A tumor suppressor, merlin is a member of ERM family proteins. It consists of N-terminal FERM domain, ${\alpha}-helical$ region, and C-terminal domain. Alternative splicing of merlin's mRNA generates two isotypes of merlin. Isotype I, which has exon17 at the C-terminus instead of exon16 in isotype II, is known to have tumor suppressor activity. Like other ERM proteins, the C-terminal domain of merlin isotype I interacts to its FERM domain. That of isotype II, however, was reported not to bind FERM domain despite the large common part of C-terminal domain, which possibly binds FERM domain. Here, we show the binding of FERM domain to both C-terminal domains of merlin's two isotypes by isothermal titration calorimetry. These results support that merlin isotype II also can form a closed conformation or a multimer by intramolecular or intermolecular interactions using their FERM domain and C-terminal domain.

This paper investigates the modelling of coupled soil-structure interaction problems by domain decomposition techniques. It is assumed that the soil-structure system is physically partitioned into soil and structure subdomains, which are independently modelled. Coupling of the separately modelled partitioned subdomains is undertaken with various algorithms based on the sequential iterative Dirichlet-Neumann sub-structuring method, which ensures compatibility and equilibrium at the interface boundaries of the subdomains. A number of mathematical and computational characteristics of the coupling algorithms, including the convergence conditions and choice of algorithmic parameters leading to enhanced convergence of the iterative method, are discussed. Based on the presented coupling algorithms a simulation environment, utilizing discipline-oriented solvers for nonlinear structural and geotechnical analysis, is developed which is used here to demonstrate the performance characteristics and benefits of various algorithms. Finally, the developed tool is used in a case study involving nonlinear soil-structure interaction analysis between a plane frame and soil subjected to ground excavation. This study highlights the relative performance of the various considered coupling algorithms in modelling real soil-structure interaction problems, in which nonlinearity arises in both the structure and the soil, and leads to important conclusions regarding their adequacy for such problems as well as the prospects for further enhancements.
The purpose of this study was to examine mothers' ideal and actual parenting behaviors toward their infants in three parenting domains; social, didactic, and limit setting. A total of 264 mothers of young children under age three from Seoul, Korea completed Parental Style Questionnaires (PSQ). Mothers' self report on their ideal and actual parenting were explored as a function of child sex, age, and birth order. As expected, there were significant differences between mothers' ideal and actual behaviors in all three parenting domains: Mothers' ideal behaviors such as social interaction, didactic interaction and limit setting were higher than those of their actual behaviors. For mothers' ideal parenting, results revealed neither significant main effects nor interaction effects. However, the Parenting Domain x Birth-Order 2-way interaction and the Parenting Domain x Child Age 2-way interaction were significant for mothers' actual behaviors. Specifically, mothers reported more social and didactic behaviors with their first-born than later born children, but not for limit setting behavior. It was also found that higher limit setting behaviors were apparent for their 2- and 3-year-old than 1-year old children whereas lower social interactions were found for 3-year-old than for 1-year-old. In light of universality and uniqueness, mothers' parenting behavior toward young children has been discussed.
The role of domain-specific causal mechanism information and domain-general conditional probability in young children's causal reasoning on physics and psychology was investigated with the participation of 121 3-year-olds and 121 4-year-olds recruited from seven child care centers in Seoul, Kyonggi Province, and Busan. Children watched moving pictures on physical and psychological phenomena, and were asked to choose an appropriate cause and justify their choice. Results showed that young children's causal reasoning differed depending on domain-specific mechanism. In addition, their causal reasoning on physics and psychology differed by the developmental level of causal mechanism. The interaction of domain-specific mechanism and domain-general conditional probability influenced children's causal reasoning : evident conditional probability between domain-appropriate cause and effect helped children make more inferences based on domain-specific causal mechanism.

