Development and Practical Use of RT-PCR for Seed-transmitted Prune dwarf virus in Quarantine

  • Among imported plants, seeds are the items that have many latent pathogens and are difficult to inspect. Also, they are the import and export items whose market is expected to expand. The biggest problem with seeds is viruses. Prune dwarf virus (PDV) is the virus that is commonly inspected in Prunus cerasifera, P. persica, P. armeniaca, P. mandshurica, P. cerasus, P. avium or P. serotina seeds. In this study, two RT-PCR primer sets, which can promptly and specifically diagnose plant quarantine seed-transmitted PDV, were developed; and nested PCR primers, where products amplify 739 and 673 nucleotides (nt), and an nested PCR-product, 305 nt, can be obtained as these products are amplified again, were developed. Also, a modified-positive control plasmid was developed, where the restriction enzyme XhoI, which can identify the contamination of samples from the control, was inserted. The method developed in this study has detected PDV in 18 cases since 2007, and is expected to continuously contribute to the plant quarantine in Korea.

Dormancy and Germination Characteristics of Round-Leaved Sundew (Drosera rotundifolia L.) Seeds Native to Korea (자생 끈끈이주걱(Drosera rotundifolia L.) 종자의 휴면과 발아특성)

  • This research was performed to develop mass propagation method of round-leaved sundew (Drosera rotundifolia L.) for improving horticultural usefulness. Seeds were collected around Hwaseong-si, Gyeonggi-do in October of 2011 and experimented upon while being dry-stored at 4 ± 1.0℃ (darkness). Seed length of a major and minor axis were 1.58 ± 0.060 and 0.21 ± 0.016 ㎜, respecrively, and weight of 1,000 seeds was 6.24 ± 0.172 ㎎. Seeds were thus classified as ‘dwarf seeds’. Regarding the dormancy type, since round-leaved sundew seeds were dormancy broken and germinated at 20~30℃ under the light condition after wet-chilling treatment for 12 weeks, seeds were estimated to have physiological dormancy. Germination conditions of dormancy broken seeds were found to be 20℃ and light condition (54.7%), but germination decreased at higher temperature. Percent germination (PG), germination energy (GE), mean germination time (MGT) and T50 were effectively improved by chemical treatment such as GA3 200 ㎎/L + kinetin 20 ㎎/L and wet-chilling treatment for 14 weeks. In conclusion these optimal conditions were thought to be a useful method for raising seedling uniformly.

Dormancy and Germination Characteristics of Alpine Modest Primrose (Primula modesta var. hannasanensis T.Yamaz.) Seeds (설앵초의 종자 휴면과 발아특성)

  • This research was performed to develop mass propagation method of Alpine modest primrose (Primula modesta var. hannasanensis T.Yamaz.) for improving the conservation and utilization of the species. Seeds were collected on August 2011 in Jeju-city and well-selected seeds were dry-stored at $4{\pm}1.0^{\circ}C$. Seed size ranged $0.44{\pm}0.07{\times}0.61{\pm}0.04mm$, and weight of 1,000 seeds was $51.78{\pm}0.021mg$. Thus the seed was classified as 'dwarf seeds'. As the result of dormancy characteristics, moisture content of freshly matured seeds increased rapidly by water-soaking treatment and seeds did not germinate at $20^{\circ}C$ for 4 weeks under the light condition. Therefore, seeds were estimated to have physiological dormancy. Germination conditions of dry-stored seeds were found to be $20^{\circ}C$ and light condition, and seeds did not germinated under dark condition regardless of all temperature regimes. Therefore, Alpine modest primrose seed was considered as photoblastic type. Percent germination (PG) and germination energy (GE) were greatly improved by soaking the seeds in $GA_3$ and kinetin for 24 hours. Especially, 200~500 mg/L $GA_3$ treatment resulted in the highest PG (95.5%) and GE (98.3%). So, chemical treatment such as $GA_3$ was thought to be a useful method for raising seedling uniformly.

