Some pharmacological studies with Cycleanine, a diphenylbisbenzylisoquinoline alkaloid from Stephania hernandifolia

  • Stephania hernandifolia belonging to the family Menispermaceae is the biggest storehouse of diphenylbisbenzylisoquinoline (DBBI) alkaloids. Exhaustive chemical processing of the bulb of S. hernandifolia by the application of modern separation techniques yielded a DBBI alkaloid which was identified as cycleanine using spectroscopic methods (UV, IR, $^1HNMR$. $^{13}CNMR$, Mass). Cycleanine showed significant anti-inflammatory activity against carrageenin induced paw oedema, comparable to that produced by diclofenac sodium, used as standard drug. It exhibited potent analgesic effects against chemical and thermal noxious stimuli. It was also found to possess anticonvulsive activity in the strychnine induced convulsion model.

  • Pharmacological activities of Paeoniae Radix alba were studied with water extract (F-I), parched preparation (F-II) and 25% ethanol-treated preparation (F-III) of it. The results were summerized as follows; 1) Analgesic actions were recognized with F-I, F-II and F-III. 2) Sedative actions were recognized with F-I and F-III. 3) Prolonged effect of sleeping time were recognized significantly with F-I and F-III. 4) Antipyretic actions were recognized with F-1, F-II and F-III in rabbits. 5) F-I, F-II and F-III showed the antagonic action against contraction of isolated-ileum induced by acetylcholine, barium chloride and histamine in mice and guinea-pigs.

  • Micro injection molding analysis for microneedle fabrication was performed in the present study. The dimensions of width and thickness for microneedle are 600um, 500um, respectively. A delivery system based on guidelines for traditional injection molding was designed for four-cavities molding system. To investigate the effects of processing conditions in the mirconeedle fabrication, injection molding analysis using commercial code was performed. It was shown that the total injection time has a significant effect on the fabrication of microneedles.

  • The prolonged mechanical grinding process may enhance the bioavailability of the drugs due to the change of solid state such as micronization and decrease of crystallinity. A series of attempts to enhance the bioavailability of insoluble drugs have been made by the fine grinding technique using a planetary mill. The objective of the present study is to investigate the possibility of improving the dissolution properties of poorly water- soluble drugs such as diphenyl hydrantoin (phenytoin) and diphenyl dimethyl dicarboxylate (DDB) based on the molecular interaction between drug and additives during pharmaceutical processing to be related with the bioavailability behavior.   • Regarding chemical changes in oriental drugs after food processing such as roasting, fermentation, and extrusion, fifty commonly-used medicinal plants were investigated. As a result, Aurantii nobilis Pericarpium (a tangerine peel from Citrus unshu Markovich) showed remarkably different HPLC profiles after being roasted. An increased peak was isolated by repeated chromatography and identified as 5-hydroxymethyl furfral (5-HMF) by means of instrumental analyses. The 5-HMF content of Aurantii nobilis Pericarpoum reached its maximum level after being roasted for 30 min at 225$^{\circ}C$ (49.2 mg/g extract, ca 42 times of increase over untreated control). Although there were no significant changes in in vitro biological activity such as antioxidative, anti-dementia, anti-hypertension, anti-coagulation, or cytotoxicity, before and after roasting process, our results suggested that simple heat treatment might improve the value of the above oriental drug since 5-HMF has been known to possess inhibitory activities toward nitric oxide formation, tyrosinase, and sickling of red blood cells.

A Study on Attitudes Toward Food Safety Issues in Korea - Focus on the Public Official Related to Food Hygiene - (식품안전성에 대한 기본인식 조사 - 식품위생 관련 공무원을 중심으로 -)

  • A survey was conducted to investigate the attitudes of public officials related to food hygiene toward food safety. The official groups were divided into four sub-groups based on their area, agency, gender and age. Response of each group to the survey was statistically analysed. The survey's results showed that most of them (87.0%) were satisfied with a domestic level of food hygiene. But, some of then (29.8%) considered unsatisfactoryly on safety of the food they eat. Their response to food safety did not statistically significant between each group of area, agency and age but, in case of gender, female group showed stastically more negative attitude to food safety than male one (P<0.05). All groups chose residues of chemical substances such as pesticides and food additives as the most potential food risk factor followed by foodborne pathogens, heavy metals and animal drug residues. The results are not consistent with the scientific judgement. Therefore, more education and information were needed fro these groups. They pointed out food manufacturer as a responsible group for poor food hygiene (48.7%). In addition, food manufacture and processing were selected as main business types with the lowest level in the food hygiene but official, working in the area of the central government and Seoul metropolitan city, pointed out food services establishments as the poorest hygiene one (P<0.01). This results suggested that education and information to let mind of responded groups change, working in this part, and governmental financial support are needed to improve hygiene level of food manufacture and processing (70.3%). They also chose HACCP as the most effective way for improving the level of food hygiene followed by Recall, PL (Product Liability), monitoring, labeling and increasing of number of company with good manufacture and processing (GMP).

