Flexural and Impact Resisting Performance of HPFRCCs Using Hybrid PVA Fibers (하이브리드 PVA 섬유를 이용한 HPFRCCs의 휨 및 충격 성능 평가)

  • HPFRCCs (high-performance fiber reinforced cementitious composites), which is relatively more ductile and has the characteristic of high toughness with high fiber volume fractions, can be used in structures subjected to extreme loads and exposed to durability problems. In the case of PVA (polyvinyl alcohol) fiber, it is noted by former studies that around 2% fiber volume fractions contributes to the most effective performance at HPFRCCs. In this study, flexural tests were carried out to evaluate the flexural behavior of HPFRCCs and to optimize mix proportions. Two sets of hybrid fiber reinforced high performance specimens with total fiber volume fraction of 2 % were tested: the first set prepared by addition of short and long PVA fibers at different combination of fiber volume fractions, and the second set by addition of steel. In addition, in order to assess the performances of the HPFRCCs against to high strain rates, drop weight tests were conducted. Lastly, the sprayed FRP was applied on the bottom surface of specimens to compare their impact responses with non-reinforcing specimens. The experimental results showed that the specimen prepared with 1.6% short fibers (REC 15) and 0.4% long fiber (RF4000) outperformed the other specimens under flexure, and impact loading.

Preimplantation Genetic Diagnosis for Single Gene Disorders (단일 유전자 질환에 대한 착상전 유전진단)

  • Preimplantation genetic diagnosis (PGD) has become an assisted reproductive technique for couples who are at risk that enables them to have unaffected baby without facing the risk of pregnancy termination after invasive prenatal diagnosis. The molecular biology and technology for single-cell genetics has reached an extremely high level of accuracy, and has enabled the possibility of performing multiple diagnoses on one cell using whole genome amplification. These technological advances have contributed to the avoidance of misdiagnosis in PGD for single gene disorders. Polymerase chain reaction (PCR)-based PGD will lead to a significant increase in the number of disorders diagnosed and will find more widespread use, benefiting many more couples who are at risk of transmitting an inherited disease to their baby. In this review, we will focus on the molecular biological techniques that are currently in use in the most advanced centers for PGD for single gene disorders, including biopsy procedure, multiplex PCR and post-PCR diagnostic methods, and multiple displacement amplification (MDA) and the problems in the single cell genetic analysis.

Estimation of the relationship between below-ground root and above-ground canopy development by measuring dynamic change of soil ammonium-N concentration in rice

  • In the early part of rice growth, root volume primarily limits the amount of plant-accessible nitrogen (N). Therefore, knowledge of the root development is important for modeling N uptake of rice. The timing when the volume of rhizosphere cover the whole soil is also important to carry out timely top dressing. However, information about initial root expansion and associated N uptake is limited due to intrinsic technical difficulties in assessing below-ground processes. Some studies, however, showed a close relationship between below-ground root and above-ground leaf development, suggesting a possibility that above-ground attributes could serve as surrogates for the root processes. In this study, we investigated the relationship between below-ground and above-ground development of rice. Field experiments were conducted where we cultivated Koshihikari (a leading cultivar in Japan) for four different cropping schedules in 2012. In 2016, Gimbozu (HEG4) and three flowering time mutant lines of Gimbozu (X61 (se13), HS276 (ef7), DMG9 (se13, ef7)) were examined for a single season. Experiments were performed with three replications in a completely randomized design. We monitored ammonium-N concentration ([NH4+-N]) in soil solution by repeatedly taking samples from a porous tubing (10-cm long) vertically inserted at the most distant point from surrounding rice hills. Samples were taken in triplicate (= triplicate tubes) and every three days from transplanting in each experimental unit. For above-ground attributes, leaf area index (LAI) was measured in 2012, whereas soil coverage ratio was estimated by image processing in 2016. Results showed that [NH4+-N] increased gradually after transplanting and then rapidly decreased from a certain day. This distinct drop in [NH4+-N] informed us the timing at which the rice root system reached the point of porous tubing and thus essentially covered the whole soil volume. The LAI at the dropping point was about 0.43 regardless of the cropping schedules in 2012 experiment. In 2016, the coverage ratio at the N dropping point was within the range of 0.12 to 0.19 for four genotypes having different growth durations. In addition, the coverage ratios at seven weeks after the transplanting showed a good correspondence to LAI across the four genotypes. We therefore conclude that both LAI and coverage ratio may serve as robust indicators for root development and might be useful to estimate the timing when the root system fully cover the soil volume. Results obtained here will also contribute to develop models that can predict not only above-ground canopy development but also associated below-ground processes.

