Understanding Drug-Protein Interactions in Escherichia coli FabI and Various FabI Inhibitor Complexes

  • Many ligands have been experimentally designed and tested for their activities as inhibitors against bacterial enoyl-ACP reductase (FabI), ENR. Here the binding energies of the reported ligands with the E. coli ENR-$NAD^+$ were calculated, analyzed and compared, and their molecular dynamics (MD) simulation study was performed. IDN, ZAM and AYM ligands were calculated to have larger binding energies than TCL and IDN has the largest binding energy among the considered ligands (TCL, S54, E26, ZAM, AYM and IDN). The contribution of residues to the ligand binding energy is larger in E. coli ENR-NAD+-IDN than in E. coli ENR-$NAD^+$-TCL, while the contribution of $NAD^+$ is smaller for IDN than for TCL. The large-size ligands having considerable interactions with residues and $NAD^+$ have many effective functional groups such as aromatic $\pi$ rings, acidic hydroxyl groups, and polarizable amide carbonyl groups in common. The cation-$\pi$ interactions have large binding energies, positively charged residues strongly interact with polarisable amide carbonyl group, and the acidic phenoxyl group has strong H-bond interactions. The residues which have strong interactions with the ligands in common are Y146, Y156, M159 and K163. This study of the reported inhibitor candidates is expected to assist the design of feasible ENR inhibitors.

유기산이 Escherichia coli O157:H7, Salmonella 및 Listeria monocytogenes의 증식에 미치는 영향 (Inactivation of Escherichia coli O157:H7, Salmonella and Listeria monocytogenes by Organic Acid)

  • The inhibitory effect of the food processing agent on growth of Escherichia coli O157:H7, Salmonella Enteritidis, and Listeria monocytogenes was performed with organic acid, and combination of citric acid, acetic acid, propionic acid and vanillic acid. The minimun inhibitory concentration(MIC) of propionic acid was 5,000 ppm in E. coli O157:H7, 2,500 ppm in Salmonella Enteritidis and Listeria monocytogenes. MIC of citric acid was 10,000 ppm in E. coli O157:H7 and Salmonella Enteritidis, 2,500 ppm in Listeria monocytogenes. MIC of acetic acid was 2,500ppm, while in vanillic acid was 5,000 ppm in Escherichia coli O157:H7, Salmonella Enteritidis, and Listeria monocytogenes. MIC of combined organc acid in E. coli O157:H7 were 2,500ppm in PC, 1,250 ppm in PA, PV, CA, CV and AV. MIC of combined organc acid in Salmonella Enteritidis were 2,500 ppm in PC, PA, PV, CA, and CV, 1,250 ppm in AV. MIC of combined organc acid in Listeria monocytogenes were 1,250 ppm in all treatment group. MIC of combined treatment of three organc acid in E. coli O157:H7, S. Enteritidis and L. monocytogenes were 1,250 ppm in PCA, PCV, PAV and CAV. The inhibitory effect of organc acid in E. coli O157:H7, S. Enteritidis and L. monocytogenes could be confirmed from the result of this experiment. Therefore, it was expected that the food process would increase or maintain by using organic acid.

ENTEROCOCCUS FAECALIS 추출물이 다형핵 백혈구의 METALLOPROTEINASE-8 분비에 미치는 영향에 관한 연구 (Effect of Sonicated Extracts of Enterococcus faecalis on the Production of Matrix Metalloproteinase-8 by Human Polymorphonuclear Neutrophils)

  • Enterococcus faecalis의 추출물을 성인의 말초혈액으로부터 얻은 인간의 백혈구를 이용해 Matrix Metalloproteinase-8 (MMP-8)의 생산에 미치는 영향에 대해서 알아보았다. Enterococcus faecalis를 배양한 뒤 초음파 분쇄를 하여 추출물을 얻어냈다. 이중 일부는 적정한 농도의 Ca(OH)_2$를 처리하였다. 이후 E. faecalis 추출물군, E. faecalis 추출물에 $Ca(OH)_2$ 를 처리한 군으로 분리하여 다형핵백혈구를 자극하여 MMP-8의 생산량을 측정하였다. MMP-8는 처리하지 않은 PMN group에서도 나타났고, PMN을 E. coli LPS로 자극시킨 경우 증가하였다 (p < 0.05) SEF group 들에서는 MMP-8의 생성량이 SEF 의 농도가 증가함에 따라 감소하였다 (p < 0.05). 따라서 Enterococcus faecalis는 PMN의 MMP-8 생산 억제 효과를 가진다고 볼 수 있다. $Ca(OH)_2$로 처리한 SEF 군의 경우 각기 다른 SEF 농도에서도 모두 MMP-8 생성량이 아무 처리도 하지 않은 PMN 군보다 많았다 (p < 0.05) . 하지만 각 군들 사이에 통계적 차이는 없었다 (p > 0.05).

