Effect of Selected Inoculant Applications on Chemical Compositions and Fermentation Characteristics of High Moisture Rye Silage

  • The aim of this study was to investigate the effect of isolated lactic acid bacteria (LAB) on the quality of high moisture rye silage. Rye forage (Secale cereale L.) was harvested at the heading stage (27.3% of dry matter (DM)) and cut into approximately 3-5 cm lengths. Then, the forage divided into 4 treatments with different inoculants: 1) No additives (CON); 2) Lactobacillus brevis strain 100D8 at a 1.2 × 105 colony-forming unit (cfu)/g of fresh forage (LBR); 3) Leuconostoc holzapfelii strain 5H4 at a 1.0 × 105 cfu/g of fresh forage (LHO); and 4) Mixture of LBR and LHO (1:1 ratio) applied at a 1.0 × 105 cfu/g of fresh forage (MIX). About 3 kg of forage from each treatment was ensiled into a 20 L mini-bucket silo in quadruplicate for 100 days. After silo opening, silage was collected for analyses of chemical compositions, in vitro nutrient digestibilities, fermentation characteristics, and microbial enumerations. The CON silage had the highest concentrations of neutral detergent fiber and acid detergent fiber (p = 0.006; p = 0.008) and a lowest in vitro DM digestibility (p < 0.001). The pH was highest in CON silage, while lowest in LBR and MIX silages (p < 0.001). The concentrations of ammonia-N, lactate, and acetate were highest in LBR silage (p = 0.008; p < 0.001; p < 0.001). Propionate and butyrate concentrations were highest in CON silage (p = 0.004; p < 0.001). The LAB and yeast counts were higher in CON and LHO silages compare to LBR and MIX silages (p < 0.001). However, the mold did not detect in all treatments. Therefore, this study could conclude that L. brevis 100D8 and Leu. holzapfelii strain 5H4 can improve the digestibility and anti-fungal activity of high moisture rye silage.

Iron sulfate and molasses treated anthocyanin-rich black cane silage improves growth performance, rumen fermentation, antioxidant status, and meat tenderness in goats

  • Objective: This study investigated the effects of feeding anthocyanin-rich black cane treated with ferrous sulfate and molasses on animal performance, rumen fermentation, microbial composition, blood biochemical indices, and carcass characteristics in meat goats. Methods: Thirty-two Thai-native×Anglo-Nubian crossbred male goats (14.47±2.3 kg) were divided equally into two groups (n = 16) to investigate the effect of feeding diet containing 50% untreated anthocyanin-rich black cane silage (BS) vs diet containing anthocyaninrich black cane silage treated with 0.03% ferrous sulfate and 4% molasses (TBS) on average daily gain (ADG) and dry matter intake (DMI). At the end of 90 d feeding trial, the goats were slaughtered to determine blood biochemical indices, rumen fermentation, microbial composition, and carcass characteristics differences between the two dietary groups. Results: Goats fed the TBS diet had greater ADG and ADG to DMI ratio (p<0.05). TBS diet did not affect rumen fluid pH; however, goats in the TBS group had lower rumen ammonia N levels (p<0.05) and higher total volatile fatty acid concentrations (p<0.05). Goats in the TBS group had a higher (p<0.05) concentration of Ruminococcus albus but a lower (p<0.05) concentration of methanogenic bacteria. The TBS diet also resulted in lower (p<0.05) thiobarbituric acid-reactive substances concentration but higher (p<0.05) total antioxidant capacity, superoxide dismutase, catalase, glutathione peroxidase, and glutathione reductase concentrations in blood plasma, while having no effect on plasma protein, glucose, lipid, immunoglobin G, alanine transaminase, and aspartate aminotransferase. Meat from goats fed the TBS diet contained more intramuscular fat (p<0.05) and was more tender (p<0.05). Conclusion: In comparison to goats fed a diet containing 50% untreated anthocyanin-rich black cane silage, feeding a diet containing 50% anthocyanin-rich black cane silage treated with 0.03% ferrous sulfate and 4% molasses improved rumen fermentation and reduced oxidative stress, resulting in higher growth and more tender meat.

