The Effect of Strength by Changing Tool Shape in the Friction Stir Welding (마찰교반용접에서 툴의 형상이 접합부 강도에 미치는 영향에 관한 연구)

  • As demand regarding a recent energy-saving rises, the using ratio of the aluminum plate in manufacturing of a railroad vehicle has been increasing. The aluminum structure to be applied to a railroad vehicle is divided to single skin and double skin, and the main aluminum product is mainly Al 6005 extrude and Al 5083 rolled in domestic market. The Al 6005 alloy is applied heat treatment in order to improve the strength of material. Therefore there is the disadvantage that the strength of welding zone decreases compare with base material's if you apply to fusion welding like MIG(metal inert gas) welding. In this paper we tried to apply friction stir welding to solve these problems. In this study we investigated how tensile strength and fatigue strength were changed in case of changing the shoulder diameter of thread tool.

Sausage Waves in a Plasma Cylinder with a Surface Current

  • Linear sausage oscillations of a cylinder embedded in a plasma with an azimuthal magnetic field, created by a current on the surface of the cylinder, are studied. Such a plasma configuration could be applied to modelling demonstrate that the lowest radial harmonic of the sausage mode is in the trapped regime for all values of the parallel wave number. In the long-wavelength limit, phase and group speeds of this mode are equal to the Alfven speed in the external medium. It makes the oscillation period to be determined by the ratio of the parallel wavelength, e.g., double the length of an oscillating loop, to the external Alfven speed, allowing for its seismological estimations. The application of the results obtained to the interpretation of long-period (longer than 20-30 s) oscillations of emission intensity detected in solar coronal structure, gives reasonable estimations of the external Alfven speed. Cutoff values of the parallel wavenumber for higher radial harmonics are determined analytically. Implications of this finding to the observational signatures of fast magnetoacoustic wave trains guided by the plasma non-uniformity are discussed.

  • Prime editors (PEs), which are CRISPR-Cas9 nickase (H840A)-reverse transcriptase fusion proteins programmed with prime editing guide RNAs (pegRNAs), can not only edit bases but also install transversions, insertions, or deletions without both donor DNA and double-strand breaks at the target DNA. As the demand for in-locus tagging is increasing, to reflect gene expression dynamics influenced by endogenous genomic contexts, we demonstrated that PEs can be used to introduce the hemagglutinin (HA) epitope tag to a target gene locus, enabling molecular and biochemical studies using in-locus tagged plants. To promote genome-wide in-locus tagging, we also implemented a publicly available database that designs pegRNAs for in-locus tagging of all the Arabidopsis genes.

An Analytical Study of the Effect of Inclined Angle of Road on Turn-over Accident of a High-speed Coach running on a Curved Road under Cross-wind Condition (횡풍이 작용하는 속도로의 회전구간에서 도로의 편경사각이 주행차량의 전복사고에 미치는 영향에 관한 분석연구)

  • Kyeonggi Provincial Government is considering double decker bus service to solve the problem of heavy rush hour traffic. However, the height-to-width ratio is more than 1.16 times larger than that of a general high-speed single decker bus, and the center of gravity is higher. This could cause driving stability problems, such as turnover and breakaway from the lane, especially under strong side-wind conditions at high speed. In this numerical study, the driving characteristics of a model double decker bus were reviewed under side-wind and superelevation conditions at high driving speed. The rolling, pitching, and yawing moment of the model bus were calculated with CFD numerical simulation, and the results were compared to the recovery angular moments of the model bus to evaluate the dynamic stability under given driving conditions. As the model vehicle moves on a straight level road, it is stable under any side-wind conditions. However, on a curved road under side-wind conditions, it could reach unstable conditions dynamically. There is a chance that the bus will turn over when it moves on a curved road with a radius of gyration less than 100 m under side-wind (15 m/s). However, there is a very small chance of breakaway from the lane under any driving conditions.

  • Electrode surface reaction on three carbon materials(glassy carbon, synthesized graphite, graphite foil) in 0.5 M K2SO4 electrolyte is investigated by impedance spectroscopy during anodic polarization. The double layer capacitance of the graphite foil electrode is relatively higher than that of other two materials. The change of capacitance parameter C due to chemical adsorption on glassy carbon and synthesized graphite(PVDF graphite) is observed in 0.5 M K2SO4 solution at anodic polarization. In general, the faradic impedance on glassy carbon depends on anodic polarization, and the change of impedance parameter on graphite foil at anodic polarization is not remarkable, because this reaction is controlled by field transport.

  • The paper presents a new short-term dynamic displacement estimation method based on an acceleration and a geophone sensor. The proposed method combines acceleration and velocity measurements through a real time data fusion algorithm based on Kalman filter. The proposed method can estimate the displacement of a structure without displacement sensors, which is typically difficult to be applied to earthquake or fire sites due to their requirement of a fixed rigid support. The proposed method double-integrates the acceleration measurement recursively, and corrects an accumulated integration error based on the velocity measurement, The performance of the proposed method was verified by a lab-scale test, in which displacement estimated by the proposed method are compared to a reference displacement measured by laser doppler vibrometer (LDV).