Expression and Characterization of catA1 (catechol 1,2-dioxygenase I1) of Acinetobacter lwoffii K24 in Escherichia coli

  • Catechol 1,2-dioxygenase $I_1$ (CD $I_1$) gene of Acinetobacter Iwoffii K24, $catA_1$ was expressed in Escherichia coli and was partially purified by using a MonoQ column. Expressed CD $I_1$ had the same molecular weight as purified CD $I_1$ from A. Iwoffii K24 on SDS-PAGE. Expressed CD $I_1$ was also identified by Western blotting and peptide sequencing of N-terminal and internal regions. When compared with purified CD $I_1$ of A. Iwoffii K24, expressed CD $I_1$ had similar substrate specificities and the effects of compounds on enzyme activity. N-terminal amino acid sequence of CD I expressed in E. coli was the same as that of purified CD $I_1$, suggesting that CD $I_1$ may be under the same posttranslational processing in E. coli and A. Iwoffii K24.

Therapeutic effect of oregano essential oil on subclinical bovine mastitis caused by Staphylococcus aureus and Escherichia coli

  • Oregano (Origanum vulgare) is an aromatic herb belonging to the Lamiaceae family. This study evaluated the therapeutic effect of oregano essential oil (OEO) on clinical bovine mastitis caused by Staphylococcus (S.) aureus and/or Escherichia (E.) coli. Thirty-two quarters from 18 lactating cows with subclinical mastitis were selected from a dairy farm and were divided into 4 groups (8 quarters/group): negative control (CON), positive control (GEN) and two OEO-treated groups (OEO-1 and OEO-2). Each group was intramammarily treated with saline, 10 g/tube of gentamicin ointment, and single and double doses of 0.9 mL OEO ointment twice a day for three days, respectively. After the treatments, physical udder conditions were greatly improved in GEN and OEO groups. In CON group, somatic cell counts (SCCs), number of white blood cells (WBC) and bacteria were increased. However, in OEO groups, SCCs and number of WBC were significantly decreased and S. aureus and E. coli were not detected in milk as compared with those before the treatments as well as GEN group. These results suggest that OEO may be a useful alternative to antibiotics for the control of subclinical bovine mastitis caused by S. aureus and/or E. coli.

Interaction of Naegleria fowleri Trophozoites with Escherichia coli and MRSA by N-acetylglucosamine and Galactose

  • Naegleria fowleri is a free-living amoeba causing primary amoebic encephalitis. In this study, we analyzed how the N-aceytlglucosamine (GlcNAc) and D-galactose affected the interaction between Naegleria fowleri and methicillin-resistant Staphylococcus aureus (MRSA) or Escherichia coli O157:H7, and the interaction with bacteria when monosaccharides were treated with N. fowleri for a longer pre-incubation time. When GlcNAc was treated with N. fowleri for 1 hr, the E. coli association was almost the same as that of the control not treated with GlcNAc until the concentration of GlcNAc was 25 mM. However, the E. coli association was reduced by approximately 91% with 100 mM GlcNAc. E. coli invasion into N. fowleri showed statistical significance only in the group treated with 100 mM GlcNAc. The interaction when treated with galactose showed a very different pattern in the 50 mM galactose group than when treated with GlcNAc. In the MRSA interaction, a statistically significant decrease in association (76.3% by GlcNAc and 88.7% by galactose) and invasion (3.6% by GlcNAc and 9.3% by galactose) was shown by the concentration of two 100 mM monosaccharides. The group treated with monosaccharides at the same time showed almost no difference in all interactions from the group treated with monosaccharides at the same time. Taken together, it suggested that the effect of monosaccharides on the interaction of several Gram-negative or positive bacteria and the evidence that the interaction could be enhanced by longer pre-incubation time.

Mannose-resistant Hemagglutination(MRHA) 및 Colonization Factor Antigen I(CFA I)을 표현하는 대장균의 장관내 존재와 설사증 발현과의 관계 (Gastrointestinal Carriage of Escherichia coli with Hemagglutination Activity and Colonization Factor Antigen I and its Relation to Diarrhea)

  • Colonization factor antigen I(CFA I) has been shown to be one of several virulence factors that promote attachment of enterotoxigenic E. coli(ETEC) to small intestinal epithelial cells of humans. The ability of ETEC to produce mannose-resistant hemagglutination(MRHA) of human blood group A has been used to detect CFA I. To determine gastrointestinal carriage in Korean children of E. coli with MRHA and CFA I, 116 strains of E. coli from diarrheal children admitted to Hanyang University Hospital were examined for MRHA of human erythrocytes and the presence of CFA I. Of 45 ETEC strains, 18(40%) gave a positive MRHA($MRHA^+$) and eight(18%) were positive for CFA I(CFA $I^+$). ETEC with CFA I were all heat-stable enterotoxin(ST) producers and two of these strains were of serogroups $O_{25}$. Of 17 classic enteropathogenic E. coli(EPEC), 7(41%) were $MRHA^+$ but all were negative for CFA I(CFA $I^-$). Of 30 enteroadherent E. coli(EAEC) strains, 11(37%) were $MRHA^+$ and one was CFA $I^+$. Of 24 nonpathogenic E. coli, 4(17%) were $MRHA^+$ but all were CFA $I^-$. It was shown that MRHA was common in all strains of E. coli, CFA I was limited only to ST producing ETEC and EAEC; although MRHA is a useful screening procedure, serologic tests seem to be necessary to comfirm CFA I production. CFA I was associated with a lower proportion of ETEC isolates in Korea than has been reported for other locations.