Fatty acids composition and lipolysis of Parmigiano Reggiano PDO cheese: effect of the milk cooling temperature at the farm

  • Objective: The aim was to study the influence of cooling milk at 9℃ at the farm versus keeping it at 20℃ on Parmigiano Reggiano cheese lipolysis. Methods: A total of six cheesemaking trials (3 in winter and 3 in summer) were performed. In each trial, milk was divided continuously into two identical aliquots, one of which was kept at 9℃ (MC9) and the other at 20℃ (MC20). For each trial and milk temperature, vat milk (V-milk) and the resulting 21 month ripened cheese were analysed. Results: Fat and dry matter and fat/casein ratio were lower in MC9 V-milk (p≤0.05) than in MC20. Total bacteria, mesophilic lactic acid and psychrotrophic and lipolytic bacteria showed significant differences (p≤0.05) between the two V-milks. Regarding cheese, fat content resulted lower and crude protein higher (p≤0.05) both in outer (OZ) and in inner zone (IZ) of the MC9 cheese wheels. Concerning total fatty acids, the MC9 OZ had a lower concentration of butyric, capric (p≤0.05) and medium chain fatty acids (p≤0.05), while the MC9 IZ had lower content of butyric (p≤0.05), caproic (p≤0.01) and short chain fatty acids (p≤0.05). The levels of short chain and medium chain free fatty acids (p≤0.05) were lower and that of long chain fatty acids (p≤0.05) was higher in MC9 OZ cheese. The principal component analysis of total and free fatty acids resulted in a clear separation among samples by seasons, whereas slight differences were observed between the two different milk temperatures. Conclusion: Storing milk at 9℃ at the herd affects the chemical composition of Parmigiano Reggiano, with repercussion on lipolysis. However, the changes are not very relevant, and since the cheese can present a high variability among the different cheese factories, such changes should be considered within the "normal variations" of Parmigiano Reggiano chemical characteristics.

Antifungal and carboxylesterase-producing bacteria applied into corn silage still affected the fermented total mixed ration

  • Objective: This study investigated the effects of corn silage as a source of microbial inoculant containing antifungal and carboxylesterase-producing bacteria on fermentation, aerobic stability, and nutrient digestibility of fermented total mixed ration (FTMR) with different energy levels. Methods: Corn silage was used as a bacterial source by ensiling for 72 d with an inoculant mixture of Lactobacillus brevis 5M2 and L. buchneri 6M1 at a 1:1 ratio. The corn silage without or with inoculant (CON vs MIX) was mixed with the other ingredients to formulate for low and high energy diets (LOW vs HIGH) for Hanwoo steers. All diets were ensiled into 20 L mini silo (5 kg) for 40 d in quadruplicate. Results: The MIX diets had lower (p<0.05) acid detergent fiber with higher (p<0.05) in vitro digestibilities of dry matter and neutral detergent fiber compared to the CON diets. In terms of fermentation characteristics, the MIX diets had higher (p<0.05) acetate than the CON diets. The MIX diets had extended (p<0.05) lactic acid bacteria growth at 4 to 7 d of aerobic exposure and showed lower (p<0.05) yeast growth at 7 d of aerobic exposure than the CON diets. In terms of rumen fermentation, the MIX diets had higher (p<0.05) total fermentable fraction and total volatile fatty acid, with lower (p<0.05) pH than those of CON diets. The interaction (p = 0.036) between inoculant and diet level was only found in the immediately fermentable fraction, which inoculant was only effective on LOW diets. Conclusion: Application of corn silage with inoculant on FTMR presented an antifungal effect by inhibiting yeast at aerobic exposure and a carboxylesterase effect by improving nutrient digestibility. It also indicated that fermented feedstuffs could be used as microbial source for FTMR. Generally, the interaction between inoculant and diet level had less effect on this FTMR study.

Effects of Italian ryegrass silage-based total mixed ration on rumen fermentation, growth performance, blood metabolites, and bacterial communities of growing Hanwoo heifers

