability to utilize alcohol as a fuel source. In this regard, Molybdenum-based electrocatalysts (Mo-ECs) have emerged as a substitution for expensive Pt and Ru-based co-catalyst electrode materials in DAFCs, owing to their unique electrochemical properties useful for alcohol oxidation. The catalytic activity of Mo-ECs displays an increase in alcohol oxidation current density by several folds to 1000-2000 mA mgPt-1, compared to commercial Pt and PtRu catalysts of 10-100 mA mgPt-1. In addition, the methanol oxidation peak and onset potential have been significantly reduced by 100-200 mV and 0.5-0.6 V, respectively. The performance of Mo-ECs in both acidic and alkaline media has shown the potential to significantly reduce the Pt loading. This review aims to provide a comprehensive overview of the bifunctional mechanism involved in the oxidation of alcohols and factors affecting the electrocatalytic oxidation of alcohol, such as synthesis method, structural properties, and catalytic support materials. Furthermore, the challenges and prospects of Mo-ECs for DAFCs anode materials are discussed. This in-depth review serves as valuable insight toward enhancing the performance and efficiency of DAFC by employing Mo-ECs.

The thermal effect on electrical capacitance sensor for two-phase flow monitoring

  • One of major errors in flow rate measurement for two-phase flow using an Electrical Capacitance Sensor (ECS) concerns sensor sensitivity under temperature raise. The thermal effect on electrical capacitance sensor (ECS) system for air-water two-phase flow monitoring include sensor sensitivity, capacitance measurements, capacitance change and node potential distribution is reported in this paper. The rules of 12-electrode sensor parameters such as capacitance, capacitance change, and change rate of capacitance and sensitivity map the basis of Air-water two-phase flow permittivity distribution and temperature raise are discussed by ANSYS and MATLAB, which are combined to simulate sensor characteristic. The cross-sectional void fraction as a function of temperature is determined from the scripting capabilities in ANSYS simulation. The results show that the temperature raise had a detrimental effect on the electrodes sensitivity and sensitive domain of electrodes. The FE results are in excellent agreement with an experimental result available in the literature, thus validating the accuracy and reliability of the proposed flow rate measurement system.

  • This paper proposes a product data model that can express and enforce integrity constraints on product structure during engineering changes (ECs). The model adopts and extends an active object-oriented database model in order to Integrate IC data and their integrity constraints. Tightly integrated with product structure, It will enable designers to maintain and exchange consistent EC data throughout the product life cycle. In order to properly support operations for ECs, the model provides the data, operations, and Event-Condition-Action rules for nested ECs and simultaneous EC applications to multiple options. in addition, the EC objects proposed In the model integrate the data and Integrity constraints into a unified repository. This repository enables designers to access all EC data and integrity constraints through the product structure and relationships between EC objects. This paper also describes a prototype product data management system based on the proposed model In order to demonstrate its effectiveness.

  • Canada British Columbia 주의 Prince Rupert 와 Port Hardy(503km) 를 1주일에 2회 왕복하던 Queen of the North 호가 2006년 3월 22일 Gill Island 북쪽 해상에서 좌초되었다. 사고 후, 선박에 탑재된 Electronic Chart System(ECS)을 수거하여 선박의 항해자료를 회수키로하고, 2006년 6월 15일 해양탐사로봇을 이용하여 해저 약 453m 지점에 착저된 선박으로부터 ECS 컴퓨터를 수거하였고 Maker인 Transas사는 수거된 ECS의 Data를 복원하여 선주사에 재공하였다. 이에 따라 캐나다 교통안전국 (Transportation Safety Board of Canada)는 재현 결과물을 통하여 사고원인을 규명한 사건이다.

