Sensitization of Children to Storage Mites in Kutahya, Turkey

  • Specific IgE against Acarus siro, Glycphagus domesticus, Tyrophagus putrescentiae, and Lepidoglyphus destructor have been investigated by ELISA in sera of 92 children. Of them, 41 were found to be specific IgE positive ($\geq$ 0.35 IU/ml) against at least one of house dust mite species, Dermatophagoides pteronyssinus and Dermatophagoides farinae, by an immunoblot. In 65.9% of the dust mite-sensitized children, specific IgE against at least one of these mite species was found. Sensitization levels, including co-sensitization cases were found to be 35.7% against A. siro, 24.4% against T. putrescentiae, 31.7% against L. destructor, and 26.8% against G. domesticus. In non-sensitized children, dust mite sensitization level was found to be 25.5%. Breakdown of sensitization by individual species in this group was; against A. siro and T. putrescentiae at 7.8%, against L. destructor at 13.7%, and against G. domesticus at 9.8%. When all children were reckoned, 43.5% was found to be sensitized against at least one storage mite species, with sensitizations against A. siro at 18.5%, T. putrescentiae at 26.1%, L. destructor at 21.7%, and G. domesticus at 17.4%. In dust samples collected from the dwellings of children, distribution of species was found to be A. siro (17%), G. domesticus (23%), T. putrescentiae (29%), L. destructor (25%), and unidentified (6%). In Fisher's chi-square test on SPSS program, there was a relationship between dust mite sensitization and storage mite sensitization (P < 0.05), but no meaningful relationship was found on the basis of individual mite species.

A Survey of House Dust Mite Allergen Contamination in House (일부 주택에서 집먼지 진드기 알러젠 조사)

  • House dust mite was the most important cause of allergic asthma and rhinitis. More than 70% of Korean children and about 50% of adult with respiratory allergy were sensitive to house dust mite. This experiment was indoor environment in house and house dust mite existence inquiry. From 23rd December 2005 to 28th February 2006, dust samples were collected from the bedclothes by a vacuum cleaner and air sampler And the levels of Der f1 and Der p1 were determined by ELISA((Enzyme-Linked Immunosorbent Assay). The results were as follows : 1 The dust mites allergy contaminations of bedclothes in house were higher than international standards(2000 ng/g). 2. In type of dust mite, the Korean house the almost have the D. farinae other than D. pteronyeeinus of mite. 3. The Der f1 and Der p1 levels per gram of dust from the bedclothes were 2074.99 ng on average, but they did not exist in air. The concentrations of house dust mite were significantly high in the bedclothes. This results suggest that the bedclothes have enough concentrations of dust mites to develop the sensitization. The control of indoor environment should be emphasized to prevent the sensitization by the repeated exposure to dust mite.

The Relationship between House Dust Mite Sensitization and Month Birth Distribution in Children with Respiratory Allergy (호흡기 알레르기 환아에서 집먼지진드기 감작과 출생 월 분포의 연관성)

  • Purpose : It has been suggested that the exposure to aeroallergens during early infancy after birth is important in the subsequent development of sensitization and allergic diseases. In Korea, the level of house dust mites as one of the important aeroallergens is known to be the highest in autumn. The aim of this study was to test whether the distribution of month of birth bears a relationship to the presence of mite sensitization in children with respiratory allergy. Methods : Skin prick tests and methacholine provocation tests were performed on 1,327 patients with chronic respiratory symptoms who visited Seoul National University Children's Hospital from January 1995 to May 2002. An analysis of patients' month of birth distribution according to the presence of mite sensitization was performed. Results : Atopic subjects who had at least one positive skin test numbered 864(65.1%); and non-atopic subjects numbered 463(34.9%). Among atopic subjects, 787(59.3%) had positive skin tests to mites and 77(5.8%) had positive skin test only to minor allergens. A significantly greater than expected number of mite atopic subjects were born in the months between August and November(P=0.03), however, the birth month of non-atopic subjects didn't show a consistent seasonal preference. Asthma patients numbered 543(40.9%). Among these, atopic asthmatics numbered 421(77.5%) and non-atopic asthmatics, 122(22.5%). Dust-mite atopic asthmatics numbered 387(91.9%) out of 421 atopic asthmatics. Dust-mite atopic asthmatics were born significantly higher in the season lasting from August to November in comparison to non-atopic asthmatics(P=0.002). Conclusion : Month of birth seems to be related with sensitization to allergens. Our results show that August to November is the risk period for the development of mite sensitization in Korea.

