A Case of Spondylodiscitis with Spinal Epidural Abscess Due to Brucella

  • Brucellosis, a zoonosis with worldwide distribution, is a systemic infection caused by facultative intracellular bacteria of the genus Brucella, which can involve multiple organs and tissues. We report an uncommon case of spondylodiscitis with epidural abscess due to Brucella in a male stockbreeder. Diagnosis was based on clinical history, and supported by Brucella serology and magnetic resonance imaging. Clinical and radiological improvement were observed with a combined antimicrobial therapy of doxycycline, rifampicin, and gentamycin.

  • Recently we performed video-assisted thoracoscopic[VAT] examination and bullectomy under local anesthesia. Of the 10 patients undergoing VAT examination under local anesthesia with primary spontaneous pneumothorax, 8 patients underwent VAT bullectomy under local anesthesia using endo-GIA; 7 patients discharged within 24 hours after operation; 1 patient had an air leak after operation, so chemical pleurodesis with doxycycline was performed and discharged postoperative day 3. There have been no recurrence to date[60-120 days after operation]. We think spontaneous pneumothorax can be treated on an out-patient basis.

Ehrlichia ewingii infection in a dog from South Korea - A case report

  • The report describes a case of natural E. ewingii infection confirmed by microscopic examination and molecular analyses in a domestic dog with acute lameness. After the diagnosis, the dog was successfully treated with oral doxycycline. To authors' knowledge, this is the first case report of natural E. ewingii infection in dogs in South Korea.

Effects of Six Antibiotics on the Activity of the Photosynthetic Apparatus and Ammonium Uptake of Thallus of Porphyra yezoensis

  • The modern integrated fish-seaweed mariculture has been tested to reduce the environmental impacts of an intensive fed culture. To obtain the best seaweed bioremediation performance, the effects of therapeutants used for fish disease control on the selected seaweed species should be considered. As a selected seaweed, Porphyra yezoensis was tested with six commercial antibiotics including erythromycin thiocyanate_A, erythromycin thiocyanate_B, oxytetracycline, doxycycline, pefloxacin, and amoxicillin trihydrate under the batch incubation at a photon flux density of 10 $\mu$mol ${\cdot}m^{-2}\;{\cdot}\;s^{-1}$ at 15$^{\circ}C$. Among the tested commercial antibiotics, erythromycin thiocyanate_A, erythromycin thiocyanate_B, oxytetracycline, and doxycycline showed decreases in Fv/Fm, the photochemical efficiency of photosystem II, with a dose-dependant and time-dependant manner. From the quenching analysis of chlorophyll fluorescence, three differential patterns were observed in the antibiotics-treated Porphyra: (1) high nonphotochemical quenching (NPQ) and low photochemical quenching (qP) in the cases of Erythromycin thiocyanate_B and amoxicillin trihydrate, (2) high NPQ and high qP in the case of pefloxacin and (3) low NPQ and low qP in the case of oxytetracycline. These results indicated that antibiotics affected in various ways on the photosynthetic apparatus, reflecting differential lesion sites of antibiotics. In addition, the rates of ammonium uptake also decreased with a decrease of Fv/Fm in P. yezoensis thalli treated with erythromycin thiocyanate_B and oxytetracycline. Therefore, the four antibiotics mentioned could affect the bioremediation capacity of the selected seaweed species in the integrated fish-seaweed mariculture system due to the decrease of photosynthetic activity and the simultaneous decrease of ammonium uptake.

벌꿀중의 잔유항생물질 및 Propionic Acid 분석011 관한 조사연구 (A Study on Analysis of Residual Antibiotics and Prop Acid in Honey)

