Clinical study of Endocardial Cushion Defect [7 Cases Report] (심내막상 결손증의 임상적 고찰7례 보고)

  • Seven patients had undergone repair of endocardial cushion defect from Jan. 1977 to Dec. 1984 at National Medical Center. Most patients had no associated anomalies except one who had PFO, and mortality case was absent. Five patients had partial ECD and two had complete ECD [Rastelli type A]. In P-ECD patients, the atrial septal defect was closed with patch in all cases and mitral cleft was approximated with 2-3 direct stitches. In two cases of C-ECD, atrial and ventricular septal defect was closed with single patch in one case and atrial septal defect was closed with patch but ventricular septal defect was closed with patch but ventricular septal defect was closed it direct suture in the other case. Atrioventricular cleft was approximated with 2-3 direct sutures. Postop. transient A-V block was noted in 2 cases but returned to regular sinus rhythm after 2 to 6 months.

Electron Capture Dissociation Mass Spectrometry for Gaseous Protonated Melittin Ions and Its Single Amino Acid Substituted Variants

  • This study demonstrated the sensitivity of electron capture dissociation mass spectrometry (ECD-MS) to probe subtle conformational changes in gaseous melittin ions induced by the substitution of an amino acid. ECD-MS was performed for triply and quadruply-protonated melittin and its variants obtained by a single amino acid substitution, namely, D-Pro14, Pro14Ala, and Leu13Ala. Although native triply-protonted melittin showed only a few peptide backbone cleavage products, the D-Pro14 and Pro14Ala variants exhibited extensive backbone fragments, suggesting the occurrence of a significant structural or conformational change induced by a single amino acid substitution at Pro14. On the contrary, the substitution at Leu13, namely Leu13Ala (+3), did not cause significant changes in the ECD backbone fragmentation pattern. Thus, the sensitivity of ECD-MS is demonstrated to be good enough to probe the aforementioned conformational change in melittin.

  • Although artifacts in tomographic imaging can arise from a number of sources, this case presents an artifact caused by using inadequate acquisition parameters which do not match the collimator with which a camera equipped in Tc-99m ECD brain SPECT. Whenever the collimator in tomographic studies is changed, we should take care to use suitable parameters of acquisition protocol according to the collimator with which a camera is fitted. Other ways to prevent an error of this type is to use a point source.

  • To determine levels of PCBs in food, beef, pork, chicken, egg, mackerel, yellow croaker, anchovy, common squid and little neck clam were chosen and collected at markets in Seoul, Busan and Kwangju. Among 209 PCB congeners, 7 congeners (#28, #52, #101, #118, #138, #153 and #180) were selected as target compounds that were known as indicator congeners. Samples were homogenized, treated in alkali solution for 1 hour, and extracted with organic solvents. After extraction, extracts were cleaned up by sulfuric acid, purified on silica gel column chromatography, analyzed by dual column-HRGC/ECD and then confirmed by HRGC/MSD. As results, PCBs were detected in fish samples ranged from 0.0002 to 0.001 mg/kg. Both PCB #101 and PCB #118 were the major contributors among 7 congeners.

  • A simultaneous and rapid gas chromatographic determination of organophosphorus, organo-chlorine, carbamate, and pyrethroid pesticides in polished rice was developed. The pesticides were extracted from samples with acetone by automated soxhlet apparatus and this extract was evaporated to dryness. The residue was dissolved in hexane, the solvent was applied to a Sep-Pak florisil catridge, was eluted with 50% ethyl acetate in n-hexane, and was injected to dual GC-ECD/NPD system. This simple method affords a high recovery of hydrophilic pesticides, allows rapid analysis, and is cheap to perform. Except for dichlorvos and captan, recoveries of 46 pesticides were over 60%.

Complete genome sequence of Flavivirga eckloniae ECD14T isolated from a seaweed Ecklonia cava (감태(Ecklonia cava)에서 분리한 Flavivirga eckloniae ECD14T의 유전체 서열 분석)

  • The genome of Flavivirga eckloniae $ECD14^T$ isolated from a seaweed Ecklonia cava was sequenced. The genome comprises a single circular 5,665,358 bp chromosome with a G + C content of 33.9%, 4,647 total genes, 4,595 protein-coding genes, 44 pseudo genes, and 52 RNA genes. CRISPER genes and sequences were not found and there were some phage remnants and transposons. This strain contains alginate lyase and ${\beta}$-glucosidase genes responsible for the degradation of seaweed polysaccharides.

