Inhibitory Effect of Nicardipine on hERG Channel

  • Drug-induced long QT syndrome is known to be associated with the onset of torsades de pointes (TdP), resulting in a fatal ventricular arrhythmia. QT interval prolongation can result from blocking the human ether-a-go-go-related gene (hERG) channel, which is important for the repolarization of cardiac action potential. Nicardipine, a Ca-channel blocker and antihypertensive agent, has been reported to increase the risk of occasional serious ventricular arrhythmias. We studied the effects of nicardipine on hERG $K^+$ channels expressed in HEK293 cells and Xenopus oocytes. The cardiac electrophysiological effect of nicardipine was also investigated in this study. Our results revealed that nicardipine dose-dependently decreased the tail current of the hERG channel expressed in HEK293 cells with an $IC_{50}$ of 0.43 ${\mu}M$. On the other hand, nicardipine did not affect hERG channel trafficking. Taken together, nicardipine inhibits the hERG channel by the mechanism of short-term channel blocking. Two S6 domain mutations, Y652A and F656A, partially attenuated (Y652A) or abolished (F656A) the hERG current blockade, suggesting that nicardipine blocks the hERG channel at the pore of the channel.

Development of ICT Teaching-Learning materials for Motorcycle Safety Education (이륜차 안전교육을 위한 ICT 교수-학습 자료 개발)

  • The purpose of this study was to develop ICT(Information and Communication Technology) teaching-learning materials for motorcycle safety education. Physical subjects which are relevant with motorcycle dynamics were introduced to increase the safety awareness through integrated learning such as the combination of technology and science. The results of this study were as follows : (1) Utilizing the physics basic theories which are relevant to motorcycle dynamics, the lesson plan of motorcycle ICT teaching-learning materials, which were included to plan and instruction action plan, was developed. (2) Flash animation or movie was developed to apply physics knowledge to technology in driving motorcycle. This is able to improve the attitude of driving motorcycle of students. (3) Through the contents of appendix of motorcycle safety driving section in this web-site developed here, junior and senior high school students would be able to strengthen their cognitive domain of motorcycle by themselves. (4) Using the web-site of this study, both teachers and students would be able to freely interact through an online bulletin board.

Flexible Decision-Making for Autonomous Agent Through Computation of Urgency in Time-Critical Domains (실시간 환경에서 긴급한 정도의 계산을 통한 자율적인 에이전트의 유연한 의사결정)

  • Autonomous agents need considerable computational resources to perform rational decision-making. The complexity of decision-making becomes prohibitive when large number of agents are present and when decisions have to be made under time pressure. One of approaches in time-critical domains is to respond to an observed condition with a predefined action. Although such a system may be able to react very quickly to environmental conditions, predefined plans are of less value if a situation changes and re-planning is needed. In this paper we investigate strategies intended to tame the computational burden by using off-line computation in conjunction with on-line reasoning. We use performance profiles computed off-line and the notion of urgency (i.e., the value of time) computed on-line to choose the amount of information to be included during on-line deliberation. This method can adjust to various levels of real-time demands, but incurs some overhead associated with iterative deepening. We test our framework with experiments in a simulated anti-air defense domain. The experiments show that the off-line performance profiles and the on-line computation of urgency are effective in time-critical situations.

  • The Dryness and the Dampness is the congential nature. the Water and Fire is the acquired action. the skin is related to the physiological function of the lung, so the skin is the domain of the lung. the lung belongs to the dryness-metal(燥金). And the Dryness and the Dampness in the six pathogenic factors exert a important pathogenic influence upon the skin. Psoriasis is a chronic skin condition charactered by red, silver scaly patches in the affected areas, it is caused by excessive growth of the top layer of the skin(epidermis). Scalp psoriasis affects at least two thirds people who have psoriasis. Generally, the scalp will have plaque psoriasis characterized by elevated, inflamed, lesions covered with silvery white scales. The lesion of psoriasis is head, elbow and knee that are yang-channels in Oriental medicine. The syndrome falls with in the purview of the dryness-metal(燥金). In time, psoriasis takes a turn for the worse in the winter season. In space, many of people in comparison to the average in the world have psoriasis in high latitude that is dry and cold, for instance, the Scandinavian peninsula. Deficiency of yin-fluid and blood course often the dryness-syndrome, for instance, psoriasis, etc. So psoriasis is one of the dryness-syndrome in Oriental medicine.

An Exploratory Study on Development of Korean Media Educational Model (한국형 미디어교육 모형의 개발에 대한 탐색적 연구)

  • This study suggested appropriate media educational model for the Korean situation based on the two previous research results concerning Korean media mental model which have abstracted from elementary schoolers and parents respectively living in and around Seoul. This looked through typical media educational model being in effect in Western countries such as Buckingham's creation-centered model, Hobbs' action-centered model, Potter's analysis-centered model and Vanhoozer's worldview-centered model, synthesized all the strengths of each model, and finally modified and reorganized this new model to reflect demanders' needs. Newly developed demander-centered educational model is a kind of circulation model consisting of a chain of the steps: worldview and viewpoints, use(access), analysis and evaluation, reflection, and social act; Each step borrowed the essential contents of each domain of objectives and tried to reflect the specific situations for Korean demanders. The needs for media use etiquette against addiction and cyberbullying were applied to the step of 'worldview and viewpoint', the needs for the educational status of books as media to 'use(access)', and the reality of highly used digital media to 'social act.'