The Sensitization Rates for Inhaled Allergens by Skin Prick Test among Some Farmers in Gyeonggi Province, South Korea (피부 단자 검사로 평가한 경기도 일부 농업인의 흡입 알레르겐 감작률)

  • Objectives: Farmers are known to be exposed to a variety of allergens related to the work environment. This study was conducted to determine the sensitization rates as well as South Korea that they are sensitized to certain allergens farmers through the skin prick test. Methods: By targeting a total of 1143 people living in the rural town of Gyeonggi Province, it was conducted a questionnaire containing demographic and occupational risk factors and underwent skin prick tests with 15 types of allergens(including positive and negative controls). Multivariable logistic regression analysis was used to analyze the association between occupational risk factors and skin prick test positivity. Results: Except for the 30 people whose result is invalid, positive rate of the skin prick test was 18.6% in 1,113 people. The species of house dust mite, Dermatophagoides pteronyssinus and Dermatophagoides farinae was the highest at 8.7% and 8.6%. After adjusted by age, gender, smoking and education level, odds ratio of flower plant farmers is 4.467(95% CI: 2.094-9.527) and fruit farmer is 2.275(95% CI: 1.096-4.721). In addition, the rate of sensitization to grass pollen mixture of the flower plant farmers is significantly higher(15.9%, p<0.001) than other allergens. Conclusions: Even farmers, the rate of sensitization to allergens related to the general environment, such as house dust mite is relatively dominant. However, given the presence of potential cross-reactivity between the allergens or distribution showed that the unique aspects of allergen sensitization in the flower growers, occupational cause is not be completely ruled out.

  • Purpose: Aeroallergens are important causative factors of allergic diseases. Previous studies on aeroallergen sensitization rates investigated patients groups that had visited pediatric allergy clinics. In contrast, we investigated sensitization rates in a general population group of elementary school to teenage students in Incheon, Jeju, and Ulsan. Methods: After obtaining parental consent, skin-prick tests were performed on 5,094 students between March and June 2010. Elementary school students were tested for 18 common aeroallergens, whereas middle and high school students were tested for 25 allergens. The 25 allergens included Dermatophagoides pteronyssinus, Dermatophagoides farinae, pollen (birch, alder, oak, Japanese cedar, pine, willow, elm, maple, Bermuda grass, timothy grass, rye grass, orchard grass, meadow grass, vernal grass, mugwort, Japanese hop, fat hen, ragweed, and plantain), and mold (Penicillatum, Aspergillus, Cladosporium, and Alternaria). Results: The sensitization rates in descending order were 25.79% (D. pteronyssinus ), 18.66% (D. farinae ), 6.20% (mugwort), and 4.07% (willow) in Incheon; 33.35% (D. pteronyssinus ), 24.78% (D. farinae), 15.36% (Japanese cedar), and 7.33% (Alternaria) in Jeju; and 32.79% (D. pteronyssinus), 30.27% (D. farinae), 10.13% (alder), and 8.68% (birch) in Ulsan. The dust mite allergen showed the highest sensitization rate among the 3 regions. The sensitization rate of tree pollen was the highest in Ulsan, whereas that of Alternaria was the highest in Jeju. The ragweed sensitization rates were 0.99% in Incheon, 1.07% in Jeju, and 0.81% in Ulsan. Conclusion: The differences in sensitization rates were because of different regional environmental conditions and distinct surrounding biological species. Hence, subsequent nationwide studies are required.

Distribution of Allergen Reactivity in Serum of Allergy Patients

  • This study was conducted to evaluate the distribution, frequency, sensitization rate, and types of specific allergen in allergic patients. We analyzed allergens of 12,882 allergic patient's serum referred for Medical Laboratory using MAST Allergy Screen Test (Inhalation/food panel) from April, 2010 to March, 2011. Allergen reactivity were not detected in 833/12,882 (6.5%) allergic patients and the other allergic patients had two or more types of allergens of food and inhalant, respectively. In the distribution of the allergen food 73.4% (9,450/12,882 patients) was much higher than the pollen 26.6% (3,432/12,882 patients). The sensitization rate to each allergen showed garlic 26.7%, egg white 21.5% of food allergens, mugwort 24.9%, ragweed short 19.6% of pollen allergens, Candida albicans 10.5%, Alternaria spp. 7.9% of fungus allergens, and showed high sensitized rate to Dermatophagoides farinae 90.2%, Dermatophagoides pteronyssinus 77.8%, house dust 57% of indoor allergens, respectively. The specific allergens of food allergen, fungus allergen, and indoor allergen were more frequent in young child than adult, but pollen allergens were frequent in adult than young child. In seasonal distribution, the food allergen were frequent in summer and autumn, the pollen allergen in autumn, fungus allergen in spring and in winter, and indoor allergen in autumn and in winter. In conclusion, the mite of Dermatophagoides farinae and Dermatophagoides pteronyssinus, and house dust showed the highest sensitization rate in specific allergen of allergic patients. The allergens showed difference feature according to the age, region, and types of allergen.

Microstructure of Faecal Pellets and Silk of the Two Spotted Spider Mite, Tetranychus urticae (Tetranychidae: Acarina) (점박이응애 분비물의 미세구조)

  • Asthma and allergic rhinitis due to outdoor spider mites are major health problems worldwide. The sensitization route to spider mites has not yet been well elucidated. We examined the microstructure of faecal pellets and silk of the two-spotted spider mite, Tetranychus urticae, using a field emission scanning electron microscope. Black and white pellets of the two-spotted spider mites faeces contain a large amount of plant pigment waste products. Black faecal pellets are strawberry-shaped. White faecal pellets are silken threads. These pellets are likely to be the source of allergens of relevant mites because desiccated faeces particles probably disintegrate and become incorporated into dust particles more readily than whole bodies or encased internal organs. We conclude that the importance of spider mites in respiratory allergy needs emphasis.

