Strain Improvement and Genetic Characterization of Tautomycetin Biosynthesis in Streptomyces spp.

  • TMC (Tautomycetin) is a liner polyketide immunosuppressive antifungal compound produced by Streptomyces spp. Inhibition of T cell proliferation with TMC was observed highly efficient at 100-fold lower than those needed to achieve maximal inhibition with cyclosporin A. To elucidate the biosynthetic pathway of TMC, a genomic DNA library was constructed using a E. coil-Streptomyces shuttle cosmid vector, pOJ446. The DNA libraries were screened by colony blot hybridization using several polyketide ${\beta}-ketosynthase$ (KS) probes amplified from TMC-producing Streptomyces genomic DNA using polymerase chain reaction (PCR), of which the degenerate primers were designed based on the highly conserved sequences present in KS domains of various type I polyketide synthase genes in Streptomyces species. This library construction and screening approach led to the isolation of several positive cosmid clones representing type I polyketide biosynthetic gene clusters. In addition, a Streptomyces regulatory gene called afsR2 (a global regulatory gene stimulating antibiotic production in both S. coelicolor and S. lividans) was successfully integrated into the TMC-producing Streptomyces chromosome via E. coil-Streptomyces heterologous conjugation mehtod. The more detailed results of production improvement and genetic characterization of TMC-producing Streptomyces spp. will be discussed.

LPG 및 Gasoline 겸용 차량의 엔진 점화시기 변환 제어시스템 개발 (The Development of the Ignition Spark Timing Conversion System for LPG/Gasoline Bi-fuel Vehicle)

  • In a bi-fuel engine using gasoline and LPG fuel, with the current ignition timing for gasoline being used, the effective performance could not be taken in LPG fuel supply mode. The ignition timing in LPG fuel mode must be advanced much more than that of gasoline mode for the compensation of its lower flame speed, due to engine torque drop. This study aims to develop the control system for ignition spark timing conversion which is composed of hardwares and control algorithm for gasoline/LPG engine. We propose the control system which can advance the ignition spark timing in LPG fuel mode more than used in gasoline fuel mode. The advance of ignition timing is achieved by change of the ignition dwell time of coil igniter. The engine torque and F/E(Fuel-Economy) in LPG fuel mode are measured to evaluate the difference of engine performance between before and alter changing ignition spark timings. The engine torque and F/E are increased respectively, which proves the developed control system is effective so much for gasoline and LPG bi-fuel engine.

의료용 무선 에너지 전달 장치의 개발 (Development of Transcutaneous Energy Transmission System for Medical Instrument)

  • 완전 이식형 인공심장 개발의 일환으로 무선에너지 전송장치를 개발하였다. 본 연구에서는 피하에 이식된 2차 코일과 피부표면에 놓인 1차 코일 간의 자기 유도의 원리에 의한 전기에너지 전송시스템을 개발하였으며, 이식될 소자의 소형화 및 전송효율의 향상을 목표로 하였다. 클래스 E 공진 증폭기를 이용하여 인공심자의 구동에 요구되는 40 Watt 전송시 75% 이상의 전송 효율을 얻었다. 또한 동물 실험을 실시하여, 체내 성능을 검증하였고 체내 이식부의 생체 적합성도 확인하였다.

  • In this paper, we designed the cylindrical type light source that had a electromagnetic principle of inductively coupled plasma, and measured its electrical-optical properties. Using the principle of transformer, electrically equivalent circuit of cylindrical type light source was analyzed. According to the parameters of electromagnetic induction which were diameter of coil with 0.3~1.2 mm, number of turns with 4~12 turns, distance with 40~120 mm and RF power with 10~150 W, the electrical and optical properties were measured. When diameter of coil was 0.3 mm, number of turns was 8 turns and distance was 40 mm, the highest brightness of 29,730 $cd/m^2$ was shown with RF power 150 W. The relationship between electromagnetic induction and plasma discharges was shown by mode transition from E-mode to H-mode.

재조합된 대장균의 세포성장에 미치는 저해제의 영향 (Effects of Inhibitor Concentrations on the Growth of Recombinant E. chli)

  • 재조합 대장균의 성장과 그 부산물 생성 관계를 알기위하여 탄소원을 glucose로하여 d-cycloserine, acetic acid 첨가에 따른 각각의 영향들을 검토하였다. 세포벽에 자극을 주는 d-cycloserine을 초기 농도 $0.1g/\ell$ 로 첨가하면 첨가하지 않는 경우에 비해 세포 성장이 93% 증가 했고 ${\alpha}$-amylase 생성은 5 5%정도 분비가 촉진 되였다. 그러나 d -cycloserine 초기 농도 $0.2g/\ell$로 첨가하면 세포와 a-amylase 아주 적게 생성되었다. 또한 d-cycloserine을 첨가하면 부산물에 lactic acid 생성에는 영향이 없으나 acetic acid는 적게 생성되었다. 그러므로 d­c cycloserine과 같은 자극제를 적당히 첨가하면 세포 성장 및 ${\alpha}$-amylase 분비가 촉진되지만 너무 많이 첨가 면 저해 작용이 얼어남을 알 수 있다. 저해제 acetic acid는 세포 성장에 작용을 주는 결과를 보였고 acetic acid의 농도가 높지 않는 상 태에서 lactic acid의 생성은 높은 양으로 생성 되엣 다. 이상의 결과에서 lactic acid를 적게 나오게 하 는것은 세포 수율에 큰영향을 주며 acetic acid가 척게 생성되면 성장에 좋은 영향을 주고 있다. 이상의 전체적인 결과는 세포 성장과 부산물 생성 상태를 고려할때 d -cycloserine은 초기 농도 $0.1g/\ell$ 정도 첨가하고 저해제 acetic acid의 생성을 적게 하는 배양방법을 검토 할 필요가 있다고 생각된다.

