  • Effects of phenobarbital on the pharmacokinetics of griseofulvin were studied in rats. Phenobarbital was administered orally for five days at the dose of 75mg/kg/day. Absolute bioavailability of oral griseofulvin was significantly(p<0.005) reduced but total clearance(CL$_s$ was not changed by phenobarbital pretreatment. Absorption rate constant(K$_a$ and maximum plasma concentration(C$_{max}$) were significantly(p<0.05) reduced, and time to reach maximum plasma concentration(T$_{max}$) of griseofulvin was significantly(p<0.05) increased by phenobarbital pretreatment. Changed pharmacokinetics of griseofulvin seemed not to be due to induced enzyme activity by phenobarbital but to reduced GI absorption of griseofulvin.

In vivo Brain-to-blood Efflux Transport of Choline at the Blood-brain Barrier

  • The purpose of this study was to clarify the efflux transport system of choline from brain to blood across the blood-brain barrier (BBB) in rats using the brain efflux index (BEI) method. $[^3H]$Choline was micro-injected into parietal cortex area 2 (Par2) of the rat brain, and was eliminated from the brain with elimination halflife of 45 min. The BBB efflux clearance of $[^3H]$choline was about 124 mL/min/g brain, which was determined from combination of an elimination rate constant $(1.54X10^{-2}min^{-1})$ and the distribution volume in the brain (8.05 mL/g brain). The efflux of $[^3H]$choline was inhibited by unlabeled choline in a dose-dependent manner and was significantly inhibited by cationic substrates, such as hemicholinium-3 and tetraethylammonium (TEA). These results suggest that the BBB may act as an efflux pump for choline to reduce the excessive choline concentration in the brain interstitial fluid.

  • Effects of phenobarbital pretreatment on the pharmacokinetics of enterohepatic recirculating griseofulvin were investigated by comparing normal to bile duct cannulated rats and also the effects of enhanced endogeneous bile flow on the absorption of griseofulvin were studied by means of in situ recirculation method in phenobarbital-pretreated rats. Phenobarbital was administered orally for five days at the dose of 75 mg/kg/day. The influence of phenobarbital pretreatment on the absorption rate constant, area under the plasma concentration-time curve, maximum plasma concentration of orally administered griseofulvin was not found in bile duct cannulated rats. Decreased absorption clearance and apparent partition coefficient of griseofulvin in accordance with the amount of endogeneous bile juice seemed to be due to the decrease of thermodynamic activity of griseofulvin as bile forms the micelle with griseofulvin.

Synthesis of Poly(N-methylol Methacrylamide/Vinyl Sulfonic Acid) Hydrogels for Heavy Metal Ion Removal

  • In this study, poly(N-methylol methacrylamide) (NMMAAm) and poly(N-methylol methacrylamide/vinyl sulphonic acid) (NMMAAm-VSA) hydrogels were synthesized by $^{60}Co-{\gamma}$ ray irradiation at an ambient temperature. The graphs belonging to the gelation percent- percent-dose and swelling curves were drawn by using data which were obtained from water and different pH solutions. Characterization of hydrogels was performed by FTIR and DSC-TGA analysis. Heavy metal ion ($Ni^{2+}$, $Co^{2+}$) removal capacities of hydrogels were investigated in aqueous solutions, which had different concentrations (100-1500 mg/L). In metal ion removal studies, pH value of aqueous medium was kept constant at 5.0. Maximum metal ion removal values were obtained for NMMAAm-VSA (1:3 mole ratio) hydrogels. Metal ion removal capacities of NMMAAm-VSA (1:3 mole ratio) hydrogels were found as 82 mg/g and 98 mg/g for $Ni^{2+}$ and $Co^{2+}$ ions, respectively.

Drug Interaction between Nimodipine and Cyclosporine in Rabbits (가토에서 니모디핀과 싸이크로스포린과의 약물상호작용)

  • The purpose of this study was to report the pharmacokinetic changes of cyclosporine after oral administration of cyclosporine, 10 mg/kg, in rabbits coadministered or pretreated twice per day for 3 days with nimodipine, dose of 5 mg/kg. The area under the plasma concentration-time curve (AUC) of cyclosporine was significantly higher in rabbits pretreated with nimodipine than that in control rabbits (p<0.01), showing about 149% increased relative bioavailability. The peak plasma concentration (C$_{max}$), elimination half-life (t$_{1}$2/) and MRT of cyclosporine were increased significantly (p<0.05) in rabbits pretreated with nimodipine compared with those in control rabbits. This findings could be due to significant reduction of elimination rate constant and total body clearance by pretreated with nimodipine. The effects of nimodipine on the pharmacokinetics of oral cyclosporine were more considerable in rabbits pretreated with nimodipine compared with those in control rabbits. The results suggest that the dosage of cyclosporine should be adjusted when the drug would be coadministered chronically with nimodipine in a clinical situation.n.

  • The main focus of this paper is on the study of the radiation effect upon the electrical property of a A-Kind insulator which is one of the organic insulators. The varnished cloth is so typical of the A-Kind insulators as to be selected for a sample. AC and Impluse voltage is applied to the variable Gamma ray irradiated dose samples with the constant time duration and the dielectric breakdown voltage are measured from the sample. The experimental data of the dielectric breakdown voltage are considerably decreased. The above conclusion is useful for the selection and application of the orgainc insulators under the irradiation effects.

