Shape optimization for partial double-layer spherical reticulated shells of pyramidal system

  • Triangular pyramid and Quadrangular pyramid elements for partial double-layer spherical reticulated shells of pyramidal system are investigated in the present study. Macro programs for six typical partial double-layer spherical reticulated shells of pyramidal system are compiled by using the ANSYS Parametric Design Language (APDL). Internal force analysis of six spherical reticulated shells is carried out. Distribution regularity of the stress and displacement are studied. A shape optimization program is proposed by adopting the sequence two-stage algorithm (RDQA) in FORTRAN environment based on the characteristics of partial double-layer spherical reticulated shells of pyramidal system and the ideas of discrete variable optimization design. Shape optimization is achieved by considering the objective function of the minimum total steel consumption, global and locality constraints. The shape optimization of six spherical reticulated shells is calculated with the span of 30m~120m and rise to span ratio of 1/7~1/3. The variations of the total steel consumption along with the span and rise to span ratio are discussed with contrast to the results of shape optimization. The optimal combination of main design parameters for six spherical reticulated shells is investigated, i.e., the number of the optimal grids. The results show that: (1) The Kiewitt and Geodesic partial double-layer spherical reticulated shells of triangular pyramidal system should be preferentially adopted in large and medium-span structures. The range of rise to span ratio is from 1/6 to 1/5. (2) The Ribbed and Schwedler partial double-layer spherical reticulated shells of quadrangular pyramidal system should be preferentially adopted in small-span structures. The rise to span ratio should be 1/4. (3) Grids of the six spherical reticulated shells can be optimized after shape optimization and the total steel consumption is optimized to be the least.

Biological and Molecular Characterization of a Korean Isolate of Orthotospovirus chrysanthinecrocaulis (Formerly Chrysanthemum Stem Necrosis Virus) Isolated from Chrysanthemum morifolium

  • Biological and molecular characterization of a Korean isolate of Orthotospovirus chrysanthinecrocaulis (formerly known as chrysanthemum stem necrosis virus, CSNV) isolated from Chrysanthemum morifolium was determined using host range and sequence analysis in this study. Twenty-three species of indicator plants inoculated mechanically CSNV-Kr was investigated for determination of host range. CSNV-Kr induced various local and systemic symptoms in the inoculated plant species. CSNV-Kr could not infect three plant species and induced symptomless in systemic leaves in Nicotiana tabacum cultivars, though the plant samples reacted positively with the antiserum to CSNV by double-antibody sandwich-enzyme-linked immunosorbent assay. The complete genome sequence of CSNV-Kr was determined. The L RNA of CSNV-Kr consists of 8,959 nucleotides (nt) and encodes a putative RNA-dependent RNA polymerase. The M RNA of CSNV-Kr consists of 4,835 nt and encodes the movement protein (NSm) and the glycoprotein precursor (Gn/Gc protein). The S RNA of CNSV-Kr consists of 2,836 nt and encodes NSs protein and N protein. The Gn/Gc and N sequence of CSNV-Kr were compared with those of previously published CSNV isolates originating from different countries at nucleotide and amino acid levels. The Gn/GC sequence of CSNV-Kr shared 98.8-99.5% identity with CSNV isolated from other countries and the N sequence of CSNV-Kr shared 98.8-99.6% identity. No particular region of variability could be found in either grouping of viruses. All of the CSNV isolates did not show any relationship according to geographical origins and isolation hosts, suggesting no distinct segregation of the CSNV isolates.

  • Delta-integration vectors were constructed for the purpose of achieving homologous integration of the hirudin expression cassette into the chromosome of Saccharomyces cerevisiae. A double $\delta$ system truncated with the unnecessary bacterial genes, and consequently having a reduced insert size for integration, showed a four-fold increase in transformation efficiency at given DNA concentrations, and as a result, the constructed recombinant yeast strain had a 1.3-fold enhancement in hirudin expression level compared with a single $\delta$ system.

Effective Family Shuffling Method Using Complementary DNA Fragments Produced by S1 Nuclease

  • An efficient method for the in vitro reassembly of homologous DNA sequences is presented. The proposed method involves obtaining single strands of homologous genes and hybridizing them to obtain partially hybridized heteroduplex DNA; cleaving the single-stranded regions of the heteroduplex DNA using S1 nuclease to generate double-strand DNA fragments; denaturing the double-strand DNA fragments to generate single-strand DNA fragments; conducting a series of polymerase chain reactions (PCR) using the single-strand DNA fragments as internal primers and a mixture of homologous DNA as templates to obtain elongated reassembled DNA; and finally, amplifying the reassembled DNA by a PCR using terminal primers. As a result, DNA reassembly could be achieved between homologous genes with a sequence similarity as low as 78%.

