The purpose of this study was to isolate lactic acid bacteria, useful in the fermentation industry from Hahyangju Nuruk. Five strains were isolated, and identified as Lactobacillus based on growth inhibition by 10% (v/v) alcohol at pH 4.0. Isolated strains were identified to species, and named Lactobacillus plantarum L-3, L. sakei L-10, and L. curvatus strains L-8, L-11, and L-12. Morphological characteristics, physiological data, carbohydrate fermentation patterns, and 16S rRNA sequence data, were all used to characterize the bacterial isolates. L. plantarum L-3 showed the highest lactic acid productivity of all isolates, but grew only poony in the presence of 10% (v/v) alcohol at pH 4.0. The other strains exhibited lower lactic acid productivity than did L. plantarum L-3 and did not grow in the presence of 10% (v/v) alcohol at pH 4.0. The optimal temperature and pH for lactic acid production were $30^{\circ}C$ and pH 6.0 7.0, respectively. The lactic acid productivity of L. plantarum L-3, L. sakei L-10 and the three L. curvatus strains L-8, L-11, and L-12 were (% v/v of culture supematant) 1.55, 1.0, 1.06, 1.0, and 0.99, respectively, at $30^{\circ}C$ and pH 6.0. While L. plantarum L-3 suffered growth inhibition in the presence of 10% (v/v) alcohol, growth of the other strains was inhibited at 8% (v/v) alcohol.

Maize which has very high omega-6 fatty acid content has been used as a main feed grain for Hanwoo beef production to increase marbling, and thus omega-6 to omega-3 fatty acids ratio in Hanwoo beef is expected to be biased. To elucidate the current status of omega fatty acids ratio in Hanwoo beef, fatty acid profiles of neutral lipid and phospholipid fraction were analyzed separately using 55 Hanwoo steers' longissimus dorsi muscle slaughtered at Pyeongchang, Korea from Oct. to Nov. 2015. In addition, an association study was conducted to evaluate associations between single nucleotide polymorphism (SNP) markers from references and omega fatty acid profiles in phospholipid of Hanwoo beef samples using analysis of variance (ANOVA). In neutral lipid fraction, composition of saturated and monounsaturated fatty acids was higher and polyunsaturated fatty acids was lower compared to those in phospholipid fraction. The mean n-6/n-3 ratios of Hanwoo were $56.059{\pm}16.180$ and $26.811{\pm}6.668$ in phospholipid and neutral lipid, respectively. There were three SNPs showing statistically significant associations with omega fatty acid content. GA type of rs41919985 in fatty acid synthase (FASN) was significantly associated with the highest amount of C20:5 n-3 (p = 0.031). CC type of rs41729173 in fatty acid-binding protein 4 (FABP4) was significantly associated with the lowest amount of C22:2n-6 (p = 0.047). AG type of rs42187261 in FADS1 was significantly linked to the lowest concentration of C20:4 n-6 (p = 0.044). The total n-6/n-3 ratio of the steer which has all four SNP types in above loci (27.905) was much lower than the mean value of the total n-6/n-3 ratio in phospholipid of the 55 Hanwoo steers ($56.059{\pm}16.180$). It was found that phospholipid and neutral lipid of Hanwoo have very high n-6/n-3 ratios compared to the reported data from different cow breeds. Four SNPs in genes related with fatty acid metabolism showed significant associations with the fatty acid profile of phospholipid and may have potential as SNP markers to select Hanwoo steers in terms of n-6/n-3 balance in the future.