  • Seeds of five radish varieties were soaked in water for three hours and cellulose was coated. The seeds were classified as three groups in UV light; non-fluorescent(NF), partly fluorescent(PF), and fully fluorescent(FF) seeds. Germination rate was less in the order of NF>PF>FF seeds. The seed coat structure of NF seeds was dense and showed round shape, while those of PF and FF seeds were wrinkled or ruptured. The cotyledon and hypocotyl of NF seeds were normal, while those of PF and FF seeds were dwarf and showed some scars in the cotyledons.

Effects of Windbreak Fences Composed of Natural Vegetation on Dwarf Siberian Pine (Pinus pumila) Seedlings (식생을 이용한 방풍책이 눈잣나무 유묘에 미치는 영향)

  • In this study, the effects of windbreak fences composed of natural vegetation on one-year-old seedlings were analyzed to develop restoration methods for an endangered subalpine species, the dwarf Siberian pine (Pinus pumila (Pall.) Regel). One-year-old seedlings were planted in 2016 by sowing seeds that had been collected from the Daecheongbong area on Mt. Seoraksan, South Korea, in 2014. The area near Daecheongbong was selected as the experimental site, and treatment and control plots (2m×2m) were installed at the site. To analyze the effects of wind protection, windbreak fences were constructed in the treatment plots using hairy Korean rhododendrons (Rhododendron mucronulatum Turcz. var. ciliatum Nakai) from the surrounding area and weather stations were installed to investigate atmospheric temperature, humidity, and wind speed. In all control plots without windbreak fences, dwarf Siberian pine seedlings were killed by strong winds seven months after planting. In contrast, the average survival rate of the seedlings in treatment plots was 96.7% after seven months, 64.2% after two years, and 45% after three years, with most (85.3%) of the seedlings showing good initial root establishment. Accordingly, windbreak fences composed of natural vegetation are suitable for promoting the early establishment of seedlings in the restoration of dwarf Siberian pine stands.

Effect of Wet Cold and Gibberellin Treatments on Germination of Dwarf Stone Pine Seeds (저온습윤 및 지베렐린 처리가 눈잣나무의 종자발아에 미치는 영향)

  • In South Korea, Pinus pumila (Pall.) Regel (dwarf stone pine) has been designated as a critically endangered species by the Korea Forest Service. We have difficulties in obtaining the seeds of P. pumila because P. pumila grows only in the Daecheongbong area (1550–1700 m above sea level) of Mt. Seorak and almost all of its cones are damaged by birds and rodents. For establishing an ex situ conservation stand of P. pumila, this study was conducted to figure out the effects of wet cold (cold stratification, prechilling) and GA3 treatment on the germination of P. pumila seeds. After cold stratification (1, 2, 3, 4, 5 months), prechilling (1, 2, 3, 4, 5 months) and GA3 treatment (0, 100, 500, 1,000, 2,000, 3,000 ㎎/L), seeds were placed on petri-dishes at 25℃ under light condition. The percentage of germination, mean germination time and the germination rate were investigated. The results showed that both of the cold stratification and prechilling were effective in improving germination performances. However, there were no significant differences in performances between the two cold treatments. Within each treatment, the germination performances improved with the period of treatment. However, after three months of treatment, the performances showed no significant improvement. The gibberellin treatment was also effective in improving seed germination of P. pumila. The percentage of germination reached 79.0% in the seeds treated with 100 ㎎/L of GA3. However, the germination performances decreased at high concentration of GA3 treatments (over 2000 ㎎/L). In conclusion, cold stratification (over 3 months) or 100 ㎎/L of GA3 treatment was considered to be the appropriate method for seedling production of P. pumila.

  • 1) Germination rate was rather irregular than decreasing as increasing dose of radiation and there were no differences between Kyong-Sam and Chuong-Bang of Chinese cabbage. 2) In R1 generation, abnormal leaves from seedling of irradiated seeds were observed. These were more apparent in X-ray irradiation than in thermal neutron. 3) Seedling height was inhibited with increasing dose of X-ray and thermal neutrons. Growth inhibition was more remarkable in X-ray than in thermal neutron. Kyong-Sam demonstrated more sensitivity than Chyong-Bang in both X-ray and thermal neutron. 4) Seedling height produced from seeds subjected to thermal neutrons showed small variation around its mean value, while in X-irradiation there was a greater deviaton from the mean value. 5) Fertility was decreased as increasing with dose, while the frequency of abortive pollen was increased. There were variability of the fertility and frequency of abortive pollen among plants or branches of a plant. 6) The mutants were obtained more in thermal neutron irradiation than in X-ray. The types of mutations obtained in Chinese radish of R2 generation were abnormal leaf, densely glowing leaf, degeneration in growing point and dwarf. The maximum frequency of phenotypic mutations was abnormal leaf mutation.