  • A sensitive and simple analytical system for the simultaneous determination of residual oxytetracycline, tetracycline, and doxycycline in honey was described, and that the analytical method for determination of residual propionic acid in honey was established. Experimental subjects were purchased four kinds of honey, native kind honey, acaccia honey, mixed floral honey, chestnut honey in Kyung Sang Nam Do. Several microbiological methods are available to determine tetracycline antibiotecs(TCs) in foods but their precision apears to be variable and the specificity is questionable. These methods are considered to be not suitable for analysis of tetracycline antibiotics in honey because honey itself has bacteriostatic action. For determination of tetracycline antibiotics in honey, therefore the High Performance Liquid Chromatography(HPLC) method was applied, and the propionic acid were determined by Gas Chromatography(5.C). Ethylacetate, as an extract solvent, was found to be suitable for seperation of TCs in honey, but methanol and acetone were not. The recoverly rate of Oxytetracycline(OTC), Tetracycline(TC), Doxycycline(DC) from honey spiked at a level of 10 $\mu $g/g were 97%, 89%, and 91%, respectively. The cailbration curve in TCs was linear expression from 2$\mu $g/ml to 10$\mu $g/ml. As the results of analysis, the residual tetracycline antibiotics were not detected in the 100 samples of honey. The recovery rate of propionic acid from honey spiked at level of 10$\mu $g/g was 98.3% , and the calibra lion curves were linear expression from 21$\mu $g/ml to 101$\mu $g/ml. As the results of analysis, the residual propionic acid was not detected in the 100 samples of honey. Retention time(min) of OTC, DC, and TC were 3.35, 4.61, and 5.30 minutes at the conditions of table 2, respectively, and retention time(min) of propionic acid was 3.50 minutes at the conditions of table 3. The residual TCs and propionic acid were not detected in the 100 samples of honey, but there is a possibility that antibiotics or propionic acid will be to remain in honey if they are used during product period in order to prevent putrefaction of honey-bee.

Treatment-failure tularemia in children

  • Purpose: Tularemia is an infection caused by Francisella tularensis. Its diagnosis and treatment may be difficult in many cases. The aim of this study was to evaluate treatment modalities for pediatric tularemia patients who do not respond to medical treatment. Methods: A single-center, retrospective study was performed. A total of 19 children with oropharyngeal tularemia were included. Results: Before diagnosis, the duration of symptoms in patients was $32.15{\pm}17.8days$. The most common lymph node localization was the cervical chain. All patients received medical treatment (e.g., streptomycin, gentamicin, ciprofloxacin, and doxycycline). Patients who had been given streptomycin, gentamicin, or doxycycline as initial therapy for 10-14 days showed no response to treatment, and recovery was only achieved after administration of oral ciprofloxacin. Response to treatment was delayed in 5 patients who had been given ciprofloxacin as initial therapy. Surgical incision and drainage were performed in 9 patients (47.5%) who were unresponsive to medical treatment and were experiencing abcess formation and suppuration. Five patients (26.3%) underwent total mass excision, and 2 patients (10.5%) underwent fine-needle aspiration to reach a conclusive differential diagnosis and inform treatment. Conclusion: The causes of treatment failure in tularemia include delay in effective treatment and the development of suppurating lymph nodes.

The Effects of Peroxiredoxin III on Human HeLa Cell Proliferation

  • Background: Peroxidases (Prx) of the peroxiredoxin family reduce hydrogen peroxide and alkyl hydroperoxides to water and alcohol respectively. Hydrogen peroxide is implicated as an intracellular messenger in various cellular responses such as proliferation and differentiation. And Prx I activity is regulated by Cdc-2 mediated phosphorylation. This work was undertaken to investigate the proliferation role of peroxiredoxin III as a member of Prx family in Prx III overexpressed HeLa cell line. Methods: To provide further evidence of proliferation, we selected Prx III stably expressed HeLa Tet-off cell lines. Cell proliferation was examined by using proliferation reagent WST-1 in the presence or absence of doxycycline. Prx III, 2-cys Prx enzymes exist as homodimer. The activation of Prx III heterodimer with induced and endogenous Prx III was examined by immunoprecipitation. Results: Immunoprecipitation analysis of the induced and endogenous Prx III with anti-myc showed that the induced wild type (WT) and dominant negative (DN) Prx III from HeLa Prx III Tet-off stable cell heterodimerized with endogenous Prx III each other. And the expression level of induced Prx III was examined after addition of doxycycline. By 72 hr, the expression level of induced Prx III was diminished gradually and the half-life of the induced wild type Prx III was approximately 17 hr. The proliferation experiment demonstrated that the relative proliferation value of induced and endogenous WT Prx III stable cell has no changes but the DN Prx III induced HeLa Tet-off stable cells were lower than endogenous Prx III. Conclusion: In conclusion, the HeLa dominant negative Prx III Tet-off stable cells were decreased the proliferation.