This paper introduces a software system and process model for constructing domain-specific relation extraction datasets semi-automatically. The system uses a set of terms such as genes, proteins diseases and so forth as inputs and then by exploiting massive biological interaction database, generates a set of term pairs which are utilized as queries for retrieving sentences containing the pairs from scientific databases. To assess the usefulness of the proposed system, this paper applies it into constructing a genic interaction dataset related to Alzheimer's disease domain, which extracts 3,510 interaction-related sentences by using 140 gene names in the area. In conclusion, the resulting outputs of the case study performed in this paper indicate the fact that the system and process could highly boost the efficiency of the dataset construction in various subfields of biomedical research.
Several members of tumor necrosis factor receptor (TNFR) superfamily that these members activate caspase-8 from death-inducing signaling complex (DISC) in TNF ligand-receptor signal transduction have been identified. In the extrinsic pathway, apoptotic signal transduction is induced in death domain (DD) superfamily; it consists of a hexahelical bundle that contains 80 amino acids. The DD superfamily includes about 100 members that belong to four subfamilies: death domain (DD), caspase recruitment domain (CARD), pyrin domain (PYD), and death effector domain (DED). This superfamily contains key building blocks: with these blocks, multimeric complexes are formed through homotypic interactions. Furthermore, each DD-binding event occurs exclusively. The DD superfamily regulates the balance between death and survival of cells. In this study, the structures, functions, and unique features of DD superfamily members are compared with their complexes. By elucidating structural insights of DD superfamily members, we investigate the interaction mechanisms of DD domains; these domains are involved in TNF ligand-receptor signaling. These DD superfamily members play a pivotal role in the development of more specific treatments of cancer.
The increasing capabilities of the computers enable us to utilize various numerical schemes for the time-domain simulations concerned with 3-dimensional free-surface wave problems. There are still difficulties to solve such kind of problems, however. That's because long time simulations with large computational domain are needed in time-domain analysis. So, we need faster and more efficient numerical schemes to get the solutions practically for these problems. In this paper, a high-order spectral/boundary-element method is used for the numerical investigation of physics involved in wave-body interaction. This method is one of the most efficient numerical methods by which the nonlinear gravity waves can be simulated and hydrodynamic forces also can be calculated in time-domain. To get the robust study in these topics, various numerical tests are performed and compared with others' works.

In this work the Multi-Domain model of an electric vehicle is developed. The electric domain model consists on the traction drive and allows including faults associated with stator winding. The thermal model is based on a spatial discretization. It receives the power dissipated in the electric domain, it interacts with the environment and provides the temperature distribution in the induction motor. The mechanical model is a half vehicle model. Given that all models are obtained using the same approach (Bond Graph) their integration becomes straightforward. This complete model allows simulating the whole system dynamics and the analysis of electrical/mechanical/thermal interaction. First, experimental results are aimed to validate the proposed model. Then, simulation results illustrate the interaction between the different domains and highlight the capability of including faults.
The serine-threonine kinase AKT plays a pivotal role in tumor progression and is frequently overactivated in cancer cells; this protein is therefore a critical therapeutic target for cancer intervention. We aimed to identify small molecule inhibitors of the pleckstrin homology (PH) domain of AKT to disrupt binding of phosphatidylinositol-3,4,5-trisphosphate (PIP3), thereby downregulating AKT activity. Liposome pulldown assays coupled with fluorescence spectrometry were used to screen flavonoids for inhibition of the AKT PH-PIP3 interaction. Western blotting was used to determine the effects of the inhibitors on AKT activation in cancer cells, and in silico docking was used for structural analysis and optimization of inhibitor structure. Several flavonoids showing up to 50% inhibition of the AKT PH-PIP3 interaction decreased the level of AKT activation at the cellular level. In addition, the modified flavonoid showed increased inhibitory effects and the approach would be applied to develop anticancer drug candidates. In this study, we provide a rationale for targeting the lipid-binding domain of AKT, rather than the catalytic kinase domain, in anticancer drug development.

It has been proposed that chitin synthase 3 (CHS3)-nediated chitin synthesis during the vegetative cell cycle is regulated by chitin synthase 4 (CHS4) of Saccharomyces cerevisiae. To investigate direct protein-protein interaction between the coding products of these two genes, a domain of Chs3p that is responsible for interaction with Chs4p was identified, using the yeast two-hybrid system. This domain of 54 amino acids, termed MIRC3-4 (Maximum Interacting Region of Chs3p with Chs4p), is well conserved among CHS3 homologs of various fungi. Some mutations in MIRC3-4 resulted in a decrease in the enzymatic activity and chitin contents. Chs3p carrying those mutations exhibited weak interactions with Chs4p, when assayed by the yeast two-hybrid system. Surprisingly, all the mutants were sensitive to Calcofluor regardless of changes in enzymatic activities or chitin contents. This report deals with a core region in MIRC3-4 that affects the interaction with Chs4p.
