in vitro에서 B(a)P이 착상전 마우스 배자에 미치는 독성학적 영향에 관한 연구 (Toxicological Effects of B(a)P on Preimplantation Mouse Embryos in Vitro)

  • Effects of B(a)P on preimplantation mouse embryos in vitro were studied. Preimplantation mouse embryos were exposed to a concentration of 0.3, 1, 3 and 10 $\mu$M B(a)P for 72 hrs. The toxicological effects of B(a)P were evaluated by morphological observation of embryos up to the blastocyst stage, and by measuring DNA, RNA and protein synthesis by radioactive precursor incorporation. At 1 $\mu$M B(a)P did not affect preimplantation development but interfered with hatching and ICM formation. Suppressing effect of ICM formation was dose dependent. At the eight cell stage, the developmental rate was decreased at above 3 $\mu$M of B(a)P. At the blastocyst stage, attachment and trophoblast outgrowth were diminished at the 10 $\mu$M of B(a)P and ICM formation was decreased at 1 $\mu$M of B(a)P. Inner cell number of blastocyst was decreased dose dependently. So, number of ICM was one of the most sensitive and toxicological end point. The RNA incorporation rate of 0.1 $\mu ^3$H-uridine was dosedependent and the protein incroporation of 0.5 $\mu Ci ^{35}$S-methionine showed a significant decrease after 48 hrs. But the DNA incorporation rate of methyl-$^3$H thymidine was not affected. Our results suggested that B(a)P did not affect the DNA replication but transcription was inhibited by dose dependent manner. There delay of development during the blastocyst stage was mainly due to the inhibition of RNA synthesis followed by protein synthesis.

Haloperidol 이 심혈관계에 미치는 영향 (Experimental Studies on the Cardiovascular Effects of Haloperidol in Cat and Rabbit)

  • Haloperidol, a butyrophenone, was synthetized by Janssen and introduced for the treatment of psychosis. Although structurally different from the phenothiazines, the butyrophenones share many of their pharmacological properties, such as inhibition of conditioned avoidance response, blocking effect of amphetamine reaction, producing catalepsy, antishock effect and protection against the lethal effects of catecholalmines. Chlorpromazine can lower the arterial blood pressure through its adrenergic blocking activity, its direct effect in relaxing vascular smooth muscle, its direct effect in depressing the myocardium and its action in a complex manner on the central nervous system. In the case of haloperidol, however, was not clarified the mechanism of lowering the blood pressure. The present paper describes the effects of haloperidol on cardiovascular system to investigate the mechanisms of its actions on the arterial blood pressure. The results are followings; 1. In anesthetized cats, intravenous administration of haloperidol and chlorpromazine in the dose of 0.1mg/kg produced a slight decrease in the blood pressure, which followed by complete recovery within $30{\sim}60$ minutes. In the dose of 3mg/kg, however, both produced an abrupt and marked decrease of the blood pressure, which followed by delayed recovery. 2. Haloperidol in the dose ranges of 0.1mg to 3.0mg/kg tended to produce the heart rate slowing in the cats, while chlorpromazine has no effect on the rate. 3. Following administration of haloperidol or chlorpromazine, epinephrine reversal in the arterial blood pressure was observed in the cat, however the responses of norepinephrine and acetylcholine were little affected. 4. In the isolated rabbit atrium the contractility was depressed by haloperidol in the doses over 0.5mg per 100ml, but the rate was not affected. In contrast, the epinephrine-induced contractility was not depressed after haloperidol treatment. However, the increased rate of atrium by epinephrine was partially blocked after haloperidol. 5. In the isolated rabbit aortic strip, epinephrine-induced contraction was blocked by haloperidol. With the above results, it may be concluded that the hypotensive effect of haloperidol was largely due to ${\alpha}$-adrenergic blocking properties and the direct effect in depressing the myocardium as well as its action on central nervous system.

Nonmonotonic Effects of Chronic Low-Dose Di(2-ethylhexyl) Phthalate on Gonadal Weight and Reproduction

  • Endocrine disruptors have been concerned in toxicology but now challenged as physiological point especially concerned with exposing dose and period. In this study the low-dose chronic administration of di(2-ethylhexyl) phthaltae (DEHP) during reproductive period was examined to evaluate the possible roles. Adult male and female CD-1 mice were exposed to DEHP with drinking water containing $133{\mu}g/L$ and $1,330{\mu}g/L$ DEHP in water according to OECD 433 guide line and sacrificed just after weaning. The weights of uterus and ovary were decreased by drinking of $1,330{\mu}g/L$ DEHP water. There was not adverse effects on either accumulated mating rate and mating rate depend on estrus stage, pregnancy duration, and sex ration at birth. However, the accumulated rate of successful delivery and litter size were significantly high at $1,330{\mu}g/L$ DEHP water. The number of epididymal sperm was significantly increased by drinking of $1,330{\mu}g/L$ DEHP water. In addition, the number of follicles (primary, secondary, tertiary) were more many than control at $1,330{\mu}g/L$ DEHP water drunk mother. Though further studies are needed to identify what are the mechanism of DEHP in folliculogenesis and spermatogenesis. From this study we firstly report the effect of low-dose chronic administration of DEHP with drinking could change the ovarian follicle population size and spermatogenesis rate. Put together, those finding is different from previous high-dose effects and suggest the physiological role of DEHP in gonads and uterus.