  • This study utilized Italian ryegrass silage (IRGS) - based total mixed ration (TMR) as feedstuff and evaluated its effects on rumen fermentation, growth performance, blood parameters, and bacterial community in growing Hanwoo heifers. Twenty-seven Hanwoo heifers (body weight [BW], 225.11 ± 10.57 kg) were randomly allocated to three experimental diets. Heifers were fed 1 of 3 treatments as follows: TMR with oat, timothy, and alfalfa hay (CON), TMR with 19% of IRGS (L-IRGS), and TMR with 36% of IRGS (H-IRGS). Feeding high levels of IRGS (H-IRGS) and CON TMR to heifers resulted in a greater molar proportion of propionate in the rumen. The impact of different TMR diets on the BW, average daily gain, dry matter intake, and feed conversion ratio of Hanwoo heifers during the growing period did not differ (p > 0.05). Furthermore, the blood metabolites, total protein, albumin, aspartate aminotransferase, glucose, and total cholesterol of the heifers were not affected by the different TMR diets (p > 0.05). In terms of rumen bacterial community composition, 264 operational taxonomic units (OTUs) were observed across the three TMR diets with 240, 239, and 220 OTUs in CON, L-IRGS, and H-IRGS, respectively. IRGS-based diets increased the relative abundances of genera belonging to phylum Bacteroidetes but decreased the abundances of genus belonging to phylum Firmicutes compared with the control. Data showed that Bacteroidetes was the most dominant phylum, while Prevotella ruminicola was the dominant species across the three TMR groups. The relative abundance of Ruminococcus bromii in the rumen increased in heifers fed with high inclusion of IRGS in the TMR (H-IRGS TMR). The relative abundance of R. bromii in the rumen significantly increased when heifers were fed H-IRGS TMR while P. ruminicola increased in both L-IRGS and H-IRGS TMR groups. Results from the current study demonstrate that the inclusion of IRGS in the TMR is comparable with the TMR containing high-quality forage (CON). Thus, a high level of IRGS can be used as a replacement forage ingredient in TMR feeding and had a beneficial effect of possibly modulating the rumen bacterial community toward mainly propionate-producing microorganisms.

항생제 대체제로서 생균제가 이유자돈의 성장능력 및 영양소 이용률, 설사 빈도, 면역 반응에 미치는 영향 (Evaluation of Probiotics as an Alternative to Antibiotic on Growth Performance, Nutrient Digestibility, Occurrence of Diarrhea and Immune Response in Weaning Pigs)

  • 본 실험은 이유자돈 120두를 이용하여 생균제의 첨가가 이유자돈의 성장 능력, 영양소 소화율, 혈중 요소태 질소, 설사 빈도, 면역 반응에 미치는 영향에 대해 조사하였다. 이유자돈 사료에 항생제 대체제인 생균제의 첨가는 전 사양 기간에 걸쳐 사양 성적이 항생제 첨가구(PC)에 비해 유의적으로 낮게 나타났다. 항생제 첨가구(PC)를 제외하고 비교하였을 때에는 비록 유의적인 차이는 발생하지 않았지만 생균제 첨가구가 항생제 무첨가구(NC)에 비해 일당증체량, 사료효율에서 높은 경향을 보였으며, 이는 항생제 첨가 효과에는 미치지 못하지만 항생제 대체제로서 그 가능성을 보인 것이라고 하겠다. 특히 생균제 A 첨가구는 0~5 주차 사료 효율(P<0.05)에서 항생제 첨가구(PC)와 비슷한 성적을 나타내어, 항생제 대체제로서 이유자돈의 성장을 개선 시킬 가능성이 가장 높다고 하겠다. 본 실험에서는 5주동안 이유자돈의 사료에 생균제를 첨가하여 사양성적을 보았는데, 생균제의 급여기간을 좀 더 늘리면 생균제의 처리효과가 더욱 뚜렷하게 나타날 수도 있으리라 생각된다. 이유자돈 사료에 생균제의 첨가가 건물, 조단백질, 조지방, 조회분 소화율에 어떠한 영향도 미치지 않았으며(P>0.10), 질소 축적율은 생균제 첨가구에서 항생제 첨가구(PC)에 비해 높게 나타났으나 일관된 효과를 보이지는 않았다. 혈중 요소태 질소 농도에서는 2주차와 4주차에서 유의적인 차이가 나타났다(P<0.05). 또한 생균제 B 첨가구가 다른 처리구에 비해 혈중 요소태 질소 농도가 가장 낮게 나타났다. 설사 발생 빈도 측정 결과, 항생제 첨가구(PC)에서 설사 발생 빈도가 유의적으로 낮게 나타났으며, 생균제 C 첨가구에서 다음으로 낮게 나타났다(P=0.18). 면역 반응 실험 결과, $CD^{4+}$ T-cell 수의 경우 생균제 C 첨가구에서 다른 처리구에 비해 가장 낮게 나타났으며(P<0.01), $CD^{8+}$ T-cell 수와 $CD^{4+}:CD^{8+}$ 비율의 경우에는 모든 처리구에서 유의적인 차이가 발생하지 않았다(P>0.10). 결론적으로 이유자돈사료 내 생균제의 첨가가 이유자돈의 성장 능력, 질소축적율, 혈중 요소태 질소, 설사 빈도, 및 면역 능력에 일정한 개선 효과를 갖고 있지만, 항생제 첨가 효과에는 미치지 못한다고 하겠다. 본 실험을 통해 이유자돈 사료에 생균제의 첨가는 항생제 대체제로서 일정 부분 개선효과가 있는 것으로 사료된다.