  • Current driving capability of MCT (MOS Controlled Thyristor) is determined by turn-off capability of conducting current, that is off-FET performance of MCT. On the other hand, having a good turn-on characteristics, including high peak anode current ($I_{peak}$) and rate of change of current (di/dt), is essential for pulsed power system which is one of major application field of MCTs. To satisfy above two requirements, careful control of on/off-FET performance is required. However, triple diffusion and several oxidation processes change surface doping profile and make it hard to control threshold voltage ($V_{th}$) of on/off-FET. In this paper, we have demonstrated the effect of $V_{th}$ adjustment ion implantation on the performance of MCT. The fabricated MCTs (active area = $0.465mm^2$) show forward voltage drop ($V_F$) of 1.25 V at $100A/cm^2$ and Ipeak of 290 A and di/dt of $5.8kA/{\mu}s$ at $V_A=800V$. While these characteristics are unaltered by $V_{th}$ adjustment ion implantation, the turn-off gate voltage is reduced from -3.5 V to -1.6 V for conducting current of $100A/cm^2$ when the $V_{th}$ adjustment ion implantation is carried out. This demonstrates that the current driving capability is enhanced without degradation of forward conduction and turn-on switching characteristics.

Study on the Applicability of Reflection Method using Ultrasonic Sweep Source for the Inspection of Tunnel Lining Structure - Physical Modeling Approach - (터널 지보구조 진단을 위한 초음파 스윕 발생원의 반사법 응용 가능성 연구 - 모형실험을 중심으로 -)

  • Reflection method using ultrasonic source has been attempted to obtain the information about tunnel lining structures composed of lining, shotcrete, water barrier and voids at the back of lining. In this work, two different types of sources, i.e. single-pulse source and sweep source, can be used. Single-pulse source with short time duration has the frequency content whose amplitudes tend to be concentrated around the dominant frequency, whereas sweep source with long time duration denotes a flat distribution of relatively larger amplitude over a broad frequency band, although the peak to peak amplitude of single-pulse source wavelet is equivalent to that of sweep source one. In traditional seismic application, a single-pulse source(weight drop, dynamite) is typically used. However, to investigate the fine structure, as it is the case in the tunnel lining structure, the sweep wavelet can be also a desirable source waveform primarily due to the higher energy over a broad frequency band. For the investigation purposes of sweep source, a physical modeling is a useful tool, especially to study problems of wave propagation in the fine layered media. The main purpose of this work was using a physical modeling technique to explore the applicability of sweep source to the delineation of inner layer boundaries. To this end, a two-dimensional physical model analogous to the lining structure was built and a special ultrasonic sweep source was devised. The measurements were carried out in the sweep frequency range 10 ∼ 60 KHz, as peformed in the regular reflection survey(e.g. roll-along technique). The measured data were further rearranged with a proper software (cross-correlation). The resulting seismograms(raw data) showed quitely similar features to those from a single-pulse source, in which high frequency content of reflection events could be considerably emphasized, as expected. The data were further processed by using a regular data processing system "FOCUS" and the results(stack section) were well associated with the known model structure. In this context, it is worthy to note that in view of measuring condition the sweep source would be applied to benefit the penetration of high frequency energy into the media and to enhance the resolution of reflection events.