마늘즙액의 대장균 생육 저해 작용 (Antibacterial Activity of Garlic Extract against Escherichia coli)

  • 마늘이 상처를 입으면 마늘중에 존재하는 alliin이 alliinase에 의해 allicin으로 분해되며 allicin은 마늘의 주된 항미생물 작용 물질로 알려져 있다. 이 성분에 의한 미생물번식 저해작용을 크게 받는 E. coli는 1%의 마늘즙액이 들어있는 TSB에서 사멸효과가 나타났으며 농도가 높을수록 사멸속도는 더욱 빨랐다. 마늘즙액에 의한 미생물생육 저해효과는 pH에 따라서도 다르게 나타나는데 pH 7.2에 비해 5.2와 6.2에서 저해작용이 강하게 나타났다. 초기 접종균수가 $10^{6}\;CFU/ml 이상일 때는 번식에 대한 저해효과가 나타나지 않았으나 $10^{5}\;CFU/ml 이하일 때는 저해효과가 나타나서 항미생물 작용에 대한 초기 미생물 수도 중요한 변수였다. 유리 SH기를 가진 cysteine이나 glutathione을 첨가하면 마늘즙액의 번식 저해효과로부터 E. coli를 보호하였다. 결과적으로 E. coli에 대한 마늘즙액의 번식 저해효과는 마늘즙액의 농도뿐만 아니라 pH, cystein이나 glutathione같은 SH화합물의 존재여부, 접종균수에 의해서 영향을 받음을 알 수 있었다.

Biological Synthesis of Baicalein Derivatives Using Escherichia coli

  • Two baicalein derivatives, baicalin and oroxylin A, were synthesized in this study. These derivatives exhibit diverse biological activities, such as anxiolytic and anticancer activities as well as memory enhancement. In order to synthesize baicalin from aglycon baicalein using Escherichia coli, we utilized a glycosyltransferase that regioselectively transfers glucuronic acid from UDP-glucuronic acid to the 7-hydroxy group of baicalein. To increase baicalin productivity, an araA deletion E. coli mutant, which accumulates UDP-glucuronic acid, was used, and ugd, which converts UDP-glucose to UDP-glucuronic acid, was overexpressed. Using these strategies, approximately $720.3{\mu}M$ baicalin was synthesized from $1,000{\mu}M$ baicalein. Oroxylin A was then synthesized from baicalein. Two O-methyltransferases (OMTs), ROMT-15 and POMT-9, were tested to examine the production of oroxylin A from baicalein. E. coli harboring ROMT-15 and E. coli harboring POMT-9 produced reaction products that had different retention times, indicating that they are methylated at different positions; the structure of the reaction product from POMT-9 was consistent with oroxylin A, whereas that from ROMT-15 was 7-O-methyl baicalein. Using E. coli harboring POMT-9, approximately 50.3 mg/l of oroxylin A ($177{\mu}M$) was synthesized from 54 mg/l baicalein ($200{\mu}M$).

Treatment of pigs with enrofloxacin via different oral dosage forms - environmental contaminations and resistance development of Escherichia coli

  • Background: Antibacterial agents play important roles in the treatment of bacterial infections. However, the development of antimicrobial resistance (AMR) and carry-over of substances into the environment are several problems arising during oral treatment of bacterial infections. We assessed AMR development in commensal Escherichia coli (E. coli) in enrofloxacin treated and untreated animals. In addition, we examined fluoroquinolone in the plasma and urine of treated and untreated animals, and in sedimentation dust and aerosol. Methods: In each trial, six pigs were treated with enrofloxacin via powder, granulate or pellet forms in two time periods (days 1-5 and 22-26). Four pigs served as untreated controls. The minimum inhibitory concentration (MIC) was determined to evaluate AMR development. Analysis of enro- and ciprofloxacin was performed with high performance liquid chromatography. Results: Non-wildtype E. coli (MIC > 0.125 ㎍/mL) was detected in the pellet treated group after the first treatment period, whereas in the other groups, non-wildtype isolates were found after the second treatment period. E. coli with MIC > 4 ㎍/mL was found in only the pellet trial. Untreated animals showed similar susceptibility shifts several days later. Bioavailability differed among the treatment forms (granulate > pellet > powder). Enro- and ciprofloxacin were detected in aerosols and sedimentation dust (granulate, powder > pellet). Conclusions: This study indicates that the kind of the oral dosage form of antibiotics affects environmental contamination and AMR development in commensal E. coli in treated and untreated pigs.