장기 중선량률의 감마선 피폭 기간에 따른 실험동물의 생물학적 영향 연구 (Biological Effects of Different Chronic Medium-Dose-Rate Gamma Radiation Period Exposed on Mice)

  • Recently, chronic gamma radiation exposure on biological effects in middle dose-rates have become a serious concern. We investigated the biological effects of middle dose chronic exposure to gamma ray. Fifty male 6-week-old specific free Balb/c mice were randomly divided into five groups (four groups irradiated and one non-irradiated control group). Gamma radiation exposed in Gamma phytotron on Advanced Radiation Technology Institute (Jeongeup, Korea). Irradiation was carried out for 1 or 2 weeks using gamma rays at dose rates of 45 and $50mGy\;h^{-1}$ with total doses 7.56 Gy ($45mGy\;h^{-1}$, 1 week), 8.4 Gy ($50mGy\;h^{-1}$, 1 week), 15.12 Gy ($45mGy\;h^{-1}$, 2 weeks) and 16.8 Gy ($50mGy\;h^{-1}$, 2 weeks). After irradiation, immediately we sacrificed and counted body and organ weights. Moreover we counted spleen cell numbers. Compared with control non-irradiated group, all irradiated groups of body and spleen weights showed significant decreased. However, no significant alteration was observed between same irradiated period groups. In spleen cell numbers, reduced compared to the control group. However, significant alteration was observed between same irradiated period groups ($45mGy\;h^{-1}$, $50mGy\;h^{-1}$). These results demonstrated biological effects according to the radiation dose rate and irradiated period.

이산화염소에 의한 페놀제거 및 살균 (Disinfection & Removal of Phenol by Chlorine Dioxide)

  • The effects of chlorine dioxide on the oxidation of phenol and disinfection were studied in the various test water conditions. With the 0.3mg/l of chlorine dioxide dose, the spiked phenol(initial concentration: 0.1mg/l) was completely oxidized within 10 minute. The removal rate of phenol was much faster in distilled water than in ground water and filtered water. The applied dose of chlorine dioxide concentrations higher than 0.2mg/l was sufficiently enough for the complete oxidation of phenol. However, with 0.1mg/l of dose, chlorine dioxide can oxidize only 20% of the spiked phenol. The reactive substances present in test water may influence the chlorine dioxide demand in water. pH effect of oxidation rate was also investigated. Increasing the pH, the removal rate of phenol was found to be increased. The disinfection test of chlorine and chlorine dioxide were conducted and compared. The lethal effect for the both disinfectants are similarly powerful. The time for 99% inactivation of E. coli was obtained within 120 sec with the 0.2mg/l of each dose.

근접조사용 Ir-192 마이크로선원의 디자인과 선량 특성 (The Dose Characteristics of Designed Ir-192 Micro-source for Brachytherapy)

  • 근접조사용 선원으로 직경이 0.5 mm에 길이 3.5 mm인 Ir-192선원을 고안하고, 선량알고리즘을 개발하였다. 선원을 0.1 mm 두께로 균등분할하고 총 875개의 미소선원으로부터 조직에 도달되는 선량을 계산하고 선량분포를 조사하였다. 선원을 감싸는 캡슐용기의 두께는 0.3 mm이며, 선량기준점인 Ir선원의 Air-kerma strength(S$_{k}$ ) 는 mCi당 4.108U를 얻었으며 선원 측방의 1 cm에서 1.154 Uh$^{-1}$ (1.3167${\times}$$10^{-3}$cGy/mCi-sec)의 조직선량률을 얻었다. Air-kerma strength로부터 조직내 선량률상수(Λ)는 Ir-192에너지 스펙트럼과 실험치인 조직내 산란보정계수를 적용하여 1.154 cGy h$^{-1}$U$^{-1}$ 얻었다. 선량분포의 특성은 선원 축방향으로 56%의 상대선량률을 보였고, 케이블측의 선량감쇠는 50%를 보였다. 마이크로선원의 중심에서 반경 1 cm 주위의 선량분포와 5 cm의 선량분포를 비교한 결과 반경이 큰 경우의 측방선량에 비해 축 방향의 선량이 약 2% 감쇠함을 보았다. 선원자체의 필터효과는 점선원에 비해 88.3%의 선량률을 얻어 약 12%의 감쇠를 보였으며, 캘슐벽의 필터효과는 약 3% 정도로 미미하게 나타났으며 기준점에서 선량필터효과는 약 15.6% 감쇠되는 것으로 나타났다. Ir-192 선원과 같이 에너지가 낮은 선원을 사용하는 경우 선량분포특성에 따라 아프리케이터의 직경을 크게 하여 선원 축 방향의 선량저하를 피할 수 있음을 알 수 있었다.