Bi-2212/Cu-Ni 튜브로 제작한 초전도 한류소자의 단락사고시험 결과 (Fabrication and Fault Test Results of Bi-2212/Cu-Ni Tubes for Superconducting Fault Current Limiting Elements)

  • For the development of superconducting fault current limiters (SFCLs), fault current limiting elements were fabricated out of Bi-2212 bulk tubes and tested. The SFCL elements consisted of tube shaped Bi-2212 bulks and metal shunts for the stabilizers. Firstly, the Bi-2212 bulk tubes were processed based on a design of monofilar coils in order to acquire large resistance and high voltage rating. 300 mm-long Bi-2212 tubes were designed to have the current path of 410 cm in length with 24 turns and 41 mm in diameter. The processed monofilar coil, as designed, had 300 A $I_c$ at 77 K. The fabricated superconducting monofilar coils were affixed to Cu-Ni alloy as that of stabilizers. The Cu-Ni alloys were processed to have the same shape of the superconducting monofilar coils. The Cu-Ni coil had resistivity of 32 ${\mu}{\Omega}$-cm at 77 K and 37 ${\mu}{\Omega}$-cm at 300 K. The metal shunts were attached to the outside of the Bi-2212 monofilar coil by a soldering technique. After the terminals made of copper were attached to both ends of the superconductor-metal shunt composite, the gap between the turns and the surface of the elements was filled with an epoxy and a dense mesh made of FRP in order to enhance the mechanical strength. The completed SFCL elements went through fault tests, and we confirmed that the voltage rating of 143 $V_{rms}$ (E =0.35 $V_{rms}$/cm) could be accomplished.

브라운관의 자기장 영향 분석용 저자기장 제어 장치의 설계 및 제작 (Development and Construction of low Magnetic Field Control System for Analysis of Magnetic Field Effect in the Deflection Yoke)

  • 컴퓨터, TV, 의료용기기 등의 모니터로 사용되는 브라운관의 자기장 영향에 따른 특성을 분석하기 위한 자기장 발생 및 측정장치를 개발하였다. 한 변의 길이가 2 m인 정육각형 형태의 3-축 자기장 발생장치, 컴퓨터, 전원공급기 및 자기장 측정기 등을 이용하여 전세계 각 나라별 지구자기장(Earth Magnetic Field, EMF)을 발생시킬 수 있는 대형 3-축 코일 시스템을 설계하고 제작하였다. 각 코일의 코일상수는 X축(E-W)는 30.31 $\mu$T/A, Y축(Ver)는 29.73 $\mu$T/A 및 Z축(N-S)는 30.51 $\mu$T/A이고, 자기장 분해능은 0.01 $\mu$T이었다. 자기장 균일도는 사용되는 편향요크(Deflection York, DY)의 크기를 고려하여 중심에서 12 cm 이내의 공간에서 측정하였고, 균일도는 1 % 내외 이었다.

유도전동기의 구속운전에 따른 전기 및 발열 특성 연구 (A Study on the Electrical and Heat Generation Characteristics of an Induction Motor under Restrained Operation)

  • In this study, we determined the failure rate and fire status of electric motors widely used in domestic and industrial devices and analyzed the associated fire risks by identifying the electrical and temperature characteristics of electric motors under the normal and restrained operation modes in industrial sites and laboratories. A 2.2kW motor used for driving a conveyor during the vulcanization process in a rubber product manufacturing plant was employed as the study object and was exposed to a high- temperature environment as this motor is widely used in industrial sites. The current amplitude was 4.45-4.50 A during normal operation and 38.2-41.5 A during restrained operation due to the pinching of products and semi-finished products (i.e., 8.5 times higher than that during normal operation). The leakage current amplitude was 0.33 mA during both operation modes. The temperature of the workplace in summer was 42.38℃, indicating a poor environment for the installed motor. In the laboratory, the current and temperature of the coil inside a 3.7kW motor were measured under the restrained operation mode as performing measurements of the coil inside the motor in industrial sites is challenging. The current amplitude during normal operation was 3.5 A, whereas that during restrained operation for 30 s was 51.7-58.6 A, which is 14.8-16.7 times higher than that of normal operation. Moreover, the temperature of the motor coil increased from 22.9℃ to 101℃. Based on the experimental data, we derived the temperature increase formula according to the restrained operation time by performing a regression analysis and verified the time at which the temperature exceeded the stipulated limit for the insulation grade. The findings presented in this paper can be utilized to establish fire-prevention measures and perform safety management of motors of the same type or with a similar capacity.

  • Contactless systems use electromagnetic fields to transport power from railway substation to the traction system. In Korea Railroad Research Institute, KRRI, the RIPS (Railway Inductive Power Supply), has been developed. The RIPS is based on a contactless transformer with a fixed coreless primary and a moving core, compared with E shape and U shape, in the secondary. The primary coil is supply with 20 kHz current which produces a magnetic flux in the secondary core. Further this flux induces a current in the secondary coil.

  • This paper deals with the effect of teeth number variation in permanent Magnet DC motor. As teeth number varies, both flux density distribution and winding pitch are influenced, which is closely related to torque-speed characteristics, output power, and efficiency. In this study, motor design carried out using finite element method, and prototype motors were manufactured to test their performance analysis. In spite of torque ripple due to cogging effect better characteristic of machine using small teeth number was recorded than the motors with large number of rotor teeth. One of that reasons is caused by adopting large coil-length due to large number of teeth, i. e. large coil-pitch.
