Industrial pig breeding has used the Duroc breed and terminal sires in a three-way crossbred system in Korea. This study identified the gene variation patterns related to carcass quality in crossbred pigs ([Landrace × Yorkshire] × Duroc) using whole-exome sequencing (WES). This study used crossbred pigs and divided them into two groups (first plus grade, n = 5; second grade, n = 5). Genomic DNA samples extracted from the loin muscles of both groups were submitted for WES. A set of validated single-nucleotide polymorphisms (SNPs: n = 102) were also subjected to the Kompetitive allele-specific polymerase chain reaction (KASP) to confirm the WES results in the loin muscles. Based on the WES, SNPs associated with meat quality were found on chromosomes 5, 10, and 15. We identified variations in three of the candidate genes, including kinesin family member 5B (KIF5B), GLI family zinc finger 2 (GLI2), and KIF26B, that were associated with meat color, marbling score, and backfat thickness. These genes were associated with meat quality and the mitogen-activated protein kinase (MAPK) and Hedgehog (Hh) signaling pathways in the crossbred pigs. These results may help clarify the mechanisms underlying high-quality meat in pigs.
The objective of this study was to investigate effects of supplementation levels of L-leucine in low-lysine diets on growth performance and meat quality parameters in finishing Duroc pigs. A total of fifty-four pigs with an average initial weight of 74±1kg were allotted to one of three dietary treatments. Each treatment had six replications of three pigs per replicate. The treatment diets included 1)a corn-soybean meal basal diet containing lysine as low as 0.45% (CON), 2) basal diet plus 1.5% L-leucine (LEU 1.5), and 3) basal diet plus 3.0% L-leucine (LEU 3.0). No difference was found in ADG, ADFI and Feed/Gain among treatments. In carcass parameters, dressing precent and back-fat thickness were not affected by L-leucine levels, however, longissimus dorsi area tended to increase by supplementation of L-leucine. Marbling score was significantly higher (P<0.05) in LEU 1.5 and LEU 3.0 than in CON. In meat quality parameters, CIE L* of meat color significantly increased (P<0.05) and crude fat tended to increase by supplementation of L-leucine. However, there was no difference in water holding capacity, cooking loss and shear force value. This study suggests that dietary supplementation of L-leucine in low-lysine diets at the end of finishing period can produce high marbled pork which is preferred by Korean consumers.
Five regions and 258 pigs were selected for this study: North (Beijing), Central (Wuhan), South (Guangzhou), Southwest (Chongqing), Northeast (Harbin). Five typical genetics of growing-finishing pig were selected: Landrace${\times}$Large White${\times}$Beijing Black, Duroc${\times}$Landrace${\times}$Large White, Duroc${\times}$Large White${\times}$Landrace, Landrace${\times}$Rongchang, Landrace${\times}$Harbin White, respectively at each sites. The basal diet was a corn-soybean meal containing sufficient nutrients to meet requirements. Carcass fat-free lean gain was determined by dissecting and analyzing chemical composition of the carcass. Cubic function fitted lean moistures to live weights better than other functions. Exponential function fitted lean lipids to live weights equally to allometric function. Carcass fat-free lean gain of Duroc${\times}$Large White${\times}$Landrace, Landrace${\times}$Large White${\times}$Beijing Black, Duroc${\times}$Landrace${\times}$Large White, Landrace${\times}$Harbin White, Landrace${\times}$Rongchang from 20 to 100 kg of average body weight was 259 g/d, 261 g/d, 311 g/d, 220 g/d, 200 g/d, respectively. All are lower than intermediate fat-free lean gain in NRC (1998).
Pigs may need to be exploited as xenotransplantation donors due to the shortage of human organs, tissues and cells. Porcine endogenous retroviruses (PERVs) are a significant obstacle to xenotransplantation because they can infect human cells in vitro and have the potential for transmission of unexpected pathogens to humans. In this research, 101 pigs, including four commercial breeds (23 Berkshire, 13 Duroc, 22 Landrace and 14 Yorkshire pigs), one native breed (19 Korean native pigs) and one miniature breed (10 NIH miniature pigs) were used to investigate insertional variations for 11 PERV loci (three PERV-A, six PERV-B and two PERV-C). Over 60% of the pigs harbored one PERV-A (907F8) integration and five PERV-B (B3-3G, B3-7G, 742H1, 1155D9 and 465D1) integrations. However, two PERV-A loci (A1-6C and 1347C1) and one PERV-B locus (B3-7F) were absent in Duroc pigs. Moreover, two PERV-C loci (C2-6C and C4-2G) only existed in Korean native pigs and NIH miniature pigs. The results suggest that PERV insertional variations differ among pig breeds as well as among individuals within a breed. Also, the results presented here can be used for the selection of animals that do not have specific PERV integration for xenotransplantation research.

This study evaluated whether dietary supplementation of tapioca could alleviate the negative effects of palm kernel meal (PKM) on growth performance and meat quality in growing and finishing pigs. In experiment 1, 120 73-dold crossbred growing pigs [$(Yorkshire{\times}Landrace){\times}Duroc$], with an average body weight of $31.7{\pm}4.5kg$, were used in a 3-week trial. In experiment 2, 120 108-d-old crossbred finishing pigs [$(Yorkshire{\times}Landrace){\times}Duroc$], with an average BW of $52.6{\pm}4.2kg$ were used in a 10-week trial. Treatments were: CON, a corn-soybean meal-based diet; PKM, 8% PKM, and TPKM, 8% PKM and 10% tapioca. No difference was observed in growth performance or meat quality among treatments in growing pigs. In finishing pigs, no difference was observed in growth performance or meat quality among CON and TPKM dietary treatments. Finishing pigs fed PKM decreases in final BW and ADG compared with those fed CON. Meat quality was not affected by dietary treatments. In conclusion, dietary supplementation of tapioca alleviated anti-nutritional effect of PKM on growth performance in growing and finishing pigs. Thus, the PKM with tapioca could be an available alternative energy source to reduce the cost of pig diets.