Drug Interaction of Cimetidine and Isoniazid (시메티딘과 이소니아짓의 약물 상호작용)

  • Pharmacokinetic interaction of cimetidine and isoniazid was investigated in the rabbits. Isoniazid was administered orally at a dose of 30mg/kg to six rabbits after 10, 20, and 30mg/kg pretreatment of cimetidine twice a day for 10days. Concentration of the free and the total isoniazid in the blood and the urine was determined by spectrophotometer. Relative bioavailability and biological half-life($t\frac{1}{2}{\beta}$) were increased significantly by cimetidine pretreatment. Overall elimination rate constant and total clearance of isoniazid were decreased significantly by cimetidine pretreatment. The ratio of metabolites to isoniazid in the blood and the urine was decreased significantly by cimetidine pretreatment. Relative bioavailability, INAH to metabolites ratio in the blood and decrease in total clearance were highly correlated with the does of cimetidine pretreated. This result might be due to the inhibition of isoniazid metabolism in the liver by cimetidine pretreatment.

  • Polyimide films have excellent thermal stability, reliable mechanical properties and low dielectric constant. Therefore, this material is widely used in many industrial fields such as microelectronics, flexible circuits, semiconductor products and aerospace materials. In space applications, earth-orbiting hardware operates in environments that generally include neutral particles, charged particles such as trapped protons and electrons, solar protons, and cosmic rays. Under these conditions, polyimide films were changed in the optical, electrical and mechanical properties. Therefore, in this study, we evaluated the effects of electron beam irradiation on polyimide. The O-H functional groups were created on the polyimide film surface in the results of FT-IR spectra. And it was found that the dielectric constants were changed as a function of electron beam dose.

The Comparison of Absolute Dose due to Differences of Measurement Condition and Calibration Protocols for Photon Beams (6MV 광자선에서 측정 조건의 변화와 측정법의 차이에 의한 절대 선량값의 비교)

  • The absolute absorbed dose can be determined according to the measurement conditions; measurement material, detector, energy and calibration protocols. The purpose of this study is to compare the absolute absorbed dose due to the differences of measurement condition and calibration protocols for photon beams. Dosimetric measurements were performed with a farmer type PTW and NEL ionization chambers in water, solid water, and polystyrene phantoms using 6MV photon beams from Siemens linear accelerator. Measurements were made along the central axis of 10cm $\times$ 10cm field size for constant target to surface distance of 100cm for water, solid water and polystyrene phantom. Theoretical absorbed dose intercomparisons between TG21 and IAEA protocol were performed for various measurement combinations of phantom, ion chamber, and electrometer. There were no significant differences of absorbed dose value between TG21 and IAEA protocol. The differences between two protocols are within 1% while the average value of IAEA protocol was 0.5% smaller than TG21 protocol. For the purpose of comparison, all the relative absorbed dose were nomalized to NEL ion chamber with Keithley electrometer and water phantom, The average differences are within 1%, but individual discrepancies are in the range of - 2.5% to 1.2% depending upon the choice of measurement combination. The largest discrepancy of - 2.5% was observed when NEL ion chamber with Keithley electrometer is used in solid water phantom. The main cause for this discrepancy is due to the use of same parameters of stopping power, absorption coeficient, etc. as used in water phantom. It should be mentioned that the solid water phantom is not recommended for absolute dose calibration as the alternative of water, since absorbed dose show some dependency on phantom material other than water. In conclusion, the trend of variation was not much dependent on calibration protocol. However, it shows that absorbed dose could be affected by phantom material other than water.

Effects of Arc Number or Rotation Range upon Dose Distribution at RapidArc Planning for Liver Cancer (간암환자를 대상으로 한 래피드아크 치료계획에서 아크수 및 회전범위가 선량분포에 미치는 영향)

  • In this paper, we evaluated the performance of 3D CRT, IMRT and three kind of RA plannings to investigate the clinical effect of RA with liver cancer case. The patient undergoing liver cancer of small volume and somewhat constant motion were selected. We performed 3D CRT, IMRT and RA plannings such as 2RA, limited triple arcs (3RA) and 3MRA with Eclipse version 8.6.15. The same dose volume objectives were defined for only CTV, PTV and body except heart, liver and partial body in IMRT and RA plannings. The steepness of dose gradient around tumor was determined by the Normal Tissue Objective function with the same parameters in place of respective definitions of dose volume objectives for the normal organs. The approach between the defined dose constraints and the practical DVH of CTV, PTV and Body was the best in 3MRA and the worst in IMRT. The DVHs were almost the same among RAs. Plans were evaluated using Conformity Index (CI), Homogeneity Index (HI) and Quality of coverage (QoC) by RTOG after prescription with dose level surrounding 98% of PTV in the respective plans. As a result, 3MRA planning showed the better favorable indices than that of the others and achieved the lowest MUs. In this study, RA planning is a technique that is possible to obtain the faster and better dose distribution than 3D CRT or IMRT techniques. Our result suggest that 3MRA planning is able to reduce the MUs further, keeping a similar or better targer dose homogeneity, conformity and sparing normal tissue than 2RA or 3RA.