에너지 및 단백질 공급 수준에 따른 비유 중기 유산양의 유생산성에 미치는 영향 (Effects of Different Dietary Energy and Protein Levels on Milk Production in Saanen Dairy Goats in Mid Lactation)

  • 본 시험은 산양유 생산을 위해 유기사료를 이용하여 유산양의 적정 에너지 및 단백질 공급 수준과 조농비율을 결정하기 위하여 비유중기 Saanen종 유산양 4두($57.5{\pm}10.8\;kg$ BW)를 이용하여 $4{\times}4$ 라틴방각법으로 실시하였다. 대사에너지 공급수준과 단백질 공급수준을 10.59 (T1), 10.14 (T2), 9.45 (T3) 및 8.75 (T4) MJ/kg과 11.90 (T1), 12.73 (T2), 15.19 (T3) 및 16.60% (T4)로 달리하였으며, 조농비율은 49:51 (T1), 59:41 (T2), 74:26 (T3) 및 90:10 (T4)의 네 처리구로 나누었다. 사료 섭취량 및 체중은 모든 처리구 사이의 유의한 차이가 없었다. 유량은 T3구에서 가장 높았으며, T2 및 T4구와 비교하여 유의하게 높게 나타났다 (p<0.05). 유지방함량과 생산량은 T4구에서 다른 처리구와 비교하여 가장 낮았다 (p<0.05). 유단백질과 유당 생산량은 T1 (61.29 및 70.95 g/일)과 T3 (60.52 및 75.82 g/일)구에서 T2 (50.49 및 59.64 g/일)와 T4 (52.49 및 59.82 g/일)구와 비교하여 유의하게 높게 나타났다. 결과적으로 유산양의 유생산성을 극대화시키기 위해서 에너지 공급수준은 최소 9 MJ/kg 이상과 단백질 공급수준은 최소 15% 이상을 공급해 주어야할 것으로 기대된다.

전분박을 이용한 돈분발효퇴비의 시용이 감자의 생육과 수량에 미치는 영향 (Effects of Pig Manure Composting Using Starch Pulp Treating on Growth and Yield Characteristics of Potato Cropping)

  • 농산폐기물인 전분박을 톱밥대용 부자재로 사용하여 제조된 전분박을 이용한 퇴비를 감자재배에 적용시켜 지상부생육 및 괴경수량에 미치는 영향을 조사하였다. 전분박 발효퇴비는 돈분과 톱밥 및 탈수 전분박(수분 65%내외)을 일정한 비율로 배합하여 퇴적송풍식으로 조제된 것으로 전분박 발효퇴비의 처리내용은 대조구(퇴비 무비구), 1 : 1 : 0 처리구(돈분 : 톱밥 : 전분박의 혼합비), 1 : 0,75 : 0.25 처리구 및 1 : 0.5 : 0.5 처리구 4수준으로 수행하였다. 초장은 생육초기에는 톱밥혼합 돈분발효퇴비와 전분박혼합 돈분발효퇴비가 화학비료 단용처리구에 비해 컸으나, 생육중반이후에는 차이를 보이지 않았으며, 경직경 및 지상부건물중은 퇴비시용구가 무비구에 비해 양호한 경향을 보였으나 통계적 유의성은 없었다. 총괄괴경수는 1 : 0.5 : 0.5 처리구가 가장 많았으며, 1 : 1 : 0 처리구, 1 : 0.75 : 0.25 처리구는 비슷하였다. 80g 미만의 괴경수량은 각 처리간에 일정한 경향을 보이지 않았으나 81-120g 및 120g이상의 괴경수량은 무비구에 비해 퇴비처리구가 현저히 높았다. 상서율도 전분박 1 : 0.75 : 0.25 및 1 : 0.5 : 0.5 처리구에서 86%내외로 높게 나타났다. 더뎅이병 이병율 및 발병도는 전분박이 많이 혼합된 1 : 0.5 : 0.5 처리구에서 높은 경향을 보였으나 통계적인 유의성은 없었다. 그러나 앞으로 전분박을 흔합하여 돈분발효퇴비를 제조했을 때 나타날 수 있는 문제점으로 더뎅이병 이병을 및 발병도를 가장 고려해야 될 것으로 생각된다. 엽중 N, P, K, Ca 등의 함량은 무처리에 비해 퇴비처리구가 많았으며 처리구 간에는 통계적으로 유의성 있는 차이가 없었다. 결과적으로 전분박을 혼합하여 제조한 퇴비가 감자 생육에 미치는 영향은 톱밥혼합퇴비에 비해 긍정적인 영향을 미치는 것으로 보인다.