For the Exp. 1, twelve Duroc $\times$ Yorkshire $\times$ Landrace pigs (6.12 $\pm$ 0.24 kg average initial body weight and 21 d average age) were used in an 8-d metabolic assay to determine the effects of replacing spray-dried plasma protein (SDPP) with spray-dried egg Protein containing egg yolk antibody (SDCEP) on nutrient and amino acids digestibility in weaning Pigs. Experimental animals were fed diets containing SDPP, SDCEP (spray-dried commercial egg protein) and SDAEP. Protein content in the SDPP, SDCEP and SDAEP were 58.20, 45.83 and 41.85%, respectively. Pigs fed the SDPP diet tended to increase the apparent digestibility of dry matter and nitrogen compared to pigs fed the SDAEP diets without significant differences. The ileal digestibility of lysine and methionine for the SDAEP were greater than those for the SDPP, however, there are no significant differences between groups. For Exp.2, 36 Duroc $\times$ Yorkshire $\times$ Landrace pigs (4.11 $\pm$ 0.05 kg average initial body weight and 14 d average age) were used in a 10-day growth assay to determine the effects on growth performance and nutrient digestibility of replacing SDPP with SDAEP in early-weaning pigs. Experimental animals were fed diets containing CON (corn-dried whey-SBM based diet), SDAEP3 (corn-dried whey-SBM based diet + 3% SDAEP), SDAEP6 (corn-dried whey-SBM based diet + 6% SDAEP). The average daily weight gain of the pigs fed the SDAEP3 diet was higher than that for the pigs fed the CON and SDAEP6 diets (p < 0.05). SDAEP3 significantly increased the digestibility of dry matter and nitrogen compared to the CON and SDAEP6 diets (p < 0.05).
The association between the promoter region genotypes of the porcine adipocyte fatty acid binding protein (A-FABP) gene and carcass traits in commercial pigs was examined. Interestingly, only two different genotypes (-406T/T and T/C) for the A-FABP gene were detected using Hinf-RFLP for the substitution mutation T-406C in the A-FABP promoter in commercial pigs, and no -406C/C homozygotes were detected. The reason for the lack of the -406 C/C genotype is due to the breeding system in which only Duroc, which has a high frequency of -406C/- in this locus among the three breeds involved in commercial pig production, is typically used as a terminal sire. The pigs containing the genotype -406C/- were significantly associated with an increase in intramuscula. fat content and carcass weight (p<0.05), but there was no association with the other carcass traits tested (fat composition, color score, texture score, moisture, and separation score between muscles). This study suggests that the -406C/- genotype of the porcine A-FABP gene may not only be a useful molecular marker for intramuscular fat, but may also contribute to the improvement of meat quality by the production of well-marbled pigs by breeding animals containing this genotype, especially Duroc, as a terminal sire for commercial pigs.
This study was carried out to investigate the effects of imported pellet frozen semen on sperm motility and NAR acrosomes after thawing, and the farrowing rates, litter sizes and preweaning body weights after artificial insemination. A total of 28 sows of Landrace, Large White and Duroc were inseminated at the Chungnam Provincial Animal Breeding Station. The results obtained are summarized as follows: 1. Landrace andLarge White had higher sperm motility than Duroc by about 20% and had higher NAR acrosomes by about 10%. 2. The farrowing rates of Landrace, Large White and Duroc were 63.6, 55.6 adn 50.0%, respectively. The number of pigs born alive per litter were larger in Landrace and Large White as compared with Duroc (p<.01). 3. Duroc had the highest mean pig weight at birth, followed by Landrace and Large White (p<.01). The mean pig weights at 21 days and 56 days had no significant differences between the breeds.
The objective of this study was to investigate the relationship between fertility and protein pattern change using in vitro fertilization, analysis of sperm characteristics and two-dimensional gel electrophoresis in different pig types. In results, the viability and mitochondria integrity of sperm were higher significantly (p<0.05) but the portions of acrosome reaction was lower significantly (p<0.05) in Duroc and $F_1$ (potbellied ${\times}$ PWG miniature pig) than PWG miniature. On in vitro fertilization to investigate fertility, the fertility of $F_1$ semen war higher significantly (p<0.05) than in Duroc and PWG miniature pig. On the other hand, protein patterns showed similar function among the different boar semen. Especially, the heat shock 70 kDa 1-like and G patch domain-containing protein 4 were significantly (p<0.05) higher expressed in $F_1$ than in Duroc and PWG miniature pig. The proteins associated with mitochondria in Duroc were significantly (p<0.05) higher expressed than in $F_1$ and PWG miniature pig. The developmental rates to blastocyst stage of oocytes fertilized with sperm of $F_1$ pig were significantly (p<0.05) higher than in PWG miniature pig. However, phosphoglycerate kinase 2 and zinc finger protein 431 were significantly (p<0.05) higher expressed in PWG miniature pig than in $F_1$ and Duroc pigs. In conclusion, the results of the present study indicate that different proteins were expressed in different pig types, and were associated with a sperm functions and embryo development.