DDVP, Malathion 및 Diazinon유제의 경시변화 특성 (Decomposition Characteristics of DDVP , Malathion and Diazinon Emusifiable Concentrates)

  • 유기인계 살충제 유제의 경시변화 특성을 구명하기 위하여 3종의 농약원제(DDVP, malathion 및 diazinon), 3종의 용매(xylene, cyclohexanone 및 DMF), 조제된 유화제 9종 및 분해방지제 epichlorohydrin을 재료로 조성과 수분함량을 달리하는 시험용 유제를 제제한 다음 $54^{\circ}C$에서 15일간, $38^{\circ}C$에서 9일간 각각 고온학대처리하여 유효성분의 경시변화를 조사하고 상호비교하였다. DDVP원제는 $54^{\circ}$에서 초기에 크게 분해되었으나 경시적으로 분해속도가 완만해졌다. 한편 malathion과 diazinon은 점차 분해가 촉진되었으며, diazinon의 경시변화가 가장 심하였다. 유제중 유효성분의 경시변화는 사용한 용매에 따라 큰 차이가 있었으며, 대체로 용매의 극성이 증가할수록 경시변화가 현저하게 증가하였다. 유화제를 첨가한 유제는 수분함량과 경시변화율이 증가하였으며, 유화제의 종류에 따라 경시변화의 차이가 심하였다. 유제중의 수분함량이 증가할수록 경시변화율이 증가하였으며, 약제에 따라 차이가 있어서 DDVP가 수분함량증가에 대하여 가장 안정하였으며, diazinon이 가장 불안정하였다. Malathion과 diazinon유제는 epichlorohydrin에 의한 분해방지 효과가 탁월하였으나, DDVP유제에서는 그 효과가 매우 작았다. 고온학대처리조건이 달라도 경시변화경향은 유사하였으나, $38^{\circ}C$에서 90일간 보관한 유제의 경시변화율이 $54^{\circ}C$에서 15일간 보관한 것보다 1-2배 크게 나타났다.

중증 장애인을 위한 생활환경 제어장치개발에 관한 연구 (A Study on Development of ECS for Severly Handicaped)

  • 본 논문은 음성기반 생활환경 제어장치 (Enviropmet Control System: ECS)의 개발과 적용에 대해 다룬다. 구체적으로 개발 내용은 음성인식을 통한 생활환경의 제어 장치와 음성 지원 이동형 휠체어 리프트 제어장치를 구현하였다. 구현된 시스템은 경수 손상 사지마비 환자에게 적용되었고 활용의 문제점과 장치의 사용이 장애인 및 가족에게 미치는 신체적 정신적 영향을 평가 고찰하였다. 음성 인식 시스템은 HMM기반 실시간 음성 인식 시스템을 구성하였고 임상의 적용을 평가하기 위하여 가전 기구를 제어하도록 간략화된 음성인식 모듈을 구현하여 54세 남자로 완전 제 4 경수 손상인을 대상으로 1주간 관찰하였다. 그리고 설문 조사를 통하여 Beck 우울 척도와 오락 및 사회적 관계성의 변화를 평가하였다. 또한 음성 지원 이동형 휠체어 리프트의 전동기 구동 제어 장치를 개발하고 구조 역학 해석을 통해 기계적 안정성을 평가하고 시제품을 제작 성능 시험을 하였다. 시스템의 실험 결과 95%이상의 인식률을 나타내었다. 설문조사 결과 장애인의 장치에 대한 만족도는 높은 편이고 간호의존도와 사회적 관계성의 증진을 확인할 수 있었다. 이에 더불어 간병인의 부담과 우울 경향의 감소가 있었다. 또한 음성 지원 이동형 휠체어 리프트는 구조 해석 결과, 상단 휠체어 바퀴 지지대의 재료로서의 피로 수명이 우수하고 무게 중심 이동이 안정성을 가짐을 확인하였다. 본 논문은 실시간 음성 인식 시스템과 이동형 휠체어 리프트를 이용한 장애자를 위한 생활환경 제어장치 구성의 한 예를 제시하였고, 실험을 통하여 실시간 음성인식의 중요한 활용 분야인 재활 의학 분야에서 한국형 생활환경 제어장치의 개발의 필요성을 확인하였으며 상용화를 모색의 발판이 될 것이다.