A Method to Recover 2D barcodes Contaminated with Dust (2D 바코드의 분진 오염 극복 방법)

  • Food printers must use food ink cartridges approved by the Ministry of Food and Drug Safety (MFDS). A 2D bar code is used to read whether the ink cartridge is authentic. However, since the dye is diverged by heat pressure and printed, the barcode is contaminated. In this paper, we propose a pre-processing algorithm to solve the problem of barcode contamination by food coloring dust in a latte art printer. The algorithm is based on various morphological operations. We apply this algorithm before reading contaminated barcode images with a general QR code reader. It has been confirmed that, as compared with the existing QR code reader, the contamination rate that can be perceived is increased from 25% to 40% and even at a contamination rate of 45%, the recognition rate reaches 50%.

  • Posttraumatic stress disorder (PTSD) is well known to have a limited response to drug treatment. Many recently published clinical care guidelines recommend trauma-focused psychotherapies such as cognitive processing therapy (CPT) and prolonged exposure therapy (PE) as first-line treatment and medication such as serotonin reuptake inhibitors and venlafaxine as second-line treatment. Current review introduces the session composition and contents of CPT and presents various CPT studies that show therapeutic effect for civilian and veterans/military with PTSD. In order for clinicians to help effectively patients with PTSD, it is necessary to learn and actively use evidence-based trauma-focused psychotherapies including CPT and PE.

The Effects of steam heat processing of Helianthus tuberosi Rhizoma on Blood glucose lowering (국우(菊芋) 증자가 혈당강하작용에 미치는 영향)

  • Objective : This study was designed to evaluate the hypoglycemic effects of Helianthus tuberosi Rhizoma extracts and its optimum Heat processing conditions Methods : We investigated the Salivary ${\alpha}$-amylase, pancreas ${\alpha}$-amylase and ${\alpha}$-glucosidase inhibitory activities of extracts from Steam Heated Helianthus tuberosi Rhizoma Ext. The inhibitory activities of a 50% EtOH extract of Steam Heated Helianthus tuberosi Rhizoma Ext against ${\alpha}$-glucosidases were evaluated in this study. Inhibiting these enzymes involved in the absorption of disaccharides significantly decreases the postprandial increase in blood glucose level after a mixed carbohydrate diet. Furthermore, the postprandial blood glucose lowering effect of Steam Heated Helianthus tuberosi Rhizoma Ext. was compared to a known type 2 diabetes drug(Acarbose(R)) in a mice model. Steam Heated Helianthus tuberosus L. Ext significantly reduced the blood glucose increase after glucose loading. Results : The results were confirmed by real-time PCR that after treated with Streptozotocin in L6 cells, induced expression of GLUT4, after the steamed Helianthus tuberosus L. Ext. treated, observed its expression was increased. Steam Heated Helianthus tuberosus L Ext treated 4 hours in L6 cells, cytotoxicity was measured in MTT assay. Its toxicity were 5.7%, 9% and 11.3% at the treatment concentration $12.5{\mu}g/m{\ell}$, $25{\mu}g/m{\ell}$, the $50{\mu}g/m{\ell}$ respectively. Conclusions : Overall, the results of this study indicate that Hypoglycemic effect of Helianthus tuberosi Rhizoma caused by the Steam heat treatment, the optimum Heat processing condition is steamming at $121^{\circ}C$ for 30 min, and it will provide the basis for developing a useful dietary supplement for controlling postprandial hyperglycemia.

Effect of Processing Treatment on Physicochemical Characteristics of Brown Rice Varieties with Different Amylose Content (아밀로오스 함량이 다른 현미의 품종별 가공 처리에 따른 품질 특성 비교)

  • The quality characteristics of brown rice polished with grains of four rice varieties (Hwaseonchal, Baegjinju, Ilpum and Goami) were examined. For the physical properties of native and fried brown rice starches, the gelatinization temperatures and enthalpies (determined with differential scanning calorimetry) of brown rice starches decreased with increasing amylose content. The pasting viscosities of native brown rice starches in distilled water were higher than those in $HgCl_2$ solution (as an ${\alpha}$-amylase inhibitor). The highest viscosity of gelatinized starch was observed for the starch of Ilpum rice variety relative to other varieties. Cystal packing arrangement of brown rice starches subjected to deep-fat frying converted A-type (typically observed for cereal starches) into V-type (formed by retrogradation and recrystllization of amylose molecules).
