Effects of 3-methylcholanthrene on the Expression of Drug Metabolizing Enzyme Genes in Monkey Liver (원숭이 간 약물대사효소 유전자 발현에 미치는 3-methylcholanthrene 영향)

  • In order to understand the mechanism of the regulation of drug metabolizing enzyme gene expression, we have studied the induction of CYP1A1 and GST$\alpha$, $\mu$, $\pi$ enzymes in Japanese monkey and rhesus monkey after the treatment with 3-methylcholanthrene (3MC) and di-n- butyl phthalate (DBP) and bisphenol A (BPA). The levels of mRNA were measured by RT-PCR in brain, intestine and liver. In the case of adult monkey, treatment with 3MC induced CYP1A1 mRNA in liver by 10-fold. The treatment with DBP induced CYP1A1 mRNA. Effects of 3MC and DBP on GST mRNA expression was not clear. But GST$\mu$ was slightly inhibited by the treatment with 3MC and DBP. GST$\pi$ was not induced by the treatment with 3MC and DBP in liver. GST$\alpha$ was slightly induced by the treatment with 3MC and DBP in liver. In the case of fetus monkey, the basal levels of fetus CYP1A1 mRNA and GSTs mRNA were relatively low compared to adult monkey. As the age of monkey increased, the basal levels of CYP1A1 mRNA were also increased. 3MC induced the expression of CYP1A1 mRNA in liver. The levels of GST$\mu$ and GST$\alpha$ were not changed by the treatment with 3MC and DBP. GST$\pi$ was slightly induced by the treatment with 3MC and DBP.

The Effect to Drug Metabolizing Enzyme Cytochrome P450 3A4 by Chungyulyak (청열약 수종의 Cytochrome P450 3A4 효소활성도에 미치는 영향)

  • In this study, the author experimented the influence of five herbal medicines, which are Lonicera japonica Thunb, Paeonia suffruticosa Andr., Fraxinus rhynchophylla Hance, Gardenia jasminoides Ellis, Scutellaria baicalensis George which are called 'Chungyulyak(淸熱藥)' on drug metabolizing enzyme cytochrome P450 3A4 in Human Liver Microsome. Above all, the reason for this study is that herbal medicines can be assumed that herbs might have interactions with drugs, other herbs, alcohol and chemicals whether those are much better synergy effects than expected effects when the medicine was treated alone or not. As a result, we showed that all of five traditional herbal medicines had no CYP 3A4 inhibition effect on 10, 20, 30, 40, $50{\mu}g/m{\ell}$ doses in Human Liver Microsome. However, this result are mostly not enough to prove that PMT has a CYP 3A4 inhibition effect. Moreover, it is not that those rates showed that those herbal medicines have CYP 3A4 induction effect. In conclusion, the result could support that those herbal medicines are more safe than chemical drugs even if this is the basic step to prove that result. Therefore, more specific studies to support this result, which are Kinetic study, cell and animal study then finally until clinical research, are required.

Activity Changes in Phase II Drug-metabolizing Enzymes UDP-Glucoronosyl Transferase and Glutathione S-Ttansferase to Crude Oil Exposure in Mussel and Rockfish (원유의 노출이 담치와 조피볼락의 phase II 약물대사효소 UDP-glucoronosyl transferase 및 glutathione S-transferase의 활성에 미치는 영향)

  • This study examined effects of crude oil on the phase II drug-metabolizing enzymes UDP-glucuronosyl transferase (UDPGT) and glutathione S-transferase (GST) in mussel Mytilus edulis and rockfish Sebastes schlegeli, a representative bivalve and a culture fish, respectively. This work also intended indirectly to evaluate the post impact recovery from the massive oil tanker spillage accidents occurred during the summer of 1995 in the sea area off Yosu City, Chonnam. For these, enzyme activities of UDPGT and GST were examined in the fish and mussel following laboratory exposure to fresh crude oil, weathered oil, field-obtained oil residues, or in the field biota samples. Decreased GST activity was observed in rock fish following exposure to oil-soluble fraction (OSF) of fresh oil. A similar diminished GST activity was also observed after OSF of artificially weathered oil. OSF of field oil residues retrieved from the spillage area approximately 1 year later also exerted a slight inhibition of GST to rockfish. There was neither a change in UDPGT in rockfish, nor were there changes in mussel in both enzymes to any oil fractions. We could not observe any difference in the two enzymes either in rockfish or mussel sampled from the field during $1.5{\sim}2.0$ years post spillage, indicating that their enzyme systems might had been recovered by the sampling time. In conclusion, it seems that the inhibition of GST activity in rockfish is a biomarker response to crude oil exposure. The results, however, must be interpreted with care, as the inhibition nay reflect various factors such as oil concentration, duration and water temperature.

Regulatory Mechanism of Cytochrome P450IIE in the Rat with Hepatic Injury and Ketosis (간장장해와 Ketosis시에 Cytochrome P450IIE의 Regulatory Mechanism에 관한 연구)

  • In order to elucidate the alteration of drug-metabolizing enzymes and mechanism in the animal with hepatic injury and ketosis, the regulation of P450IIE was studied in the rats with heaptic injury caused by CCl$_4$ and with ketosis caused by streptozotocin and high-fat diet. P450IIE expression in liver was examined by the combination of enzyme activities, Western immunoblot, and mRNA Northern blot analyses using specific polyclonal antibody and cDNA probe for P450IIE. Enzyme activity and amounts of immunoreactive P450IIE were rapidly decreased in a time-dependent manner after a single dose of CCl$_4$ . However, the decreases in P450IIE enzyme activity and immunoreactive protein by CCl$_4$ were not accompanied by a decline in its mRNA level. The data thus suggested a post-translational reduction of P450IIE by CCl$_4$. The enzyme activities (aniline hydroxylase) in hepatic microsomes were elevated about 2-3-fold by streptozotocin and feeding with a high fat diet. This increases in enzyme activities were also accompanied by 3-fold increases in immunoreactive P450IIE protein and its mRNA. Our data thus indicated that P450IIE induction during the ketosis appears to be due to pretranslational activation.