Objective: Leukocyte cell-derived chemotaxin 2 (LECT2) is associated with several physiological processes including inflammation, tumorigenesis, and natural killer T cell generation. Chicken LECT2 (chLECT2) gene was originally identified as one of the differentially expressed genes in chicken kidney tissue, where the chickens were fed with different calcium doses. In this study, the molecular characteristics and gene expression of chLECT2 were analyzed under the stimulation of toll-like receptor 3 (TLR3) ligand to understand the involvement of chLECT2 expression in chicken metabolic disorders. Methods: Amino acid sequence of LECT2 proteins from various species including fowl, fish, and mammal were retrieved from the Ensembl database and subjected to Insilco analyses. In addition, the time- and dose-dependent expression of chLECT2 was examined in DF-1 cells which were stimulated with polyinosinic:polycytidylic acid (poly [I:C]), a TLR3 ligand. Further, to explore the transcription factors required for the transcription of chLECT2, DF-1 cells were treated with poly (I:C) in the presence or absence of the nuclear factor ${\kappa}B$ ($NF{\kappa}B$) and activated protein 1 (AP-1) inhibitors. Results: The amino acid sequence prediction of chLECT2 protein revealed that along with duck LECT2 (duLECT2), it has unique signal peptide different from other vertebrate orthologs, and only chLECT2 and duLECT2 have an additional 157 and 161 amino acids on their carboxyl terminus, respectively. Phylogenetic analysis suggested that chLECT2 is evolved from a common ancestor along with the actinopterygii hence, more closely related than to the mammals. Our quantitative polymerase chain reaction results showed that, the expression of chLECT2 was up-regulated significantly in DF-1 cells under the stimulation of poly (I:C) (p<0.05). However, in the presence of $NF{\kappa}B$ or AP-1 inhibitors, the expression of chLECT2 is suppressed suggesting that both $NF{\kappa}B$ and AP-1 transcription factors are required for the induction of chLECT2 expression. Conclusion: The present results suggest that chLECT2 gene might be a target gene of TLR3 signaling. For the future, the expression pattern or molecular mechanism of chLECT2 under stimulation of other innate immune receptors shall be studied. The protein function of chLECT2 will be more clearly understood if further investigation about the mechanism of LECT2 in TLR pathways is conducted.

This research was conducted to improve and establish Bokto direct seeding technology use of silicate in rice. Bokto direct seeding technology has recently released to alternative new method for transplanting method of the rice plant. In recent rice production has serious problem due to higher cost and labor shortage in Korea. In particular rice has been faced on open market to internationally due to WTO, FTA agreement etc. Thus rice growers in Korea have been more concentrated on cost reduction using by direct seeding technology such as dry seeding and water seeding while those methods have severe constraints like poor seedling, more weed occurrence, lodging and poor grain quality. In recent a new technology for improvement of these problem has released to farmer's field and proven to alternative rice growing technology of transplanting. In this technology there was additional application of silicate which has known as soil improvement and this research was obtained in the following results. At the application of silicate, sand, and soil there was high in seedling establishment by 90~100% in pot experiment but there was 55.0% seedling stand of the untreated control. Under dry condition seedling stand was 0% at the untreated control while 65~100% high in application of silicate, sand, and soil. Meanwhile, under rainy condition untreated control there was 85% of seedling stand but soil application was 20% of seedling establishment due to blocking of oxygen and physical soil blockage.

Drought tolerant species from 26 Korean native plants were selected using different physiological indicators. Arundinella hirta, Solanum carolinense and Carpesium divaricatum were withered after 8days of the stopping of irrigation. Plants except Kummerowia striata, Lespedeza cuneata and Ulmus parvifolia were withered in over 80% at 9-10days of the irrigation stopping. K. striata was withered after 10days, and L. cuneata and U. parvifolia were withered in over 90% after 11days of the stopping of irrigation. As stopping experiment of irrigation, A. hirta, S. carolinense, C. divaricatum, K. striata, L. cuneata and U. parvifolia were proved to be drought tolerant species. Among those plant species, transpiration rate of Cassia mimosoides var. nomame Makino was high as 0.042ml/㎠·4hr. However, unit transpiration rate of U. parvifolia and L. cuneata were 0.005 and 0.010ml/㎠·4hr, respectively. In testing of physiological indicators, leaf area and transpiration rate were different among plant species. Unit transpiration rate of U. parvifolia was lower compared with other plant species. L. cuneata, U. parvifolia, Kummerowia striata, Arundinella hirta and C. divaricatum were high in relative water content and low in relative water loss. As this results, L. cuneata and U. parvifolia. were identified as drought tolerant species.