Effect of Work and Urea-Molasses Cake Supplementation on Live Weight and Milk Yield of Murrah Buffalo Cows

  • Two experiments were carried out to investigate the effect of Murrah buffalo cows pulling sledges under field conditions on milk production and milk composition. In Exp. 1, 24 buffaloes in the fourth month of lactation were used. They were allotted to four treatments according to a $2{\times}2$ factorial arrangement: work or no work, and with or without urea-molasses cake supplementation (700 g/animal/day). Feeds consisted of 20 kg fresh elephant grass (18% DM), 2 kg rice bran per day and rice straw ad lib. The animals worked in pairs three hours per day (work done: $3464{\pm}786kJ/d$) five days a week for three months. Three teams worked in the morning and the others worked in the afternoon in the same day. The following day the working times were switched. In Exp. 2, 16 lactating Murrah buffalo cows in the sixth month of lactation were allotted to two groups (work and no work). They were fed with fresh ruzi grass (Brachiaria ruziziensis) ad lib. supplemented with 2 kg rice bran and 700 g urea-molasses cake. The working regime was similar to that of the first experiment (work done: $3753{\pm}879kJ/d$) and they worked for two months. In the first experiment, there was a small but significant drop (p<0.05) in milk yield from 3.5 to 3.0 kg/day due to work, but there was no supplementation effect. The working buffaloes lost 5.2 kg whereas the non-working animals gained 9.7 kg during the three months (p<0.05). Supplementation increased live weight by 9.9 kg as compared to -5.4 kg for those not supplemented (p<0.05). Milk composition was not affected by the treatments. In the second experiment, daily milk production was similar for both treatments and approximately 3 kg. No significant differences were found in milk composition or in live weight changes for working and non-working groups, respectively. It was concluded that work may cause a reduction in milk yield and a loss of live weight on a poor rice straw diet but that an appropriate supplementation can alleviate this situation.

A Study on the Reliability Improvement of Compartment Leak Test in Surface Vessels (함정 격실기밀 평가 방안에 대한 신뢰성 향상 연구)

  • Generally, surface vessels have many compartments for operation and living quarters, and each compartment is an important space for the ship's survivability. During ship construction, a compartment leak test is necessary and is carried out on each vessel. However, the current test method is in doubt when looking at the actual test results. The reason is that only one pressure gauge is used for the measurement to check the air, so an uncomprehended phenomenon is detected during group compartment leak tests. From this point of view, an improved test device and method are needed. In this study, a multi-channel data acquisition device with multiple pressure sensors is proposed to detect each compartment's pressure variation or pressure drop. This test is a more confidential compartment leak test than the current method, and the test device can show real-time pressure detection values of each of the pressure sensors, which are installed in each compartment, including unmanned space.

Studies about the bioactive component analysis and an oral glucose tolerance test of Add-Omit-Saenghyeoryunbu-eum(AO-SHU) for confirmation of diabetes therapy (가감생혈윤부음(加減生血潤膚飮)의 당뇨병 치료효과 확인을 위한 생리활성성분 분석과 경구포도당부하 연구)

  • Objectives : Instrumental chemical analysis was utilized to investigate the effect of Add-Omit-Saenghyeoryunbu-eum(AO-SHU) on diabetic treatment. One of the most exciting, yet also controversial, arguments is the safety and biological mechanisms of the natural medicine on human body. Therefore, the aim of this study is to provide a better understanding on bioactive chemical components, hazards of heavy metal contamination and biological mechanism of the diabetic medicine composed of 12 different natural herbs. Methods : To study bioactive compound and metallic component in the diabetic medicine in detail, LC-MS/MS (Liquid Chromatography-Mass/Mass), GC (Gas Chromatography) and ICP (Inductively Coupled Plasma) were utilized to characterize the extract of the diabetic medicine and the result was compared with 18 marker substances selected from literature survey. In addition, in vitro assay experiments including GPR 119 activity and human DGAT-1 inhibition, and OGTT (Oral Glucose Tolerance Test) were performed to verify the effectiveness of this medicine on diabetic treatment. Results : Out of 18 marker substances, 9 bioactive compounds were identified from LC-MS/MS analysis which include Citruline, Catalpol, Berberine, Ginsenoside Rb1, Ginsenoside Rg1, Oleanolic acid, β-Sitosterol, Mangiferin, and Schizandrin. ICP study on 245 residual pesticides revealed that 239 species were not detected but 6 species, Dimethomorph, Trifloxystrobin, Pyraclostrobin, Isoprocarb, Carbaryl and Flubendiamide, while the amounts are trace levels, below permitted concentrations. The biological activity was observed in vitro assay and Oral Glucose Tolerance Test(OGTT), which are consistent with a preliminary clinical test result, a drop in blood sugar level after taking this herbal medicine. Conclusions : Instrumental chemical analysis using LC-MS/MS, GC, and ICP was conducted successfully to identify bioactive compounds in AO-SHU for the treatment of diabetes, finding 9 bioactive compounds. Furthermore, in vitro assay experiments and OGTT show that AO-SHU has its biological activities, which imply that it can be a candidate for the future diabetes remedy.