Enzymatic Activities of Allergen Extracts from Three Species of Dust Mites and Cockroaches Commonly Found in Korean Home

  • Allergen extracts from dust mites and cockroaches commonly found in Korean homes were used to evaluate their enzymatic activity as they are believed to influence allergenicity. Allergen extracts were prepared from 3 dust mite species (Dermatophagoides farinae, D. pteronyssinus, and Tyrophagus putrescentiae) and 3 cockroach species (Blattella germanica, Periplaneta americana, and P. fuliginosa) maintained in the Korea National Arthropods of Medical Importance Resource Bank. Proteins were extracted in PBS after homogenization using liquid nitrogen. The activities of various enzymes were investigated using the API Zym system. No significant difference in phosphatase, lipase, or glycosidase activity was observed among the 6 allergen extracts, but much difference was observed in protease activity. Protease activity was assessed in more detail by gelatin zymography and the EnzChek assay. Extract from T. putrescentiae showed the highest protease activity, followed by those of the cockroach extracts. Extracts from D. farinae and D. pteronyssinus showed only weak protease activity. Gelatinolytic activity was detected mainly in a 30-kDa protein in D. farinae, a 28-kDa protein in D. pteronyssinus, a > 26-kDa protein in T. putrescentiae, a > 20-kDa protein in B. germanica, and a > 23-kDa protein in P. americana and P. fuliginosa. The information on various enzymatic activities obtained in this study may be useful for future studies. In particular, the strong protease activity found in cockroach extracts could contribute to sensitization to cockroach allergens, which is known to be associated with the development of asthma.

Influence of the Adjuvants and Genetic Background on the Asthma Model Using Recombinant Der f 2 in Mice

  • Der f 2 is the group 2 major allergen of a house dust mite (Dermatophagoides farinae) and its function has been recently suggested. To determine the optimal condition of sensitization to recombinant Der f 2 (rDer f 2) in murine model of asthma, we compared the effectiveness with different adjuvants in BALB/c and C57BL/6 mice. Mice from both strains sensitized with rDer f 2 by intraperitoneal injection or subcutaneous injection on days 1 and 14. The dosage was $20{\mu}g$. Freund's adjuvants with pertussis toxin (FP) or alum alone were used as adjuvants. On days 28, 29, and 30, mice were challenged intranasally with 0.1% rDer f 2. We evaluated airway hyperresponsivenss, eosinophil proportion in lung lavage, airway inflammation, and serum allergen specific antibody responses. Naive mice were used as controls. Airway hyperresponsiveness was increased in C57BL/6 with FP, and BALB/c with alum (PC200: $13.5{\pm}6.3$, $13.2{\pm}6.7$ vs. >50 mg/ml, p<0.05). The eosinophil proportion was increased in all groups; C57BL/6 with FP, BALB/c with FP, C57BL/6 with alum, BALB/c with alum ($24.8{\pm}3.6$, $20.3{\pm}10.3$, $11.0{\pm}6.9$, $5.7{\pm}2.8$, vs. $0.0{\pm}0.0$%, p<0.05). The serum allergen specific IgE levels were increased in C57BL/6 with FP or alum (OD: $0.8{\pm}1.4$, $1.1{\pm}0.8$, vs. $0.0{\pm}0.0$). C57BL/6 mice were better responders to rDer f 2 and as for adjuvants, Freund's adjuvant with pertussis toxin was better.

Changes of Causative Allergen in Patients with Allergic Diseases in Chonbuk Province (전북지역 알레르기 환자에서 원인 알레르겐의 변화)

  • Background: The prevalence of allergic disease has increased substantially over the past decades. The reasons for these trends are complex but include increased environmental allergen exposure. Environmental control measures are of particular importance in the prevention and management of allergic disease. Thus, we purposed to evaluate the changes of causative allergens in allergic patients in Chonbuk provincial area that began industralized recently. Method: To evaluate the allergens epidemiologically, we performed skin prick tests and RAST in 201 allergic patients from February, 1984 to February, 1985 and 339 patients from February, 1997 to February, 1998. Results: The Following results were obtained. 1) Total sensitization rates did not change significantly(74.2% vs 75.8%, p=0.664). 2) Positive reaction rates to pollens were similar(35.8% vs 30.1%, p=0.17). 3) Positive rates to HDM(house dust mite) were decreased(66.1% vs 56.3%, p=0.02). 4) Positive rates to fungi and animal danders were decreased significantly(p<0.001). Conclusion : Environmental allergens associated with hygine in Chonbuk provincial areas such as HDM and fungi are decreased significantly during 13 years.