Objective: The objective of this study was to identify candidate genes that play important roles in skeletal muscle development in ducks. Methods: In this study, we investigated the transcriptional sequencing of embryonic pectoral muscles from two specialized lines: Liancheng white ducks (female) and Cherry valley ducks (male) hybrid Line A (LCA) and Line C (LCC) ducks. In addition, prediction of target genes for the differentially expressed mRNAs was conducted and the enriched gene ontology (GO) terms and Kyoto encyclopedia of genes and genomes signaling pathways were further analyzed. Finally, a protein-to-protein interaction network was analyzed by using the target genes to gain insights into their potential functional association. Results: A total of 1,428 differentially expressed genes (DEGs) with 762 being up-regulated genes and 666 being down-regulated genes in pectoral muscle of LCA and LCC ducks identified by RNA-seq (p<0.05). Meanwhile, 23 GO terms in the down-regulated genes and 75 GO terms in up-regulated genes were significantly enriched (p<0.05). Furthermore, the top 5 most enriched pathways were ECM-receptor interaction, fatty acid degradation, pyruvate degradation, PPAR signaling pathway, and glycolysis/gluconeogenesis. Finally, the candidate genes including integrin b3 (Itgb3), pyruvate kinase M1/2 (Pkm), insulin-like growth factor 1 (Igf1), glucose-6-phosphate isomerase (Gpi), GABA type A receptor-associated protein-like 1 (Gabarapl1), and thyroid hormone receptor beta (Thrb) showed the most expression difference, and then were selected to verification by quantitative real-time polymerase chain reaction (qRT-PCR). The result of qRT-PCR was consistent with that of transcriptome sequencing. Conclusion: This study provided information of molecular mechanisms underlying the developmental differences in skeletal muscles between specialized duck lines.
In order to extend shelf life for rockfish fillets by modified atmosphere packaging (MAP), different package atmospheres were compared in the product quality preservation. Firstly, CO2 solubility was measured at 0, 5, 10, and 15℃ to be incorporated into the mathematical model to predict the volume and CO2 concentration of the package at expected storage temperature. The CO2 solubility given in Henry's constant decreased with temperature to be fitted with a linear equation. Then air packaging as control and four MAP conditions of 100 g fillets were prepared and stored for duration of 5 days at 10℃ to compare them in the quality preservation effect. Four MAP conditions employed were CO2(60):O2(30):N2(10), CO2(60):O2(0):N2(40), CO2(30):O2(30):N2(40) andCO2(30):O2(0):N2(70). MAP conditions with high CO2 concentration inhibited total aerobic bacteria, and the conditions containing O2 led to low TVB-N. MAP of CO2(60):O2(30):N2(10) was found to be the best condition for rockfish fillet preservation considering total aerobic bacteria growth and nitrogenous volatiles production.
Sorghum has been consumed as one of the important staple food in the semiarid tropics of Africa and Asia. Sorghum is rich in starch, protein, essential vitamins and minerals and grows relatively well in dry climate regions when it compared with other staple food crops. Sorghum has taken an increased interest due to several studies that report about the beneficial effects of sorghum on human health. In the present study, we investigated the antioxidative and activity of extract of milling by-products (hull and bran) of Korean sorghum cultivar, 'Hwanggeaumchal' as well as its grain. Hull extract showed the highest total polyphenol contents ($29.7{\pm}0.2mg\;GAE/100g$) and major four pigments content ($322.6{\pm}14.5mg/100g$). From results of 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonate (ABTS) radical scavenging activity, hull extract ($IC_{50}$, $6.3{\pm}0.1{\mu}g/mL$) was also showed the strongest antioxidative effects. Bran and grain showed similar polyphenol, pigments contents and antioxidative effects. We determined cell viability by MTT assay and evaluated the anti-inflammatory activity by measuring nitric oxide (NO) of hull, bran and grain methanol extract (0.5% HCl v/v) on RAW 264.7 cells. Hull extract treatment was significantly decreased NO production with dose-dependant manner. Apigeninidin as one of the major pigment of hull was showed inhibitory activity against NO production without cytotoxicitiy. Therefore, sorghum milling by-products can be used as a good source of antioxidative and anti-inflammatory agents.