Effects of Yuldahanso-tang and Chungsimyonja-tang on Cytochrome P450 Activities (열다한소탕과 청심연자탕의 Cytochrome P450 활성 연구)

  • Objectives : The purpose of this study is to investigate the inhibitory or inductive potentials of Yuldahanso-tang (YDT) and Chungsimyonja-tang (CST), herbal formulas for Taeeumin, on cytochrome P450 (CYP450) drug metabolizing enzyme. The mechanisms for the herbal formula-drug interaction has not been well reported in spite of the chance for co-administration with conventional drugs. Methods : To evaluate the interaction potential of YDT-drug or CST-drug, the fluorescence-based enzyme assays on CYP450 isozymes including CYP3A4, CYP2C19, CYP2D6 and CYP2E1 were established in vitro. The inhibitory effects of herbal formulas were characterized with $IC_{50}$ values. Results : YDT showed inhibitory effects on CYP2D6 and CYP2E1-mediated metabolism, while it exhibited week inhibition on CYP3A4 and CYP2C19 relatively. CST exerted relatively weak inhibitory effects on the four CYP450 isozymes compared to that of YDT. Conclusions : These results suggest that the herbal formula-drug interaction could be occur when YDT are co-administered with drugs mediated by CYP2D6 or CYP2E1.

Effect of Spices on hepatic microsomal enzyme function in mice

  • The effect of twenty two spices on liver microsomal monooxygense activity was tested as measured by alteration of hexobarbital (HB) narcosis and strychnine mortality in mice. Oral administration of seven spices for 7 consecutive days caused a significant shortening of the duration of HB-induced sleeping time. The treatment of mice with a single i. p. injection of 9 spices resulted in a significant prolongation of the sleeping time. White pepper, dill and fennel reduced the toxicity of strychnine. These results strongly indicated that some spices might affect the activity of liver microsomal drug metabolizing enzyme (DME) function.

Effects of Hydroxylated Flavonoids on the Ethoxyresorufin O-deethylase and Benzo($\alpha$)pyrene Hydroxylase

  • In order to understand the mechanism of action of flavonoids on the drug metabolizing enzyme, cytochrome P450IA1, this study was undertaken to examine the effect of chrysin, morin, myricetin and aminopyrine on the activities of ethoxyresorufin O-deethylase and benzo(.alpha.) pyrene hydroxylase in the liver. In the isolated perfused rat liver that was pretreated with 3-methylcholanthrene (3MC), chrysin, morin, myricetin and aminopyrine inhibited the activity of ethoxyresorufin O-deethylase with concentration dependent manner. The isolated liver perfusion with chrysin, morin, myricetin and aminopyrine showed inhibition on the induction of ethoxyresorufin O- deethylase by 3MC. And also, in mouse liver hepa I cells, 3MC-stimulated the benzo(.alpha.)pyrene hydroxylase activity which was inhibited by chrysin, morin, myricetin and aminopyrine. These results strongly suggested that hydoxylated flavonoids interfered not only the induction of cytochrome P45OIA1 enzymes by 3MC but also the interaction of substrates and enzyme.

  • A triterpenoid(melianone) and a limonoid(28-deacetyl sendanin) were isolated from the chloroform fraction of Meliae toosendan Fructus, the rippen fruits of Melia toosendan SIEB. et ZUCC.(Meliaceae) and were applied to serial experiments to clarify the liver protective activities. We found that the compounds promoted slightly the drug metabolizing enzyme activities and decreased serum transaminase activities which was elevated by carbon tetrachloride intoxication. And also they slightly increased the secretion of bile juice in rat.

Isolation of Hepatic Drug Metabolism Inhibitors from the Rhizomes of Curcuma zedoaria

  • The methanolic extract of the Rhizome of Curcuma zedoaria exhibited a significant prolongation of hexobarbital (HB)-induced hypnosis. Through liquid chromatography of an ether soluble fraction. monitoring by bioassay, three sequiterpenes, germacrone (A), curzerenone (B) and germacrone epoxide (C) were isolated as active consituents. A single treatment (100-200 mg/kg, i.p.) of each compound showed not only a significant prolongation of HB-induced sleeping time but also a significant inhibition of aminopyrine N-demethylase activity in mice, and further exhibited a typical type I binding spectra with oxidized rat hepatic cytochrome P-450 induced by phenobarbital. All of the compounds provoked a sleep episode at a subhypnotic dose of HB, implying that they possess CNS depressant properties.

  • Meliae toosendan Fructus is the fruit of Melia toosendan $S_{IEB}$. et $Z_{UCC}$. (Meliaceae), which is written in oriental terminology as clearing heat and drying dampness, and also explained using liver, stomach and small intestine for channels entered. Among the five fractions prepared from methanol whole extractive of the herb, the chloroform fraction which suggests the presence of triterpenoid, flavonoid and alkaloid stimulated the activities of drug metabolizing enzymes and bile secretion and lowered the serum transaminase activities of liver damaged by carbon tetrachloride.