한국 토종 단모종 고양이에서 Mycoplasma Haemofelis 감염 증례 (A Case of Mycoplasma haemofelis Infection in a Korean Domestic Shorthair Cat)

  • 6개월령의 한국토종단모고양이가 발열, 빈호흡, 식욕부진, 체중감소로 내원하였다. 고양이는 10일 전 Fellocel$^{(R)}$ 백신을 접종받은 이후 증상을 보였다. 신체검사 결과 공막의 황달과 호흡곤란을 호소했다. 혈액혈구검사와 혈액화학검사에서 빈혈, 혈소판감소증, 백혈구증가증, 고빌리루빈혈증이 관찰되었다. 혈액도말 검사와 기타 검사를 통해 심한 용혈을 확인할 수 있었고 고양이 용혈성 빈혈을 감별진단 하기위해 Erhlichia, Hemobartonella, FeLV, FIV, Anaplasma PCR 검사를 의뢰하였다. PCR 검사 결과 Mycoplasma haemofelis 특이 208 bp의 양성밴드가 확인되었고, 다른 질병은 음성으로 진단되었다. 고양이는 4주 동안 Doxycycline을 사용하였고 급성기 1주 동안에는 prednisolone을 같이 사용하였다. 임상증상은 약물투여 4일째부터 호전되기 시작하였고 이후 PCR 재검을 통해 완치를 확인하였다. 이것은 M. haemofelis 감염 고양이의 진단을 확인하고 완치한 국내 첫 증례이다.

병용 항균제의 어류질병 세균에 대한 시험관내 항균활성 (In vitro Antimicrobial Activity in Combination of Antibacterials Against Fish-pathogenic Bacteria)

  • 최근 육상의 넙치 양식장에서는 두 가지의 서로 다른 특성을 가지는 세균성질병이 같이 감염되는 혼합감염증이 빈번하게 발생하고 있는데, 혼합감염증은 서로 다른 원인균의 특성 때문에 단일 종류의 약제만으로는 치료효과를 거두기가 어려운 실정이다. 그러므로 본 연구는 약제의 병용 투여에 의해 이들의 상승작용을 이용함으로써 합병증에 대한 치료효과를 높이기 위한 기초자료를 얻고자 실시하였다. 어류 세균성질병의 원인세균인 Vibrio anguillarum, Edwardsiella tarda, Streptococcus sp., Staphylococcus epidermidis을 선정하여 병용 항균제의 시험관내 활성을 평가하는 대표적 방법인 Checkerboard법을 적용하여 fractional inhibitory concentration (FIC)지수를 구하여 상승효과를 나타내는 병용 항균제를 찾아내었다. (OTC+LM), (TC+FF), (OTC+FF)의 병용 항균제는 V. anguillarum에 대하여, (SNF+FF), (TC+FF), (SNF+OXA), (OTC+FF)의 병용 항균제는 E. tarda에 대하여, (CFX+OL), (OTC+OL), (TC+OL), (OTC+LM), (OTC+SPM), (OTC+EM), (DOXY+OL), (TC+SPM)의 병용 항균제는 Streptococcus sp.에 대하여, (CFX+EM), (FF+EM), (DOXY+OL), (CFX+OL)의 병용 항균제는 S. epidermidis에 대하여 시험관내 상승적 상호작용을 나타내었다.

액체크로마토그래피를 이용한 벌꿀 중 테트라싸이클린계 항생물질의 정량분석 및 잔류조사 (Determination and survey of tetracyclines residue in honey by high performance liquid chromatography)

  • Oxytetracycline, tetracycline, chlortetracycline and doxycycline in honey were separated by solid phase extraction (SPE) and determined with high performance liquid chromatography (HPLC) with UV/Visible detector. Analysis was carried out using following conditions: XTerra $C_8$ column $(3.9\times150mm\;i.d. 5{\mu}m)$, mobile phase composed of 0.01M oxalic acid : methanol : acetonitrile (820 : 80 : 100, v/v/v), isocratic pump at a flow rate of 0.9 ml/min. and $50{\mu}l$ of injection volume, UV/Visible detector with wavelength of 360nm. The calibration curves of four tetracyclines showed linearity $(\gamma^2>0.999)$ at concentration range of $100\~1,000 ng/ml$. The recoveries in fortified honey represented more than $70\%$ with low coefficient of variation $(<10\%)$ for concentration range of four tetracyclines. The detection limits for oxytetracycline, tetracycline, chlortetracycline and doxycycline were 13.8, 14.6, 26.2 and 24.9ng/g in acacia honey. respectively. We also monitored tetracyclines residue in domestic honey [n : 38, acacia (20), wild flower (18) ] and foreign honey [n=22, legally distributed (13), illegally distributed (9)] using modified Charm II screening and HPLC confirmation methods. Seven of the 60 samples $(11.7\%)$ were suspect positive using modified Charm II screening test. Chlortetracycline residue was found in one foreign honey (illegally distributed) tested at concentrations of 0.22 ppm. Conclusively, for more effective control of tetracyclines used in beekeeping should be further survey for residues in honey and also national guidelines (maximum residue limit : MRL) and methods should be obligatory.