The effects of different feeding times, using a diet containing 800 ppm betaine, on production, blood profile, and the short-chain fatty acid (SCFA), was investigated using 240 Cherry Valley (Anas platyrhynchos) meat ducks exposed to a scorching heat wave. The animals were randomly assigned to four groups, each of which was replicated three times with twenty ducks per replicate. The experimental period was 42 days for each group. Four groups were assigned into C (heat wave control group without betaine), T1 (ad libitum group fed a diet containing 800 ppm betaine), T2 (diet-restricted group fed twice daily between 05:00-10:00 and 17:00-20:00, using a diet containing 800 ppm betaine), and T3 (night-restricted group, fed from 17:00-10:00, with a diet containing 800 ppm betaine). At 42 days, body weight increased in order of T2, T1, T3 compared to the heat wave control group although. However, these differences were not found between the groups of T1 and T3. The heat wave control group, provided an ad libitum diet without betaine (C), showed an elevated feed conversion ratio compared to the groups fed a diet containing betaine. However, these differences were not found between the groups of T1, T2 and T3. RBC and platelet profiles except for PLT and MPV showed statistically significant differences between study groups fed a diet containing betaine. T2 presented significantly higher blood electrolytes $Na^+$ and $Cl^-$ than the other groups. T2 also showed a blood gas level that was generally higher than the other groups. Total SCFA, acetic acid and propionic acid concentration has been the increasing trend in T2, but butyric acid, isobutyric acid and valeric acid concentration has been the decreasing trend in T2 compared to the other groups. It is concluded that the feeding-restricted group, fed two times daily between 05:00-10:00 and 17:00-20:00, with a diet containing 800 ppm betaine may improve growth performance in meat ducks exposed to a scorching heat wave.

Transparent exopolymer particles (TEP) are formed by aggregation of polysaccharide products excreted by phytoplankton and have sticky character like gel. They play important role in the production of marine snow in water column. To study the distribution pattern of TEP concentration and its role in carbon cycle in the surface ocean, we measured pH, Total alkalinity (TA), and chlorophyll-a in addition to physical characteristics of seawater within the surface water column. TEP concentrations ranged from nearly undetectable values to $338{\mu}g\;Xeq\;l^{-1}$. They were considerably lower than previously reported values from costal sites, but showed similar values observed in other oceanic region during phytoplankton bloom periods. The spatial distribution of TEP concentrations were similar to those of chlorophyll-a, which indicate that the production of TEP were closely related to phytoplankton. Calculated total dissolved inorganic carbon ($TCO_2$) from the pH and TA was normalized to 35 psu of salinity ($NTCO_2$) and showed negative linear relationship with temperature. Biological drawdown of $NTCO_2$ ($NTCO_{2bio}$) was estimated from the difference between theoretical $NTCO_2$ values and observed $NTCO_2$. In the warm region located south of $40^{\circ}N$ along the $132.5^{\circ}N$ meridional lines, $NTCO_{2bio}$ showed negative value and TEP concentrations were high. This suggested that negative $NTCO_{2bio}$ may be attributed to the biological processes. At the stations located between 44 and $46^{\circ}N$, TEP concentrations showed high concentration at the chlorophyll-a maximum layer within the water column while they showed low concentration in the surface layer. Carbon content of TEP constituted about 40% of $NTCO_{2bio}$ at the chlorophylla maximum layer. In this study, we could not observe any positive and negative relationship between TEP concentration and $NTCO_2$ or pH. It is obvious that we should consider the importance of TEP in the biological carbon cycling processes within surface layer.