Effects of Organic or Inorganic Acid Supplementation on Growth Performance, Nutrient Digestibility and White Blood Cell Counts in Weanling Pigs

  • Four experiments were conducted to investigate the effect of organic or inorganic acid supplementation on the growth performance, nutrient digestibility, intestinal measurements and white blood cell counts of weanling pigs. In growth trial (Exp I), a total of 100 crossbred pigs ({$Landrace{\times}Yorkshire$}${\times}$Duroc), weaned at $23{\pm}2$ days of age and $7.25{\pm}0.10kg$ average initial body weight (BW), were allotted to 5 treatments by body weight and sex in a randomized complete block (RCB) design. Three different organic acids (fumaric [FUA], formic [FOA] or lactic acid [LAA]) and one inorganic acid (hydrochloric acid [SHA]) were supplemented to each treatment diet. Each treatment had 5 replicates with 4 pigs per pen. During 0-3 wk, average daily gain (ADG), average daily feed intake (ADFI) and feed efficiency (G/F ratio) were not significantly different among treatments. However, pigs fed LAA or SHA diet showed improved ADG by 15 or 13% respectively and 12% greater ADFI in both treatments compared to CON diets. Moreover, compared to organic acid treatments, better ADG (p = 0.07) and ADFI (p = 0.09) were observed in SHA diet compared to pigs that were fed the diet containing organic acids (FUA, FOA or LAA). However, during 4-5 wk, no differences in ADG, ADFI and G/F ratio were observed among treatments. Overall, ADG, ADFI and G/F ratio were not affected by acidifier supplementation. Although it showed no significant difference, pigs fed LAA or SHA diets showed numerically higher ADG and ADFI than pigs fed other treatments. In metabolic trial (Exp II), 15 pigs were used to evaluate the effect of acidifier supplementation on nutrient digestibility. The digestibility of dry matter (DM), crude protein (CP), crude fat (CF), crude ash (CA), calcium (Ca) and phosphorus (P) was not improved by acidifier supplementation. Although the amount of fecal-N excretion was not different among treatments, that of urinary-N excretion was reduced in acidsupplemented treatments compared to CON group (p = 0.12). Subsequently, N retention was improved in acid-supplemented groups (p = 0.17). In anatomical trial (Exp III), the pH and $Cl^-$ concentrations of digesta in gastrointestinal (GI) tracts were not affected by acidifier supplementation. No detrimental effect of intestinal and lingual (taste bud) morphology was observed by acidifier supplementation particularly in inorganic acid treatment. In white blood cell assay (Exp IV), 45 pigs were used for measuring white blood cell (WBC) counts. In all pigs after LPS injection, WBC counts had slightly declined at 2 h and kept elevating at 8 h, then returned to baseline by 24 h after injection of lipopolysaccharide (LPS). However, overall WBC counts were not affected by acidifier supplementation. In conclusion, there was no difference between organic and inorganic acidifier supplementation in weanling pigs' diet, however inorganic acidifier might have a beneficial effect on growth performance and N utilization with lower supplementation levels. Furthermore, inorganic acidifier had no negative effect on intestinal measurements and white blood cell counts in weanling pigs. These results suggested that inorganic acidifier might be a good alternative to organic acidifiers in weanling pigs.