The Installation Effect and Optimal Pipe Sizes of an Anti-Wind Net by Computational Analysis (전산 해석에 의한 파풍망의 설치 효과와 최적 파이프 규격)

  • This study was carried out to(1) visualize the installation effect of an anti-wind net; (2) evaluate structural stability of typical anti-wind nets in Jeju; and (3) present the optimal specification of pipes in an anti-wind net for maximum instant wind velocities of 40 m/s and 45 m/s. The analyses were done for anti-wind nets with a mesh of 4 mm and a height of 3 m by using CFX and ANSYS. The results showed that the wind went down due to flow resistance when passing through an. anti-wind net. The anti-wind net with the supporting pipe being installed every two main columns was certainly unstable because the main column not sustained by the supporting pipe became cantilever. With regard to the position of a fixing point of the supporting pipe, von Mises stress on pipes was certainly increased as vertical positions of the supporting pipe were changed to be too lower or higher than an adequate position but there was little difference according to horizontal positions. The adequate vertical position was $2{\sim}2.5\;m$ high from the ground. For a maximum instant wind velocity of 40 m/s, the optimal specification of pipes was a main column of ${\varphi}48.1{\times}2.1$ t@2,000, cross beams(bottom and top) of ${\varphi}26.7{\times}1.9\;t$, cross beams(center) of ${\varphi}33.5{\times}2.1$ t/2ea and a supporting pipe of ${\varphi}31.8{\times}1.5$ t@2,000. In case of a maximum instant wind velocity of 45 m/s, the optimal specification of pipes with structural stability was a main column of ${\varphi}48.6{\times}3.25$ t@2,000, cross beams(bottom and top) of ${\varphi}26.7{\times}1.9\;t$, cross beams(center) of ${\varphi}48.1{\times}2.1$ t/2ea and a supporting pipe of ${\varphi}31.8{\times}1.5$ t@2,000.

The Effects of Jingansikpung-tang, derived from Anal Therapy on CHT-induced Brian Edema and Neurological Disturbance Symptom in Rats (항장요법(肛腸療法)에의한 진간식풍탕(鎭肝熄風湯)이 CHT로 유발된 백서(白鼠)의 뇌부종 및 신경 장애에 미치는 영향)

  • Objectives: We identified the effects of Jingansikpung-tang, (JGSPT), derived from therapy on closed head trama(CHT)-induced brain edema and neurologial disturbance symptom in rats. Methods : We used a drop device and induced CHT. We divided the animals into 3 groups. They were JGSPT 1 group (Administered JGSPT 96.5mg/kr through anal far one hour after using CHT) and JGSPT 2 group(Administered JGSPT 386mg/kg through anal for one hour after using CHT) Then, we tested neurological severity score(NSS), water content of brain and haematological changes. Result : aNSS increased significantly in JGSPT 1 and JGSPT 2 as compared to the controlled group, and the change of water content which was pulled out of the right hemisphere from the left hemisphere decreased in JGSPT 1 and JGSPT 2 as compared to the controlled group. The lactate level in serum decreased in JGSPT 1 and JGSPT 2 as compared to the controlled group, which showed no efficacy. Glucose level in serum increased in JGSPT 1 and JGSPT 2 as compared to the controlled group, only JGSPT 2 showed no efficacy. Conclusions : JGSPT, caused by Anal Therapy and effect on CHT-induced brain edema and neurological symptom.