  • The present study covers the ultrasonic patterning process to replicate micro-patterns on a polymer substrate. The ultrasonic patterning process uses ultrasonic waves to generate frictional heat between an ultrasonic horn and the polymer substrate, from which the surface region of the polymer substrate is softened sufficiently for the replication of micro-patterns. The ultrasonic patterning process can divided into two categories according to the direction of vibration transmission: direct patterning and indirect patterning. The direct patterning uses a patterned horn, and the ultrasonic vibration is transferred directly from the patterned horn to the substrate. On the contrary, the indirect patterning process uses a plain horn, and the micro-patterns are engraved on a mold that is located below the substrate. Thus, the micro-patterns are replicated as an indirect manner. In this study, these direct and indirect patterning processes are compared in terms of the replication characteristics. Additionally, the possibility of double-side patterning is also discussed in comparison with the conventional single-side patterning process.

  • In order to provide natural images with a specified depth through three-dimensional display system, the stereo images should be similar to those projected from real environment as much as possible. Even when two persons see an identical scene, the binocular Parallax between two images of an object varies as a function of one's inter-pupilary distance (IPD). In this study, we investigated whether individual differences, such as IPD and accommodative vergence, would affect the perception of three dimensional scene provided by stereo-images. Results showed that a person's IPD is correlated with the limit of screen and binocular parallax for single vision, and affects the perceived depth of an object on fixation. More specifically, with longer IPD the limit of screen and binocular parallax for single vision is decreased, and the perceived depth is reduced. These results suggest that the screen and binocular parallax of an object should be calibrated with regard to users IPD to provide natural stereo-images with a specified depth and to Prevent double vision.

Construction, Investigation and Application of TEV Protease Variants with Improved Oxidative Stability

  • Tobacco etch virus protease (TEVp) is a useful tool for removing fusion tags, but wild-type TEVp is less stable under oxidized redox state. In this work, we introduced and combined C19S, C110S and C130S into TEVp variants containing T17S, L56V, N68D, I77V and S135G to improve protein solubility, and S219V to inhibit self-proteolysis. The solubility and cleavage activity of the constructed variants in Escherichia coli strains including BL21(DE3), BL21(DE3)pLys, Rossetta(DE3) and Origami(DE3) under the same induction conditions were analyzed and compared. The desirable soluble amounts, activity, and oxidative stability were identified to be reluctantly favored in the TEVp. Unlike C19S, C110S and C130S hardly impacted on decreasing protein solubility in the BL21(DE3), but they contributed to improved tolerance to the oxidative redox state in vivo and in vitro. After two fusion proteins were cleaved by purified TEVp protein containing double mutations under the oxidized redox state, the refolded disulfide-rich bovine enterokinase catalytic domain or maize peroxidase with enhanced yields were released from the regenerated amorphous cellulose via affinity absorption of the cellulose-binding module as the affinity tag.

The Study of Trnascriptional Regulated Gene, $hrp^{2+}$, in Yeast

  • This study was designed to clone the SNF2/SW12 helicase-related genes from the fission yeast Schizosaccha-romyces pombe and thereafter to elucidate the common functions of the proteins in this family. The $hrp^{2+}$gene was cloned by polymerase chain reaction amplification using degenerative primers from conserved SNF2 motifs within the ERCC6 gene, which encodes a protein involved in DNA excision repair. Like other SNF2/SW12 family proteins, the deduced amino acid sequence of Hrp2 contains DNA-dependent ATPase/7 helicase domains as well as the chromodomain and the DNA binding domain. This configuration is similar to that of mCHD1 (mouse chromo-ATPase/helicase-DNA-dinding protein 1), suggesting that Hrp2 is a S. pombe homolog of mCHD1, which is thought to function in altering the chromatin structure to control the gene expression. To characterize the function of Hrp2, 4 Uracil-Hrp2 fusion protein, it was purified near homogeneity by affinity chromatography on $Ni^{2+}$-NTA agarose, DEAE-Sepharose ion exchange arid Sephacryl S-200 gel filtration chromatographies. The purified fusion protein exhibited DNA-dependent ATPase activity, which was stimulated by both double-stranded and single-stranded DNA. To determine the steady-state level of $hrp^{2+}$ transcripts during growth, cells were cultured in medium and collected at every 2hr to prepare total RNAs. The northern blot analysis showed that the level of $hrp^{2+}$ transcripts reached its maximum before the cells entered the exponential growth phase and then decreased gradually, This result implies that Hrp2 may be required at early stages of cell growth.h.