Mitigation of wind-induced vibrations of bridge hangers using tuned mass dampers with eddy current damping

  • To mitigate vibrations, tuned mass dampers(TMD) are widely used for long span bridges or high-rise buildings. Due to some durability concerns, such as fluid degradation, oil leakage, etc., the alternative solutions, such as the non-contacted eddy current damping (ECD), are proposed for mechanical devices in small scales. In the present study, a new eddy current damping TMD (ECD-TMD) is proposed and developed for large scale civil infrastructure applications. Starting from parametric study on finite element analysis of the ECD-TMD, the new design is enhanced via using the permanent magnets to eliminate the power need and a combination of a copper plate and a steel plate to improve the energy dissipation efficiency. Additional special design includes installation of two permanent magnets at the same side above the copper plate to easily adjust the gap as well as the damping. In a case study, the proposed ECD-TMD is demonstrated in the application of a steel arch bridge to mitigate the wind-induced vibrations of the flexible hangers. After a brief introduction of the configuration and the installation process for the damper, the mitigation effects are measured for the ambient vibration and forced vibration scenarios. The results show that the damping ratios increase to 3% for the weak axis after the installation of the ECD-TMDs and the maximum vibration amplitudes can be reduced by 60%.

Derivatization and Residual Determination of MCPP in Soil Leachate (토양 침투수중 MCPP의 유도체화 및 잔류분석)

  • Analytical methods for the determination of the derivatives of the herbicide (${\pm}$)-2-(4-chloro-2-methylphenoxy)propionic acid (MCPP) by capillary column gas chromatography with mass spectrometer (GC-MS) and electron-capture detection (GC-ECD) were studied. A successful procedure was introduced for the ester preparation using $H_2SO_4$, as the catalyst and the alcohol 2,2,2-trichloroethanol (TCE) or 2,2,2-trifluoroethanol (TFE). The identificaiton and elucidation of MCPP by GC-MS spectrometry following the esterification with diazomethane, $BF_3$/methanol, $H_2SO_4$/methanol, TCE, TFE, or pentafluorobenzyl bromide (PFB) were carried out. A comparison of the response-sensitivities among those MCPP esters was made with GC-ECD. Although the methylation product of MCPP was confirmed by GC-MS, its low sensitivity to the ECD limited the detection of MCPP. TCE, TFE, and PFB derivatization methods resulted in a high rate of MCPP esterifications and very sensitive ECD molecular responses. Based on efficiency, convenience, worker safety, and least sample contamination, TFE esterificaiton was considered as the superior method for MCPP analysis to the other methods of derivatization. An accurate method is described for quantifying MCPP in soil leachates by GC-ECD at very low concentrations without the requirement of a complicated clean-up process. As a result, MCPP residues at concentrations of less than $0.1{\mu}g$ in 100ml soil leachate were detected.

Development of Analytical Method for Fipronil Residues in Agricultural Commodities Using GC-ECD/MS (GC-ECD/MS를 이용한 농산물 중 Fipronil의 잔류 분석법 개발)

  • BACKGROUND: An analytical method was developed using GC-ECD/MS to precisely determine the residue of fipronil, a phenylpyrazole insecticide used to control a wide range of foliar and soil-borne pests.METHOD AND RESULTS: Fipronil residue was extracted with acetone from representative samples of five raw products which comprised hulled rice, soybean, Kimchi cabbage, green pepper, and apple. The extract was diluted with saline water, and fipronil was partitioned into n-hexane/dichloromethane (20/80, v/v) to remove polar co-extractives in the aqueous phase. Florisil column chromatography was additionally employed for final purification of the extract. Fipronil was separated and quantitated by GC-ECD using a DB-17 capillary column. Accuracy of the proposed method was validated by the recovery from crop samples fortified with fipronil at 3 levels per crop in each triplication.CONCLUSION: Mean recoveries ranged from 86.6% to 106.0% in five representative agricultural commodities. The coefficients of variation were less than 10%. Limit of quantitation of fipronil was 0.004 mg/kg as verified by the recovery experiment. A confirmatory technique using GC/MS with selected-ion monitoring was also provided to clearly identify the suspected residue. Therefore, this analytical method was reproducible and sensitive enough to determine the residue of fipronil in agricultural commodities.