  • In this paper we studied an implementation of QR decomposition-based RLS algorithm using modified Givens rotation method. Givens rotation can be obtained with a sequence of the CORDIC operations. In order to reduce the computing time of QR decomposition we restricted the number of iterations of the CORDIC operation per a Givens rotation and used double-rotation method to remove the square-root in the scaling factor.

  • In this paper, we analyze and simulate performance of space time block coded (STBC) binary pulse amplitude modulation-direct sequence (BPAM-DS) ultra-wideband (UWB) systems with double binary turbo code in indoor environments for various ubiquitous healthcare (u-healthcare) applications. Indoor wireless channel is modeled as a modified Saleh and Valenzuela (SV) model proposed as a UWB indoor channel model by the IEEE 802.15.SG3a in July 2003. In the STBC encoding process, an Alamouti algorithm for real-valued signals is employed because UWB signals have the type of real signal constellation. It is assumed that the transmitter has knowledge about channel state information. From simulation results, it is shown that the STBC scheme does not have an influence on improving bit error probability performance of the BPAM-DS UWB systems. It is also confirmed that the results of this paper can be applicable for u-healthcare applications.

  • Fault location algorithms based on the current distribution factors under the one-phase to earth fault condition of a double-circuit line are presented. The derivation method for current distribution factors is showed, to calculate fault current, fault resistance and the zero sequence current of other parallel circuit which are unknown. As the proposed algorithms 1,2,3 embodies an accurate location by the voltage and the current of the relaying point.

Double-Dwell Hybrid Acquisition in DS-UWB System

  • In this paper, we analyze the performance of double-dwell hybrid initial acquisition in DS-UWB system via detection, miss, false alarm probabilities and mean acquisition time. In the analysis, we consider the effect of the acquisition sequence, and deployment scenario of the abundant multipath components over the small coverage of the piconet in DS-UWB system. Based on the simulation, we obtain various performance on the mean acquisition time by varying the parameters, such as the total number of hypotheses to be searched, subgroup size, and dwell time. Then, we suggest the optimum parameter set for the initial acquisition in DS-UWB system.

  • New binuclear Ni(Ⅱ) and Cu(Ⅱ) complexes with various alkyl derivatives of 1,2-bis(1,3,6,8,10,13-hexaaza-1-cyclotetradecyl) ethane, in which two fully saturated 14-membered hexaaza macrocyclic subunits are linked together by an ethylene chain, have been synthesized by the one step template condensations of formaldehyde with ethylenediamine and appropriate primary alkyl amines in the presence of the metal ions. Each macrocyclic subunit of the double-ring macrocyclic complexes contains one alkyl pendant arm and has a square planar geometry with a 5-6-5-6 chelate ring sequence. The visible spectra and oxidation properties indicate that the metal-metal interaction of the binuclear complexes are not significant. Synthesis, characterization, and the properties of the complexes are presented.

Photodamage to Double-stranded DNA by Xanthone Analogues Increases Exponentially with Their HOMO Energies

  • DNA photodamage mediated by photosensitizers are believed to play an important role in solar UVA carcinogenesis. We investigated the relationship between the DNA-damaging abilities of photoexcited xanthone analogues (as photosensitizers) and their highest occupied molecular orbital (HOMO) energies. DNA damage was examined using /sup 32/P-labeled DNA fragments obtained from the p53 tumor suppressor gene. These compounds induced DNA photodamage in a similar manner, and the extents of DNA damage were following order: xanthone> thioxanthone > acridone. Photoexcited xanthone caused nucleobase oxidation specifically at 5'-G of GG sequence in double-stranded DNA. An oxidative product of 2'-deoxyguanosine, 8-hydroxy-2'-deoxyguanosine (8-OHdG), was detected, and the amount was decreased by DNA denaturation. These findings suggest that photoexcited xanthone generates 8-OHdG at 5'-G of GG in double-stranded DNA through electron transfer. The calculated HOMO energies of these photosensitizers decreased in the following order: xanthone> thioxanthone > acridone. This study has demonstrated that DNA-damaging abilities of these photosensitizers increased exponentially with an increase in their HOMO energies.