A study on distribution of plant communities around chunji in a crater on mt. paektu (백두산 분화구내 천지 주변의 식물 군락 분포에 관한 연구)

  • We investigated major plant communities and seed dispersal patterns in areas a(alt. 2,263 ~2,350m),b(alt. 2,350~2,420m), c(alt. 2,420m) with levels of a crater on mt. paektu. Thirty two species distributed around chunji consisted of 24 perennial herbs, 1 biennial herb and 7 alpine dwarf shurbs. Their fruit types were quite diverse; capsule(11), achene(10), grain(4), legume(2), follicle(2), schizocarp(2) and drupe(1). in area a, adjancent to the chunji(lake) water surface, gramineae sp., cyperaceae sp., chrysanthemum zawadskii and bistorta ochotensis were occured as members of the major plant communities. Species of gramineae and cyperaceae seem to have become dominant because their seeds float well. Astragalus membranaceus is legume and endures the winter cold and summer drought. in area c, alpine dwarf shrubs (e. g.rhododendron redowskianum, rhododendron aureum, empetrum nigrum) were dominant. In area c, compared to a and b, the weather is drier because of strong wind and lower water holding capacity of the soil. Therefore the pattern of plants distributing around chunji is different from that in western slope on mt.paektu. environmental factor such as wind, temperature and water may contribute to the pattern.

Systematic Development of Tomato BioResources in Japan

  • Recently, with the progress of genome sequencing, materials and information for research on tomato (Solanum lycopersicum) have been systematically organized. Tomato genomics tools including mutant collections, genome sequence information, full-length cDNA and metabolomic datasets have become available to the research community. In Japan, the National BioResource Project Tomato (NBRP Tomato) was launched in 2007, with aims to collect, propagate, maintain and distribute tomato bioresources to promote functional genomics studies in tomato. To this end, the dwarf variety Micro-Tom was chosen as a core genetic background, due to its many advantages as a model organism. In this project, a total of 12,000 mutagenized lines, consisting of 6000 EMS-mutagenized and 6000 gamma-ray irradiated M2 seeds, were produced, and the M3 offspring seeds derived from 2236 EMS-mutagenized M2 lines and 2700 gamma-ray irradiated M2 lines have been produced. Micro-Tom mutagenized lines in the M3 generation and monogenic Micro-Tom mutants are provided from NBRP tomato. Moreover, tomato cultivated varieties and its wild relatives, both of these are widely used for experimental study, are available. In addition to these bioresources, NBRP Tomato also provides 13,227 clones of full-length cDNA which represent individual transcripts non-redundantly. In this paper, we report the current status of NBRP Tomato and its future prospects.

Morphological and Progeny Variations in Somaclonal Mutants of 'Ilpum' (Oryza sativa L.) ('일품'벼 체세포변이체의 표현형과 후대변이)

  • A total of 424 plants was regenerated from the seed-culture of a rice cultivar, 'Ilpum'. The regenerated plants were grown in a greenhouse. The 297 plants with high fertility were selected among 424 plants. The harvested seeds from each plant were planted to each line at experiment field in 2008 and 2009. The each line was evaluated for the agronomic and morphological traits, also. The 64 lines (21.5%) showed significant differences in agronomic and morphological traits from donor cultivar 'Ilpum' among 297 lines. The heading date different from donor cultivar 'Ilpum' showed highest frequency in 297 lines, and accounts for 9.1% (29 lines). The phenotype of opaque endosperm and rolling leaf account for 1.7% and 1.3% in 297 lines, respectively. The genetic segregation was observed in dwarf/semi-dwarf, rolling leaf and opaque endosperm at $S_1$ generation, but not in $S_2$ generation. These results suggest that the mutant derived from a tissue-culture will be one of the promising genetic resources, due to its wide variation and high frequency of mutation, comparatively.