OMA of model steel structure retrofitted with CFRP using earthquake simulator

  • Nowadays, there are a great number of various structures that have been retrofitted by using different FRP Composites. Due to this, more researches need to be conducted to know more the characteristics of these structures, not only that but also a comparison among them before and after the retrofitting is needed. In this research, a model steel structure is tested using a bench-scale earthquake simulator on the shake table, using recorded micro tremor data, in order to get the dynamic behaviors. Beams of the model steel structure are then retrofitted by using CFRP composite, and then tested on the Quanser shake table by using the recorded micro tremor data. At this stage, it is needed to evaluate the dynamic behaviors of the retrofitted model steel structure. Various types of methods of OMA, such as EFDD, SSI, etc. are used to take action in the ambient responses. Having a purpose to learn more about the effects of FRP composite, experimental model analysis of both types (retrofitted and no-retrofitted models) is conducted to evaluate their dynamic behaviors. There is a provision of ambient excitation to the shake table by using recorded micro tremor ambient vibration data on ground level. Furthermore, the Enhanced Frequency Domain decomposition is used through output-only modal identification. At the end of this study, moderate correlation is obtained between mode shapes, periods and damping ratios. The aim of this research is to show and determine the effects of CFRP Composite implementation on structural responses of the model steel structure, in terms of changing its dynamical behaviors. The frequencies for model steel structure and the retrofitted model steel structure are shown to be 34.43% in average difference. Finally, it is shown that, in order to evaluate the period and rigidity of retrofitted structures, OMA might be used.

Learning Relational Instance-Based Policies from User Demonstrations (사용자 데모를 이용한 관계적 개체 기반 정책 학습)

  • Demonstration-based learning has the advantage that a user can easily teach his/her robot new task knowledge just by demonstrating directly how to perform the task. However, many previous demonstration-based learning techniques used a kind of attribute-value vector model to represent their state spaces and policies. Due to the limitation of this model, they suffered from both low efficiency of the learning process and low reusability of the learned policy. In this paper, we present a new demonstration-based learning method, in which the relational model is adopted in place of the attribute-value model. Applying the relational instance-based learning to the training examples extracted from the records of the user demonstrations, the method derives a relational instance-based policy which can be easily utilized for other similar tasks in the same domain. A relational policy maps a context, represented as a pair of (state, goal), to a corresponding action to be executed. In this paper, we give a detail explanation of our demonstration-based relational policy learning method, and then analyze the effectiveness of our learning method through some experiments using a robot simulator.

  • To build autonomous agents who can make a decision on behalf of humans in time-critical complex environments, the formulation of operational knowledge base could be essential. This paper proposes the methodology of how to formulate the knowledge base and evaluates it in a practical application domain. We analyze threat data received from the multiple sensors of Aircraft Survivability Equipment(ASE) for Korean helicopters, and integrate the threat data into the inductive model through compilation technique which extracts features of the threat data and relations among them. The compiled protocols of state-action rules can be implemented as the brain of the ASE. They can reduce the amounts of reasoning, and endow the autonomous agents with reactivity and flexibility. We report experimental results that demonstrate the distinctive and predictive patterns of threats in simulated battlefield settings, and show the potential of compilation methods for the successful detection of threat systems.

Structure and Regulation of a Complex Promoter Region from an Alkali-tolerent Bacillus sp.

  • A DNA fragment from an alkali-tolerent Bacillus sp., conferring strong promoter activity, was subcloned into the promoter probe plasmid pPL703 and the nucleotide sequence of this promoter region was determined. The sequence analysis suggested that this highly efficient promoter region containing the complex clustered promoters comprised three kinds of promoters (P1, P2 and P3), which are transcribed by $\sigma^B (formerly \sigma^{37}), \sigma^E(formerly \sigma^{29}) and \sigma^A (formerly \sigma^{43})$ RNA polymerase holoenzymes which play major rules at the onset of endospore formation, during sporulation and at the vegetative phase of growth, respectively. S1 nuclease mapping experiments showed that all three promoters had staggered transcription initiation points. The results of chloramphenicol acetyltransferase assay after the subcloning experiments also indicated that the expression of these clustered promoters was correlated with the programs of growth and endospore development. Promoter P1, P2 and P3 were preceded by 75% AT, 79% AT and 81% AT regions, respectively, and a partial deletion of AT-rich region prevented transcription from promoter P1 in vivo. Two sets of 5 -AGTGTT-3 sequences and inverted repeat sequences located around the promoter P1 were speculated as the possible cis acting sites for the catabolite repression in B. subtilis. In vivo transcripts from these sequence regions may be able to form a secondary structure, however, the possibility that a regulatory protein induced by the excess amount of glucose could be bound to such a domain for crucial action remains to be determined.

Urban Redevelopment, Displacement and Anti-Gentrification Movements (재개발, 이주, 젠트리피케이션 반대운동)

  • The displacement of business tenants is of growing concern in urban redevelopment. This research focuses on why business tenants have not collectively taken action against urban redevelopment and explores how their rights can be improved. It met the research aim through a case study on one urban redevelopment area in Yongsan. Although urban redevelopment plans create many problems for business tenants, there is no effective mechanism in place to enable them to get involved in the decision-making procedures of urban planning. They are collectively and individually powerless in urban politics, so they do not have enough opportunities to change the Korean urban redevelopment system. The findings from this research suggest that many tenants have been apathetic to anti-gentrification movements due to their powerlessness in the urban redevelopment domain. In order to overcome structural injustice, it is important to improve business tenants' awareness of 'the right to the city' and enforce strong legislation to safeguard tenants.