Dose- and Time-Related Effects of Pilocarpine Mouthwash on Salivation

  • Purpose: Pilocarpine as a salivation stimulant in pill form has mostly been used to relieve oral dryness for xerostomic patients but its use may often be limited due to variable side effects from systemic absorption. Therefore, the purpose of this study was to investigate the effects of pilocarpine mouthwash on salivation according to the variable concentration and duration for healthy volunteers. Related adverse effects and subjective assessment on its effects on salivation were also examined. Methods: This study was performed as placebo-controlled, double-blind, randomized clinical trial. Thirty healthy volunteers (male=23, mean age=22.2 years) were randomly allocated to 6 groups with the different concentration of pilocarpine mouthwash (placebo, 0.1%, 0.5%, 1.0%, 1.5%, and 2.0%). The whole experiment consisted of 3 sessions according to the duration of mouthwash, i.e., 1, 3, and 5 minutes with the mean wash-out period ${\geq}2$ days between the sessions. Unstimulated whole saliva was collected before and after gargling with a mouthwash. Results: Salivation of the higher concentration groups ${\geq}1%$ significantly increased than those of lower concentration group. The application period of mouthwash did not cause any changes of salivary flow rate at the higher concentrations ${\geq}1.0%$. The lower concentrations of 0.5% and 0.1% had no effects on salivation even after 5-minute mouthwash. There was no significant difference between blood pressure and pulse rate before and after use of mouthwash. Conclusions: From the results of the current study, pilocarpine mouthwash with at least 1.0% concentration more than a minute might be clinically effective in salivation without any serious side effects. Dose of mouthwash rather than duration seems to be a critical factor to salivation.

전신 PET/CT 검사에서 공간선량률 측정 (Measurement of the Spatial Dose Rates During PET/CT Studies)

  • 전신 PET/CT 검사에서 $^{18}F-FDG$의 방사성의약품을 투여한 환자가 방사선원이 되기에 환자로부터 종사자의 피폭선량 예측을 위한 PET 검사실에서 환자 주변의 공간선량률을 측정 분석하였다. 연간 개인피폭선량은 대학병원의 핵의학 분야에 근무하는 종사자가 방사선종양학과와 소규모병원의 진단방사선 분야에 비해 검사 중에 환자로부터 방출되는 공간선량률에 의하여 개인피폭선량이 높게 나타났다. 그리고 PET/CT 검사에서 $^{18}F-FDG$를 이용하는 경우에 $^{99m}Tc$ 보다 공간선량률이 $4{\sim}6$배 정도로 훨씬 높고, 촬영실 전체에 공간선량률이 분포함을 알 수 있었다. 따라서 방사성의약품 투여 후 안정실이나 PET 검사 중에 촬영실 내에서는 항상 방사선 방어의 기본인 시간은 짧게, 거리는 멀리, 차폐를 고려하여 PET 검사를 수행하는 것이 매우 중요하고, 환자로부터의 공간선량률에 따른 종사자의 개인피폭선량을 줄이기 위한 최선의 노력이 필요하다.

  • To simulate the effect of high dose-rate radiation on semiconductor devices, device modeling work has been performed especially in the area of photo-current generation by a PIN diode. The resultant analytical values were compared with experimental ones that were specially designed and performed to benchmark the simulation results. Initial results showed 27.85% error between the simulation and the experiment. The error can be further reduced by improvement both in simulation and in related experiments. The developed technique from the study can be applicable to radiation dosimetry and to analysis on the radiation effects in electronics.

집속 이온빔 가공변수에 따른 Au 에칭 특성 연구 (The ocused Ion Beam Etching Characteristic of Au)

  • Focused Ion Beam(FIB) systems is a useful tool for the fabrication of micro-nano scale structures. In this study, the effects of FIB etching on the Au microstructure are systematically investigated. As the fabrication parameters, ion dose, dwell time and beam overlap ratio are studied. First, the increases of Ga ion dose makes the milling yield higher and the sidewall of milling profile steeper. Dwell time is found to have little effects on the milling profile due to the relatively large milling area of $1\times1{\mu}m^2$ used in this study. However, beam overlap significantly affects not only milling rate but also milling profile. As the beam overlap ratio changes from positive to negative, the development of regular cross-stripe patterns at the bottom with low milling rate is observed.
