  • Pharmaceutical technology has primarily focused on the development of the best dosage forms depending on the route of administration. The design of dosage forms is greatly influenced by the route of administration. Due to a variety of advantages such as avoidance of first-pass effect, abundant blood supply and easy access to the absorption site, the oral cavity has frequently been selected as a site for drug delivery. Since the oral cavity is relatively unique from the anatomical and physiological viewpoint, one should always consider these conditions when designing the drug delivery systems for the oral cavity. In this regard, the current review paper was prepared to summarize the essential features of the drug delivery systems utilized in the oral cavity, along with the introduction of various dosage forms developed to date.

  • The polymeric matrices made with poly(D,L-lactide-co-glycolide) were prepared using copolymer of poly(D,L-lactide) and poly(ethylene glycol) for application of drug delivery systems. Catalyst made use of stannous actoate. Particle size were differ greatly$(435.3{\pm}11.2{\sim}2284.1{\pm}188.5)$ that nanoparticle made use of according to solvent of various kinds. Polymer could a sharp distinction with copolymerized among LE-1, LE-2 and LE-3 of PLA and PEG of content that to examine $^1H-NMR$ of copolymer make refine and reprecipitation. Drug delivery effect at PLGA nanoparticle : PLA amount more then proved highly drug delivery amount that each LE-1, LE-2, LE-3, drug and solvent was 40mg, 20mg and 10mg. Drug delivery effect proved higher 20mg that change(10mg, 20mg, 40mg) at drug feeding amount with LE-2. The first a lot of drug proved delivery. LE-3 most lactide content proved much delivery since biodegradable on PLGA copolymer result from lactide. Also biodegradable rate was highest at LE-3 much of lactide content, because influence at biodegradable effect of lactide by inclusive of soft PEG.

  • In many biodegradable polymers recently investigated, poly(lactic acid)(PLA) or poly(lactic-co-glycolic acid)(PLGA) have extensively been utilized as drug delivery systems for sustained release drug delivery. Recently, there has been increased interest in electrospinning, which can produce fibers that are sub-micron in diameter. Effect of pH on Swelling Property of Hyaluronic Acid Hydrogels for Smart Drug Delivery Systems

  • Hyaluronic acid(HA) hydrogels were synthesized by immersing HA microbeads in phosphate buffered saline solutions having different pH levels to assess the effect of pH on the swelling ratio of HA hydrogels for smart drug delivery systems. No beads were formed when the HA solution(below pH 9) was crosslinked with divinyl sulfone(DVS) because DVS is a basic solution. The variation regarding the size of the microbead was not significant, suggesting that the bead size is not a function of pH(10 ~ 14). However, the pore size of the microbeads decreased with increasing pH from 10 to 14, leading to the surface smoothness and dense network as a result of higher crosslinking. The swelling ratio of hydrogels increased when the pH rose from 2(acidic) to 6(neutral). Afterwards, it decreased with further increasing pH(basic). The lower swelling ratio may be due to the lack of ionization of the carboxyl groups. On the other hand, a higher swelling ratio is likely due to the increased electrostatic repulsions between negatively charged carboxyl groups on different chains. Experimental results suggested that pH-responsive HA hydrogels can be applicable to the controlled drug delivery systems.

Doxorubicin-loaded PEI-silica Nanoparticles for Cancer Therapy

  • Targeted anticancer drug delivery systems are needed to enhance therapeutic efficacy by selectively delivering drugs to tumor cells while minimizing off-target effects, improving treatment outcomes and reducing toxicity. In this study, a silica-based nanocarrier capable of targeting drug delivery to cancer cells was developed. First, silica nanoparticles were synthesized by the Stöber method using the surfactant cetyltrimethylammonium bromide (CTAB). Increasing the ratio of EtOH in the solvent produced uniformly spherical silica nanoparticles. Washing the nanoparticles removed unreacted residues, resulting in a non-toxic carrier for drug delivery in cells. Upon surface modification, the pH-responsive polymer, polyethyleneimine (PEI) exhibited slow doxorubicin release at pH 7.4 and accelerated release at pH 5.5. By exploiting this feature, we developed a system capable of targeted drug release in the acidic tumor microenvironment.

  • Many nanomaterials are being harnessed as critical components in various systems for biomedical applications including diagnosis, imaging, and drug delivery. Those systems necessitate biocompatibility and low toxcity within effective dose range while achieving enough efficacy. Even though many nanomaterials enjoy successful demonstrations in bioapplications, lack of biocompatibility and high cytotoxicity often become hurdles for practical bioapplications. On the other hand, it is important to achieve enough efficiency based on chemically well-defined systems with efforts to understand mechanism at molecular level. Here, we developvarious biocompatible nanomaterials based on simple procedure using dextran as both reducing agent and surface coating. Dextran is one of the popular biocompatible polymers that have been used for drug delivery and biosensors. Dextran coated nanomaterials showed excellent colloidal stability, flexible surface chemistry for conjugation of bioactive molecules and low cytotoxicity with successful demonstrations in various bioapplications.

  • The therapy of injecting a fixed amount of a prescribed drug for a predetermined time is an effective treatment in relieving pain during anticancer treatments. Due to recent medical technology development, cancer is currently classified as a disease that can be managed in the patient's lifetime. If patients were able to use a drug delivery system that was portable, sustainable and had an accurate flow control, they would be able to inject medication whenever they need. In this study we developed a piezoelectric micropump for a drug delivery system by designing a pump chamber, check valve and diaphragm. We also developed a driving circuit that consumes low power and to which we applied a variety of signals. We fabricated a portable drug delivery system with this piezoelectric micropump and driving circuit. In addition, through a performance test, we confirmed that the system can precisely control the drug flow rate.

  • The Rapid Prototyping (RP) technology has advanced in many application areas. In this research, two different types, cylinder and scaffold, of implantable Drug Delivery System (DDS) were fabricated using Nano Composite Deposition System (NCDS), one of the RP systems. The anti-cancer drug (5-fluorouracil, 5-FU), biodegradable polymer (PLGA(85: 15)), and bio ceramic (Hydroxyapatite, HA) were used to form drug-polymer composite material. Both types of DDS were evaluated in vivo environment for two weeks. For evaluation, the cumulative drug release and shape stability were measured. Test results showed that the scaffold DDS provide higher cumulative drug release and has better stability than cylinder DDS.

  • Apoptosis is a natural cell suicide mechanism to maintain homeostasis. However, many of the diseases encountered today are caused by aberrant apoptosis where excessive apoptosis leads to neurodegenerative disorders, ischemic heart disease, autoimmune disorders, infectious diseases, etc. A variety of antiapoptotic agents have been reported to interfere with the apoptosis pathway. These agents can be potential drug candidates for the treatment or prevention of diseases caused by dysregulated apoptosis. Obviously, world-wide pharmaceutical and biotechnology companies are gearing up to develop antiapoptotic drugs with some products being commercially available. Polymeric drug delivery systems are essential to their success. Recent R&D efforts have focused on the chemical or bioconjugation of antiapoptotic proteins with the protein transduction domain (PTD) for higher cellular uptake with antibodies for specific targeting as well as with polymers to enhance the protein stability and prolonged effect with success observed both in vivo and in vitro. All these different fusion antiapoptotic proteins provide promising results for the treatment of dysregulated apoptosis diseases.
